    Light quality affects plant growth and the functional component accumulation of fruits. However, there is little knowledge of the effects of light quality based on multiomics profiles. This study combined transcriptomic, ionomic, and metabolomic analyses to elucidate the effects of light quality on metabolism and gene expression in tomato fruit. Micro-Tom plants were grown under blue or red light-emitting diode light for 16 h daily after anthesis. White fluorescent light was used as a reference. The metabolite and element concentrations and the expression of genes markedly changed in response to blue and red light. Based on the metabolomic analysis, amino acid metabolism and secondary metabolite biosynthesis were active in blue light treatment. According to transcriptomic analysis, differentially expressed genes in blue and red light treatments were enriched in the pathways of secondary metabolite biosynthesis, carbon fixation, and glycine, serine, and threonine metabolism, supporting the results of the metabolomic analysis. Ionomic analysis indicated that the element levels in fruits were more susceptible to changes in light quality than in leaves. The concentration of some ions containing Fe in fruits increased under red light compared to under blue light. The altered expression level of genes encoding metal ion-binding proteins, metal tolerance proteins, and metal transporters in response to blue and red light in the transcriptomic analysis contributes to changes in the ionomic profiles of tomato fruit.

    Although several reports exist on the use of X-ray analysis in vegetables and fruits to examine internal disorders, cavities, and porosity, information on X-ray analysis of qualities, such as texture, is lacking as well as information on X-ray analysis of legumes. Therefore, this study aimed to perform X-ray analysis with sensory and rheometer tests in cooked vegetable soybean (edamame). Edamame is popular worldwide due to its deliciousness and nutritional value. Vascular structures and cracks around them were clearly visualized using X-ray phase-contrast computed tomography (CT) imaging. In addition, we observed the fine structure of the seed coat, which could be important for seed development, germination, and processing. The density in the edamame beans declined as the boiling time increased, promoting a reduction in hardness described in sensory and rheometer tests. The reduction in density proceeded from the gap between cotyledons, the opposite side of the hypocotyl, and the crack. Collectively, the findings show that the high-resolution X-ray phase-contrast CT imaging conducted in a nondestructive manner may help in effectively evaluating the quality of vegetables and in observing the internal structures related to plant development.

    Polyols such as sorbitol and ribitol are a class of compatible solutes in plants that may play roles in tolerance to abiotic stresses. This study investigated the effects of water stress on sorbitol biosynthesis and metabolism and sorbitol and ribitol accumulation in tomato (Solanum lycopersicum L.). Water stress induced by withholding water and by using polyethylene glycol as a root incubation solution to mimic water stress, and NaCl stress were applied to wild-type (WT) and three genetically-modified lines of tomato (cv. Ailsa Craig), a control vector line TR22, and 2 sorbitol dehydrogenase (sdh) antisense lines TR45 and TR49. Sorbitol and ribitol content, as well as the enzymatic activities, protein accumulation, and gene expression patterns of the key sorbitol cycle enzymes aldose-6-phosphate reductase (A6PR), aldose reductase (AR), and sorbitol dehydrogenase (SDH), were measured in mature leaves. In response to the stresses, both sorbitol and ribitol accumulated in leaf tissue, most significantly in the sdh antisense lines. A6PR, characterised for the first time in this work, and AR both exhibited increased enzymatic activity correlated with sorbitol accumulation during the stress treatments, with SDH also increasing in WT and TR22 to metabolise sorbitol, reducing the content to control levels within 3 days after re-watering. In the sdh antisense lines, the lack of significant SDH activity resulted in the increased sorbitol and ribitol content above WT levels. The results highlighted a role for both A6PR and AR in biosynthesis of sorbitol in tomato where the high activity of both enzymes was associated with sorbitol accumulation. Although both A6PR and AR are aldo-keto reductases and use NADPH as a co-factor, the AR-specific inhibitor sorbinil inhibited AR only indicating that they are different enzymes. The determination that sorbitol, and perhaps ribitol as well, plays a role in abiotic responses in tomato provides a cornerstone for future studies examining how they impact tomato tolerance to abiotic stresses, and if their alteration could improve stress tolerance.

    To elucidate the molecular mechanism of juvenility and annual flowering of fruit trees, FLOWERING LOCUS C (FLC), an integrator of flowering signals, was investigated in apple as a model. We performed sequence and expression analyses and transgenic experiments related to juvenility with annual flowering to characterize the apple FLC homologs MdFLC. The phylogenetic tree analysis, which included other MADS-box genes, showed that both MdFLC1 and MdFLC3 belong to the same FLC group. MdFLC1c from one of the MdFLC1 splice variants and MdFLC3 contain the four conserved motives of an MIKC-type MADS protein. The mRNA of variants MdFLC1a and MdFLC1b contain intron sequences, and their deduced amino acid sequences lack K- and C-domains. The expression levels of MdFLC1a, MdFLC1b, and MdFLC1c decreased during the flowering induction period in a seasonal expression pattern in the adult trees, whereas the expression level of MdFLC3 did not decrease during that period. This suggests that MdFLC1 is involved in flowering induction in the annual growth cycle of adult trees. In apple seedlings, because phase change can be observed in individuals, seedlings can be used for analysis of expression during phase transition. The expression levels of MdFLC1b, MdFLC1c, and MdFLC3 were high during the juvenile phase and low during the transitional and adult phases. Because the expression pattern of MdFLC3 suggests that it plays a specific role in juvenility, MdFLC3 was subjected to functional analysis by transformation of Arabidopsis. The results revealed the function of MdFLC3 as a floral repressor. In addition, MdFT had CArG box-like sequences, putative targets for the suppression of flowering by MdFLC binding, in the introns and promoter regions. These results indicate that apple homologs of FLC, which might play a role upstream of the flowering signals, could be involved in juvenility as well as in annual flowering. Apples with sufficient genome-related information are useful as a model for studying phenomena unique to woody plants such as juvenility and annual flowering.

    The effect of light quality on flowering in everbearing strawberry (Fragaria × ananassa Duch.), a long-day plant, was investigated under night illumination conditions. Two everbearing strawberry cultivars, ‘Natsuakari’ and ‘Suzuakane,’ were tested. Flowering was promoted under far-red light, but it was almost inhibited under red light at 30/25°C (day/night temperature) in a natural light type phytotron. In another treatment, plants were grown under night illumination in a glass house from September. In this case, flowering was reduced under red light conditions compared with under an incandescent lamp. In everbearing strawberry cultivation, flowering is markedly promoted and plants may die due to self-topping under high temperature and long-day conditions. However, the mortality rate due to self-topping was lower under red light in this study. These results reveal the flowering response of everbearing strawberry cultivars to red and far-red light under relatively high temperatures, and the possibility of using red light to maintain vegetative growth by regulating flowering.

    Soil contains various essential and nonessential elements, all of which can be absorbed by plants. Plant ionomics is the study of the accumulation of these elements (the ionome) in plants. The ionomic profile of a plant is affected by various factors, including species, variety, organ, and environment. In this study, we cultivated various vegetable crop species and cultivars under the same field conditions and analyzed the level of accumulation of each element in the edible and nonedible parts using ionomic techniques. The concentration of each element in the edible parts differed between species, which could be partly explained by differences in the types of edible organs (root, leaf, seed, and fruit). For example, the calcium concentration was lower in seeds and fruit than in other organs because of the higher dependency of calcium accumulation on xylem transfer. The concentration of several essential microelements and nonessential elements in the edible parts also varied greatly between cultivars of the same species, knowledge of which will help in the breeding of vegetables that are biofortified or contain lower concentrations of toxic elements. Comparison of the ionomes of the fruit and leaves of tomato (Solanum lycopersicum) and eggplant (S. melongena) indicated that cadmium and boron had higher levels of accumulation in eggplant fruit, likely because of their effective transport in the phloem. We also found that homologous elements that have been reported to share the same uptake/transport system often showed significant correlation only in a few families and that the slopes of these relationships differed between families. Therefore, these differences in the characteristics of mineral accumulation are likely to affect the ionomic profiles of different families.

    Flower bud formation in shoot apex cultures was investigated using vegetative buds from the leafing and dormant stages in Gentiana triflora Pall. var. japonica Hara and Gentiana scabra Bunge var. buergeri Maxim. A few plantlets had flower buds 105 days after starting the in vitro culture. When buds from the dormant stage were used for the culture, 6 % or less of the plantlets had flower buds in all lines. After 165 days, more than 50 % of plantlets from buds from the leafing stage had flower buds, while flower bud formation was lower in plantlets from buds at the dormant stage. There were no flower buds found on plantlets from buds at the dormant stage during the culture period in Gentiana scabra.<br> The effects of flower buds and of plant growth regulators on shoot formation were examined using in vitro node culture. The growth of shoots at the upper nodes from plantlets with flower buds was poorer than that from plantlets without flower buds. By contrast, shoots at the lower nodes from plantlets with flower buds were rather long. In culture medium containing forchlorfenuron and naphthalene acetic acid (NAA), many shoots formed from the nodal segments of plantlets without flower buds. Plant growth regulators also enhanced shoot formation from the nodal segments of plantlets with flower buds. The lengths of the shoots were similar between plantlets with and without flower buds. These results suggest that plantlets with flower buds are still useful for node culture, especially with the addition of forchlorfenuron and NAA.<br>

    Effect of low temperature on sorbitol metabolism was investigated in peach (Prunus persica Batsch var. vulgaris Mixim.) leaves. Sorbitol content in leaf discs was increased after 16-hr incubation at 10/4℃ whereas it remained constant at 18℃. The conversion of [^<14>C] glucose to [^<14>C] sorbitol was also hastened by the low temperature treatment. Northern blot analysis indicated that the level of mRNA encoding putative S6PDH was higher under low temperature than control. These results suggest that sorbitol accumulation is induced by low temperature and the accumulation is due to an increase in sorbitol biosynthesis from glucose that is related to the expression of the S6PDH gene.

    Changes in indole-3-acetic acid (IAA) contents of melon (Cucumis melo L. cv. Prince) fruits were plotted during their developmental stages. The IAA content of mesocarp was almost constant during fruit development but that of seeds increased for 20 days after pollination, then decreased significantly. Hence, seeds contained much more IAA than the mesocarp throughout fruit development. The seeds may effuse IAA to the enlarging mesocarp, so that its IAA content remain constant.<BR>Exogenous application of IAA on the activities of sucrose-metabolizing enzymes, which are closely related to the sink activity of fruits, stimulated activities of the cell wallbound and soluble acid invertases, but not those of sucrose-and sucrose phosphate synthases. Thus, endogenous IAA may play an important role in the induction of cell wallbound and soluble acid invertases.<BR>We postulate that exported IAA from seeds to mesocarp stimulate cell wallbound and soluble acid invertase activities, thereby strengthening its sink activity during fruit development.

    The formation of nitrosylleghemoglobin (LbNO) was examined in cowpea and pea nodules in relation to the inhibition of nitrogen fixation by nitrate. Leghemoglobin was of the ferrous type and was mainly converted to LbNO in cowpea nodules when the acetylene-reducing activity decreased to 45% of control values as a result of the supply of nitrate. In nodules of nitrate-treat-ed pea plants, leghemoglobin was also of the ferrous type and LbNO was a minor component of leghemoglobin. The levels of LbNO isolated from nodules corresponded to the levels of LbNO calculated from equilibrium constants for LbNO and the concentration of nitrite in nodules. The dissociation rate constants for LbNO from both cowpea and pea were much smaller than those for Lb0_2 Or LbCO, as is also the case in soybean. These results indicate that the inhibition of the functions of leghemoglobin, due to the accumulation of LbNO, induces a decrease in nitrogen fixation in cowpea nodules, and that the inhibition of nitrogen fixation in pea nodules is not related to the formation of LbNO.

    Flowering control with light quality is now attracting attention as LEDs become more popularized and genetic researches make more advancement. In some model plants used in experiments, a mechanism of flowering in long-day plants promoted by far-red light has been revealed. In addition, a research concerning effects of light quality in the flowering control of cut flowers conducted in Tohoku district reveled that the long-day treatment with far-red light is effective in promoting flowering of Eustoma and stock.

    The activity and gene expression of sorbitol-6-phosphate dehydrogenase (S6PDH), NAD^+-dependent sorbitol dehydrogenase (NAD-SDH), sucrose phosphate synthase (SPS), sucrose synthase (SS) and soluble acid invertase (S-AIV), in developing 'La France' pear leaves in relation to the metabolism of sorbitol and sucrose as translocated sugars, were investigated. The levels of activity and mRNA of S6PDH were significantly higher in mature leaves than in folded and young ones. S6PDH activity was more than ten times higher than SPS activity. SPS activity remained at an almost constant level throughout the developmental stages; it was undetectable except in the very young leaves although its mRNA was present. NAD-SDH and SS activities were higher in young leaves than in mature ones; the levels of mRNAs, however, exhibited an inverse patterns to that of activities. The levels of both activity and mRNA of S-AIV were significantly higher in folded and young leaves than mature ones. The distinctly higher expression of S6PDH than that of SPS in mature leaves suggests that the former is more important than the latter in relation to CO_2 assimilation (source activity). Thus, the most important change in the transition from sink to source of the pear leaf is the increasing expression of S6PDH gene with leaf maturation.

    Sorbitol-6-phosphate dehydrogenase (S6PDH) is known to be a key enzyme in the biosynthesis of sorbitol, an early product of photosynthesis, common to fruit trees of the Rosaceae family. Effects of low temperature and abscisic acid (ABA) on the expression of the S6PDH gene were investigated in apple leaves. In leaf-disc experiments, the expression of S6PDH was enhanced by an ABA treatment, as well as by low temperature and high-salinity stresses. The level of S6PDH mRNA increased 8h after the addition of ABA; the highest level of S6PDH mRNA resulted on exposure to 10μM ABA. The level of S6PDH mRNA in leaves of apple trees growing in an orchard increased with a decrease in temperature in the fall while ABA content increased. This induction may partly be a stress-response to low temperature, a prerequisite for freezing tolerance during the coming winter. Southern blot analysis revealed that S6PDH is a single-copy gene in the apple genome, indicating that it is a unique, multifunctional one, for sorbitol biosynthesis under various stress responses, as well as for the translocation of photosynthates.

    To clarify how ABA and IAA may mediate sugar uptake during fruit development, the kinetics of sugar accumulation by excised discs of melon (Cucumis melo L. cv. Prince) was studied. Uptake kinetics for glucose and fructose in discs of young and premature fruit, 14 and 28 days after pollination, respectively, and for sucrose in young tissues have biphasic (curvilinear) concentration curves. This indicates the presence of both carrier-mediated and simple diffusional uptake systems. The carrier-mediated system occurs at lower sugar concentrations for the three sugars in young fruit, but for glucose and fructose in premature fruits ; it was not detectable in mature fruit 42 days after pollination. The Km value for carrier-mediated sugar uptake ranged from 6 to 14 mM and did not change with fruit development and among sugars. ABA at 10^<-5>M enhanced the simple diffusional uptake of the three sugars, whereas IAA at the same concentration stimulated the carrier-mediated system of the three sugars but not the simple diffusional uptake system for sucrose. The roles of ABA and IAA on the carrier-mediated and simple diffusional uptakes systems for sugars seem to be very important during the growth and maturation of the melon fruit.

    プラズマは固体、液体、気体のいずれとも異なる特有の性質を持っており、物質の第4の状態とされている。プラズマを照射した水(プラズマ水)には6種類以上の活性酸素種(ROS)および活性窒素種(RNS)が含まれるため、機能性成分の増加など植物への様々な効果が期待されるが、園芸学分野における報告は希少である。特に効果については既知の分子種のみならず植物への機能が未知の分子種が混在しており、新たな効果が期待できることから、プラズマ水の植物への未知なる効果を明らかにし、園芸学分野において新規利用技術を開発することができれば初めての知見となる。プラズマは大気を原料として作成され、プラズマ水に含まれる分子種は最終的には水や硝酸イオンに変換されて植物に利用されるため環境に優しく、生成時の消費エネルギーは少ないためカーボンニュートラルにも貢献し得る。そこで本研究では、プラズマ水が果菜類および葉菜類の生産と品質に及ぼす影響を調べるとともに、その生理機構を解明し、新たな利用技術を開発することを目的とした。 本研究では葉菜類(コマツナ)と果菜類(トマト)を用いて、プラズマ水の生育や品質への影響について調査する。本年度は、まずコマツナへのプラズマ水の効果について検証した。その結果、葉身長はプラズマ水処理により有意に短かくなったが、葉柄長は対照区と比較して有意差が認められなかった。そして、葉全長(葉柄と葉身)はプラズマ水処理により有意に短かくなった。葉数はプラズマ水処理による影響は認められなかったが、地上部新鮮重がプラズマ水処理により有意に小さくなった。以上の結果から、本研究の環境下において、プラズマ水処理はコマツナの生育を抑制することが考えられた。一方で、葉色はプラズマ水処理により有意に濃くなった。さらに、抗酸化能の指標であるDPPHのラジカル消去活性はプラズマ照射区で増加する傾向であった。

    慣行の切り花類の栽培では,開花調節は白熱電球によって行われていたが,白熱電球は寿命が短く,電気エネルギーの光エネルギーへの変換効率が低いことから,代替として長寿命で省電力の発光ダイオード(LED)を光源とする栽培が普及しつつある.しかし,白熱電球が幅広い波長の光を放射するのに対して,LEDは単色光を放射することから,代替することにより白熱電球で栽培した場合と異なる開花反応を示すことが予想される.また,白熱電球による開花調節の原理としては,フィトクロムに基づく赤色光と遠赤色光が制御要因であるとされている.一方,青色光も花成制御に関わっており,長日植物のモデル植物であるシロイヌナズナでは開花が促進されることが知られているが,LEDが普及するまでは栽培用に簡易に利用できる青色光の光源がなかったことから,開花調節に利用されてこなかった.そこで本課題ではまず,青色LEDによる開花調節に焦点を当てる. さらに,害虫防除は一般には農薬散布によって行われ,また,光の殺虫効果は波長が短いほど強く、紫外線BやCのように波長が短い紫外線だけにあるとされてきた.しかし最近,従来の常識に反して、可視光である青色光に種々の昆虫に対する致死効果が発見されたため,青色光による害虫防除を着想した.以上により,青色光による開花の制御機構を解明するとともに殺虫効果を検証し,青色LEDによって開花と害虫を同時に制御する全く新しい栽培体系を開発することを目的とする.

    Useful components are reduced in tomato compared to wild tomato. In this study, introgression lines of tomato, in which part of the chromosome of a cultivated species was replaced with the chromosome of a wild species was used to understand the mechanism in high accumulation of useful components. L-ascorbic acid (L-AsA) is an abundant antioxidant in plants and plays an important role in detoxifying reactive oxygen species (ROS) produced during stress, and amino acids are the main components of tomato's umami and have health functionality. One IL line with high accumulation was screened, respectively. In the ascorbic acid high-accumulation line, comprehensive expression analysis of biosynthetic genes was performed to clarify a part of the ascorbic acid high-accumulation mechanism. Regarding the high amino acid accumulation line, the relationship between various amino acid contents and the expression of biosynthetic genes was investigated.

    Blossom end rot in tomato fruit is a physiological disorder that causes enormous damage in the world and Japan, and its mechanism of occurrence has not been fully elucidated. Therefore, in this project, a genetic approach using chromosomal fragment substitution lines that stably show resistance to blossom end rot was performed with ionomic analysis of all ions containing calcium and analysis to clarify the dynamics of calcium. Furthermore, transcriptome analysis and the regulation of gene expression were performed to obtain the evidence. Through these approaches, we conducted basic research aimed at elucidating the mechanism of occurrence of blossom end rot and providing breeding materials for overcoming it.

    In Japan, most strawberry cultivars grown for fruit production are June-bearing, single-cropping type strawberries. Unlike June-bearing strawberry cultivars, everbearing strawberry cultivars are suitable for summer cropping. However, fruits production decreases and fruits quality were poor under high temperature in summer. To improve the fruits production and fruits quality, everbearing strawberry plants were grown under high temperature and detailed factors which lower the production and quality were investigated. There is not a correlation between rate of fruit set and fruit quality. And good tasted fruits have high sucrose content even under high temperature.

Therefore, the purpose of this study was to accelerate the identification of the function of useful genes by applying virus-induced gene silencing to horticultural crops. In this study, we analyzed the physiological role and horticultural importance of genes to be controlled in fruit trees, vegetables, and flowers. In addition, we tried to develop viral vectors applicable to various horticultural crops, and to examine the mechanism of infection related to the use of cucumber mosaic virus with potentially general versatility because of wide host range.

    Ionomic analysis, high-throughput elemental analysis, was performed in three diverse fruit crops to represent element concentrations in edible parts and to compare element concentrations between edible parts and vegetative organs and between species. Nineteen elements containing essential elements for human health, toxic elements, and elements with radioisotopes from nuclear reactors were analyzed in apple, Japanese pear, eggplant, and edamame. The distributions of elements were shown as basic information needed for biofortification, and high mobilities depending on species were found in some elements whose mobilities are believed to be low. Classification of element profiles into the leaf and the others containing the edible part except with the calyx in principal component analysis revealed the similarity in element distribution across the species. The present study may contribute to the research progress in the ionomics studies of fruit crops.

    In this study, we examined the use of rapeseed for activating the agriculture in the Tohoku region. We conducted a survey to study at three locations of rapeseed cultivation farmland of Minamisoma and katsurao of Sendai, Miyagi Prefecture, Fukushima Prefecture.We have investigated the trend of radioactive material by the sampling of soil and plants. With respect to radioactive cesium dynamics and rapeseed cultivation, it was found that three of fact.Rapeseed to a certain extent absorb radioactive cesium in the soil.When extracting the oil by oil extraction from rape seeds, cesium hardly shifts in oil. It is concentrated in remaining focused in the remaining. In the case of cultivating rapeseed in were decontaminated area can be said to be safe. As a result, in the radioactive cesium contaminated areas, there is no problem in rapeseed cultivation if decontamination is complete.

    Recently, functional components become important in fruit in addition to size and sweetness. Glucose-realted metabolites that were studied in this project are different from popular functional components such as vitamins and polyphenols. There could be a novel component that is not of antioxidants such as vitamins and polyphenols. In the results obtained in tracer experiments using sea buckthorn, the metabolite was metabolically inactive compared to other sugars. Ripening and environmental stress were investigated because those may affect synthesis and accumulation of the metabolites. The results suggested that the plant was a climacteric type and the accumulation decreased during ripening.

    In plant, it is rare that long-day variety and short-day variety exist in the same species and strawberry (Fragaria ×ananassa Duch.) is one of such a plant. In strawberry cultivars, long-day cultivars can produce fruits through the year so this characteristic is called “everbearing". This characteristic is very important in agricultural production. Because strawberry is an octoploid plant, inheritance in strawberry is complex. Thus, we studied “everbearing" characteristic using F. vesca which is a diploid plant and model plant of Fragaria. Self- and cross-pollinated lines were bred and grown under long-day condition. Some of these lines exhibited everbearing characteristic.

    The effects of light quality on the flowering of Gypsophila paniculata were investigated using light-emitting diodes (LEDs). Flowering and flower budding were observed under long-day conditions with far-red (FR), while no flowering was observed under short-day conditions. Promotion of flowering and flower budding was increased under FR supplemented with red R compared to FR alone. Although generally R inhibits flowering in long-day plants, a certain intensity of R may be necessary for effective promotion of flowering. In contrast, blue (B)supplementation of FR was not effective in inducing flowering and flower budding.

    永年性作物である果樹の生産において、盛果期を安定的に維持する事は重要な課題である。しかし、様々な環境要因、例えば土壌や気象条件、あるいは人為的な影響である栽培管理状況によっては早期から老化が誘導され、盛果期が十分な期間維持できない場合がある。果樹は、発芽から開花、枯死までのライフサイクルを1年で完結する1年生植物と異なり、数十年の歳月をかけて成長し、老化する。今回、希少かつ有用な、メンデリズムに従う表現型として若年老化する(成長の鈍化、新しい組織の形成の抑制、粗皮化などの老化現象が1-2年で生じる)ナシが交雑実生として見出されたので、これを利用し、果樹の老化機構を解明するとともに、盛果期の維持に活用可能な知見を得ることを目的とする。 今年度は引き続き、若年老化樹における形態学的、生理学的特徴を明らかにするための解析を行った。すなわち、チュウゴクナシYaliとセイヨウナシBartlettの交雑実生から分離した若年老化系統と野生型の表現型を示す個体から葉と枝を採取して、形態観察のための固定サンプルを調製するとともに、各種成分分析を行うために凍結試料を用意した。形態観察ではパラフィン切片を作成して顕微鏡観察するとともに、粗皮化に関わるリグニンなどを検出するための染色を行った。また、老化と関連する可能性のある、可溶性および不溶性炭水化物やタンパク質組成をHPLCや電気泳動等を用いて解析した。その結果、リグニン化の過程における変異が老化の進展を調節している可能性が示唆された。

    長日性花卉に白色光あるいは赤色光で長日処理しても開花が促進されない。この反応は、長日性のモデル植物であるシロイヌナズナにおいて遠赤色光受容体遺伝子PHYAのはたらきであることが示されている。そこで本研究では、人工光で栽培可能なモデル植物ではなく、太陽光の下で育てなければならない長日性花卉(シュッコンカスミソウ)を用いて、発光ダイオード(LED)による長日処理が開花を促進するかどうか調べた。その結果、開花は赤色光で促進されず遠赤色光で促進されたので、遠赤色光LEDはシュッコンカスミソウの長日処理光源として実用性が高いとみられた。遠赤色光による開花促進はPHYAの下流ではたらく花成早化遺伝子CONSTANSによって起こることが示されている。 そこで、平成20年度には、昼/夜の温度としてシュッコンカスミソウの適温域(25/20℃)を設けて、花芽形成と開花を実用的なレベルまで促進する遠赤色光の強さと照射時間を調べた。比較対照として、赤色光と青色光のLEDを用いた。また、遠赤色光による開光促進には、CONSTANSのシグナルがSOC1を介してはたらく経路が機能していると示唆された。さらに、シュッコンカスミソウを人工気象器内で栽培し、白色光を8時間照射した後、遠赤色光を8時間照射して、CONSTANSと、その下流で機能して開花を促進するFLOWERING LOCUS Tの相同遺伝子を単離し、塩基配列を確定した。以上の成果をまとめて研究発表し、論文、著書として著した。

    The role of acid invertase for cell enlargement and its regulation of gene expression were investigated using the systems of fruit enlargement of Japanese pear and of petal enlargement of rose. (1) Two full length vacuolar type acid invertase cDNA (PsV-AIV1, PsV-AIV2) were cloned from Japanese pear fruit. PsV-AIV1 related the early growth concerning active cell division and PsV-AIV2 related mainly cell enlargement with sugar accumulation. (2) The enlargement of rose petal was initiated by forming high turgor pressure which was formed by converting sucrose to hexose via the rising of invetase activity with increase of gene expression. The rising of activity was induced by auxin treatment. The regulation of NAD-SDH gene expression and the accumulation of sorbitol were investigated using straw berry fruit. (1) The full length cDNA of NAD-SDH of strawberry fruit (FaSDH) was cloned and the activity also detected. However, relating sorbitol-6-phosphate dehydrogenase the gene existed but the protein and activity were not detected. The kinetics of NAD-SDH activity revealed that sorbitol was synthesized by the reduced reaction of fructose. (2) NAD-SDH activity was stimulated by fructose, sorbitol or auxin treatment. However, there were not clear changes of mRNAs by them. (3) The mRNA of NAD-SDH were composed of the pre-mature mRNA containing intron and the mature mRNA. The stimulation of NAD-SDH activity by fructose, sorbitol or auxin depended on the stimulation of splicing rate after transcription. As described above, it was indicated that acid invertase and sorbitol dehydrogenase play an important role for the cellular enlargement and sugar accumulation and their activities were regluated by the substrates and phytohormone.

    Functional genomics is lately developing because all DNA sequences of the genome have been clarified in Arabidopsis thaliana that is the model plant. On the other hand, functional genomics in horticultural crops is not easy because there are so many species involved in horticultural crops. Therefore, an advanced research by using some model horticultural crops is needed. Arabidopsis thaliana is not useful as the model of the fruit trees and the fruit vegetables. Tomato is not only commercially important but also useful as a model horticultural plant because its transformation is comparatively easy ; its physiological information is abundant ; there is expressed sequence tag (EST) containing more than 100,000 sequences. However, there are some problems, which are the large plant size and the long generation time. Micro-Tom is a miniature tomato that is almost genetically identical to the cultivation tomatoes. Micro-Tom could be a good model plant because of its small size (5-10 cm of plant height), short generation time, and high transformation efficiency. Thus, Micro-Tom was used in this experiment as a model horticultural crop for molecular genetics. A high degree of availability of Micro-Tom as a model experimental plant was confirmed through the analysis of proton pump, pathogen response, etc. in this research. Moreover, the research on various horticultural crops such as the fruit vegetables and fruit trees was also carried out. In the future, the information and the technique of functional genomic developed using Micro-Tom are expected to be applied widely in various horticultural crops.

  24. テロメアの解析に基づく果樹における幼若性の分子機構の吟味

    果樹では幼若相の長さが数年になり、育種上の大きな障害となっている。果樹の実生を最適環境下で連続的に生育させる、あるいは実生の枝を接ぎ木するなどの、成長速度を高める処理によって開花期までの時間を短縮できることから、成長点における積算の細胞分裂回数が幼若相の決定要因ではないかと考えられている。一方、テロメアは染色体の末端部分の塩基配列のことであり、7塩基のくり返し配列からなっており、細胞分裂回数に従って短くなることが知られている。以上のように、果樹の幼若性は細胞分裂回数に関連しており、テロメアはその細胞分裂回数の指標となり得ることから、本研究を行った。 本年度の成果として、まずエージングとテロメア長の関係について精査した点を上げることができる。昨年度の成果に引き続きリンゴおよび同じバラ科樹木であるサクラにおいて、数年から数十年間におよぶ細胞分裂の積算数において差のある組織間でも、テロメア長に差異がなく、したがって幼若性とテロメア長との関連も考えられないことが明らかとなった。その過程で、非常に長期間、テロメア長が維持されており、この点がヒトなどと大きく異なることが示された。次に、このテロメア長の維持機構やテロメアによる染色体末端防御機構を明らかにするために、形質転換が比較的容易でモデル園芸作物として利用可能なトマトから、テロメア結合タンパク質を単離し、その性質を明らかにした。恐らくテロメアは、通常の加齢に加えて環境ストレスによっても影響を受け、植物の正常な発育に関与することが予想される。したがって、並行して行われた切り花の老化や根粒の硝酸態窒素による老化、あるいは環境ストレスによる糖代謝への影響など、園芸学上、重要な現象と、テロメアおよびテロメア結合タンパク質との関連が予想される。

    Many organisms, including certain plants, accumulate water-soluble organic compounds of low molecular weight that have no inhibitory effects on metabolism. These compounds are referred to as compatible solutes and allow them to tolerate certain types of environmental stress not only by adjusting osmotic pressure in cells but also by stabilizing the quaternary structures of complex proteins. Recent studies have shown that enhancement of tolerance to environmental stresses in plants can be achieved by genetic engineering of the biosynthesis of compatible solutes. All these studies, however, deal with herbaceous plant species. We selected Japanese persimmon (Diospyros kaki), which is one of the major fruit crops in Japan and susceptible to damage from salt and drought stresses, as a candidate for the first woody plant species to be genetically engineered for tolerance to environmental stress by transforming the genes involved in compatible solutes. Chimeric genes constructed from the choline oxidase gene of Arhrobacter globiformis and the cDNA for sorbitol-6-phosphate dehydrogenase of apple (Malus x domestica) were integrated into the persimmon genome to confer the ability to produce glycinebetaine and sorbitol, respectively. Tolerance to salt stress of transformed persimmon plants were determined by measuring the ratio of the variable (Fv) to the maximum (Fm) fluorescence of chlorophyll in leaves under NaCl stress. The rate of decline in Fv/Fm under NaCl stress was lower in transgenic persimmon plants producing glycinebetaine or sorbitol than control transformed and non-transformed lines. The possibility of using these transgenic persimmon as a rootstock was tested.

    Tomato floral mutant, Leafy inflorescence (lfi), does not develop a flower but indeterminant inflorescences with vegetative leaves and leaf-like organs. Many vegetative meristem-like primodia were successively differentiated in immature inflorescences of lfi plants. However, the fusion of inflorescence shoots was observed in developed lfi inflorescences. In addition, the leaf-like organs occasionally developed into fleshy fruit-like bodies. These characteristics suggest that the lfi inflorescence have a potential to form carpels. lfi plants also showed a late-flowering phenotype. When floral identity genes, TOFL and TDR4, were cloned by RT-PCR, a 12-bp deletion was observed in TOFL cDNA from lfi plants. The deletion was in a region highly conserved among several LFY/FLO homologues, suggesting the deletion resulted in lfi phenotype. The expression of flower organ identity genes, TDR5, TDR6, and TAG1, was inhibited in lfi inflorescences, suggesting the important role of TOFL in the induction of flower organ identity genes. However, those genes were also expressed during development of fleshy fruit-like bodies. Thus, there is the other pathway to induce the flower organ identity genes.

    The purpose of this research is to make clear the sink-source relation of fruit by investigating the properties and gene expression of enzymes which play an important role for sugar metabolism and the vacuolar function to accumulate sugars. 1. Sorbitol dehydrogenase was purified from Japanese pear fruit, and its cDNA was also cloned from fruits of apple, loquat and peach. The gene expression was investigated throughout the development of their fruits. 2. The gene of sorbitol-6-phosphate dehydrogenase expressed on source leaves unfolded enough, but not on leaves folded yet. 3. Sucrsoe synthase isoform appearing with sugar accumulation was purified and the reaction inclined to sucrose synthesis rather than sucrose cleavage. The direction of reaction changes easily by phosphorylation and non-phosphorylation. 4. Two soluble acid invertases were purified from Japanese pear fruit, and there was no difference between properties of both isoforms. 5. We isolated the cDNA clone of fructokinase which is closely relating to fructose accumulation and examined the relations between the gene expression and fructose metabolism. 6. H^+-ATPase and H^+-pyrophosphatase which supply the energy needed to uptake sugars into vacuole were purified from pear fruit, and the gene expression throughout fruit development were investigated using cDNA clone of their subunits. 7. We could prove the presence of sugar transporters across the tonoplast by sugar uptake experiment, although could not isolate their protein and cDNA. As described above, we could obtain the proteins and cDNA of some enzymes needed to accumulate sugars into fruit, analyzed their gene expressions and clarified partially the melecular mechanism of sugar accumulation.

    The important quality of fruit is flavor and in most fruit, consumer flavor preferences are dominated by sweetness. There is a difference in sweetness of hexose sugars, with fructose being approximately twice as sweet as glucose. Because many fruit accumulate approximately equal levels of the hexose isomers, fructose and glucose, metabolic engineering to enhance the proportion of fructose has the potential to enhance fruit sweetness. In this work, the expression patterns of two tomato fructokinase genes, Frk1 and Frk2, are described as well as a strategy to suppress their expression in transgenic tomato plants. In situ hybridization using Frk1 and Frk2 probes showed differential expression of these two genes in tomato fruit. Frk2 mRNA is localized specifically for starch synthesis and seed development in young tomato fruit whereas Frk1 mRNA is widely distributed. The results from northern blot and in situ hybridization suggest that Frk1 plays a primary role in fructosemetabolism in plant cells while Frk2 is localized specifically in cells that import and utilize moresugar such as starch synthesizing cells and developing seed-related cells. An antisense gene was constructed using Frk1 cDNA and E8 promoter that are cloned to pBl121 binary vector. Approximately 50 transformants were grown and fruit were harvested from them. Although the levels of endogenous Frk1 mRNA were decreased in several t ransformants, any changes in fruct okinase activity and fructose content were not detected in the transformants. Therefore transformation works using Frk1 and Frk2 with 35S promoter were carried out. As a result, decreases in fructokinase activity were observed in some transformants with Frk2 antisense genes. The changes in activity possibly affect sugar composition in fruit.

    The purpose of this project is to make clear the mechanism of sugar accumulation in fruit tissue or cells. 1. Cellular compartmentation of sugars : We investigated the localization of sugars and the change of their contents in fruit. More than 70% of sugars distributed in the vacuole and others in the apoplast in strawberry and melon fruits. With fruit maturation, the palsma membrane became leaky to sugar penetration, resultantly the sugar content in the apoplast increased. 2. The mechanism of sugar transport across the membrane and its stimulation by plant hormones : There present 2 types of carrier-mediated and simple diffusional transports to the uptake of sugars into cells. In strawberry and melon fruits, we detected the carrier-mediated transport of sucrose, glucose and fructose in young fruit, but the carrier-mediated transport of sucrose disappeared during fruit maturation. IAA stimulated the sugar uptake via the carrier-mediated transport. On the contrary, ABA stimulated the sugar uptake via the simple diffusional transport. This may be caused by the stimulation of sugar uptake into the vacuole. 3. The role of vacuolar H^+-ATPase and H^+-PPase : V-H^+-ATPase and V-H^+-PPase were prepared from the tonoplast vesicles of pear fruits, and their roles were compared each other through the developmental stage. H^+-PPase activity was higher in young fruit and H^+-ATPase activity became higher with fruit maturation. 4. Sugar transport mechanism into the tonoplast vesicle : We make clear the participation of carrier protein of facilitated diffusion of glucose and fructose in the tonoplast vesicles.

    オカヒジキ(アカザ科)の花成、花の性分化とアポミクシスの光による制御機構について生理学的、形態学的解析を行い、以下の成果を得た。 1.様々な光条件下での花成反応を調べ、1)16時間暗期の中央に挿入した光中断は花成を完全に阻害し、2)16時間暗期のときの花成は、8時間明期の通常の短日条件下よりも明期を12時間に延長した変則的な日長条件下で促進され、3)12時間明期のときの花成は、12時間暗期の中間日長条件下よりも16時間暗期の条件下で促進されること、4)短日条件下では光強度が高いほど花成は促進され、花成反応の強さと光強度はほぼ比例関係にあることを見いだした。以上の結果から、中間植物であるオカヒジキの花成は、短日植物として誘導された花成反応がトータルの光量の増加によって促進されるものであることを明らかにした。 2.短日条件下で形成された花は見かけ上雌性花であるが、1)組織切片の顕微鏡観察の結果、このような花では、胞子形成細胞が分化し、花粉母細胞が減数分裂して、coenocytic tetradまでは形成されるが、その段階で発生が停止し、液胞化、崩壊して、葯全体が萎縮するものであることを見いだした。このことから、この現象は雄性不稔の誘導と見なすのが適当であることを明らかにした。2)雄性不稔に関しては光周期の影響が大きく、光強度の影響は小さいこと、3)種々の植物ホルモン処理は雄性不稔を回復させることはないことを明らかにした。 3.短日条件下で形成された雄性不稔花の種子形成については再現性のある結果が得られず、種子形成率も著しく低かったことから、アポミクシスは行わないと判断された。

    果実の肉質は食味や貯蔵性と深い関わりがあるため、果実の軟化機構を解明することは、生産および流通において重要である。果実の成熟にともなう軟化には細胞壁の構造変化が生じており、それには細胞壁分解酵素が関与している。本研究ではわが国の代表的果樹であり、細胞壁成分の変化等の報告のあるニホンナシを用い、軟化前後での細胞壁画分におけるグリコシダーゼ活性の変動を調べると共に、数種酵素の可溶化および精製を試みた。 成熟前と成熟時のニホンナシ果実から細胞壁画分を調製し、各種グリコシダーゼ活性を測定したところ、最も顕著に活性が上昇したのはalpha-アラビノフラノシダーゼ(alpha-Ara)であり、成熟期を通して最も活性が高かったのはbeta-ガラクトシダーゼ(beta-Gal)であった。すでにセイヨウナシおよびニホンナシでは果実の軟化にともなって細胞壁からアラビノースやガラクトースが遊離することが報告されている。そこで、これらの遊離に関与していると思われる両酵素に着目して可溶化を試みた。 alpha-Araに関しては通常用いられる塩による可溶化は有効ではなかったが、キレート剤によりかなりの活性を遊離させることに成功したため、今後精製を進めることが可能になった。一方beta-Galについては3Mの塩化リチウムにより高い可溶化率が得られたので、疎水クロマトグラフィー、イオン交換クロマトグラフィー、ゲルクロマトグラフィーを用いて精製を行った。この間beta-Galは5つの活性ピークに分離した。これらの活性ピークの内の1つが成熟時に顕著に上昇し、しかも軟化前のニホンナシより調製したペクチン画分からのガラクトースの遊離に寄与することが確認されたため、このbeta-Galが軟化と関わりがあることが示唆された。今後この知見をもとに、ニホンナシ果実の軟化におけるグリコシダーゼの役割が明らかになることが期待される。

    Both sorbito-6-P dehydrogenase (S6PDH) and sotbitol dehydrogenase (SDH) are important as key enzymes to accumulate sugar into fruit tissue of apple. The purpose of this research is to make clear the role of lthese enzymes by purifying and characterizing them, and to clarify the regulation on gene expression of S6PDH by cloning the cDNA. We cucceeded to isolate both enzymes through some purification steps. Especially, the isolation of S6PDH is very worth because this was firstly isolated from plant tissues. From the properties of purified enzymes, S6PDH was certified to react toward S6P synthesis and SDH toward the convertion of sorbitol into fructose in vivo. Further, we succeeded to isolate the cDNA clone of S6PDH using apple seedling. The cDNA is composed of a open reading fram of 930 bases coding 310 mino acid. The molecular weight of S6PDH was 35,000 and corresponded to the molecular weight estimated by SDS-PAGE.Next, the gene expression of S6PDH with germination of apple seeds was studied using this cDNA probe. It was suggested that the gene expression of S6PDH activity acts under some post-transcriptional or translational regulations since m-RNA of S6PDH appeared 2 days ealier than the protein synthesis of S6PDH.Infuture, it will be expected to make clear the detail mechanism of sugar accumulation into fruit throught the study of gene expression of SDH by cloning the cDNA.
