    Congenital intestinal atresia (IA) is a birth defect characterised by the absence or closure of part of the intestine. Although genetic factors are implicated, mechanistic understanding has been hindered by the lack of suitable animal models. Here, we describe a medaka (Oryzias latipes) mutant, generated by N-ethyl-N-nitrosourea (ENU) mutagenesis, that develops IA during embryogenesis. Positional cloning identified a nonsense mutation in mypt1, encoding myosin phosphatase target subunit 1. Mutant embryos exhibited ectopic accumulation of F-actin and phosphorylated myosin regulatory light chain (Mrlc) in the intestinal epithelium, consistent with disrupted actomyosin regulation. These cytoskeletal abnormalities were accompanied by epithelial disorganisation, without notable alterations in cell proliferation, motility or apoptosis. Inhibition of myh11a, encoding smooth muscle (SM) myosin heavy chain, ameliorated the IA phenotype, whereas blebbistatin treatment completely rescued the defect, suggesting a non-contractile role prior to SM maturation. Together, these findings demonstrate that mypt1 loss disrupts intestinal morphogenesis through actomyosin dysregulation. Given the recent clinical identification of IA associated with MYPT1 variants, this medaka model offers a valuable platform to investigate the developmental and molecular basis of MYPT1-associated IA in humans.

    Despite being evolutionarily and commercially important, molluscs have been a traditionally challenging group to study, due to their difficulty in maintenance under lab conditions and the lack of a genetic toolkit. Previously, we showed that transgene expression can be attained in molluscan cells with reporter genes under a molluscan virus promoter sequence. Following up, we developed a simple, efficient and rapid transgene expression platform using primary hemocyte culture of Farrer's scallop Chlamys farreri, a marine bivalve mollusc. The protocol consists of two steps: collection and seeding of hemocytes and incubation for 1 to 4 days with DNA-reagent mixture. We evaluated seven transfection reagents for three bivalve species and found that X-tremeGENE 360 was highly efficient for DNA transfection, particularly for C. farreri. Subsequently, C. farreri hemocyte culture and transfection conditions were examined, such as culture medium, size and form of DNA, and the mixing ratio of DNA and transfection reagent. Using this protocol, we visualized the subcellular localization of four bivalve oncogenes, Cf-Mdm2-like, Cf-c-Myc-like, Cf-Mortalin-like, and Cf-Ras-like, tagged with EGFP. Our hemocyte platform provides an easy entry to study cellular and molecular biology of molluscs and can be readily adapted for advanced methods such as live imaging and DNA-protein interaction assays, making the study of molluscs more accessible to the scientific community.

    Understanding gene functions in marine invertebrates has been limited, largely due to the lack of suitable assay systems. Such a system requires investigative methods that are reproducible and can be quantitatively evaluated, such as a cell line, and a strong promoter that can drive high expression of a transgene. In this study, we established primary cell culture from a marine bivalve mollusc, Mizuhopecten yessoensis. Using scallop primary cells, we optimized electroporation conditions for transfection and carried out a luciferase-based promoter activity assay to identify strong promoter sequences that can drive expression of a gene of interest. We evaluated potential promoter sequences from genes of endogenous and exogenous origin and discovered a strong viral promoter derived from a bivalve-infectious virus, ostreid herpesvirus-1 (OsHV-1). This promoter, we termed OsHV-1 promoter, showed 24.7-fold and 16.1-fold higher activity than the cytomegalovirus immediate early (CMV IE) promoter and the endogenous EF1α promoter, the two most commonly used promoters in bivalves so far. Our GFP assays showed that the OsHV-1 promoter is active not only in scallop cells but also in HEK293 cells and zebrafish embryos. The OsHV-1 promoter practically enables functional analysis of marine molluscan genes, which can contribute to unveiling gene-regulatory networks underlying astonishing regeneration, adaptation, reproduction, and aging in marine invertebrates.

    Abstract Background Intestinal atresia (IA) is a congenital gut obstruction caused by the absence of gut opening. Genetic factors are assumed to be critical for the development of IA, in addition to accidental vascular insufficiency or mechanical strangulation. However, the molecular mechanism underlying IA remains poorly understood. Results In this study, to better understand such a mechanism, we isolated a mutant ofOryzias latipes(the Japanese rice fish known as medaka) generated by N-ethyl-N-nitrosourea mutagenesis, in which IA develops during embryogenesis. Positional cloning identified a nonsense mutation in themyosin phosphatase target subunit 1(mypt1) gene. Consistent with known Mypt1 function, the active form of myosin regulatory light chain (MRLC), which is essential for actomyosin contraction, and F-actin were ectopically accumulated in the intestinal epithelium of mutant embryos, whereas cell motility, proliferation and cell death were not substantially affected. Corresponding to the accumulation site of F-actin/active MRLC, the intestinal epithelium architecture was disordered. Importantly, blebbistatin, a non-muscle myosin inhibitor, attenuated the development of IA in the mutant. Conclusions Cytoskeletal contraction governed bymypt1regulates the integrity of the embryonic intestinal epithelium. This study provides new insight into our understanding of the mechanism of IA development in humans. Bullet Points Medakamypt1mutants display intestinal atresia. The level of phosphorylated myosin regulatory light chain was higher inmypt1mutant embryos than inwild-typeembryos.     Genome editing has had profound effects on biological experimentation and can now be applied to many organisms, including non-conventional models. However, the introduction of genome editing components is time- and labor-consuming and sometimes requires special skills for microinjection. In this study, we developed a technique to deliver exogenous proteins into eggs by injection into the mother's ovary (IMO), which leads to the delivery of CRISPR/Cas9 into the eggs of oviparous animals, including fish. To test this technique, we examined whether exogenous proteins tagged with GFP or luciferase (Luc), and fluorescent-labeled RNP (Cas9 and sgRNA complex), can be delivered into eggs by IMO. When GFP-Luc or Cas9-Luc was delivered by IMO, their incorporation into fertilized eggs was confirmed by GFP fluorescence or luciferase activity; proteins were accumulated in the yolk. Cas9-RNP (targeting tyrosinase) was also incorporated into the eggs. However, genome editing of the target gene, tyrosinase, was not observed yet. This is presumably because the RNP delivered by IMO was packed in the yolk granules and did not reach into the embryonic nuclei. Thus, this report shows that exogenous molecules including Cas9-RNP were successfully delivered into fertilized eggs by IMO. Transferring the delivered RNP into nuclei will be critical for successful genome editing via the IMO delivery system.

    BACKGROUND: The distribution of sensory organs is important for detecting environmental signals efficiently. The mechanosensory receptors of the lateral line system, neuromasts, are stereotypically distributed over the head and body surface of fish, although how neuromasts arise in these predetermined positions during development remains unclear. RESULTS: We investigated the development of the anterior lateral line (ALL) system in zebrafish head. The ALL neuromasts formed in the predetermined positions through proliferation and differentiation of (a) nonmigratory lateral line primordia, (b) migratory primordia, (c) interneuromast cells connecting preexisting neuromasts, and (d) budding primordia. We demonstrated that R-spondin2 (Rspo2), an activator of Wnt/β-catenin signaling, is required for the development of a particular set of neuromasts associated with hyomandibular cartilage. Further genetic analyses suggested that Rspo2, which emanates from the hyoid mesenchyme, acts on the adjacent neuromast progenitor cells to stimulate their proliferation through activating Wnt/β-catenin signaling. CONCLUSION: This study has revealed novel mechanisms for neuromast positioning through local tissue-tissue interactions, providing insights into the development and evolution of the vertebrate head.

    We examined the efficiency of electroporation for the delivery of plasmid into the skeletal muscle and also examined the subsequent secretion of recombinant protein into the circulation system, using zebrafish, Japanese flounder, and bubble-eye goldfish. The expression area of GFP fluorescence was markedly expanded by electroporation. Introduced plasmid was retained in the muscle cells and continued to express GFP for at least 180 days in zebrafish, indicating the long lifespan of plasmid DNA in the muscle cell. Luciferase and a fusion of growth hormone (GH) and luciferase were secreted into the blood from muscle electroporated with CMV:secNluc and CMV:GH-Luc plasmids, respectively, indicating that recombinant proteins such as peptide hormones can be supplied to the blood by plasmid electroporation into muscle. Interestingly, luciferase activity was detected from fertilized eggs laid by zebrafish females that had been electroporated with CMV:secNluc, indicating that maturing oocytes incorporated recombinant protein from the blood stream that had been secreted from the muscle. The plasmid electroporation system reported here also may work for the delivery of recombinant proteins, such as Cas9, into the oocytes. Since the GH-Luc fusion protein was detected in the lymph of the eye-sac of bubble-eye goldfish, this fish may be useful for the production of recombinant protein.

    Vitellogenin (Vtg), a yolk nutrient protein that is synthesized in the livers of female animals, and subsequently carried into the ovary, contributes to vitellogenesis in oviparous animals. Thus, Vtg levels are elevated during oogenesis. In contrast, Vtg proteins have been genetically lost in viviparous mammals, thus the yolk protein is not involved in their oogenesis and embryonic development. In this study, we identified Vtg protein in the livers of females during the gestation of the viviparous teleost, Xenotoca eiseni Although vitellogenesis is arrested during gestation, biochemical assays revealed that Vtg protein was present in ovarian tissues and lumen fluid. The Vtg protein was also detected in the trophotaeniae of the intraovarian embryo. Immunoelectron microscopy revealed that Vtg protein is absorbed into intracellular vesicles in the epithelial cells of the trophotaeniae. Furthermore, extraneous Vtg protein injected into the abdominal cavity of a pregnant female was subsequently detected in the trophotaeniae of the intraovarian embryo. Our data suggest that the yolk protein is one of the matrotrophic factors supplied from the mother to the intraovarian embryo during gestation in X. eiseni.

    Zebrafish as a vertebrate model have contributed to elucidating the functions of important factors involved in development, metabolism, and various diseases that are shared among human and animals. Zebrafish have also attracted attention as a gut microbiota model and have been studied for host-microbe interaction using diverse approaches, which involve host immune, metabolic, and behavioral responses to specific microbes. Using zebrafish as disease models for diabetes and pathogen infection models has been especially valuable for the elucidation of important factors as candidate targets for pharmaceutical development. Zebrafish offer unique advantages over mammalian models as a gut microbiota model because their transparency enables live imaging of individual microbes and immune cells within the host. Rearing zebrafish and setting up experiments for microbiome research entail consideration of aquatic-specific factors that may significantly impact the microbiota. Although fish are not considered as experimental animals in Japan, the handling of fish with ethical consideration is required, due to the international trend toward the awareness of animal welfare including fish. In this review, we provide an overview of zebrafish as an experimental animal model and summarize recent important studies using zebrafish as a host of microbes. We then discuss perspectives on zebrafish research as an alternative animal model and international ethical differences in handling fish, and review the applicability and prospect of fish models for microbiome research.

    Japanese flounder is one of the important fish species because of their commercial value, however, abnormal pigmentation is problematic in the aquaculture production. Aim of this study is to investigate the molecular mechanisms underlying the left/right asymmetric pigment cell differentiation. Genes involved in pigmentation were isolated by comparing gene expression profile of ocular side and blind side of flounder. Gene expression analysis using abnormal pigment fish with ectopic pigmentation revealed that these genes were expressed in such ectopic pigment sites and peripheries, which suggested that these genes also involved in ectopic pigmentation. In order to investigate the function of these genes, GFP-knock in strains were generated by using CRISPR/Cas9 genome editing system: gene expression can be monitored in live embryos with the GFP fluorescence.

    私達は,これまで異体類の発生とその内分泌調節について検討し,変態に伴う眼球移動や左右非対称な体色形成、骨形成はレチノイン酸(RA),甲状腺ホルモン,コルチゾル等の内分泌シグナルにより制御され,種苗の形態異常(眼位や体色異常,骨異常)は,これらシグナル回路の攪乱によって発生することを示唆している。本研究では,飼育技術と育種の両面から健苗育成に貢献することを目的として、これまでの研究を発展し,変態を調節するシグナル回路を解明した上で,種苗で頻発する無眼側黒化の発生原因となるシグナル回路の乱れを究明し、さらに無眼側黒化のリスク遺伝子を特定することをめざす。本年度は、ヒラメ・カレイ類の変態期に起こる有眼側限定的な色素胞の分化の発生制御機構を解明することに成功した。すなわち色素胞の前駆細胞は、変態期初期に幹細胞に近い状態で脊髄周辺に発生する。この時、幹細胞は、神経堤細胞のマーカー遺伝子であるSox10を発現し、このSox10の発現誘導にWntシグナルが働くことが明らかになった。脊髄周辺で分化したSox10陽性幹細胞は、脊髄から伸びる神経軸索に沿って背鰭基部に到達し、さらにそこから伸びる神経に沿って左右の皮膚に移動する。幹細胞から色素胞への分化はRA依存的であり、無眼側ではRAの分解酵素であるcyp26bが強発現して色素胞の分化が抑制され、有眼側のみ着色することを明らかにした。RA合成酵素は変態期に腸管上皮で強発現することから、腸管で合成されたRAは血流を介して体全体に拡散して色素胞の分化を誘導するが、無眼側ではcyp26bにより分解されることで色素分化に左右差が発生することが示唆された。 無眼側黒化のリスク遺伝子を特定するための基盤整備として、ヒラメのほぼ全遺伝子について、cDNAの塩基配列を解読し、塩基配列の一塩基多型の情報を得るために複数個体のcDNA塩基配列を取得した。

    私達は、ヒラメ・カレイ類の一種であるササウシノシタが海産魚の中では小型の部類であり、かつ産卵期間が長いことに着目し、海産魚のモデル生物として利用するための研究を進めている.これまでに周年産卵システムや研究用小型飼育システムを立ち上げるなど準備し、現在、養殖魚の種苗生産で問題となっている体色や骨格の形態異常の発生機序について研究を行っている。より分子レベルで踏み込んだ研究に着手するために、発生工学や遺伝学の技術を使いたいと望んでいるが、受精から産卵まで1年と世代サイクルが長いことが障壁となっている。似たように大型の養殖対象魚種はいずれも産卵までの期間が3年と長く、育種が難しいという問題がある。 私達は、上記の研究と並行して、ゼブラフィッシュを使って、成熟雌への腹腔内注射によるゲノム編集技術の開発を行っており、実用化に近づいている。このような背景から、「開発中のゲノム編集技術を使って、ササウシノシタを小型・早期成熟化し」、ゼブラフィッシュ並に便利なモデル生物に改変することを着想し、そのための計画を立案した。本研究は、養殖対象魚の育種を省スペース・時短化するための試験研究としての意義も大きい. 本年度は、3つの課題を検討した。ゲノム編集を進めるためには、ゲノムDNAとcDNAの塩基配列情報の整備が必須であるので、第一の課題では、次世代シーケンサーを使って、ササウシノシタのゲノムDNAと全遺伝子のcDNAの塩基配列の解読を試みた。2番目に、ササウシノシタの雌親魚の腹腔に注射して、プラスミドを卵母細胞に導入できるか検討した。3番目に、ササウシノシタの成長ホルモン遺伝子(GH)を標的としたCRISPR/Cas9プラスミドを作成して、雌親魚の腹腔に注射し、GH遺伝子のノックアウトを試みた。

    To apply genome editing to aquaculture fish, this study aimed at development of delivery method of CRISPR/Cas9 into oocytes by injection into female abdominal cavity. It is known that yolk protein (Vg) is taken into the oocytes by using Vg receptor binding sequence, HLTKTKDL. We found that by binding with HLTKTKDL, exogenous protein injected into the female abdominal cavity is taken into the oocytes. However, since the protein was not transferred from yolk to embryonic cytoplasm, further improvement is required for applying this method to genome editing. We also found that when conjugate of plasmid vector and lipid carrier for in vivo usage was injected into female, protein is synthesized from injected vector in the oocytes. We expect that this method would enable the genome editing by injection into female.

    Flatfish transform their body plan during metamorphosis. Flatfish are important fish as commercial target and actively produced by aquaculture, however, the abnormalities in pigmentation and malformation are the common issue around the world. In this international collaboration, we investigated the metamorphosis of flatfish comparing Japanese flounder and Mediterranean turbot. New findings include, blind-side specific degeneration of cartilage around eye was specific phenomena for flounder and not observed in turbot; genes involved in pigmentation is conserved among teleost fish but not in several tetrapods.

    Flounder transform their body plan during metamorphosis; right eye migrates to the other side and pigment cells differentiate only in the ocular side. Gene expression profile were compared between ocular and blind side in tissues around supra orbital bar at early metamorphic stage and skin and muscle tissue at later metamorphic stage in order to isolate genes expressed asymmetrically between ocular and blind side. Candidate genes were further analyzed by in situ hybridization and suggested that genes might be involved in right side specific degeneration of supra orbital bar and genes that genes might be involved in ocular side specific pigment cell differentiation.

    Flounder larvae are externally symmetric commonly to general fishes, but they transform to left-right (L-R) asymmetric body by migrating one eye to the other side of body and then differentiating chromatophores at ocular side skin. In the hatcheries for aquaculture, since abnormal pigmentation, including pigmentation of ocular side and pseudo-albinism of blind side, often appears, elucidation of the mechanism to induce abnormal pigmentation is required.     One of representative genetic traits required for aquaculture is fish character adequate for breeding, such as non-nervousness. In mammals, polymorphism of single amino acid repeat and microsatellites in promoter region that evolved in the genes related with brain function cause variation of character among strains. Medaka strains of d-rR and HNI-II have opposing character; the former being tame, while the latter being nervous. Using these two strains, this study aimed at searching the genes correlated with characters of fish strains by using bioinformatics and genome databases. Serotonin receptor, androgen receptor and dopamine receptor were selected as candidate genes. Since polymorphism in repetitive sequences were detected among two strains and wild medaka, it is highly possible that selected three genes are actually correlated with character of strains.

    Flounder transform their body plan during metamorphosis and turn into asymmetric body form with two eyes on left side and ocular-side pigmentation. Metamorphosis is triggered by thyroid hormone signal, however, little is known about molecular basis of the metamorphic morphogenesis. Here, we compared gene expression profile between ocular and blind side by RNA-seq analysis using next generation sequencer, which identified 32 ocular side dominant genes and 7 blind side dominant genes. These candidates are now under screening by in situ hybridization to confirm left-right biased gene expression. So far, most of them showed biased gene expression as expected, which include known pigmentation genes such as gch2.

    We investigated the developmental mechanism that controls external asymmetry of flatfish (eye location and skin pigmentation). It was demonstrated that the Nodal signaling pathway (NSP), which is known to control the sidedness of internal organs and neural circuit of habenulae, fixes the eye-sidedness by controlling the forebrain asymmetry formation. The dextral and sinistral flatfishes are segregated by the opposite asymmetry formation at the habenulae and optic chiasma under the control of the NSP. It was found that the precursors of adult-type melanophores that locate at the base of dorsal fin migrate in the myoseptum toward both left and right lateral sides. The asymmetry of skin pigmentation is established by the maturation of precursors to chromatoblasts during the migration in the myoseptum of the ocular side. Exposure to retinoic acid (RA) induces maturation of the precursors also in the ocular side, resulting in the coloration of blind side skin.

    Establishment of fish iPS cells is expected to enable the development of new techniques for maintaining selected aquaculture fish strains as frozen cells. This study aims as development of fundamental techniques essential for establishing fish iPS cells. We established medaka and zebrafish transgenic lines that emit GFP fluorescence under the control by oct4 promoter. We also developed cell culture system in which mulitpotency of cells can be monitored by GFP fluorescence. In addition, two possibilities on iPS cell production were suggested. First, transfection of Minicircle DNA may transform embryonic cells to iPS cells. Second, mulitpotent cells that appear in regenerating fin may be efficiently transformed to iPS cells.

    This study aimed to accumulate the basic scientific knowledge on the metamorphosis in fish to help establishing more efficient production of the flounder aquaculture which has a huge economical impact in Japan. I conducted molecular and developmental genetic studies using medaka as a model. GFP transgenic lines were generated in order to monitor in vivo thyroid hormone level in a live fish. In another approach to establish medaka as a model system for studying metamorphosis, approximately 20 candidate marker genes that are implicated in thyroid grand development and/or thyroid hormone signaling were cloned and their embryonic expression patterns were examined.
