    Abstract Spectrum characteristics of photoacoustic signals are potential indicators to assess the structural information of optical absorbers. This study investigated whether a spectrum analysis approach can evaluate microvascular diameters obtained by an annular array AR-PAM system. First, 2D numerical simulations were performed to estimate spectral changes in five absorbers with different diameters from 30 to 200 μm. Slope and intercept values from the regression line of the spectrum curve were calculated. Second, the estimated changes were validated by acquiring photoacoustic signals from tungsten wires with five different diameters. Both in numerically and experimentally, spectrum analysis showed the larger the diameter, the greater the lower frequency components, resulting in steeper slope and greater intercept values. Finally, photoacoustic signals acquired from cutaneous microvessels were analyzed. The FWHM diameter of the vessels were corresponded to the slope (r = −0.595; $p\,$ < 0.05) and intercept (r = 0.657; $p\,$ < 0.05) values, respectively.

    Abstract Photoacoustic imaging (PAI) represents an innovative method that utilizes optics for transmission and acoustic for sensing. Within the realm of PAI modalities, optical-resolution photoacoustic microscopy (OR-PAM) stands out for its ability to achieve high-resolution cellular and intracellular imaging. However, an optical testbed setup emulating the intracellular environment and accelerating the development of cellular-level OR-PAM systems has not been established. In this study, we present a testbed setup designed to mimic intracellular conditions. Assessment of this testbed setup efficacy involved embedding photoacoustic contrasts and imaging them using an OR-PAM system. The imaging results show that our testbed setup can provide instrumental in accurately measuring photoacoustic signals within an intracellular aqueous environment, avoiding risks of the contamination and degradation of the acoustic transducer.

    Abstract Background Quantitative and comprehensive visualization of urinary flow dynamics in the urethra is crucial for investigating patient‐specific mechanisms of lower urinary tract symptoms (LUTS). Although some methods can evaluate the global properties of the urethra, it is critical to assess the local information, such as the location of the responsible lesion and its interactions with urinary flow in relation to LUTS. This approach is vital for enhancing personalized and focal treatments. However, there is a lack of such diagnostic tools that can directly observe how the urethral shape and motion impact urinary flow in the urethra. Purpose This study aimed to develop a novel transrectal ultrasound imaging modality based on the contrast‐enhanced urodynamic vector projectile imaging (CE‐UroVPI) framework and validate its clinical applicability for visualizing time‐resolved flow dynamics in the urethra. Methods A new CE‐UroVPI system was developed using a research‐purpose ultrasound platform and a custom transrectal linear probe, and an imaging protocol for acquiring urodynamic echo data in male patients was designed. Thirty‐four male patients with LUTS participated in this study. CE‐UroVPI was performed to acquire ultrasound echo signals from the participant's urethra and urinary flow at various voiding phases (initiation, maintenance, and terminal). The ultrasound datasets were processed with custom software to visualize urinary flow dynamics and urethra tissue deformation. Results The transrectal CE‐UroVPI system successfully visualized the time‐resolved multidirectional urinary flow dynamics in the prostatic urethra during the initiation, maintenance, and terminal phases of voiding in 17 patients at a frame rate of 1250 fps. The maximum flow speed measured in this study was 2.5 m/s. In addition, when the urethra had an obstruction or an irregular partial deformation, the devised imaging modality visualized complex flow patterns, such as vortices and flow jets around the lesion. Conclusions Our study findings demonstrate that the transrectal CE‐UroVPI system developed in this study can effectively image fluid‐structural interactions in the urethra. This new diagnostic technology has the potential to facilitate quantitative and precise assessments of urethral voiding functions and aid in the improvement of focal and effective treatments for patients with LUTS.

    Abstract In veno-arterial extracorporeal membrane oxygenation (VA-ECMO) treatment, the mixing zone is a key hemodynamic factor that determines the efficacy of the treatment. This study aimed to evaluate the applicability of a novel ultrasound technique called vector flow imaging (VFI) for visualizing complex flow patterns in an aorta phantom under VA-ECMO settings. VFI experiments were performed to image aortic hemodynamics under VA-ECMO treatment simulated in an anthropomorphic thoracic aorta phantom using a pulsatile pump (cardiac output: 2.7 L/min) and an ECMO pump with two different flow rates, 0.35 L/min and 1.0 L/min. The cardiac cycle of hemodynamics in the ascending aorta, aortic arch, and descending aorta was visualized, and the spatio-temporal dynamics of flow vectors were analyzed. VFI successfully visualized dynamic flow patterns in the aorta phantom. When the flow rate of the ECMO pump increased, ECMO flow was more dominant than cardiac output in the diastole phase, and the speed of cardiac output was suppressed in the systole phase. Vortex flow patterns were also detected in the ascending aorta and the arch under both ECMO flow rate conditions. The VFI technique may provide new insights into aortic hemodynamics and facilitates effective and safe VA-ECMO treatment.

    Abstract High-intensity focused ultrasound (HIFU) therapy is a noninvasive treatment for cancer. Prediction of the HIFU treatment region in advance by ultrasound imaging is important for the efficacy and safety of the treatment. Acoustic radiation force (ARF) imaging has been shown to be useful in estimating the heat distribution caused by HIFU. In this study, we investigated the effect of shielding on the estimated heat distribution when the propagation of HIFU is partially shielded by an object simulating ribs, imitating an actual clinical situation, and examined the correction method of HIFU output power using ARF imaging to obtain tissue coagulation equivalent to that without shielding. As a result, it was suggested that even with partial acoustic shielding, when the HIFU output power is adjusted so that the displacement induced by ARF is equal to that without shielding, almost the same coagulated region can be obtained as in the unshielded case.

    Detecting abnormal blood flow is possible through transcranial Doppler (TCD) ultrasound by measuring blood velocity in cerebral arteries. Velocity measurements are at the highest precision when the direction of blood flow coincides with the ultrasound beam. However, because TCD is typically performed blindly (i.e., without a B-mode), a 0° interrogation angle is usually assumed. This leads to a common issue of angular mismatch. This study quantitatively shows the angular mismatch effects on the measured velocities using a TCD ultrasound flow phantom compared with the velocities measured by optical particle image velocimetry (PIV) as control. Resulting errors with and without ultrasound machine angular correction were also considered. An ultrasound phantom developed by combining polyvinyl alcohol hydrogel (PVA-H), quartz glass as a scatterer, and a gypsum plate as a skull bone was utilized to approximate the middle cerebral artery TCD measurement from the temporal window. The PVA-H and quartz glass compositions were controlled to achieve transparency and enable PIV velocity measurement. Then, TCD velocity measurement was conducted on several interrogation and mismatch angles. Comparison results revealed that without an ultrasound machine angle correction, all measurements yielded underestimation with 73.9% at the highest in the 80° interrogation window at the 130 mL/min flow. On the other hand, with the correction, the errors in almost all angles were comparatively lower; however, at 80° at the 124 mL/min flow, a maximum overestimation rate of 113.7% was found, showing a larger error magnitude. Therefore, we find that angular mismatch, especially in larger angles, leads to inaccurate velocity measurements in TCD. Our results suggest that despite angle correction, velocity errors may still occur when the interrogation angle changes.

    OBJECTIVE: The need for ultrasound flow phantoms to validate ultrasound systems requires the development of materials that can clearly visualize the flow inside for measurement purposes. METHODS: A transparent ultrasound flow phantom material composed of poly(vinyl alcohol) hydrogel (PVA-H) with dimethyl sulfoxide (DMSO) and water solution manufactured using the freezing method and mixed with quartz glass powder to exhibit scattering effects is proposed. To achieve transparency of the hydrogel phantom, the refractive index (RI) was changed to match that of the glass by modifying the PVA concentration and the ratio of DMSO to water in the solvent. The feasibility of optical particle image velocimetry (PIV) was verified by comparing an acrylic rectangular cross-section channel with a rigid wall. After the feasibility tests, an ultrasound flow phantom was fabricated to conduct ultrasound B-mode visualization and Doppler-PIV comparison. DISCUSSION: The results revealed that the PIV measured through PVA-H material exhibited 0.8% error in the measured maximum velocity compared with PIV through the acrylic material. B-mode images are similar to real tissue visualization with a limitation of a higher sound velocity, when compared with human tissue, of 1792 m/s. Doppler measurement of the phantom revealed approximately 120% and 19% overestimation of maximum and mean velocities, respectively, compared with those from PIV. CONCLUSION: The proposed material possesses the advantage of the single-phantom ability to improve the ultrasound flow phantom for validation of flow.

    PURPOSE: Tissue elasticity can be measured and mapped using color Doppler elastography. In a previous study, a binary pattern of shear waves was observed using a color flow imaging (CFI) system with matched pulse Doppler packet size as well as shear wave frequency and displacement condition. In the present study, we demonstrate the possibility of mapping shear wave velocity and resolving phantom elasticity using any commercial ultrasound machine without fulfilling that condition. METHODS: We derive a relation between Doppler autocorrelator integration time and the estimated flow velocity. The underlying principles behind the shear wave shadows captured by a typical modern ultrasound machine are investigated. The ultrasound machine measurement preset is calibrated to remove the effect of transducer array scanning delay in modifying the appearing wavenumber and thus correct the measurement error. RESULTS: The method was used to successfully measure the elasticity of a biological tissue-mimicking phantom and distinguish a stiff phantom from a soft phantom. CONCLUSION: Using this method, the elasticity of a biological tissue-mimicking phantom can be recovered with less strict constraint. As a result, it provides more flexibility to be implemented in common ultrasound machines. This method may be practically used to help identify tissue stiffness-related disease.

    Some studies suggested a correlation between tissue elasticity and diseases, such as Adhesive Capsulitis (AC) of the shoulder. One category of method to measure elasticity is by utilizing Doppler imaging. This paper discusses color Doppler shear wave elastography methods and demonstrated an experiment with biological tissue mimicking phantom. A simulation with binary pattern color Doppler shear wave elastography shows that wavelength of a shear wave with suggested magnitude is equal to four multiple of pitch strip in a color flow image. However, the larger amplitude changes the duty ratio and frequency of the pattern. An experiment with biological tissue mimicking Polyvinyl Alcohol (PVA) phantoms has shown that the binary pattern color Doppler method has successfully recovered shear wave velocity map and calculate the elasticity.Clinical Relevance-The result of experiments presents a possibility of using the method for quantitatively access the stage of tissue stiffness related disease.

    <title>Abstract</title> Visualization of cutaneous micro-vasculatures is a powerful approach assisting in the diagnosis of skin vascular disorders. These minute structures can be visualized by high-frequency ultrasound (HFUS) using ultrafast Doppler imaging. Ultrasound flow phantoms have been used as assessment tools to evaluate the performance of the ultrasound imaging system, however, to optimize the imaging system for visualization of micro-structures, flow phantom with micro-channels is required which are usually difficult to fabricate. Here, we design a simple approach for micro-flow phantom which is easy to fabricate and cast for detection of micro-circulation in superficial micro-structures. The proposed approach features (i) the micro-channels of 200-micron at the depth of 4 mm (ii) casted in the cryogel mixture of Poly-vinyl alcohol (PVA) and (iii) infused at flow speed of 30 mm/s using infusion pump. Visualization of micro-flow channel in power Doppler image obtained by HFUS ultrafast Doppler imaging reveals that the proposed micro-flow phantom could serve as a viable assessment tool for optimizing the system for in-vivo cutaneous micro-vasculature imaging.

    <title>Abstract</title> A flow velocity or tissue motion can be measured by pulse Doppler method. However, when the method is applied to a sinusoid motion or harmonic vibration, the estimation result is deviated. In this paper, the generalized theory of pulse Doppler measurement to the sinusoid motion is explained and the relation between the estimation deviation and the real value is given. Furthermore, a special case of binary velocity estimation is expanded and simulated with different parameters. This simulation result suggested that different selection of parameters may fundamentally affect the output of pulse Doppler estimation.

    <title>Abstract</title> Detecting lung tumors in early stage by reading chest X-ray images is important for radical treatments of the disease. In order to decrease the risk of missed lung tumors, diagnosis support systems that can provide the accurate detection of lung tumors are in high demand, and the use of artificial intelligence with deep learning is one of the promising solutions. In our research, we aim to improve the accuracy of a deep learning-based system for detecting lung tumors by developing a bone suppression algorithm as a preprocessing for the machine-learning model. Our bone suppression algorithm was devised for conventional single-shot chest X-ray images, which do not rely on a specific type of imaging systems. 604 chest X-ray images were processed using the proposed algorithm and evaluated by combining it with a U-net deep learning model. The results showed that the bone suppression algorithm successfully improved the performance of the deep learning model to identify the location of lung tumors (Intersection over Union) from 0.085 (without the bone suppression algorithm) to 0.142, as well as the ability to classify the lung cancer (Area under Curve) that increased from 0.700 to 0.736. The bone suppression algorithm would be useful to improve the accuracy and the reliability of the deep learning-based diagnosis support systems for detecting lung cancer in mass medical examinations.

    A designer enzyme consisting of an abiological molecule incorporated into a natural protein has been developed as an exceptionally chemoselective catalyst, highlighting that the internal space of proteins is highly beneficial for enhancing catalytic performance. However, other superior features of proteins have received less attention in designer enzymes: e.g., their use as ligands to construct abiological (multinuclear) metal centers and an intrinsic protein function that has often been traded off for a new function. Here, grafting a synthetic trinuclear zinc complex inside a human cytokine macrophage migration inhibitory factor (MIF) scaffold using solely amino-acid side chains led to a designer multi-metalloenzyme with extrinsic and intrinsic functions. The crystal structure of the designer tri-zinc enzyme verified the high accuracy of our design process based on geometry optimizations and quantum-chemical calculations. The extrinsic catalytic performance of this designer enzyme is of the highest class and comparable to that of previously reported designer zinc hydrolases. Importantly, an intrinsic function of MIF, i.e., its tautomerase activity, was maintained in this designer tri-zinc enzyme. Considering that cytokines are originally expressed in response to in vivo events, this cytokine-based designer metalloenzyme shows promising potential as a synthetic biological tool for the self-adaptive regulation of life phenomena.

    Introduction: Trans-urethral incision (TUI) is carried out as a surgical treatment for Bladder Outlet Obstruction (BOO). In the clinical practice, there are empirical patterns of the direction and the depth of incision, because mechanism of the surgical outcome to the intra-urethral flow has not been cleared. In this study, we designed models of bladder outlet and urethra and simulated the internal urine flow to study relationship between surgical incision of bladder outlet and urine flow dynamics. Method: Based on anatomical characteristic and cysto-urethroscopic image, a model of intra-urethral shape with BOO (BOO model) and the post-surgical models, incised at 6 o&#039;clock or 5 and 7 o&#039;clock direction of the bladder outlet (6 o&#039;clock model and 5 - 7 o&#039;clock model), were generated. Then the intra-urethral flow through the models was visualized and transition of fluid energy was calculated. Results: Two incise direction methods had different property in the rapid flow distribution and the occurrence of vortex, as well as the amount of energy loss thorough the urethra. Conclusion: Internal flow dynamics changes and properties by surgical incision were indicated with urine flow simulation of simplified urethral models.

    As evaluation of disease progression or prediction of prognosis of patients with Polycystic kidney disease (PKD), measurement of total kidney volume (TKV) have widely been accepted clinically. To establish automatic TKV measuring method with accuracy in PKD, we developed a segmentation algorithm of renal region. The coronal section of MRI T1 weighted images were used for the present study to capture the irregular boundary of polycystic kidney which often contacts with liver due to nephromegaly. Since biphasic luminance value of cysts and renal parenchyma was distributed, the GraphCut algorithm was utilized to discriminate renal region from the other organs. Then, the likelihood of pixels in foreground and background were calculated using Gaussian Mixture Model (GMM), and the GraphCut was performed. Segmentation results were evaluated by precision and recall, and compared with the method of the previous studies applying the region growing method and the level set method. The proposed procedure indicated high recall and precision compared to the previous studies.

    Among patients with prostate cancer, the method to detect fine cancerous lesions has been required for focal therapy to maintain QOL after therapy. In this study, we extracted cancerous and noncancerous regions from a paraffin embedded prostatic specimen of patients with prostatic cancer. The acoustic properties of prostatic tissue were measured using ultrasonic microscope. The significant differences in acoustic properties between cancerous and noncancerous regions were observed. Though the research requires further experiments for confirmation of acoustic properties in different pathological specimens, detection of fine cancerous lesion by ultrasound would be promising by interpreting the results in the lower frequency of ultrasound.

    In this paper, we propose a novel method to reconstruct a 3D-model of the luminal cavity, which conformation is calculated with the luminance value of the endoscopic video. It will make up for shortcomings of endoscopic imagery, including lack of 3D-vision and narrow field of vision.

    Memo-dynamics relating to musical recognition process in human brain were investigated by utilizing a one-channel near infrared spectroscopy (NIRS) method. During active and passive listening music, an optical absorption at 780nm-830nm in frontal- and temporal lobes were measured. The obtained time-course of parameters which associated with a cerebral blood flow showed a specific blood level pattern depending on each of musical tunes.     本研究では尿路内の形態と流れや症状との関係をより明らかにすることと,従来では困難であった尿道壁面の動き情報を数値計算に適用することを目的とし,主に【課題1】狭窄した尿路に対する様々な切開方向や深さによる流れへの影響を検討することと,【課題2】超音波診断装置による排尿時の尿道内腔の形状変化を抽出するための基礎検討を実施した. 課題1については,従来膀胱頸部硬化症などに対して実施されるTrans-Urethral Incision (TUI)という治療に則り,狭窄を模した尿道形状Computer Graphics(CG)モデルに6時方向や5,7時方向の切開を様々な深さで加え,内腔の流れをシミュレーションすることにより,切開による形態変化と症状改善との関係性を調べた.膀胱収縮力や許容される切開量によって,至適な切開方向が存在することが示唆されたが,今後膀胱機能を含めた数値計算による更なる検討が必要であると考えられた. また,課題2については,3次元超音波診断装置により下部尿路の排尿時の形態変化を計測することを前提に,排尿の超音波による観察に適したプローブ配置や撮像パラメータを明らかにする準備として,下部尿路周辺の音響的/構造的に生体等価性を持つファントムの開発に取り組んだ.下部尿路展開時の尿道内腔形状の型を作り,凍結融解法を用いたPolyvinyl alcohol (PVA) Cryogelで成型した(以下,管腔ファントム).この管腔ファントムを内腔を陰圧にして閉塞させた状態で,周囲をゼラチンで固めることにより,膀胱端側の圧力により閉塞と解放を繰り返す動的な尿道ファントムを構築した.超音波計測による動的な形態情報とカテーテルによる膀胱内圧測定を組み合わせることにより,より詳細な尿道内腔における流れの可視化が可能となり,高精度な診断や局所療法支援へと発展することが期待される.

    本研究では、前立腺部尿道の前立腺部尿道における蓄排尿機能障害の評価法として、従来困難であった尿道内部、特に蓄排尿障害の原因疾患である、膀胱頚部や前立腺部における粘弾性や形状の性状変化が流体に与える影響を数値化することを目的として、患者の尿道形状取得と、尿道の尿輸送機能評価法の確立に取り組んできた。本年度においては、下部尿路形状構築における構築精度向上のためのアルゴリズムの開発と、臨床データを用いた提案手法における臨床的有用性についての検討を実施した。 本手法における形状モデルの精度向上には、内視鏡画像における同一臓器の色彩情報を、臓器表面の血流量によって分別することが必要である。内視鏡から収集された下部尿路の色彩情報の分布をHSB色空間上に表現し、最尤法を用いて複合ガウス分布としてフィッティングすることで、収集した色情報を、低血流部位、高血流部位、非尿道臓器の3分布に分割できる手法を開発した。 次に、臨床データを用いた、本研究の臨床的有用性について検討した。提案手法によって構築した、膀胱尿道鏡映像を基にした下部尿路の形状モデルを用いて、有限体積法による定常流解析を行い、尿道内における流れを可視化した。前立腺部尿道における圧力損失を、本手法における排尿症状の指標とし、従来の臨床検査におけるデータとの関連性を検討した。その結果、圧力損失量は前立腺体積に強い相関を示し、本研究が患者の尿道内腔の正常を適切に再現していることが示唆された。また、排尿症状の改善薬剤の投与により、臨床データと同様に、提案手法による圧力損失も改善傾向を示すことも確認された。また、本手法では、尿道内における詳細な流れの振る舞いを可視化できることから、尿道内における排尿症状の責任部位を同定し、より繊細な治療法を可能にすることが示唆された。今後、医学研究として大規模な症例数における本手法の評価や治療への応用を目指した検討が実施される見込みであり、現在、臨床現場での使用を想定したソフトウェアの開発を実施している。
