    Abstract Ataxin-3, the causative protein of Machado-Joseph disease, comprises an enzymatic domain, Josephin domain (JD), and a polyglutamine repeat (polyQ). The polyQ expansion promotes the fibrillogenesis of ataxin-3, which is responsible for the disease onset. The fibrillogenesis proceeds through two distinct steps: protofibril formation, in which JD undergoes self-assembly, and subsequent fibrillation. Both polyQ elongation and metal ions accelerate fibrillation; however, the effects of these factors on each stage remain unclear. This study comprehensively analyzed the effects of metal ions on ataxin-3 protofibril formation using spectroscopic analyses. The protofibril formation of JD and a 28-residue polyQ tract (Q28) did not complete within 14 days under metal-free conditions. In contrast, the introduction of Cu²⁺ or Zn²⁺ markedly accelerated the process: JD denatured within one day, and protofibrils formed within four days. The acceleration was similar between JD and Q28, indicating that metal binding primarily affects JD. SH assays and mass spectrometry showed that Cu²⁺ binds to thiols and induces intra- and intermolecular disulfide bonds, whereas Zn²⁺ binds to thiols but does not promote disulfide formation. These observations suggest that Cu²⁺ and Zn²⁺ enhance the protofibril formation through interactions with specific JD sites, but the underlying mechanisms differ between the two metal ions.

    Abstract In this study, a validated analytical method based on liquid chromatography/electrospray ionization-selected reaction monitoring/mass spectrometry (LC/ESI-SRM/MS) was developed for dipeptides in a functional food, fermented brown rice and rice bran with Aspergillus oryzae (FBRA). The aim of this study was to screen and quantify dipeptides in the water-soluble fraction of FBRA. There are few studies focusing on dipeptides in FBRA, even though FBRA is expected to contain bioactive peptides: FBRA or its aqueous extract has shown variety of biological activities. An anti-hypertensive dipeptide was found in similar rice bran products digested by thermolysin. Dipeptides are the smallest class of peptides, and many show a variety of biological activities. Some bioactive peptides are widely found in fermented foods. However, because dipeptides are generally too polar to be retained on versatile LC columns, such as octadecylsilyl columns, dipeptides have been often overlooked and not quantified because of the analytical difficulties. The LC was performed using a porous graphitic carbon column with the mobile phases of 0.1% formic acid/0.1% trifluoroacetic acid/1% tetrahydrofuran in water and acetonitrile. The stable isotope-labeled internal standards and SRM for dipeptides made it possible to develop a validated method. Five dipeptides were identified and quantified in FBRA in much higher concentrations than related materials (malted brown rice, rice bran, brown rice, and white rice) as follows: Ile-Arg, 82.1 µg/g; Ala-Phe, 27.8 µg/g; Ala-Tyr, 31.6 µg/g; Val-Phe, 46.3 µg/g; and Ile-Tyr, 49.9 µg/g. This method was simple and robust and would be applicable for other functional foods. Graphical Abstract

    Angiotensin (Ang) peptides are the main effectors of the renin-angiotensin system (RAS) regulating diverse physiological conditions and are involved in renal and vascular diseases. Currently, quantitative analyses of Ang peptides in human plasma mainly rely on radioimmunoassay-based methods whose reported levels are quite divergent. Analyses are further complicated by the potential of Ang peptides to bind to solid surfaces, to be enzymatically decomposed during sample preparation, and to undergo post-translational modifications. A column switching-parallel LC/ESI-SRM/MS method has been developed for seven Ang peptides (Ang I, Ang II, Ang III, Ang IV, Ang 1-9, Ang 1-7, and Ang A) in human plasma. Aqueous acetonitrile (5%) containing 50 mM arginine (Arg) as a dissolving solution and a combination of protease inhibitors with formic acid were used to prevent adsorption and enzymatic degradation, respectively. Plasma samples were simply deproteinized with acetonitrile followed by clean-up with an on-line trap column via column-switching. Stable isotope dilution with [13C5,15N1-Val]-Ang peptides as internal standards was employed for quantitative analysis. The current methodology has been successfully applied to determine the plasma levels of Ang peptides in healthy participants, suggesting future applicability to studies of various diseases related to RAS.

    Biological macromolecules, such as deoxyribonucleic acid (DNA) and proteins, are exposed to higher chemical stresses during some physiological events such as increased oxidative stress from degenerative aging diseases and higher glucose stress in diabetes mellitus. Therefore, the resulting chemical modifications can provide significant information about biological events. Chemically modified DNA and abundant proteins have been analyzed by mass spectrometry, mainly for use as dosimeters of chemical exposure. However, quantitative analyses of bioactive peptides and proteins in specific diseases have relied almost exclusively on the use of immunoassay-based procedures with no concern for possible minute chemical modifications. Therefore, important information could be overlooked, or considerable misunderstandings could be created, if the modifications are not distinguished and if they alter protein functions such as phosphorylation. I believe that chemical modifications on biological macromolecules could play important roles, not only as diagnostic and therapeutic markers, but also as triggers that initiate some diseases. In this article, I first introduce my previous research on biomarkers such as DNA adducts and chemically modified proteins. I then present a novel “omics” strategy for biomarker discovery using minute chemical modifications on biological macromolecules.

    テープストリッピング法で上腕部から皮膚角質層タンパク質を非侵襲的に採取し、試料精製にFASP法を用いてトリプシン消化後にMALDI-TOF/MS分析に付した。角質層タンパク質由来の75のペプチドピーク中61ピーク(81%、K1:15、K2:11、K9:21、K10:14)がケラチンに帰属され、本法にてほぼ選択的に精製・回収が可能であることが示された。3%H2O2で30min処理したヒト上腕部から試料を得てケラチン酸化修飾を精査した。主要な酸化修飾ターゲットはK1とK10であり、K1のα-ヘリックスドメイン上のMet259、262、296、469が濃度依存的に酸化を受け、MALDI-TOF/MSで容易に分別可能であった。操作中の酸化はBHT添加で抑制された。これらのMet上の酸化修飾が酸化ストレスを量的に反映するマーカーとなるのか否かを判断するため、H2O2との処理時間との関係を精査し、いずれも酸化修飾の割合は処理時間を反映しており、酸化ストレスの指標になると考えられた。

    近年、化学物質の環境濃度のみならず、個々人の暴露(exposure)を解析する『エクスポゾーム』の重要性が示唆されている。主にターゲット分子の長期内部暴露を血中タンパク質付加体として解析されるが、環境濃度のみならず滞在時間・防御・吸収・代謝・排泄などの要因もあり、経時的かつ網羅的解析が望まれる。一方、表皮角質層を構成する疎水性タンパク質『ケラチン』は、外部刺激の防護壁として直接かつ恒常的に暴露されており、『エクスポゾーム』の記録媒体として以下の可能性を有する:①多様な化学修飾を受ける、②疎水性低分子を保持できる、③短周期で置き換わる、④非侵襲的に採取できる。そこで申請者は、従来の長期暴露のターゲット分析から経時的かつノンターゲット解析可能な新規環境エクスポゾーム解析法開発を目的とした。 ボランティア16人の表皮ケラチンの酸化修飾を精査した所、7、8月に向けた増加と梅雨期の減少、被覆部位での抑制が見られた。また、秋期には修飾比率が減少し、各条件間の有意差も見られなくなり、表皮ケラチンのターンオーバーにより酸化修飾は蓄積しないことも確認された。一方、予想に反し、日焼け止めクリームの効果に有意差は見られなかった。この結果は、有効成分の紫外線吸収剤で生じる熱やラジカルによる影響の可能性から、紫外線防御効果を相殺したものと思われる。他の化学修飾に関しては、検出限界に近い修飾比率と個人差により、マーカーとして応用可能な物はなかった。 一方、表皮脂質のin vitro実験では、UVB暴露の結果、セラミドとphosphatidylethanolamine は種類が増加し、UVBが表皮脂質種の構造に影響を及ぼす可能性が示唆された。また二重結合の位置・異性化・酸化など構造の特定までは至らなかった。 更に本手法を毛髪ケラチンにも応用し、パーマ・染色などによる酸化修飾も確認した。

    Pyridoxamine (PM) is a drug candidate for various chronic diseases because it can inhibit advanced glycation end product formation. PM might therefore prevent protein damage from lipid hydroperoxide-derived aldehydes 4-oxo-2(E)-nonenal (ONE) and 4-hydroxy-2(E)-nonenal (HNE). Upon reaction with PM, ONE and HNE yielded an identical product containing a pyrrole ring, which structure was characterized by LC-MS and NMR. When human serum albumin (HSA) was reacted with a linoleic acid hydroperoxide, ONE modified more residues than did HNE. Upon treatment with PM, the formation of ONE-modified HSA peptides, but not of HNE-modified peptides, was reduced. Concomitantly, PM-ONE adducts increased in a dose-dependent manner. The inhibition effect of PM was confirmed in the cell system subjected to oxidative stress. Therefore, PM can inhibit lipid hydroperoxide-derived damage to proteins by trapping ONE preferentially, and the resulting PM-ONE adducts can be used as a dosimeter for ONE production.

    Aggregation of Aβ40 was affected by chemical stresses: promoted by H2O2, but inhibited by MG, 4-hydroxy-2(E)-nonenal, 4-hydroxy-2(E)-nonenal, and methylglyoxal. Major modification sites were also identified as follows: Met32, N-term, His6,13,14, Arg5, Lys16, etc. Non-aggregated Aβ40 was digested by neprilysin(NEP)or insulysin(IDE)for one day incubation. However, aggregated Aβ40 was not digested even by prolonged incubation (7 days). From the reaction mixture between non-aggregated Aβ40 and NEP/IDE, several long peptides (ca. 30) were identified. The major peptides (ca. 10 amino acids) were found to include the turn region (E22DVGS26), suggesting that NEP/IDE can recognize the secondary structure, not primary structure.

    Proteins are exposed to higher physiological/chemical stresses in some biological events. The resulting chemical modifications on proteins (such as oxidation, glycation, and lipidation) can provide meaningful information about the biological events. Therefore, abundant proteins (such as albumin, hemoglobin, and keratin), that are major targets of chemical modifications,can work as molecular medical record. From the view point, following results have been obtained: i) Chemical modifications on albumin has been screened and found the difference between individuals, ii) A sensitive and simultaneous analytical method has been developed for oxidized, nitrated, lipidated, and glycated hemoglobin, iii) A sensitive and non-invasive analytical method has been developed to estimate UV irradiation-induced skin damage through keratin oxidation.

    In the reactions of angiotensin (Ang) II with lipid peroxidation-derived 4,5-epoxy-2(E)-decenal or 4-hydroperoxy-2(E)-nonenal, the major modifications have been identified at the N-terminus and His6 of Ang II. 4-Oxo-2(E)-nonenal was shown to introduce an α-ketoamide moiety at the N-terminus of Ang peptides containing not only N-terminal Asp but also Ala, Arg and Val. The resulting N-terminal α-ketoamide is converted to the D- and L-amino acids by transamination in the presence of pyridoxamine. The reaction of Ang II and the hydroxyl radical produced N-terminal cyclized-Ang II (Ang C) as well as pyruvamide-Ang II (Ang P). Ang P and Ang C were not further metabolized by aminopeptidase A, which converts Ang II to Ang III. Formation and transamination of N-terminal α-ketoamide, and N-terminal cyclization were also observed in neuropeptides including amyloid beta 1-11. Analytical conditions were optimized to detect N-terminal α-ketoamide Ang peptides in human plasma.

    Background・Aim: Reactive metabolites of drugs result in adverse drug reactions through the formation of covalent biding to biological macromolecules. Therefore, trapping assays, such as glutathione trapping method, are widely performed together with LC-MS for the screening. However, the current strategy still has problems: ex. low throughput, structural gap between trapping reagents and biological macromolecules, and metabolic degradation of the reagents. We therefore aimed to develop a high-throughput/hybrid screening system to solve the problems. Outcome: I) Using simple motif of each nucleophilic site followed by mixing as a cocktail, the structural gap and metabolic degradation were avoided. II) Concomitant use of the stable isotope labeled motifs facilitated the screening because of the characteristic doublet peaks. III) Isotope pattern dependent scanning enabled the automated MS/MS analysis using the characteristic doublet peaks.

    In the reactions of angiotensin (Ang) II with lipid peroxidation-derived bifunctional electrophiles, 4-oxo-2(E)-nonenal (ONE) or 4-hydroxy-2(E)-nonenal (HNE), the major modifications have been identified at the N-terminus, Asp^1, Arg^2, and His^6 of Ang II. The identities of ONE- and HNE-modified Ang IIs were confirmed by mass spectrometry (MS) before and after the reaction with sodium borohydride. A novel ONE-derive pyruvamide-Ang II was formed via oxidative decarboxylation of N-terminal aspartic acid. The unexpected formations of Arg- and His- modifications were confirmed using model reactions with N^α-tert-butoxycarbonyl-amino acids. To investigate the biological effects of modified Ang IIs, MS-based label-free Ang II type 1 (AT_1) receptor binding assay and aminopeptidase (AP) A enzyme assay have been developed. Using these methodologies, we have confirmed that the oxidative modifications on the N-terminus of Ang II disrupt interactions with AT_1 receptor and APA, which could affect the regulation of cardiovascular function.

    【目的】アルツハイマー病、パーキンソン病、プリオン病に代表されるコンフォメーション病は、原因蛋白質の構造変化、即ち異常構造・凝集体生成に端を発する難病であり、多様な観点から研究されている。 しかし、本疾病の診断・研究は主に、『免疫化学的手法による関連蛋白質の分析』、『分子プローブによる凝集体の検出』を分析基盤として行われ、分別困難な微少な化学修飾の可能性を無視した議論となっている。一方、申請者は、コンフォメーション病のいずれも酸化ストレスと関係が深い事、蛋白質がin vitroで容易に酸化修飾を受ける事に着目し、生体中での酸化修飾が構造変化を惹起する可能性を示唆してきた。本研究では、早期診断マーカー候補の探索を念頭に、従来法では確認が困難であった酸化修飾を分別可能とする精緻な分析基盤の構築を目的とした。 【研究実績】モデルとしてアンジオテンシン類、ケラチン、ヒト血清アルブミン、インシュリンβ鎖等を用いた。各種酸化ストレス条件下、それらの化学修飾反応生成物の同定、反応機構の解明、化学修飾体のクリーンアップ法の開発、質量分析における化学修飾反応生成物のイオン化・フラグメント特性を明らかにすると共に、これらを駆使したスクリーニング法の構築を行った。その結果、生体試料中の微量かつ微小な蛋白質・ペプチドの化学修飾変化を分別し得る分析基盤の確立が示唆された。

    胆汁酸は主に腸肝循環を行なっており、その代謝もこれを中心として営まれるとされていた。申請者らは、赤血球の細胞質画分に3-オキソ胆汁酸を3α-並びに3β-ヒドロキシ体へと還元する酵素を見いだし、通常血中には見られない3-オキソ胆汁酸を有する先天性胆道閉塞症、原発性胆汁性肝硬変症患者との関連に着目した。そこで、本酵素の特性、肝胆道疾患との関連、更に肝3-ヒドロキシステロイドデヒドロゲナーゼとの相同性を明らかにし、これら疾患の病態解明に寄与することを目的として以下の実験を行った。 1.7及び12位にカルボニル基、水酸基を持つ5α-並びに5β-3-オキソ胆汁酸を合成した。 2.カルボニル酸素の交換反応及びその還元反応を利用し、ステロイド核上水酸基に重酸素及び重水素を導入した安定同位元素標識胆汁酸を調製した。 3.1.で合成した各種3-オキソ体及びその還元成績体である3α-、3β-ヒドロキシ体のカルボキシル基及び水酸基の誘導体化法を吟味し、負イオン検出ガスクロマトグラフィー(GC)/マススペクトロメトリー(MS)における分離系及び超高感度測定法を確立した。 4.液・液抽出法、固相抽出法を組み合わせた生体試料中胆汁酸の効果的なクリーン・アップ法を確立した。 5.種々3-オキソ胆汁酸を基質として用い、本酵素の基質特異性を精査した。 6.α-、3β-ヒドロキシ体への還元酵素活性の至適pH、Km値はそれぞれ異なっており両異性体への変換には異なる酵素の関与が示唆された。 7.3α-及び3β-ヒドロキシ体いずれからも3-オキソ体が生成することから本酵素がオキシドレダクターゼであることが示唆された。

    病態あるいは薬物の治療効果・副作用など、刻々と変化する生理現象を把握するには、静的なゲノム解析よりむしろ、動的なタンパク質組成の変動解析が重要である。しかしながら、数百万種に及ぶタンパク質を対象とするプロテオーム解析では、標品が必要な『絶対定量』は事実上不可能であり、タンパク質の増減のみを見る『相対定量』のみが行われている。それ故、コントロール実験との比較でバイオマーカー候補のタンパク質を見出しても、その絶対量が不明であり、定量的議論が困難である。そこで申請者らは、次世代型プロテオーム解析基盤の開発を究極の目標として、生体試料中の未知タンパク質の同定のみならず絶対定量をも可能とする分析基盤の構築を目的とする。
