    Discoloration of soybean-based foods, particularly reddening during processing, remains poorly understood. In this study, yellow and red pigments associated with soybean reddening were extracted, purified, and characterized. The pigments showed distinct UV–Vis spectra, with a shoulder near 420 nm for the yellow pigment and an absorption maximum near 510 nm for the red pigment. Heat and pH stability experiments showed that the yellow pigment decreased during thermal treatment, accompanied by an increase in the red pigment. Mass spectrometric analyses revealed a consistent 2 Da mass difference and a common fragment ion, indicating a close structural relationship between the pigments. Both pigments were detected in soybean, soymilk, and tofu, and the red pigment increased during processing. The pigments did not correspond to known major soybean constituents. These findings suggest that soybean reddening involves the conversion of a yellow pigment into a structurally related red pigment during soybean processing.

  2. Effects of Fermentation and Oxidative Degradation on the Composition, Antioxidant Activity, ACE Inhibitory Activity, and In Vitro Neuroprotective Potential of Soybean-Derived Kefir Polysaccharide-Rich Extracts 査読有り

    Kefir is a probiotic beverage produced by symbiotic bacteria and yeasts. Polysaccharide-rich extracts from yellow and black soybeans (S and B) were obtained and subsequently fermented to produce S-F and B-F. The fermented extracts were further subjected to oxidative degradation using ascorbic acid and hydrogen peroxide to generate S-FD and B-FD. Physicochemical analyses revealed distinct differences in composition, phenolic profiles, and molecular weight among S-F, S-FD, B-F, and B-FD. Fourier transform infrared (FTIR) spectra indicated that oxidative degradation altered specific functional group intensities without disrupting the fundamental polysaccharide framework. Fermentation enhanced angiotensin-converting enzyme (ACE) inhibitory activity, and subsequent oxidative degradation further improved this effect. Both fermented and degraded extracts exhibited antioxidant activities, including 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging capacity, ferrous-ion chelating ability, and reducing power, with degraded samples showing greater activity. The effects of the extracts on SH-SY5Y human neuroblastoma cells were evaluated in vitro. No cytotoxicity was observed at concentrations up to 400 μg/mL. Treatment at 200 μg/mL increased cell viability and reduced apoptosis in rotenone (ROT)-treated cells. Multivariate analysis further indicated that oxidative degradation enhanced antioxidant and ACE inhibitory activities but may reduce the protective effects observed in SH-SY5Y cells. Overall, soybean-derived kefir polysaccharide-rich extracts show potential as functional ingredients for applications related to blood pressure regulation and antioxidant activity, while their protective effects in neuronal cell models warrant further investigation.

    This study investigated the effect of rice bran derived from broken rice feeding on the lipid classes and fatty acid composition of oil extracted from crickets (Acheta domesticus). During the final seven days prior to cricket oil extraction, crickets were fed either rice bran or control feed. The lipid classes in both the cricket oil and feed materials (rice bran or control feed) were identified using thin-layer chromatography, while the proportion of neutral lipids, phospholipids, and glycolipids was quantified by solid-phase extraction. Fatty acid composition was determined using GC-FID. Cricket oil from the rice bran-fed group exhibited a significantly higher ratio of glycolipids, which are abundant in rice bran, compared to the control group. Although both oleic and linoleic acid levels were significantly higher than in the control feed, only the oleic acid proportion was significantly increased in the cricket oil from the rice bran-fed group. Additionally, the ω-6/ω-3 ratio in each lipid fraction was significantly higher in the rice bran-fed group compared to the control group. These results suggest that dietary supplementation with rice bran derived from broken rice significantly alters the lipid profile of crickets and may represent a valuable strategy for enhancing the nutritional quality of cricket oil.

    (1) Background: Plasmalogens are vinyl ether-type glycerophospholipids that are characteristically distributed in neural tissues and are significantly reduced in the brains of individuals with dementia compared to those in healthy subjects, suggesting a link between plasmalogen deficiency and cognitive decline. Hen eggs are expected to be a potential source of dietary plasmalogens, but the details remain unclear. (2) Methods: We evaluated the fresh weight, dry weight, total lipid, neutral lipids, glycolipids, and phospholipids in the egg yolk and egg white of hen egg. Then, the molecular species of plasmalogens were quantified using HPLC-ESI-MS/MS. (3) Results: In egg yolk, the total plasmalogen content was 1292.1 µg/100 g fresh weight and predominantly ethanolamine plasmalogens (PE-Pls), specifically 18:0/22:6-PE-Pls, which made up 75.6 wt% of the total plasmalogen. In egg white, the plasmalogen content was 31.4 µg/100 g fresh weight and predominantly PE-Pls, specifically 18:0/20:4-PE-Pls, which made up 49.6 wt% of the total plasmalogen. (4) Conclusions: Plasmalogens were found to be more enriched in egg yolk than in egg white. It was found that humans are likely to ingest almost 0.3 mg of total plasmalogens from one hen egg. These findings highlight the importance of plasmalogens in the daily diet, and it is recommended to explore the impact of long-term dietary plasmalogen intake to assess its effect on human health. This provides a viewpoint for the development of new food products.

    Brain-derived neurotrophic factor (BDNF) plays an important role in neurogenesis, synaptic plasticity, and cognition. BDNF is a neurotrophin that binds to tropomyosin receptor kinase B (TrkB), a specific receptor on target cell surfaces; it acts on neuronal formation, development, growth, and repair via transcription factors, such as cAMP response element-binding protein (CREB), and it is involved in learning and memory. BDNF expression is decreased in patients with Alzheimer’s disease (AD). Exercise and the intake of several different foods or ingredients can increase BDNF expression, as confirmed with lutein, xanthophylls (polar carotenoids), and ethanolamine plasmalogen (PlsEtn), which are present at high levels in the brain. This study examined the effects of combining lutein and PlsEtn using lutein-rich Chlorella and ascidian extracts containing high levels of PlsEtn bearing docosahexaenoic acid, which is abundant in the human brain, on the activation of the BDNF–TrkB–CREB signaling pathway in the hippocampus of Sprague-Dawley rats. Although activation of the BDNF–TrkB–CREB signaling pathway in the hippocampus was not observed in Chlorella or ascidian PlsEtn monotherapy, activation was observed with combination therapy at an equal dose. The results of this study suggest that the combination of Chlorella and ascidian PlsEtn may have a preventive effect against dementia, including AD.

    Walnuts are rich sources of lipids and polyunsaturated fatty acids and are expected to promote health. There are two Japanese native walnut species: Oni walnut (Juglans ailanthifolia Carr.) and Hime walnut (Juglans subcordiformis Dode.). However, despite the fact that these Japanese native walnuts have long been consumed in local cuisine, their nutritional composition is largely unknown. This study aimed to assess the concentrations of total lipids, and fatty acid composition including polyunsaturated fatty acids, in the kernels of Oni walnut and Hime walnut. In addition, we assessed various aspects related to their nutritional and functional values, by measuring the total protein, amino acids, minerals, and total polyphenols. The concentrations of the measured compounds in the two native species were compared with those in the English walnut (Juglans regia L.), a globally recognized cultivar, and its counterpart cultivated in Japan, Shinano walnut (Juglans regia L.). The results showed that Oni walnut and Hime walnut contained significantly higher protein and minerals and lower lipid content than conventional cultivars. However, both Oni and Hime walnuts contained higher ratios of unsaturated fatty acids in total fatty acids. This study offers novel insights into the nutritional components of Oni and Hime walnuts, contributing to a deeper understanding of their nutritional value and potential applications as unique native walnut species. The findings of this study highlight the relationship between the different types of walnut species and their nutritional composition, and the value of native walnuts used in local cuisine, and will lead to new developments in functional foods from walnut species consumed around the world. It will contribute to the development of functional and processed foods by increasing the production of native walnut species, which are rich in protein, unsaturated fatty acids and minerals and by using them in local cuisines and health-promoting foods.

    Aging increases oxidative and inflammatory stress caused by a reduction in metabolism and clearance, thus leading to the development of age-associated diseases. The quality of our daily diet and exercise is important for the prevention of these diseases. Marine resources contain various valuable nutrients, and unique glycerophospholipid plasmalogens are found abundantly in some marine invertebrates, including ascidians. One of the major classes, the ethanolamine class (PlsEtn), exists in a high ratio to phospholipids in the brain and blood, while decreased levels have been reported in patients with age-associated diseases, including Alzheimer’s disease. Animal studies have shown that the administration of marine PlsEtn prepared from marine invertebrates improved PlsEtn levels in the body and alleviated inflammation. Animal and human studies have reported that marine PlsEtn ameliorates cognitive impairment. In this review, we highlight the biological significance, relationships with age-associated diseases, food functions, and healthcare materials of plasmalogens based on recent knowledge and discuss the contribution of marine plasmalogens to health maintenance in aging.

  11. Hypolipidemic and Anti-Inflammatory Effects of Curcuma longa-Derived Bisacurone in High-Fat Diet-Fed Mice 査読有り

    Turmeric (Curcuma longa) contains various compounds that potentially improve health. Bisacurone is a turmeric-derived compound but has been less studied compared to other compounds, such as curcumin. In this study, we aimed to evaluate the anti-inflammatory and lipid-lowering effects of bisacurone in high-fat diet (HFD)-fed mice. Mice were fed HFD to induce lipidemia and orally administered bisacurone daily for two weeks. Bisacurone reduced liver weight, serum cholesterol and triglyceride levels, and blood viscosity in mice. Splenocytes from bisacurone-treated mice produced lower levels of the pro-inflammatory cytokines IL-6 and TNF-α upon stimulation with a toll-like receptor (TLR) 4 ligand, lipopolysaccharide (LPS), and TLR1/2 ligand, Pam3CSK4, than those from untreated mice. Bisacurone also inhibited LPS-induced IL-6 and TNF-α production in the murine macrophage cell line, RAW264.7. Western blot analysis revealed that bisacurone inhibited the phosphorylation of IKKα/β and NF-κB p65 subunit, but not of the mitogen-activated protein kinases, p38 kinase and p42/44 kinases, and c-Jun N-terminal kinase in the cells. Collectively, these results suggest that bisacurone has the potential to reduce serum lipid levels and blood viscosity in mice with high-fat diet-induced lipidemia and modulate inflammation via inhibition of NF-κB-mediated pathways.

    The rapid development of aberrant cells outgrowing their normal bounds, which can subsequently infect other body parts and spread to other organs—a process known as metastasis—is one of the significant characteristics of cancer. The main reason why cancer patients die is because of widespread metastases. This abnormal cell proliferation varies in cancers of over a hundred types, and their response to treatment can vary substantially. Several anti-cancer drugs have been discovered to treat various tumors, yet they still have harmful side-effects. Finding novel, highly efficient targeted therapies based on modifications in the molecular biology of tumor cells is essential to reduce the indiscriminate destruction of healthy cells. Exosomes, an extracellular vesicle, are promising as a drug carrier for cancer therapy due to their good tolerance in the body. In addition, the tumor microenvironment is a potential target to regulate in cancer treatment. Therefore, macrophages are polarized toward M1 and M2 phenotypes, which are involved in cancer proliferation and are malignant. It is evident from recent studies that controlled macrophage polarization might contribute to cancer treatment, by the direct way of using miRNA. This review provides an insight into the potential use of exosomes to develop an ‘indirect’, more natural, and harmless cancer treatment through regulating macrophage polarization.

    The delivery of curcumin (CUR) using the solid dispersion system (CUR solid dispersions; C-SDs) has been shown to improve CUR bioavailability. However, it is unclear how different particle sizes of C-SDs affect the bioavailability and biological activities of CUR. Hence, we prepared C-SDs in different sizes using food-grade excipients and evaluated their bioavailability and biological activities. By pulverizing large particle sizes of C-SDs using zirconia beads, we successfully prepared C-SDs I-IV (particle size: (I) 120, (II) 447, (III) 987, (IV) 1910 nm). When administrated orally in rats, the bioavailability of CUR was increased with decreasing C-SDs size, most likely by improving its solubility in micelles. When administrated intravenously in rats, blood concentrations of CUR were increased with increasing particle size, suggesting that larger C-SDs presumably control the metabolic conversion of CUR. In RAW264 cells, more CUR was taken up by cells as their sizes reduced, and the more potent their anti-inflammatory activities were, suggesting that smaller C-SDs were taken up through a number of cellular uptake pathways. Altogether, the present study showed an evident effect of C-SDs size on their bioavailability and anti-inflammatory activities—information that serves as a basis for improving the functionality of CUR.

    Microgreens, a hypothesized term used for the emerging food product that is developed from various commercial food crops, such as vegetables, grains, and herbs, consist of developed cotyledons along with partially expanded true leaves. These immature plants are harvested between 7–21 days (depending on variety). They are treasured for their densely packed nutrients, concentrated flavors, immaculate and tender texture as well as for their vibrant colors. In recent years, microgreens are on demand from high-end restaurant chefs and nutritional researchers due to their potent flavors, appealing sensory qualities, functionality, abundance in vitamins, minerals, and other bioactive compounds, such as ascorbic acid, tocopherol, carotenoids, folate, tocotrienols, phylloquinones, anthocyanins, glucosinolates, etc. These qualities attracted research attention for use in the field of human health and nutrition. Increasing public concern regarding health has prompted humans to turn to microgreens which show potential in the prevention of malnutrition, inflammation, and other chronic ailments. This article focuses on the applications of microgreens in the prevention of the non-communicable diseases that prevails in the current generation, which emerged due to sedentary lifestyles, thus laying a theoretical foundation for the people creating awareness to switch to the recently introduced category of vegetable and providing great value for the development of health-promoting diets with microgreens.

    Iron is the most abundant mineral in the human body and plays essential roles in sustaining life, such as the transport of oxygen to systemic organs. The Fenton reaction is the reaction between iron and hydrogen peroxide, generating hydroxyl radical, which is highly reactive and highly toxic to living cells. “Ferroptosis”, a programmed cell death in which the Fenton reaction is closely involved, has recently received much attention. Furthermore, various applications of the Fenton reaction have been reported in the medical and nutritional fields, such as cancer treatment or sterilization. Here, this review summarizes the recent growing interest in the usefulness of iron and its biological relevance through basic and practical information of the Fenton reaction and recent reports.

    The concept of food and aging is of great concern to humans. So far, more than 300 theories of aging have been suggested, and approaches based on these principles have been investigated. It has been reported that antioxidants in foods might play a role in human aging. To clarify the current recognition and positioning of the relationship between these food antioxidants and aging, this review is presented in the following order: (1) aging theories, (2) food and aging, and (3) individual food antioxidants and aging. Clarifying the significance of food antioxidants in the field of aging will lead to the development of strategies to achieve healthy human aging.

  1. 食品・微生物成分による抗炎症型ミエロイド系細胞の分化と機能発現メカニズムの解析

    本研究では骨髄細胞から抗炎症型ミエロイド系細胞への分化を食品・微生物成分を用いて促進し、その抗炎症機能獲得メカニズムを明らかにすることを目的としている。ミエロイド系細胞は前駆細胞が血液を循環した後に組織へ定着し分化する樹状細胞や、マクロファージ、ミクログリアが含まれる。食品・微生物成分により前駆細胞の段階で抗炎症特性を獲得できれば、抗炎症特性を持つ細胞分化を促進でき、免疫制御へと結びけることができる。これまでに、マウス骨髄細胞の培養系を用いて、n-3系多価不飽和脂肪酸の一種、エイコサペンタエン酸がミエロイド系マクロファージの分化を促進し、その機序として核内受容体 peroxisome proliferator activated receptor γ(PPARγ)との結合が関与することを明らかにしている。また、酵母(Saccharomyces cerevisiae)由来αマンナンがミエロイド系細胞の抗炎症機能獲得を促進すること、その機序にはSpleen tyrosine kinase(Syk)やPhosphoinositide 3 kinase (PI3K)/Akt/mammalian target of rapamycin (mTOR)経路が関与することを見いだしている。本研究はミエロイド系細胞の機能獲得メカニズムに新たな知見をもたらし、科学的根拠に基づく免疫機能性食品の開発に貢献すると期待される。

    今年度は、昨年度の「ビタミンCの細胞内の代謝や細胞取り込みにおける、細胞外の物質の影響」を更に検討した。グルコーストランスポーターの発現量の異なる細胞を用い、デヒドロアスコルビン酸やアスコルビン酸等のビタミンCやその類縁体に関する物質が存在するときの細胞死、デヒドロアスコルビン酸の細胞取り込み量、デヒドロアスコルビン酸やアスコルビン酸の実験系での安定性、細胞内の還元されたアスコルビン酸の濃度の変化を確認した。更に、細胞内の活性酸素の上昇や様々な抗酸化物質の消費や酵素の活性を確認し、その値が系に存在するビタミンCの類縁体によって一定の法則に従って変化することを確認した。これらの研究結果より、既報(Science, 350, 6266, 1391-1396 (2015))で報告されている、「細胞内活性酸素の上昇が、がん細胞の細胞死に関与する」というストーリーと異なる新たな経路を見出しつつあり、更に検討を進めている。リポソームのような、細胞内のDHA濃度を上昇させる技術と組み合わせる検討も引き続き継続しており、ビタミンCを用いた新たな治療法を検討する。また、昨年度に引き続き、デヒドロアスコルビン酸とアスコルビン酸以外の代謝物の定量法の開発を液体クロマトグラフ質量分析計(LC-MS/MS)を用いて検討を進め、in vitroの細胞実験から得られたサンプルにおいてはある程度定量性のある分析条件を確立できたが、再現性の問題などがあるため引き続き信頼性のある分析法の開発を続けている。来年度は、これらの研究を継続しつつ、動物実験で得られたサンプルも分析できるように研究を遂行していく。

  3. 標的細胞への酸化型ビタミンCの送達・代謝を可能とする人工トランスポーターの開発

    今年度は、去年度に開発したビタミンCの酸化体(デヒドロアスコルビン酸、DHA)と還元体(アスコルビン酸、ASC)の分析法を用いて、まずグルコーストランスポーターの発現量が異なる細胞への取り込みを定量的に確認するとともに、細胞死との関係性を明らかにしようとした。細胞は、グルコーストランスポーターの発現量とグルコースの取り込みのモデルとしてよく使用される、胃がん細胞を用いた。これらの細胞に、DHAを添加すると、グルコーストランスポーターの発現量の高い細胞ほどDHAによる細胞死の割合が高く、更にグルコーストランスポーターの発現量依存的なDHAの細胞への取り込みを確認した。この現象は、人工トランスポーターをこのDHAの細胞取り込みに用いることの意義を高める結果である。一方、今年は去年から引き続き、人工トランスポーターの合成に集中した。前年度、目的の人工トランスポーターの合成に成功したが、その精製が課題であった。その精製のために、まず分取HPLCを用いて目的物と夾雑物の分離を試みたが、得られた目的の人工トランスポーターは極微量であり、その後の研究に使用可能な量ではなかった。そのために、合成法の改良や精製法を更に検討し、殆ど90%以上の収率で人工トランスポーターが得られる方法を確立し、細胞実験に用いることができる量を得られたととも分取HPLCでなく液液抽出で精製できることを確認した。今年度の試みによって、人工トランスポーターの合成法を確立したが、最終年度となり、それ以降予定していた実験に手をつけることができなかった。しかしながら、申請者は引き続き今後も本研究を基盤とした実験を続けていくことで、目標を達成したい。

    AD脳では抗酸化成分であるカロテノイドの濃度が健常者と比較して有意に低値であることも報告されている。しかし、血液脳関門の存在により食品として摂取したカロテノイドが効率よく脳内に移行し、その抗酸化能を発揮させることは困難である。グリコール酸/乳酸共重合体(PLGA)ナノ粒子は生分解性高分子の1つであり体内で二酸化炭素と乳酸に分解される。また、非イオン性界面活性剤であるTween-80でコーティングすることでナノ粒子表面にアポリポ蛋白が吸着し、in vivoの動物試験では血液脳関門を通過できることが報告されている。本研究では、カロテノイドを内包したTween-80装飾PLGAナノ粒子を作成し、脳内へのルテインの移行量を増加させ、ADの発症や進行を抑制することを目標とした。β-カロテン封入PLGAナノ粒子投与群では尾静脈投与後1時間経過した血漿のβ-カロテン濃度はTween-80で覆った粒子を投与した群と比較して約162倍高濃度だった。Tween-80で表面を覆わなかったPLGAナノ粒子は表面にアポE蛋白質以外にも、様々なタンパク質が接着し、臓器へ取り込まれにくくなり血中に滞留していたと考えられた。また、ナノ粒子表面をTween-80で覆った群と覆わなかった群で脳内のβ-カロテン濃度には有意差が確認されなかったが、その原因は凍結乾燥で使用したスクロースの存在が考えられる。粒子は受容体介在性トランスサイトーシスによって脳内に移行されると考えられることを目的とした。しかし、in vitroの試験でスクロースはエンドサイトーシスの阻害物質として使用されており、PLGAナノ粒子においてもスクロースの存在によってエンドサイトーシス阻害が確認されている。今後、別の凍結乾燥保護剤を使用する方法で粒子を作成しナノ粒子の脳への取り込み検証する。
