    Abstract Curcumin, a bioactive phenolic compound derived from turmeric, attenuates heavy metal toxicity, including inorganic arsenite (As(III)), via activation of the Nrf2–Keap1 signaling pathway; however, its low solubility and stability limit its bioactivity. To address these limitations, we synthesized curcumin derivatives and investigated the cytoprotective effects of a monocarbonyl analog, GO-Y015. GO-Y015 exhibited cytotoxicity comparable to curcumin at concentrations below 8 µM but significantly reduced As(III)-induced cytotoxicity. Pretreatment with GO-Y015 markedly improved cell viability under cytotoxic As(III) exposure, whereas curcumin showed minimal protective effects. Mechanistically, GO-Y015 induced robust Nrf2 activation in a concentration- and time-dependent manner, resulting in increased expression of downstream targets, including HO-1, GCLC, and MRP2. Pharmacological inhibition of Nrf2 (ML385), GSH synthesis (BSO), or MRP transporters (MK571) abolished the cytoprotective effects, indicating dependence on the Nrf2–GSH–MRP axis. Furthermore, ICP-MS analysis demonstrated that GO-Y015 significantly reduced intracellular arsenic accumulation after 2 h exposure. These findings indicate that GO-Y015 enhances arsenic detoxification and reduces cytotoxicity by promoting GSH-dependent conjugation and MRP-mediated efflux.

    <p> The natural phytotoxin coronatine, which is composed of two individual parts, coronafacic acid and coronamic acid, exhibits various promising biological activities similar to jasmonoyl L-isoleucine, which is one of the phytohormones, through interacting COI1-JAZ complex receptor. Interestingly, coronatine induces stomatal opening in which jasmonoyl L-isoleucine is not involved as an endogenous regulator. Moreover, a diastereomer of coronatine had stomatal opening activity but did not interact with COI1-JAZ complex protein. In addition, the mutated plant lacking the function of COI1 protein, retained stomatal opening activity by coronatine. From these data, we hypothesized coronatine would have a target protein other than jasmonate receptor in the guard cells. To identify the target protein, we synthesized alkyne-tagged coronatine for Raman imaging in living guard cells. </p>

    Fluorinated compounds have drawn the attention of medicinal chemists because of their potential metabolic tolerance and modulation of drug efficacy. Thus stereo- and enantioselective construction of fluorinated carbon center is an important issue. In spite of remarkable progress in the last decade, the first effective method of catalytic enantioselective fluorination by nucleophilic fluorine reagent has been reported in this year. In this article, construction of fluorinated chiral center is reviewed being categolized into three classes.

    本研究は、「有機ニトロキシルラジカル-遷移金属協奏触媒システム」に潜在する、未踏の合成化学的機能性の開発を機軸として、常温・常圧で空気中の分子状酸素を触媒的に活性化して、①アルコールおよび②フェノールの高化学選択的酸化を基点とする高次分子変換法の創出を企図して3年間の計画研究として実施するものである。以下、項目毎に2年目までの研究実績の概要を記す。 ①多座配位型AZADOを網羅的に合成し、N-保護 vic-アミノアルコールと第一級アミンとの空気酸化的縮合によるペプチド合成反応を検討した。その結果、Stahlらによって報告されていたABNO-Cu触媒を凌駕する合成効率を発揮する多座配位型AZADO-Cuハイブリッド触媒システムを見出すことに成功した。種々のアミノ酸由来のN-保護 vic-アミノアルコールを用いて基質適用性を検証した結果、本触媒システムは幅広い官能基許容性を示し、多様なジペプチド誘導体の合成に適用可能であることを確認した。また、上述した酸化的ペプチド縮合反応の検討の途上において、予期せず見出した酸化的ペプチド切断反応について検討を進め、その切断部位を特定することが出来た。 ②クロムサレン錯体と有機ニトロキシルラジカルの協奏触媒系による分子内脱芳香族的フェノールカップリング反応の不斉反応への展開と触媒機構の解明を目指した検討を行った。その結果、不斉フェノールカップリング反応の生成物の絶対立体化学を決定することに成功し、これより基質特異性とリガンドー不斉収率の相関に関する知見を統合して、本分子内脱芳香族的不斉フェノールカップリング反応の触媒機構モデルを構築した。また、本触媒システムの合成化学的有用性の拡張を企図して分子間フェノールカップリングへの適用を行い、効率的なレスベラトロール型スチルベノイドの空気酸化的二量化反応を開発することに成功した。

    Toward the total synthesis of kadcoccilactone A, synthesis of two fragments (B and DE) was achieved. 3-Butyn-1-ol was successfully converted to B ring fragment via a 12-step sequence involving the Sharpless epoxidation and a catalytic reductive cyclization of 1,6-diyne. Synthesis of the DE ring fragment was accomplished starting from a chiral building block via a key radical cascade reaction (decarboxylation/ 1,4-addition/ cyclization/ fragmentation) of arylsulfone.

    Toward total synthesis of antitumor saponin silaciloside E-1, synthesis of the A ring unit was accomplished. By incorporating the cyclic carbonate structure into the substrate, the key gold-catalyzed 6-endo cyclization of 1,6-enyne proceeded with perfect regioselectivity. An improved synthesis of CDE ring unit was also developed by exploiting the ring-contractive coupling reaction and the elimination reaction of the quaternary ammonium salt. As a result, the synthetic route was shorted by six steps.

    Toward the total synthesis of marine toxin spirolide D, the stereoselective construction of BCD fragment has been explored. Transannular spiroketalization of the triketone with a tether between the carboxyl group at C10 and hydroxyl group at C24 proceeded stereoselectively to give the desired stereoisomer in good yield. Since the reaction using acyclic triketone gave desired stereoisomer as the major product, the stereoselectivity of this process is rationalized by a preferred dipole orientation. The product was successfully converted to the BCD ring fragment through a stereoselective hydrogenation of C11-C12 double bond.

    Alkyne-tag Raman imaging was proposed as an approach for live cell imaging of small molecules by using an alkyne-tag. Structure-Raman shift/intensity relationships of various alkynes was examined by use of EdU as standard. On the basis of obtained results, simultaneous imaging of two alkyne-tagged molecules, EdU and alkyne-tagged CoQ analogue (AltQ), and semiquantitative estimation of cellular concentration of CoQ analogues have been achieved. Finally, simultaneous imaging of protonated and deprotonated carbonylcyanide p-trifluoromethoxyphenylhydrazone (FCCP) was successfully demonstrated.

    2年計画の後半となる本年度は、研究計画に基づき、クルクミンアナログGO-Y086とその結合タンパク質KSRP/FUBP2との相互作用を詳細に解析した。また、GO-Y086の抗腫瘍細胞活性発現機構の解明を目指した。全9種類のGO-Y086誘導体を合成し、KSRP/FUBP2との結合能を評価した。その結果、GO-Y086とKSRP/FUBP2が結合するためにはGO-Y086の芳香環の置換様式とリンカー部の両方が重要であることが明らかとなった。また、誘導体のがん細胞成長阻害活性とKSRP/FUBP2との結合活性は良い相関を示した。FUBPsはがん原遺伝子c-Mycの転写を活性化することが報告されている。c-Mycの発現抑制は細胞死を引き起こすことが知られているため、GO-Y086がKSRP/FUBP2の阻害を介してc-Mycの発現を抑制し、細胞毒性を示すことが考えられた。実際に、GO-Y086処理時のc-Myc発現量を、タンパク質レベルおよびmRNAレベルで調べたところ、いづれにおいても発現を顕著に抑制した。また、GO-Y086の非ラベル化体であるGO-Y030やクルクミンもc-Mycの発現を抑制した。すなわち、GO-Y086、GO-Y030およびクルクミンは、KSRP/FUBP2を介して細胞の生存に必須なc-Mycの発現を抑制し、細胞毒性を示すことが示唆された。以上より、クルクミンおよび誘導体の抗腫瘍細胞活性発現における重要な作用標的の一つがKSRP/FUBP2であることが考えられる。したがって、本結果はKSRP/FUBP2が新たな抗腫瘍剤開発の標的であることを示唆するものであり、創薬化学的に重要な知見である。
