    Preventing feather pecking (FP) in adult laying hens is important for the welfare of intensively poultry farming. Fear-related behavior in growing female layer chicks may predict FP in adult hens. In this study, in two representative laying breeds (White Leghorn [WL] and Rhode Island Red [RIR]) that have different FP frequencies, we identified a candidate gene associated with fear-related behavior in chicks and FP in adult hens. In the tonic immobility test and open-field test, the behavioral activity was lower in WL chicks than in RIR chicks (P < 0.01), suggesting that WL chicks were more fearful than RIR chicks. Based on previous studies, 51 genes that have been found to be differentially expressed in the brain between high- and low-FP populations were chosen, and their expression levels were screened in the chick diencephalon. This analysis revealed that myeloperoxidase (MPO) gene expression level was higher in WL chicks than that in RIR chicks (P < 0.05). Furthermore, STRING analysis predicted the gene network including MPO and MPO-related genes and revealed the association of these genes with fear-related behavior. These results suggest that MPO is potentially associated with fear-related behavior in growing female layer chicks and FP in adult hens.

    Hot, humid weather causes heat stress (HS) in broiler chickens, which can lead to high mortality. A recent study found that HS causes endoplasmic reticulum (ER) stress. However, the possible involvement of ER stress in HS-induced physiological alterations in broiler chickens is unclear. This study aimed to evaluate the effect of the dietary supplementation of 4-phenylbutyric acid (4-PBA), an alleviator of ER stress, in acute HS-exposed young broiler chickens. Twenty-eight 14-day-old male broiler chickens (ROSS 308) were divided into two groups and fed either a control diet or a diet containing 4-PBA (5.25 g per kg of diet feed) for 10 days. At 24 days old, each group of chickens was kept in thermoneutral (24 ± 0.5 °C) or acute HS (36 ± 0.5 °C) conditions for 2 h. The results showed that thermoneutral birds supplemented with 4-PBA exhibited no negative effects in terms of broiler body weight gain and tissue weight compared to non-supplemental birds. HS increased body temperature in both the control and 4-PBA groups, but the elevation was significantly lower in the 4-PBA group than in the control group. The plasma non-esterified fatty acid concentration was significantly increased by HS treatment in non-supplemental groups, while the increase was partially attenuated in the 4-PBA group. Moreover, 4-PBA prevented HS-induced gene elevation of the ER stress markers GRP78 and GRP94 in the skeletal muscle. These findings suggest that the 4-PBA effect may be specific to the skeletal muscle in HS-exposed birds and that 4-PBA supplementation attenuated HS-induced muscle ER stress, which could be associated with a supplementation of the body temperature elevation and lipolysis.

    Enzymes excreted by rumen microbiome facilitate the conversion of ingested plant materials into major nutrients (e.g., volatile fatty acids (VFA) and microbial proteins) required for animal growth. Diet, animal age, and health affect the structure of the rumen microbial community. Pathogenic organisms in the rumen negatively affect fermentation processes in favor of energy loss and animal deprivation of nutrients in ingested feed. Drawing from the ban on antibiotic use during the last decade, the livestock industry has been focused on increasing rumen microbial nutrient supply to ruminants through the use of natural supplements that are capable of promoting the activity of beneficial rumen microflora. Selenium (Se) is a trace mineral commonly used as a supplement to regulate animal metabolism. However, a clear understanding of its effects on rumen microbial composition and rumen fermentation is not available. This review summarized the available literature for the effects of Se on specific rumen microorganisms along with consequences for rumen fermentation and digestibility. Some positive effects on total VFA, the molar proportion of propionate, acetate to propionate ratio, ruminal NH3-N, pH, enzymatic activity, ruminal microbiome composition, and digestibility were recorded. Because Se nanoparticles (SeNPs) were more effective than other forms of Se, more studies are needed to compare the effectiveness of synthetic SeNPs and lactic acid bacteria enriched with sodium selenite as a biological source of SeNPs and probiotics. Future studies also need to evaluate the effect of dietary Se on methane emissions.

    Peptide transporter 1 (PepT1) is a transporter responsible for absorbing dipeptide and tripeptide in enterocytes and is upregulated by dipeptide in mammals. It has not been certain whether intestinal PepT1 expression is responsive to dipeptides in chickens because of the lack of in vitro study using the cultured enterocytes. This study established a primary culture model of chicken intestinal epithelial cells (IECs) in two-dimensional monolayer culture using collagen gel by which the response of chicken PepT1 gene expression to dipeptide stimuli was evaluated. The cultured chicken IECs showed the epithelial-like morphology attached in a patch-manner and exhibited positive expression of cytokeratin and epithelial cadherin, specific marker proteins of epithelial cells. Moreover, the chicken IECs exhibited the gene expression of intestinal cell type-specific marker, villin1, mucin 2, and chromogranin A, suggesting that the cultured IECs were composed of enterocytes as well as goblet and enteroendocrine cells. PepT1 gene expression was significantly upregulated by synthetic dipeptide, glycyl-l-glutamine, in the cultured IECs. From the results, we herein suggested that dipeptide is a factor upregulating PepT1 gene expression in chicken IECs.

    Efforts directed toward enhancing animals' productivity are focused on evaluating the effects of non-traditional feed additives that are safer than antibiotics, which have been banned because of their health hazards. Many studies used an amino acid that contributes to heme biosynthesis, known as 5-aminolevulinic acid (5-ALA), to promote the productivity of farm animals. However, these studies demonstrate inconsistent results. In order to develop a clear understanding of the effects of 5-ALA in farm animals, we comprehensively searched PubMed and Web of Science for studies evaluating 5-ALA effects on the performance, iron status, and immune response of different farm animals. The search retrieved 1369 publications, out of which 16 trials were relevant. The 5-ALA-relevant data and methodological attributes of these trials were extracted/evaluated by two independent researchers, based on a set of defined criteria. Samples were comprised of pigs, chickens, and dairy cows. The 5-ALA doses ranged from 2 mg to 1 g/kg of feed, and treatment duration ranged from 10 to 142 days. Overall, 5-ALA improved iron status in most studies and increased white blood cells count in 3 out of 10 studies, in addition to improving animals' cell-mediated immune response following immune stimulation with lipopolysaccharide. Inconsistent findings were reported for growth performance and egg production; however, a combination of 10 mg/kg of 5-ALA with 500 mg/kg of vitamin C promoted the highest egg production. In addition, 5-ALA improved milk protein concentration. In conclusion, 5-ALA can enhance farm animals' iron status and immune response; however, the heterogeneity of the reviewed studies limits the generalizability of the findings. Standard procedures and outcome measures are needed to confirm the benefits of 5-ALA. Attention should also be paid to any adverse effects.

    The objective of this study was to investigate the effect of 5-aminolevulinic acid (5-ALA) as a dietary supplement on milk yield and composition as well as iron status and immune response in lactating dairy cows. In this study 13 lactating Holstein cows were randomly assigned to either a control group or a treatment group supplemented with 10 mg of 5-ALA per kilogram of dry matter. During feeding, 5-ALA was mixed with a small amount of the total mixed ration and top-dressed. The experiments followed a crossover design with 2 periods. Each period consisted of an adaptation period of 12 d and a test period of 2 d. Dairy cows fed the diet supplemented with 5-ALA exhibited increased counts of white blood cells and granulocytes compared with the control group. The rate of phagocytosis and mitogen-induced proliferation of peripheral blood mononuclear cells in cows fed 5-ALA were higher than in cows fed a basal diet. However, 5-ALA did not affect iron status or plasma biochemical composition. Supplementation with 5-ALA improved milk protein and milk casein contents; however, it had no effect on milk production, milk fat, lactose, total solids, or solids-not-fat, compared with the control. We conclude that dietary supplementation of 5-ALA to lactating dairy cows may have a positive effect on milk protein synthesis and the immune response.

  11. Dietary Supplementation with Lysine and Threonine Modulates the Performance and Plasma Metabolites of Broiler Chicken.

    Here, we investigated whether the optimal threonine (Thr) to lysine (Lys) ratio in high Lys diet improves the growth performance of modern broiler chickens at finisher period and determined the possible mechanism underlying improvement in the growth performance of chickens fed with high Lys or Lys + Thr diet using metabolome analyses. Eighteen 21-day-old chickens housed in individual cages were randomly divided into three groups of six chickens fed with different diets as follows: control diet, high Lys diet (150% Lys content of National Research Council requirement), and high Lys + Thr diet (0.68 of Thr/Lys in high Lys diet). Body weight gain (BWG) increased in chickens receiving high Lys diet as compared with those fed with the control diet (P<0.05); no significant difference was observed in BWG of chickens from high Lys + Thr and high Lys groups. Feed conversion ratio (FCR) was lower in chickens fed with high Lys or high Lys + Thr diet than in those on the control diet. Serotonin concentration increased in the plasma of chickens fed with high Lys diet as compared to those fed with other diets. A negative correlation was observed between plasma serotonin concentration and FCR. These results provide the first evidence on the use of high Lys in broiler diets to reduce FCR during finisher period, which may be associated with change in plasma serotonin concentration. These findings suggest that high Lys content in finisher diet, but not high Thr + Lys diet, may affect the peripheral serotonergic metabolism and improve FCR. Thus, plasma serotonin may serve as a biomarker of FCR in broilers.

    During antral folliculogenesis, developmental competence of prospective oocytes is regulated in large part by the follicular somatic component to prepare the oocyte for the final stage of maturation and subsequent embryo development. The underlying molecular mechanisms are poorly understood. Oocytes reaching the advanced stage of follicular growth by administration of exogenous follicle-stimulating hormone (FSH) possess higher developmental competence than oocytes in FSH-untreated smaller follicles. In this study, the transcriptomic profile of the cumulus cells from cows receiving FSH administration (FSH-priming) was compared, as a model of high oocyte competence, with that from untreated donor cows (control). Ingenuity Pathway Analysis showed that cumulus cells receiving FSH-priming were rich in down-regulated transcripts associated with cell movement and migration, including the extracellular matrix-related transcripts, probably preventing the disruption of cell-to-cell contacts. Interestingly, the transcriptomic profile of up-regulated genes in the control group was similar to that of granulosa cells from atretic follicles. Interferon regulatory factor 7 was activated as the key upstream regulator of FSH-priming. Thus, acquisition of developmental competence by oocytes can be ensured by the integrity of cumulus cells involved in cell-to-cell communication and cell survival, which may help achieve enhanced oocyte-somatic cell coupling.

    There are two independent serotonin (5-HT) systems of organization: one in the central nervous system and the other in the periphery. 5-HT affects feeding behavior and obesity in the central nervous system. On the other hand, peripheral 5-HT also may play an important role in obesity, as it has been reported that 5-HT regulates glucose and lipid metabolism. Here we show that the intraperitoneal injection of 5-HT to mice inhibits weight gain, hyperglycemia and insulin resistance and completely prevented the enlargement of intra-abdominal adipocytes without having any effect on food intake when on a high fat diet, but not on a chow diet. 5-HT increased energy expenditure, O2 consumption and CO2 production. This novel metabolic effect of peripheral 5-HT is critically related to a shift in the profile of muscle fiber type from fast/glycolytic to slow/oxidative in soleus muscle. Additionally, 5-HT dramatically induced an increase in the mRNA expression of peroxisome proliferator-activated receptor coactivator 1α (PGC-1α)-b and PGC-1α-c in soleus muscle. The elevation of these gene mRNA expressions by 5-HT injection was inhibited by treatment with 5-HT receptor (5HTR) 2A or 7 antagonists. Our results demonstrate that peripheral 5-HT may play an important role in the relief of obesity and other metabolic disorders by accelerating energy consumption in skeletal muscle.

    Serotonin is synthesized by two distinct tryptophan hydroxylases, one in the brain and one in the periphery. The latter is known to be unable to cross the blood-brain barrier. These two serotonin systems have apparently independent functions, although the functions of peripheral serotonin have yet to be fully elucidated. In this study, we have investigated the physiological effect of peripheral serotonin on the concentrations of metabolites in the circulation and in the liver. After fasting, mice were ip injected with 1 mg serotonin. The plasma glucose concentration was significantly elevated between 60 and 270 min after the injection. In contrast, plasma triglyceride, cholesterol, and nonesterified fatty acid concentrations were decreased. The hepatic glycogen synthesis and concentrations were significantly higher at 240 min. At the same time, the hepatic triglyceride content was significantly lower than the basal levels noted before the serotonin injection, whereas the hepatic cholesterol content was significantly higher by 60 min after the injection. Furthermore, serotonin stimulated the contraction of the gallbladder and the excretion of bile. After the serotonin injection, there was a significant induction of apical sodium-dependent bile acid transporter expression, resulting in a decrease in the concentration of bile acids in the feces. Additionally, data are presented to show that the functions of serotonin are mediated through diverse serotonin receptor subtypes. These data indicate that peripheral serotonin accelerates the metabolism of lipid by increasing the concentration of bile acids in circulation.

    Identification of insulin-responsive glucose transporter proteins, GLUT4 and GLUT8, was attempted in chickens that characteristically are hyperglycemic and insulin resistant. Northern blot analysis using rat GLUT4 cDNA probe and RT-PCR using primers designed against the conserved regions in mammalian GLUT4 cDNA were not successful in identifying GLUT4 homologue(s) in various chicken tissues. Furthermore, GLUT4 homologues could not be detected in chicken tissues by genomic Southern blot analyses using a rat GLUT4 cDNA probe. These data, therefore, suggest that the GLUT4 homologous gene is deficient in chicken tissues. However, GLUT8, another insulin-responsive glucose transporter in the blastocyst, was identified with the aid of RACE (rapid amplification of cDNA ends) reactions in the chicken testis. Chicken GLUT8 was composed of 1449 bp with a coding region for a 482 amino acid protein. The deduced amino acid sequence was 58.8, 56.3, and 56.8% identical with human, rat, and mouse GLUT8, respectively. By RT-PCR, GLUT8 mRNA expressions were detected in chicken brain, kidney, adrenal, spleen, lung, testis, and pancreas; and barely detectable in skeletal muscle, liver, adipose tissue, and heart. Here we firstly report that GLUT8 was identified in chickens, while GLUT4, a major insulin-responsive transporter in mammals, is deficient in these animals. We propose the hypothesis that the hyperglycemia and insulin resistance observable in chickens is associated with their possible deficiency of GLUT4.

    The red-pigmented yeast, Phaffia rhodozyma, containing high concentration of astaxanthin (Ax) was evaluated as a dietary pigment source for egg yolk of laying hens fed on a practical diet mainly consisted of corn and soybean meal. In experiment 1, diets supplemented with the yeast to provide 0 to 6ppm Ax and commercially available canthaxanthin (Cx, Carophyll Red(R)) to provide 1ppm Cx, respectively, were fed for 2 or 4 weeks. In experiment 2, Ax extracted by acetone from the yeast (3ppm), commercially available Ax (Carophyll Pink(R)) (3ppm), or reagent-grade Cx (1ppm) were fed for 2 weeks.<br>No differences in laying performance, egg weight and feed intake were observed among all treatments. Roche color fan score and redness (a* value) in Chroma Meter analysis increased by addition of every Ax and Cx supplementation. The color fan score, a* value and yolk Ax concentration were increased linearly with the increase of logarithmic concentration of dietary Ax. Extracted Ax was estimated as more than 8 fold as potent as yeast-Ax in the pigmentation of yolk and as 3 fold as potent in the accumulation of Ax in yolk. It is presumed that the low potency of yeast-Ax in the pigmentation is due to low digestibility (degradability) of the yeast. Reagent-grade Cx was less potent than commercial Cx in the pigmentation of yolk.<br>These results suggest that Phaffia yeast containing high concentration of Ax has the potential as a natural source for egg yolk pigmentation in the poultry industry and the availability of Phaffia yeast can be improved by destruction of the cell wall.

    Two experiments were conducted if the meat color-modifying potential of Phaffia rhodozyma, a yeast containing high levels of astaxanthin (Ax), is influenced by the feeding duration and the dietary concentration and if the potential is enhanced by an addition of lecithin+α-tocopherol in broiler chickens. Experimental diets supplemented with cell wall-fractured yeast to provide 10, 20 and 30ppm Ax were fed to 3, 5 and 7 week-old chickens for 5, 3 and 1 weeks, respectively, prior to market in Experiment 1. In Experiment 2, Phaffia yeast-supplemented diets (20ppm Ax), which were supplemented with or without lecithin (5g/kg diet)+α-tocopherol (0.1g/kg diet), were fed to 5 week-old chickens for 3 weeks prior to market. Feeding of Phaffia yeast influenced neither performance of broilers nor edible meat yields, irrespective of either the feeding duration, the dietary concentration or the addition of lecithin+α-tocopherol. Concern-ing the color analysis of edible meats using with Minolta reflectance colorimeter, light-ness (L* value) was not essentially influenced by feeding of the yeast-supplemented diets, while yellowness (b* value) was slightly increased by the yeast feeding. On the other hand, the redness (a* value) of edible meats and abdominal adipose tissues was significantly or numerically increased by feeding of the yeast-supplemented diets, in which the increase was less extent in chickens received the yeast-supplemented diet (30ppm Ax) for 1 week prior to market than chickens received the yeast-supplemented diets (20ppm Ax) for 3 weeks. The extent of increase in a* values of edible meats was relatively dependent on the total Ax intake. Addition of lecithin+α-tocopherol did not intensify the coloration (redness) of edible meats by the feeding of yeast-supplemented diets. These results show that cell wall-fractured Phaffia yeast containing high concen-trations of Ax can be a useful source for modification of meat color and the effect is briefly due to the total Ax intake in broiler chickens.

    The present study was conducted to investigate the effect of acute heat stress (HS) of differing temperatures (36, 38 and 40°C) on plasma amino acids concentration of 3-weeks-old broiler birds. Birds were exposed to each temperature and subjected to blood sampling at 0, 30, 60 and 180min after the exposure. Exposure of chickens to 38 and 40°C caused elevation of core temperature compared with their 0min values until those maintained in the 40°C died shortly after 60min post-HS. At all temperatures, concen-trations of plasma Asp, Ser, Tyr and Cys were significantly decreased by the HS compared to their 0min level. On the other hand, plasma Trp and Trp/LNAA were increased in birds kept at 36 and 38°C at 30min post-HS before returning to their 0min concentrations. Changes in some amino acids were clearly temperature-specific. We speculate that changes in plasma Tyr concentration and Trp/LNAA ratio may be related to some of the metabolic responses together with reduced performance observed in HS birds.

    The inhibition of lipoprotein lipase (LPL)-mediated very low density lipoprotein (VLDL) hydrolysis by several kinds of detergents was investigated to determine the kinds of detergents for the estimation of triacylglycerol (TG) entry into the plasma in chickens. The rate of lipoprotein hydrolysis in vitro declined gradually with an increase of each detergent concentration. Addition of polyoxyethylene (23) lauryl ether, polyoxyethylene (20) cetyl ether, polyoxyethylene (20) oleyl ether, polyoxyethylene (20) sorbitan monopalmitate and polyoxyethylene (20) sorbitan monostealate to give the concentrations at 250ηg/ml caused extensive inhibition on LPL catalyzed hydrolysis (more than 70%) in vitro. Detergents with the competitive inhibition of chicken-LPL and Hydrophile-Lipophile Balance (HLB) at around 14 were characterized with lower Ki and marked inhibition as compared to detergents with non-competitive type inhibition. The injection of polyoxyethylene (20) sorbitan treated-detergents or Triton WR-1339 at 250mg/kg body weight augmented, to a small extent, the plasma TG concentration, while injection of other detergents induced death probably because of an acute toxicity. These findings suggest that the polyoxyethylene-treated detergents with HLB at around 14 inhibits chicken lipoprotein hydrolysis by LPL in vitro and the exploration of possible detergents without an acute toxicity potentiates the reliable estimation of TG entry into blood in chickens.

    The influence of prostaglandin E2 (PGE2) on the differentiation and intracellular protein accumulation of chick myoblasts was investigated in the cultures added with aspirin, an inhibitor of endogenous PG synthesis. With an addition of PGE2 (10-1-102μM), intracellular protein accumulation of chick myoblasts per dish was accelerated but that per DNA was not changed. Creatin kinase (CK) activity in chick myoblasts expressed as both per dish and per DNA was augmented with the increase of PGE2. These results suggest that PGE2 stimulates the differentiation of chick myoblasts and the protein accumulation in cultures during the myogenesis.

    In order to identify species difference of lipoprotein lipases (LPL) between chickens and rats, possible modification of purification of the LPLs was introduced. Crude enzyme prepared from acetone powders of adipose tissue in both the species was added with glycerol to give 30% in final concentration, and chromatographed on Heparin-Sepharose 4 B, Hydroxyapatite and Con A-Sepharose 4 B column. The fraction obtained by elution through the Con A-Sepharose 4 B was shown to yield single band on electrophoresis in sodium dodecyl sulfate (SDS-PAGE). Thus LPLs were purified by 2, 500 and 1, 400 folds compared to the original activity in chickens and rats, respectively, with no contamination of antithrombin III. These results showed that the modified purification procedure is effective in identification of species difference in LP

    Two experiments were conducted if the meat color-modifying potential of Phaffia rhodozyma, a yeast containing high levels of astaxanthin (Ax), is influenced by the feeding duration and the dietary concentration and if the potential is enhanced by an addition of lecithin+α-tocopherol in broiler chickens. Experimental diets supplemented with cell wall-fractured yeast to provide 10, 20 and 30ppm Ax were fed to 3, 5 and 7 week-old chickens for 5, 3 and 1 weeks, respectively, prior to market in Experiment 1. In Experiment 2, Phaffia yeast-supplemented diets (20ppm Ax), which were supplemented with or without lecithin (5g/kg diet)+α-tocopherol (0.1g/kg diet), were fed to 5 week-old chickens for 3 weeks prior to market. Feeding of Phaffia yeast influenced neither performance of broilers nor edible meat yields, irrespective of either the feeding duration, the dietary concentration or the addition of lecithin+α-tocopherol. Concern-ing the color analysis of edible meats using with Minolta reflectance colorimeter, light-ness (L* value) was not essentially influenced by feeding of the yeast-supplemented diets, while yellowness (b* value) was slightly increased by the yeast feeding. On the other hand, the redness (a* value) of edible meats and abdominal adipose tissues was significantly or numerically increased by feeding of the yeast-supplemented diets, in which the increase was less extent in chickens received the yeast-supplemented diet (30ppm Ax) for 1 week prior to market than chickens received the yeast-supplemented diets (20ppm Ax) for 3 weeks. The extent of increase in a* values of edible meats was relatively dependent on the total Ax intake. Addition of lecithin+α-tocopherol did not intensify the coloration (redness) of edible meats by the feeding of yeast-supplemented diets. These results show that cell wall-fractured Phaffia yeast containing high concen-trations of Ax can be a useful source for modification of meat color and the effect is briefly due to the total Ax intake in broiler chickens.

    The red-pigmented yeast, Phaffia rhodozyma, containing high concentration of astaxanthin (Ax) was evaluated as a dietary pigment source for egg yolk of laying hens fed on a practical diet mainly consisted of corn and soybean meal. In experiment 1, diets supplemented with the yeast to provide 0 to 6ppm Ax and commercially available canthaxanthin (Cx, Carophyll Red(R)) to provide 1ppm Cx, respectively, were fed for 2 or 4 weeks. In experiment 2, Ax extracted by acetone from the yeast (3ppm), commercially available Ax (Carophyll Pink(R)) (3ppm), or reagent-grade Cx (1ppm) were fed for 2 weeks.<br>No differences in laying performance, egg weight and feed intake were observed among all treatments. Roche color fan score and redness (a* value) in Chroma Meter analysis increased by addition of every Ax and Cx supplementation. The color fan score, a* value and yolk Ax concentration were increased linearly with the increase of logarithmic concentration of dietary Ax. Extracted Ax was estimated as more than 8 fold as potent as yeast-Ax in the pigmentation of yolk and as 3 fold as potent in the accumulation of Ax in yolk. It is presumed that the low potency of yeast-Ax in the pigmentation is due to low digestibility (degradability) of the yeast. Reagent-grade Cx was less potent than commercial Cx in the pigmentation of yolk.<br>These results suggest that Phaffia yeast containing high concentration of Ax has the potential as a natural source for egg yolk pigmentation in the poultry industry and the availability of Phaffia yeast can be improved by destruction of the cell wall.

    The present study was conducted to investigate the effect of acute heat stress (HS) of differing temperatures (36, 38 and 40°C) on plasma amino acids concentration of 3-weeks-old broiler birds. Birds were exposed to each temperature and subjected to blood sampling at 0, 30, 60 and 180min after the exposure. Exposure of chickens to 38 and 40°C caused elevation of core temperature compared with their 0min values until those maintained in the 40°C died shortly after 60min post-HS. At all temperatures, concen-trations of plasma Asp, Ser, Tyr and Cys were significantly decreased by the HS compared to their 0min level. On the other hand, plasma Trp and Trp/LNAA were increased in birds kept at 36 and 38°C at 30min post-HS before returning to their 0min concentrations. Changes in some amino acids were clearly temperature-specific. We speculate that changes in plasma Tyr concentration and Trp/LNAA ratio may be related to some of the metabolic responses together with reduced performance observed in HS birds.

    The inhibition of lipoprotein lipase (LPL)-mediated very low density lipoprotein (VLDL) hydrolysis by several kinds of detergents was investigated to determine the kinds of detergents for the estimation of triacylglycerol (TG) entry into the plasma in chickens. The rate of lipoprotein hydrolysis in vitro declined gradually with an increase of each detergent concentration. Addition of polyoxyethylene (23) lauryl ether, polyoxyethylene (20) cetyl ether, polyoxyethylene (20) oleyl ether, polyoxyethylene (20) sorbitan monopalmitate and polyoxyethylene (20) sorbitan monostealate to give the concentrations at 250ηg/ml caused extensive inhibition on LPL catalyzed hydrolysis (more than 70%) in vitro. Detergents with the competitive inhibition of chicken-LPL and Hydrophile-Lipophile Balance (HLB) at around 14 were characterized with lower Ki and marked inhibition as compared to detergents with non-competitive type inhibition. The injection of polyoxyethylene (20) sorbitan treated-detergents or Triton WR-1339 at 250mg/kg body weight augmented, to a small extent, the plasma TG concentration, while injection of other detergents induced death probably because of an acute toxicity. These findings suggest that the polyoxyethylene-treated detergents with HLB at around 14 inhibits chicken lipoprotein hydrolysis by LPL in vitro and the exploration of possible detergents without an acute toxicity potentiates the reliable estimation of TG entry into blood in chickens.

    In order to identify species difference of lipoprotein lipases (LPL) between chickens and rats, possible modification of purification of the LPLs was introduced. Crude enzyme prepared from acetone powders of adipose tissue in both the species was added with glycerol to give 30% in final concentration, and chromatographed on Heparin-Sepharose 4 B, Hydroxyapatite and Con A-Sepharose 4 B column. The fraction obtained by elution through the Con A-Sepharose 4 B was shown to yield single band on electrophoresis in sodium dodecyl sulfate (SDS-PAGE). Thus LPLs were purified by 2, 500 and 1, 400 folds compared to the original activity in chickens and rats, respectively, with no contamination of antithrombin III. These results showed that the modified purification procedure is effective in identification of species difference in LP

    The influence of prostaglandin E2 (PGE2) on the differentiation and intracellular protein accumulation of chick myoblasts was investigated in the cultures added with aspirin, an inhibitor of endogenous PG synthesis. With an addition of PGE2 (10-1-102μM), intracellular protein accumulation of chick myoblasts per dish was accelerated but that per DNA was not changed. Creatin kinase (CK) activity in chick myoblasts expressed as both per dish and per DNA was augmented with the increase of PGE2. These results suggest that PGE2 stimulates the differentiation of chick myoblasts and the protein accumulation in cultures during the myogenesis.

    2021年度は以下の3項目を実施し、精子幹細胞の遊走および細胞外基質分泌の制御機およびその種間共通性・相違性の検討を進めた。1)マウス未分化型精原細胞を対象として、既報のマイクロアレイもしくはRNAシークエンシングのデータセットの解析、細胞回収後のRT-PCRによる遺伝子発現解析、in situハイブリダイゼーションによるmRNA局在解析、免疫組織化学によるタンパク質局在解析をそれぞれ実施し、未分化型精原細胞で発現する細胞外基質関連遺伝子・タンパク質の一部を明らかにした。本項目の一部は研究分担者とともに実施した。2)前項目で特定した遺伝子群のうち、その分子機能が精子幹細胞の移動において特に重要と予想された遺伝子1つについて細胞種特異的な遺伝子欠損マウスを作製し、2022年以降に同遺伝子の精子幹細胞における機能を検討するための技術基盤を確立した。なお、同遺伝子は全身性遺伝子欠損の場合に胎性致死となるため、Cre/LoxPシステムを応用した未分化型精原細胞特異的な遺伝子機能欠損マウスを作製した。3)前項目で着目した遺伝子・タンパク質について、精子幹細胞におけるその発現の種間差比較を行うため、成ウシ・ニワトリ精巣の採材・固定・包埋を行った。本項目の一部は研究分担者とともに実施した。 以上、2021年の計画を予定通りに実施し、2022年以降の解析に必要な知見・技術が整った。本研究の推進により、実験・産業動物の精子幹細胞の特定・回収・培養に必要な幹細胞の細胞生物学的性質に関する知見が得られることが期待される。

    ニワトリ(鶏)の免疫学に関する教科書には、骨髄で造血幹細胞から分化したB細胞前駆細胞は、排泄腔近傍に存在するファブリキウス嚢でB細胞へと分化し、その後、脾臓やパイエル板(回腸終末部)、盲腸扁桃(盲腸基部)といった免疫臓器で機能すると記載されている(Avian Immunology 2nd Edition)。一方で、我々は、広大な表面積を有する盲腸にも成熟したB細胞が多数集積する濾胞構造が広範囲に発達していることを見出し、盲腸扁桃やパイエル板とは異なる、腸管に発達する新たな免疫臓器(Cecal patches: CP)の存在を明らかにしてきた。加えて、経時的に実施した形態学的解析を通して、CPは盲腸扁桃と同様に孵化後に形態形成されること、腸管の免疫機能に深く関わるIgA(免疫グロブリンのサブクラスの1つ)を産生する形質細胞へのB細胞からの最終分化が、盲腸扁桃やパイエル板以上に、CPで盛んに行われていることを見出し、既存の教科書には記載されていない、孵化後の鶏の腸管内で発達するCPを中心とした粘膜免疫システムの存在を提唱してきた。 <BR> そこで本研究では、孵化1, 2, 3, 5, 7 週間後の肉用鶏(Ross308系)より、盲腸を採材・固定し、CPの形態形成を組織学的に解析した。また、組織切片をIgM, IgA, IgYをマーカーとした免疫組織化学染色に供することで、抗体産生細胞の局在変化も比較解析した。その結果、CP形成が開始される時期は、盲腸扁桃の発達時期よりも少し遅く、孵化2週齢以降であることが明らかになった。このことは、盲腸扁桃とCPの形態形成を促す刺激は異なっている可能性を強く示唆していた。さらには、CPで濾胞構造が発達する時期は3週齢前後であり、同時期に濾胞内でB細胞からIgA産生細胞への最終分化が起きていることも明らかになった。

  4. 採卵鶏リポタンパク代謝の暑熱応答―近年の採卵鶏はなぜ採卵率が低下しにくいか?―

    採卵鶏の卵生産は、暑熱環境下で低下するとされていたが、近年の採卵鶏は体重や卵重は減少するものの、産卵率は低下しない、あるいは低下しにくいことが知られている。本研究では、この産卵率維持機構、すなわち、暑熱によりエネルギー摂取が低下した状態における産卵補償機構をニワトリのリポタンパク質代謝に着目して解明することにより、低エネルギー摂取における産卵の代謝調節を理解し、それを利用して通常条件下における産卵の効率化を行うことを目的として、研究を進めている。 昨年度までの研究により、現代の採卵鶏は32℃の暑熱条件下では、体重の減少が認められるものの産卵率には影響せず、卵重と卵黄重を低下させて卵形成を促していることが確認された。さらに、暑熱条件下では、トリグリセリド代謝は低下するものの、コレステロール代謝は肝臓以外では維持され、コレステロール代謝の維持が卵形成の効率化に大きく関与している可能性が推測された。そこで本年度は、卵胞へのリポ蛋白の取り込みと粒子の特徴、および卵胞膜の変動を暑熱環境下と通常環境下で比較して、飼料摂取量が減少する暑熱環境における卵胞発達の効率化を担う鍵を明らかにすることを試みた。 その結果、暑熱環境下(32℃)における鶏の卵胞では、卵黄前駆物質であるリポ蛋白の取り込み速度が上昇し、同時にリポ蛋白であるVLDLyの粒子径の減少が生じていることが明らかとなった。さらには、卵胞の顆粒膜細胞に存在するリポ蛋白レセプターが暑熱により誘導され、同時にギャップジャンクションを構成するタンパク質の遺伝子発現も変動していた。すなわち、摂取量が減少する暑熱環境下では、コレステロール代謝の維持とトリグリセリド代謝の低下によるリポ蛋白粒子の変化が起こると同時に、卵胞膜の卵黄前駆物質取り込みが活性化し、産卵率を維持しているものと推測された。

    本研究では、高血糖・インスリン抵抗性などの鳥類特異的糖代謝特性を支配する要因の一つは一酸化窒素(NO)であるとの仮説を、鳥類の細胞内代謝特性とNO応答の特異性から明示することを目的とした。本年度は、鶏グルコース輸送とインスリン情報伝達系に対するNOの作用を、NO産生阻害剤を用いてin vitroで明示することを試みた。 (1)鶏骨格筋細胞培養系におけるグルコース輸送に対するNO供与体添加の作用 2日齢のブロイラー骨格筋(浅胸筋)より単離した筋衛星細胞を、サブコンフルエントになるまで培養し、培地血清濃度を低下させることにより分化を進行させ、筋管が成熟した約5日目のステージの細胞を用いて試験を行った。NO供与体であるNOC5を骨格筋細胞の培養培地に1〜100μMの濃度で添加し、細胞へのグルコース輸送能を非代謝性グルコースである2-Deoxyglucoseの取り込み能で評価したところ、細胞のグルコース輸送能はNOC5濃度依存的に上昇する傾向が認められた。一方、インスリン存在下(5μg/ml)では、NOC5添加によるグルコース輸送能の上昇は抑制され、NOC5 100μMの添加時のみ、若干の増加が認められたに過ぎなかった。 (2)鶏骨格筋細胞培養系におけるグルコース輸送に対するNOS阻害剤添加の作用 前試験と同様の培養条件下で、培地にNO合成酵素阻害剤であるL-NAMEを0〜100μMの濃度で添加し、細胞のNO産生を抑制した時のグルコース輸送能を検討した。骨格筋細胞へのL-NAME添加は、濃度依存的にグルコース輸送能を低下させる傾向を示し、50μM以上の添加では、無添加の細胞に比べ、グルコース輸送能が有意に低下した。一方、インスリン存在下(5μg/ml)では、L-NAMEの添加により、グルコース輸送能が上昇する傾向が観察された。これは、哺乳動物の骨格筋細胞とは異なるNO応答性であった。以上の結果より、鶏糖代謝にはNOが関与していることが細胞レベルでも明示され、哺乳動物とは異なる鳥類のインスリン応答性特性にも、NOが関与していることが示唆された。

    Peroxisome proliferation-activated receptors (PPAR) are a transcription factor that modulates energy, lipid and glucose metabolism in mammals. The aim of the present study was to investigate whether chicken PPARs is involved in the regulation of energy, lipid and glucose metabolism and whether modification of th gene expression in embryo and post-hatched chicks is utilized to build the high-quality animal production. (1) Regulation of uncoupling protein and the co-factor by PPARs in chicken skeletal muscles In chickens, avUCP and avANT play important roles of energy metabolism in mitochondria of chickens. The expression of avUCP was increased with the increase of PGC-1α gene expression under the cold acclimation for 7days. In the condition of starvation, PPARs and avANT gene expressions were lineally increased until 48h of starvations. These results suggested that the factors involved in the uncoupling of mitochondria membrane (avUCP and avANT) are regulated by PPARs and/or these co-factors. (2) Physiological function of PPARs in chicken adipocyte differentiation The present studies showed that PPARγ is a key regulator of adipocyte differentiation during fatty acid-induced adipogensis in chickens. PPARβ/δ stimulated the chicken adipogenesis induced by activation of PPARγ. Thus, PPARs play a crucial role in the regulation of chicken adipogenesis. (3) Improvement in insulin-stimulated glucose transport in skeletal muscles of chickens administrated with PPARs agonist Troglitazone, a PPARγ synthetic ligand, improved tolerance to glucose and insulin administration through modulation of skeletal muscle glucose transport, with these effects being accompanied by changes in plasma adiponectin and NEFA levels. These results suggested that PPARγ is involved in the carbohydrate metabolism species-specific to chickens. (4) Development of high quality meat production system by activation of PPAR in post-hatched chicks The single intraperitoneal administration of Troglitazone, a PPARγ synthetic ligand, to newly hatched chicks resulted the reduction of fat deposition and the increase of feed utilization in chickens at 7 week of age. It is, therefore, likely that the regulation and modification of PPARs expressions and activities might be useful technique for high-quality chicken meat production.

    安全性・疾病予防性を持つ高品質畜産食品(食肉・卵)の効率生産システムの開発を目標として、低脂肪食肉・卵生産システムの開発研究の一貫として、申請者がはじめて発見した新規リポプロテインレセプターであるLRP380に着目して、その同定・機能解析・発現制御を試みた。 (1)LRP380のリガンドの検索 前年度に確立した精製法により精製したLRP380を用いて,そのリガンドを検索した。LRP380は,鶏VLDL,LDLのリポタンパク質,ビタミンAやDの結合タンパク質,RAPやapoJなどのレセプター関連タンパク質に結合した。さらに,約200kDaの巨大タンパク質と非常に強い結合性を示した。LRP380はヒトmegalinのホモログであるため,この分子はgp280(Cubilin)である可能性が示唆された。gp280は,近年マルチリガンドレセプターとして認識されており,特に腎臓尿細管における物質取り込みを制御していることが明らかとなっている。すなわち,鶏卵巣において,LRP380がCubilinと共同で,卵黄に機能性物質を取り込む役割を果たしているものと推察された。 (2)LRP380の局在性 鶏の前駆脂肪細胞および成熟した脂肪細胞のいずれにもLRP380は発現しており,細胞膜に局在していた。しかし,いずれの細胞にも検出されることから,脂肪蓄積との関与は明らかとはならなかった。 (3)LRP380の生体内における分子生物学的機能解析 in vivoトランスフェクションのためのコンストラクト作製を試みたが,全長が約15kbpと大きいため,ライゲーション効率が低く,現時点では成功していない。また,プロモーター解析のための5領域のクローニングに成功し,現在,機能解析を進めている。

    To develop an anti-obesity vaccine for domestic animals and a safety low fat meat production, we investigated 1)the regulatory mechanism of adipocyte differentiation on animal preadipocyte, 2)screening of functional nutrient for the inhibition of adipocyte hyperplasa, and 3)the effect of intraperitoneal administration with inhibitor of adipocyte proliferation on adipose tissue weight. 1)The regulatory mechanism of chicken adipocyte differentiation To induce the chicken pre-adipocyte differentiation, we firstly clarified that the addition of an exogenous fatty acid mixed with hormones (Ins, DEX, IMX), which induce the differentiation of mammalian adipoocyte, was essential for chicken adipogensis. In addition, it is determine that PPARγ is master regulator of chicken adipocyte differentiation during adipogensis. 2) Screening of functional nutrient for the inhibition of adipocyte hyperplasa The adipocyte proliferation declined with the addition of the mixture of insulin. 3-isobutyl-1-methylxanthin, dexamethason and oleic acid, and completely inhibition of cell growth occurred at the concentration of 20μg/ml insulin, 0.5mM 3-isobutyl-1-methylxanthin, 500nM dexamethason and 300μM oleic acid in the media. 3)The effect of intraperitoneal administration with inhibitor of adipocyte proliferation on adipose tissue weight In chickens with single intraperitoneal injection of these hormone and fatty acid mixture made emulsion with triolein, adipose fat pad weight was significantly lower than the control chickens whereas the body weight decreased to a small extent. In addition, the administration of our anti-obesity vaccine regulated in adipocyte hyperplsia. Unfortunately, it was not still known the powerful nutrient, which was easily accepted by consumer, for the regulation of adipocyte hyperplasia, but the new manipulation technique reducing fat deposition, called anti-obesity vaccine, was developed by the control of adipogensis in chickens.

    Chickens were characterized by blood glucose levels that are twice as high as human and have a marked resistance to insulin actions. In present study, characteristics in the insulin dependent glucose-transport and -uptake during early growing phase and the specificities in factors regulating myogenesis were investigated in broiler chickens. 1. Changes in plasma insulin concentrations in newly hatched chicks Plasma insulin concentrations were determined every day up to 10 day and days at 10, 14, and 28 in broiler chickens. Plasma insulin concentrations during 14 days post-hatch were significantly lower than 28-days old chickens. 2. Effect of insulin injection on plasma glucose concentrations during early growing period Plasma glucose concentrations at 60 min after insulin injections (10 or 40μg/kg BW) were decreased as the dose increased. The decrease of plasma glucose concentrations after insulin injections was significantly greater at 1 - 7 days old chicks than at 14 and 21 days. 3. Tissue specificity in insulin responsible glucose uptake in chicks In Soleus, EDL and Pectoralis muscles and liver, 2-deoxy-D-[^3H]-glucose uptake increased by insulin injection. The glucose uptake in insulin-infused chicks in liver and skeletal muscles was higher by 6.5 and 2 times, respectively, than those in control chicks with no insulin injection. 4. Insulin responsible glucose uptake in chicken skeletal muscles in vitro Glucose uptake into rat skeletal muscles incubated with insulin at 0.02μg/ml in vitro was 2 time higher than that of control culture. In contrast, insulin-responsible glucose uptake in chick skeletal muscles was not observed even when muscles were incubated with 50 time higher dose of of insulin used for rat skeletal muscle incubations. 5. Change in glucose transporter mRNA expressions during myogenesis in chicken myoblasts The mRNA expressions of chicken glucose transporter 1 and 3 (GLUT 1 and 3) were not changed during myogenesis, in particular in the differentiation stage from myoblast to myotubes.

  34. GLUT4の比較分子生物学―鳥類における高血糖維持機構―

    本研究では高血糖およびインスリン抵抗性を特徴とする鶏の特異的な血糖維持機構を解明する観点で、グルコースの取り込みを担うインスリン感受性グルコーストランスポーター(GLUT4)の分子生物学的追究を行った。これまで鶏(鳥類全体)においてGLUT4の存在が予測されてきたが、存在の確証は得られていない。 (1)ノーザンブロットによる検索 : 鶏のほとんど全ての組織(19部位)に対し、ラットGLUT4 cDNAプローブを用いたノーザンブロットを、非常に弱い条件(40-60%の相同性を持つ配列に結合)で行ったが、鶏の全ての組織においてシグナルは検出されなかった。 (2)RT-PCRによる検索これまでラット、マウス、ウシおよびヒトでGLUT4がクローニングされている。GLUT4は種を超えて非常に高い相同性があり、また保存されている領域も多い。そこでGLUT4遺伝子の全ての共通配列部分からオリゴヌクレオチドを設計し、骨格筋、心筋、脂肪組織および肝臓を対象としてRT-PCRを行った。得られたPCR産物全てにおいてシーケンスを行ったが、他種動物のGLUT4と相同性のあるものは得られなかった。 (3)サザンブロットによる検索鶏肝臓から抽出したゲノムDNAにおいてラットGLUT4 cDNAプローブを用いたサザンブロットを行った。ノーザンブロットと同様に非常に弱い条件で行ったが、鶏ゲノムにおいてシグナルは検出されなかった。 (4)他鳥類におけるGLUT4のクローニングカモ、ウズラの組織のノーザンブロット、サザンプロットおよびRT-PCRを行ったが、いずれの組織においても他種動物のGLUT4と相同性のあるものは得られなかった。 以上の結果から、鶏においてインスリン感受性のGLUT4は存在しない可能性が強く示された。このように、鶏の高血糖や高インスリン抵抗性の根拠を分子生物学的に提示した。

  35. 鶏脂肪細胞分化シグナル制御因子の同定とその発現制御

    脂肪過剰蓄積の実用的簡易制御法の確立を目標として、前駆脂肪細胞から脂肪細胞への分化機構とその制御因子を同定することにより、脂肪細胞数の調節・制御を試みることを目的とした。本年度は、分化制御シグナルタンパク質の同定、サブトラクションライブラリーからの分化制御遺伝子のクローニングおよび同定された因子の発現変動を観察した (1)鶏脂肪細胞の分化期におけるCa^<2+>結合性タンパク質の同定 鶏前駆脂肪細胞を分化誘導させた後の脂肪細胞のCalmodulineカラム吸着画分に特徴的に観察された80kDaのタンパク質を酵素処理し、アミノ酸配列を確認したところ、FAT/CD36のホモログであると推定された。また、タンパク量としては多くはないものの、VLDLreceptor(LR8)がwestern blotより検出された。 (2)鶏脂肪細胞の分化期に特徴的な遺伝子のサブトラクションLibraryからのスクリーニング 前年度に作成したサブトラクションライブラリーから約60個のクローンを採取し、異なるサイズのクローンを25種選出し、シーケンスを行ない、未知のものについては全長を決定した。脂肪細胞の分化に関与すると報告されている遺伝子は検出されなかったが、既知のものではacidic phosphoproteinおよびLR8が、未知ではあるものの哺乳動物で配列が決定されているのもではLysosomal amino acid transfer protein 1(LAAT1)が検出された。一方、これまでまったく機能が推測されていない遺伝子が新たに3種検出された。未知遺伝子のなかの1種(WWE related proteinと命名)は、発生に関与するタンパク質の1つであるDeltexに存在するWWE domeinを持っていた。 (3)分化シグナル制御因子の細胞生化学的追求 (1)および(2)で検出されたFAT/CD36、LR8およびWWE related proteinの分化に伴う発現変動を観察した。半定量的RT-PCRにより各mRNAの発現量を測定したところ、いずれのmRNAも分化の進行に伴って発現の上昇が認められた。さらに、FAT/CD36およびLR8についてはWestern blotおよび細胞の免疫染色を行ない、分化した脂肪細胞で強く発現していることが観察された。

    The improvement of meat production may be requested to meet the expanding demands for foods all over the world. In the present study, preparation of chicken myoblast cell line and regulation of gene expression related to the proliferation and differentiation of chicken myoblasts was conducted to construct the base line for promoting the efficient meat production system. 1. Preparation of chicken myoblast cell line a. Chicken myoblasts could be cloned by the colony formation procedure, while the proliferation could not be maintained for several generations. b. b. SV40 Large T was transfected to chicken primary myoblasts for potentiating the proliferation activity. The expression of SV40 Large T was transiently appeared in the cell clone. c. Promoter sequences of specific proteins in chicken muscle cells were subjected for cloning and ligated to plasmid (pd2EGFP-1) which expressed GFP, and then transfected to the chicken primary myoblasts. The analysis is currently continued and the preparation of chicken muscle cell line is scheduled with aid of this procedure. 2. Screening of factors regulating proliferation and differentiation in chicken myoblasts a. In the chicken myoblast culture, MyoD and Myogenin mRNA expression were peaked at 12 and 2h hr, respectively and they were followed by gradual decrease. b. Myosin, CPK activity and MyD mRNA expression increased by addition of prostaglandin E2 (PGE2), suggesting that PGE2 is a factor stimulating the differentiation of muscle cells. c. Proliferation in the myocyte growth stage and fusion of myoblasts in the terminal differentiation was stimulated by addition of beta-hydroxybutyrate (BTB), a ketone body, in the myocyte culture and these effects were modified by the amount of energy source present in the medium. d. Residual yolk present in the abdomen of post-hatched chicks appears to contain a plenty of proteins rich in metabolic function. Addition of the proteins extracted from the residual yolk was no influence on the muscle cell differentiation at the low concentration (1-100 ug) but induced the cell death at the high concentration (1 or 3 mg).

  37. Development in immunochemical control of regulatory enzymes and receptors concerned in the lipid metabolism and the application for low-fat meat production

    The present study was conducted to identify enzymes and receptors involved in fat accumulation and to control by immunochemical methods to reduce obesity domestic animals and consequently produce "Low fat meat". 1. Adipose tissue lipoprotein lipase (LPL) We could have obtained 24 positive clones of hybridoma to bind specifically with chicken LPL and 3 clones out of 24 were effective to reduce LPL activity in adipose tissues. In chickens received continuous infusion of LPL antibody (CLP-16) for 14 days, adipose fat pad weight was significantly lower than the control chickens whereas the body weight decreased to a small extent. These findings suggest that persistent administration of anti-LPL monoclonal antibody is effective mean to modify LPL activity and consequently manipulate fatness in broiler chickens. 2. Adipose tissue lipoprotein receptor (LR) It was shown following western blot analysis using antiserum of various LRs (LRP, LRP380, and LR8) that LR8 was present in chicken adipose tissues and concerned in the chicken adiposity. However, administration of LR8 antibody did not modify VLDL uptake to chicken adipocytes. 3. Hepatic lipoprotein receptor (HLR) It was not able to identify the new cDNA clone of chicken HLR though the screening of chicken liver cDNA library. However, new LDL binding proteins (100 and 130kDa) were detected in the ligand blot analysis in chicken liver membrane. The new LDL binding proteins were characterized by no-binding Ca ion and receptor associated protein (RAP), which were different from LDL receptor family obtained in mammals. 4. Plasma cholesteryl ester transfer protein (CETP) The CETP activity in chicken plasma was high before laying eggs whereas that in laying hens was not detectable. These results suggested that chicken CETP play an important role to the cholesterol metabolism. We have been trying to obtain chicken CETP cDNA clone using human CETP probe, while at present chicken CETP cDNA sequence is still not available.

  38. 鶏脂肪細胞リポプロティンリパーゼ遺伝子のクローニングとその発現制御領域の解明

    脂肪過剰蓄積のメカニズムの解明とその制御法の開発を目標として、脂肪組織での主要な脂肪蓄積調節酵素であるリポプロテインリバーゼ(LPLase)の遺伝子・蛋白質発現機構を明らかにすることを目的とした。本年度は、鶏脂肪細胞培養系の作成と脂肪組織LPLase cDNAのクローニングを行なった。 (1) 鶏脂肪細胞培養系の作成 無菌的に採取した7日齢ブロイラー雛の脂肪組織を供試し、酵素消化法により脂肪前駆細胞を調製した。鶏脂肪前駆細胞は10%FBSを含むM199(Growth medium)で増殖し、Growth mediumに10mM Glucose、200μg/ml Insulin、250nM Dexamethason、33μM biotin、17μM pantothenaeおよび1mM oleic acidを添加すると24時間後に脂肪滴を有する脂肪細胞に分化した。分化した脂肪細胞は、細胞内にトリグリセリドを蓄積し、LPLase活性およびVLDL receptor、LeptinのmRNAを発現しており、鶏の脂肪蓄積機構を分子生物学的に解析するのに適した細胞であることが確認された。 (2) 脂肪細胞LPLaseのcDNAクローニング まず、LPLase発現の活発な鶏脂肪細胞のcDNAライブラリー(ベクター:pBluescript II KS+)を作成した。次に、ブロイラー精製LPLaseのアミノ酸配列よりプローブを作成し、cDNAライブラリーを用いたコロニーハイブリダイゼーションで完全長のcDNAをクローニングすることに成功した。ブロイラーLPL cDNAは2,297bpで、490個のアミノ酸をコードしていた。また、鶏LPLのタンパク質部分の推定分子量は52,601、等電点は7.91と推定され、糖鎖付加を経て活性を持つ成熟LPLとなることが推測された。

    前年度は家畜のモデル動物として鶏を用い、その肝細胞および筋細胞の培養システムを確立した。そこで、本年度は生理活性物質を検索するための培養条件を検討し、種々の生理活性物質に対する細胞応答を観察するとともに、未知の生理活性物質の検索を試みた。 (1) 鶏筋細胞を用いた生理活性物質検索システムの確立 7×10^3cells/cm2の濃度で鶏筋芽細胞培養し、培養1日目(増殖)、培養2日目(初期分化)および培養3日目(終末分化)に生理活性物質を添加することにより、それぞれのステージに影響する生理活性物質が検索できることが示された。 (2) 鶏肝細胞を用いた生理活性物質検索システムの確立 各種アミノ酸、グルコースを添加したBME培地に、0.12μg/mlアプロチニンを添加することにより、少なくとも24時間は生理活性物質を検索するのに適した無血清培地での培養が可能となり、生理活性物質を検索するための培養実験系を確立することができた。 (3) 鶏筋細胞および肝細胞における各種生理活性物質の細胞応答 筋細胞:プロスタグランジンE2は鶏筋細胞の初期分化を刺激し、筋細胞の分化を促進させることが明らかとなった。さらに、β-ハイドロキシ酪酸は筋細胞の終末分化には影響しなかった。 肝細胞:α-リノレン酸だけではなく、リノール酸の添加でも細胞の脂肪酸合成能を抑制した。また、カプリル酸のVLDL分泌抑制作用が示唆された。 (4) 卵黄抽出タンパク質の筋細胞分化に及ぼす影響 孵化後1日齢の残存卵黄採取して0.5M NaClにより卵黄抽出タンパク質を調製し、筋細胞の増殖および分化の様相を観察したところ、残存卵黄中には増殖もしくは分化誘導因子が存在する可能性が示唆された。

    (I) Chylomicron remnant and VLDL remnant in chickens were prepared by the method using ultracentrifugation (d<1.019) following an in vitro incubation of chylomicron or VLDL prepared from chicken plasma with chicken LPL for 2h at 37゚C. (2) The chylomicron remnants and VLDL remnants yielded the almost single peak in the elution pattern on Bio-Gel A50m column (1.6 * 50cm) and the particle size were smaller than chylomicron and VLDL since the elution of remnants by Bio-Gel A50m was similar to LDL rather than chylomicron and VLDL.Chicken remnants had low triglyceride contents with high cholesterol and almost comparable Apo B100 and Apo A-I contents while lower Apo C,Apo 52kDa and Apo 20kDa contents compared to chylomicron or VLDL. (3) ^<125>I-labeled LDL,chylomicron remnant and VLDL remnant bound specificity to chicken or rat liver membranes. The specific binding of chicken ^<I25>I-labeled LDL to chicken liver membrane was inhibited by >70% with chylomicron, VLDL,VLDL remnant or LDL,while the addition of chylomicron remnant had little effect. These results demonstrate that chicken chylomicron remnant bind the site of different receptor of LDL. (4) The uptake of Chicken ^<I25>I-labeled lipoproteins to chicken primary hepatocyte was decreased to about>30% by an excess of the corresponding lipoproteins. It is concluded that the uptake of chicken lipoproteins are mediated by lipoprotein receptors. (5) In ligand blot aunalysis of chicken hepatic lipoprotein receptors, three difrerent receptor proteins (145,115 and 105 kDa) were present in chicken liver. (6) Two kinds of chicken hepatic lipoprotein receptors were purifed by a procedure with three chromatography step on Hydroxyapatite, LDL-affinity and Mono Q.The receptors (chicken hepatic apo B receptor (CHBR) -1 and CHBR-2) were characterized a molecular weight of 115kDa and 105kDa, as estimated on SDS-PAGE.On ligand blotting, both CHBR-1 and CHBR-2 bound to apo B lipoproteins.

  41. アポプロテインC-IIとC-IIIによる鶏脂肪組織リポプロテインリパーゼ反応の制御機構

    (1)鶏、ラットおよびヒトの血漿から超遠心で分離したカイロミクロンおびVLDLからMorrisettらの方法によりApoproteinを調製した。分離したApoproteinを6M尿素で可溶化後、分子量1万の限外濾過により得たApo C群をMno Q^Rに供し、0.4MまでのNaCl直鎖グラディントで溶出した。各種動物から調製したいずれの試料も、0.1〜0.2M付近にリポプロテインリパーゼ(LPLase)活性化作用を示すApo C-IIのピークが溶出した。さらに、ヒトでは、Apo C-IIIを含むと考えられる0.25M付近のピーク(Apo C-III画分)にはミルクLPLase阻害活性が確認されたが、ラットのApo C-III画分はラットLPLase活性に殆ど影響せず、鶏Apo C-III画分が鶏LPLase活性を逆に活性化した。 (2)鶏Apo C-IIIの存在の有無を確認するために、逆相カラム(Resource RPC)を用いて鶏Apo C-III画分を分離精製し、N末端分析を行った。逆相カラムではメインピークと多数のマイナ-ピークが検出された。メインピークのN末端アミノ酸配列は鶏Apo A-Iと100%の相同性を示し、Apo A-Iの部分分解物であることが明らかとなった。PMSFなどのProtease阻害剤を添加しても、同物質がメインピークであることから、鶏リポプロテインにはApc C-IIIは存在せず、かわりにApo A-Iの分解物が存在して、しかもApo A-Iの分解物は鶏LPLase活性化作用がある可能性が示唆された。 (3)トリオレインエマルジョンに鶏Apo C-IIおよびApo A-I分解物を30μg/ml添加したところ、鶏LPLaseのVmはいずれも上昇したが、Apo A-I分解物の上昇率はApo C-IIの約1/3であった。 (現在、本結果の投稿を準備中である)
