INTRODUCTION: Triple-negative breast cancer (TNBC), recognized as the most heterogeneous type of breast cancer (BC), exhibits a worse prognosis than other subtypes. Mitochondria dynamics play a vital role as mediators in tumorigenesis by adjusting to the cell microenvironments. However, the relationship between mitochondrial dynamics and metabophenotype exhibits discrepancies and divergence across various research and BC models. Therefore, this study aims to explore the role of mitochondrial dynamics in TNBC drug resistance and tumorigenesis. METHODS: The Wst-8 test was conducted to assess doxorubicin sensitivity in HCC38, MDA-MB-231 (TNBC), and MCF-7 (luminal). Confocal microscopy and FACS were used to quantify the mitochondrial membrane potential (ΔφM), mitophagy, and reactive oxygen species (ROS) production. Agilent Seahorse XF Analyzer was utilized to measure metabolic characteristics. Dynamin-related protein-1 (DRP1), Parkin, and p62 immunohistochemistry staining were performed using samples from 107 primary patients with BC before and after neoadjuvant chemotherapy (NAC). RESULTS: MDA-MB-231, a TNBC cell line with reduced sensitivity to doxorubicin, reduced ΔφM, and enhanced mitophagy to maintain ROS production through oxidative phosphorylation (OXPHOS)-based metabolism. HCC38, a doxorubicin-sensitive cell line, exhibited no alterations in ΔφM or mitophagy. However, it demonstrated an increase in ROS production and glycolysis. Clinicopathological studies revealed that pretreatment (before NAC) expression of DRP1 was significant in TNBC, as was pretreatment expression of Parkin in the hormone receptor-negative group. Furthermore, low p62 levels seem to be a risk factor for recurrence-free survival. CONCLUSION: Our findings indicated that the interplay between mitophagy, linked to a worse clinical prognosis, and OXPHOS metabolism promoted chemotherapy resistance in TNBC. Mitochondrial fission is prevalent in TNBC. These findings suggest that targeting the unique mitochondrial metabolism and dynamics in TNBC may offer a novel therapeutic strategy for patients with TNBC.

In this study, we proposed a method for fabricating diagnostic imaging nanoparticles composed of Au nanoparticles and silica shells (Au/SiO2). The proposed method consisted of two steps. The first step was the synthesis of Au nanoparticles. In sodium hydroxide (NaOH) solution, hydrogen tetrachloroaurate (III) trihydrate was reduced with tetrakis(hydroxymethyl)phosphonium chloride to synthesize Au nanoparticles with a diameter of 1.7 ± 0.3 nm. The Au nanoparticles were then coated with silica in the following step. The silica coating was achieved via a sol–gel reaction of tetraethyl orthosilicate in the presence of Au nanoparticles in water/ethanol dissolved in NaOH. The Au/SiO2 nanoparticles degraded faster in saline or phosphate-buffered saline than in water, and the X-ray imaging capability was retained.

Breast cancer resistance protein (BCRP), also known as ATP-binding cassette transporter G2 (ABCG2), is associated with chemotherapy resistance. BCRP is also implicated in breast cancer stem cells, and is reported as a poor prognostic factor. However, the relationship of BCRP levels in breast cancer tissues with chemotherapy resistance and prognosis has not been clarified. We aimed to evaluate the correlation between BCRP expression and prognosis in breast cancer using immunohistochemistry with fluorescent phosphor-integrated dots (IHC-PIDs). A total of 37 breast cancer patients with residual cancer in the primary tumor and axillary lymph nodes were evaluated. BCRP levels in breast cancer tissue and metastatic lymph nodes were quantitatively detected after neoadjuvant chemotherapy (NAC). Among these 37 patients, 24 had corresponding core needle biopsies obtained before NAC. Biomarker assay with IHC-PIDs showed high accuracy for the quantitative assessment of BCRP with low expression. High BCRP expression in the primary tumor and metastatic lymph nodes after preoperative chemotherapy was associated with worse overall survival. In conclusion, high BCRP levels may be associated with poor prognosis in patients with breast cancer, having residual tumors within the primary tumor and lymph nodes after preoperative chemotherapy. These findings provide a basis for further appropriate adjuvant therapy in these patients.

In this study, we aim to develop gold-coated Fe3O4 nanoparticles (Fe3O4@Au NPs) as theranostic agents for magnetic hyperthermia and CT imaging applications. The Fe3O4 NPs were synthesized via thermal decomposition method, and the gold was then deposited onto the surface of Fe3O4 NPs by reducing gold acetate at 190 °C. The average sizes of Fe3O4 and Fe3O4@Au NPs were 5.2 nm and 6.1 nm, respectively, which are effectively removed by the kidneys. The magnetization of Fe3O4@Au NPs (9.7 emu/g-Fe3O4) at 300 K was much smaller than that of Fe3O4 NPs (52.4 emu/g-Fe3O4). The heating efficiency of Fe3O4@Au NPs in water was sufficient to treat the tumor at 43–45 °C, and their high CT value of 851 HU was obtained. The synthesized ultrasmall Fe3O4@Au NPs showed great promise as a potential theranostic agent for magnetic hyperthermia and CT imaging applications.

The evaluation of angiogenesis inhibitors requires the analysis of the precise structure and function of tumor vessels. The anti-angiogenic agents lenvatinib and sorafenib are multi-target tyrosine kinase inhibitors that have been approved for the treatment of hepatocellular carcinoma (HCC). However, the different effects on tumor vasculature between lenvatinib and sorafenib are not well understood. In this study, we analyzed the effects of both drugs on vascular structure and function, including vascular normalization, and investigated whether the normalization had a positive effect on a combination therapy with the drugs and radiation using micro X-ray computed tomography with gold nanoparticles as a contrast agent, as well as immunohistochemical analysis and interstitial fluid pressure (IFP) measurement. In mice subcutaneously transplanted with mouse HCC cells, treatment with lenvatinib or sorafenib for 14 days inhibited tumor growth and reduced the tumor vessel volume density. However, analysis of integrated data on vessel density, rates of pericyte-covering and perfused vessels, tumor hypoxia, and IFP measured 4 days after drug treatment showed that treatment with 3 mg/kg of lenvatinib significantly reduced the microvessel density and normalized tumor vessels compared to treatment with 50 mg/kg of sorafenib. These results showed that lenvatinib induced vascular normalization and improved the intratumoral microenvironment in HCC tumors earlier and more effectively than sorafenib. Moreover, such changes increased the radiosensitivity of tumors and enhanced the effect of lenvatinib and radiation combination therapy, suggesting that this combination therapy is a powerful potential application against HCC.

The technology to accurately image the morphology of tumor vessels with X-ray contrast agents is important to clarify mechanisms underlying tumor progression and evaluate the efficacy of chemotherapy. However, in clinical practice, iodine-based contrast agents present problems such as short blood retention owing to a high clearance ability and insufficient X-ray absorption capacity when compared with other high atomic number elements. To resolve these issues, gold nanoparticles (AuNPs), with a high atomic number, have attracted a great deal of attention as contrast agents for angiography, and have been employed in small animal models. Herein, we developed novel contrast agents using AuNPs and captured changes in tumor vessel morphology with time using X-ray computed tomography (CT). First, glutathione-supported single nanometer-sized AuNPs (sAu/GSH) (diameter, 2.2 nm) were fabricated using tetrakis(hydroxymethyl)phosphonium chloride as a reducing agent. The sAu/GSH particles were intravenously injected into mice, remained in vessels for a few minutes, and were then excreted by the kidneys after 24 h, similar to the commercial contrast agent iopamidol. Next, the Au/GSH and lactoferrin (sAu/GSH-LF) (long axis size, 17.3 nm) complex was produced by adding lactoferrin to the sAu/GSH solution under the influence of a condensing agent. On intravenously administering sAu/GSH-LF to mice, the blood retention time was 1-3 h, which was considerably longer than that observed with iopamidol and sAu/GSH. Moreover, we succeeded in imaging morphological changes in identical tumor vessels for several days using X-ray CT with sAu/GSH-LF.

In addition to genomic signaling, Estrogen receptor alpha (ERα) is associated with cell proliferation and survival through extranuclear signaling contributing to endocrine therapy (ET) resistance. However, the relationship between extranuclear ERα and ET resistance has not been extensively studied. We sought to measure extranuclear ERα expression by immunohistochemistry using phosphor-integrated dots (IHC-PIDs) and to assess its predictive value for ET resistance. After quantitative detection of ERα by IHC-PIDs in vitro, we developed "the nearest-neighbor method" to calculate the extranuclear ERα. Furthermore, tissue sections from 65 patients with HR+/HER2- BC were examined by IHC-PIDs, and the total ERα, nuclear ERα, extranuclear ERα PIDs score, and ratio of extranuclear-to-nuclear ERα (ENR) were measured using the novel method. We demonstrate that quantification of ERα using IHC-PIDs exhibited strong correlations to real-time qRT-PCR (r2 = 0.94) and flow cytometry (r2 = 0.98). High ERα ENR was significantly associated with poor overall survival (p = 0.048) and disease-free survival (DFS) (p = 0.007). Multivariate analysis revealed that the ERα ENR was an independent prognostic factor for DFS [hazard ratio, 3.8; 95% CI, 1.4-11.8; p = 0.006]. Our automated measurement has high accuracy to localize and assess extranuclear ERα. A high ERα ENR in HR+/HER2- BC indicates decreased likelihood of benefiting from ET.

    More details Close

In breast cancer, the prognosis of human epidermal growth factor receptor 2 (HER2)-positive patients (20-25%) has been dramatically improved by the clinical application of the anti-HER2 antibody drugs trastuzumab and pertuzumab. However, the clinical outcomes of HER2-negative cases with a poor prognosis have not improved, and novel therapeutic antibody drugs or diagnostic molecular markers of prognosis are urgently needed. Here, we targeted protease-activated receptor 1 (PAR1) as a new biomarker for HER2-negative patients. The developed anti-PAR1 antibody inhibited PAR1 activation by matrix metalloprotease 1 and thereby prevented cancer-cell migration and invasion. To estimate PAR1 expression levels in HER2-negative patient tissues using the antibody, user-friendly immunohistochemistry with fluorescence nanoparticles or quantum dots (QDs) was developed. Previously, immunohistochemistry with QDs was affected by tissue autofluorescence, making quantitative measurement extremely difficult. We significantly improved the quantitative sensitivity of immunohistochemistry with QDs by using an autofluorescence-subtracted image and single-QD imaging. The immunohistochemistry showed that PAR1 expression was strongly correlated with relapse-free survival time in HER2-negative breast cancer patients. Therefore, the developed anti-PAR1 antibody is a strong candidate for use as an anticancer drug and a prognostic biomarker for HER2-negative patients.

In this study, we aimed to develop a method for improving the antitumor effect of an angiogenesis inhibitor (bevacizumab). In 2019, conditions for low-dose radiation therapy to be used in combination therapy with bevacizumab administration were examined. In addition, the tumor-bearing mice administered with gold nanoparticles (AuNPs) were irradiated with radiation, and radiosensitization effect of AuNPs on tumor shrinkage was investigated. In 2020, to evaluate the histological effect of combination therapy of angiogenesis inhibitor and radiation therapy, we developed techniques to investigate the "morphological change of tumor blood vessels" and "functional change of tumor blood vessels". In 2021, those evaluation technologies developed in 2020 were applied to the evaluation of the antitumor effect of mice in combination with angiogenesis inhibitors and radiation therapy.

We prepared ultra-small gold nanoparticles with a diameter of approximately 3 nm by coating with reduced glutathione. In animal experiments using mice, it was confirmed that the nanoparticles could be excreted in urine with a half-life of less than one hour in blood. The initial plan was then slightly modified to integrate multiple gold nanoparticles on a protein basis rather than iron oxide nanoparticles. As a result, biodegradable nanoparticles with 3~4 gold nanoparticles bound on protein particles of about 10~20 nm in diameter were successfully synthesized. The results of CT imaging analysis showed that synthesized nanoparticles had different blood convection times depending on the size and properties of the scaffold protein. The gold nanoparticles can be used as a contrast agent to delineate tiny blood vessels in tumors.

We aimed to evaluate the correlation between BCRP expression and prognosis in breast cancer tissue samples using immunohistochemistry with fluorescent phosphor-integrated dots (IHC-PIDs). A total of 37 breast cancer patients with residual cancer in the primary tumor and axillary lymph node were evaluated. BCRP levels in breast cancer tissue (PT) and metastatic lymph nodes (LN) were quantitatively detected after preoperative chemotherapy (NAC) and core needle biopsy (CNB). Biomarker assay with IHC-PIDs showed high accuracy for quantitative assessment of BCRP with low expression. High BCRP expression in Primary tumor and lymphnode after NAC was associated with worse overall survival (log-rank, p=0.0089). In conclusion, high BCRP levels are associated with poor prognosis in breast cancer patients with residual tumor in the primary tumor and lymph node after preoperative chemotherapy. These findings provide a basis for further appropriate adjuvant therapy in these patients.

Micro cancers in mouse models of colorectal cancer liver metastasis (CRCLM) and hepatocellular carcinoma (HCC) were detected using high-resolution CT imaging device with gold nanoparticle contrast agent. The in vivo CT imaging showed that there is a difference between the EPR effects of the micro cancers in HCC and in CRCLM. We succeeded to obtain pathological sections of the micro cancers and immunohistochemistry of them were performed using angiogenesis markers of CD31 and αSMA. As the result, it was suggested that differences in angiogenesis affect drug delivery in early stage of cancer.

Hepatocellular carcinoma (HCC), particularly originated from the background of non-alcoholic steatohepatitis (NASH), has been on the rise in recent years. The carcinogenic mechanisms of NASH-HCC is difficult to understood because of its complications such as diabetes mellitus and hyperlipidemia. Then, early diagnosis of NASH-HCC is very important. We attempted to detect early lesions in NASH-HCC model mice by using gold nanoparticle contrast agents. By using the EPR (Enhanced Permeability and Retention Effect) of gold nanoparticles, it was actually successfully detected a micro-cancer less than 1 mm. Furthermore, pathological analysis according to the degree of progression was possible by removing those small lesions as pathological sections.

Bone metastasis is a high frequency as a metastatic site of cancer like breast, lung and prostate cancer. It is important to diagnose and medical treatment of the bone metastasis at early stage because of many clinically problematic symptoms such as pain and hypercalcemia caused by pathological fractures. Using a cancer-bearing mouse model that causes voluntarily bone metastasis, we measured the bone density of the osteocortex and bone marrow. The researh results showed us that at the beginning of the bone metastasis, the bone marrow metastasis was occurred ahead to the osteocortex metastasis. It is considered useful for postoperative periodic examination because it can be measured at low cost and with no exposure. In this study we could diagnose early bone marrow metastasis and it seemed to be effective in elucidation of the mechanism of invasion and propagation of bone metastasis.

Assessing breast cancer tissues for expression of cancer stemness biomarkers, such as breast cancer resistance protein (BCRP) which is a member of ABC transporter, may provide critical information for patient prognosis. We developed a novel immunohistochemical method with new fluorescent nanoparticle, phosphor integrated dot (PID), by single-particle imaging using tissue samples from patients with clinical information. We successfully calculated the number of PID particles in a cell and defined it as nano patho score that reflects the level of BCRP protein expression in cancer cells. The BCRP nano patho score in 13 cases was correlated proportionally with the score of Ki67 as assessed by immunohistochemical method (R =0.62). But BCRP nano patho score were not correlated with breast cancer recurrence rate in breast cancer patients who were treated with preoperative chemotherapy

We developed new method for quantitative imaging of HER2/HER3 dimer by using fluorescent nanoparticle probes conjugated with antibodies. We performed detecting HER2/HER3 hetero-dimers by using the combination of antibody-based fluorescent probes which induce fluorescent resonance energy transfer (FRET). Because the efficiency of FRET is extremely sensitive to the distance between the pair of fluorophores, this method can be suitable for our purpose. We have applied this method for detection of HER2/HER3 hetero-dimers expressed on the membranes of human breast cancer cell lines. The amount of HER2/HER3 hetero-dimers and localization on cultured-cell lines have been evaluated with quantitative sensitivity. The imaging data showed HER2 and/or HER3-derived unique localization, and hot-spot on cancer cells. We also developed the evaluation method of the effect of antibody-drugs, especially anti-HER2 dimerization inhibitor, by FRET technique.

Contrast agents are often used to enhance the contrast of X-ray computed tomography (CT) imaging of tumors to improve diagnostic accuracy. However, because the iodine-based contrast agents currently used in hospitals are of low molecular weight, the agent is rapidly excreted from the kidney. This leads to nonspecific enhancement of contrast images for tissues. Here, we created gold (Au) nanoparticles as a new contrast agent to specifically image tumors with CT using an enhanced permeability and retention (EPR) effect. Au has a higher X-ray absorption coefficient than does iodine. Au nanoparticles were supported with polyethylene glycol (PEG) chains on their surface to increase the blood retention The developed Au nanoparticles were injected into tumor-bearing mice, and the distribution of Au was examined with CT imaging, transmission electron microscope (TEM) imaging. The results show that specific localization of the developed Au nanoparticles in the tumor

In this study, we aim to develop the new method to measure the level of expression of sorafenib-targeting proteins by fluorescent nanoparticle quantitatively with a high degree of accuracy. We combined pegylated fluorescent nanoparticle with sorafenib via linker, and confirmed by infrared spectroscopy that synthesized nanoparticle had predictable structure. From the results of experiments using cultured cells, bond between sorafenib and fluorescent nanoparticle did not decrease the inhibitory capacity of cell proliferation, which sorafenib had originally. This showed that combination via linker did not decrease EFGR-binding capacity of sorafenib. In pathological analyses using paraffin-embedded cells, it was difficult to adjust photographing conditions, which was left open.

Nanomedicine molecular science focuses on molecular reactions in the cells, which constitute the living organism and control its biological activities. The cellular environment, however, differs from the chemical reaction environment. The goal of this project is to develop molecular reaction parameters to quantify and examine such reactions. The studies are part of a scientific initiative focusing on cells and have the following main objectives: establishing the theoretical basis of molecular reactions in the cell, and understanding the cellular environment and intracellular chemical reaction mechanisms. They include topics related to the origin of cellular functions, which provide knowledge indispensable for molecular nanomedicine; topics required for establishing molecular nanomedicine; topics that may help integrate the research projects and promote constructive interactions among researchers; and topics that encourage original ideas to solve difficulties in the field.

Luminescent semiconductor nanoparticles (QDs) are expected to be used as a fluorescent marker for medical diagnostics. Among various methods for reducing harm of Cd contained in QD, encapsulating, or coating of contrast agents with materials inert to living bodies is a good candidate. Meanwhile, materials that have both properties of fluorescence and X-ray absorption will be used for imaging targets by multiple techniques. The present work developed methods for preparing the QDs coated with silica (QD/SiO2), the QD/SiO2 particles on which Au nanoparticles were immobilized (QD/SiO2/Au particles), and the QD/SiO2/Au particles coated with silica (QD/SiO2/Au/SiO2 multi-layered particles).

We prepared GFP-fused EB1-expressing human breast cancer cells. The growth rate of the microtubule plus-end was measured using the positions of EB1 assembly over time. Analysis of the dynamics of the EB1-GFP comet showed that the microtubule growth rates decreased when the cells were treated with anticancer drugs. We also measured the microtubule growth rates in tumor-bearing mice by using an original stereotaxic instrument. Thus, alteration in EB1-GFP dynamics is an effective indicator for the effects of anticancer drugs.

We succeeded in real-time fluorescent imaging of sentinel lymph nodes of various gastrointestinal tracts in pig model using QDs as a tracer. After that, we analyzed fluorescent specificities in varied conditions and revealed the range of a signal detection through fat tissue or other organs. Furthermore, we developed novel organic material nanoparticles as a safety fluorescent probe for sentinel lymph node biopsy(SLNB) with the idea of clinical application. We revealed that these particles were suitable for SLNB and their fluorescent specificities were similar to the QDs.
