    Social behavior is essential for health, survival, and reproduction of animals; however, the role of astrocytes in social behavior remains largely unknown. The transmembrane protein CD38, which acts both as a receptor and ADP-ribosyl cyclase to produce cyclic ADP-ribose (cADPR) regulates social behaviors by promoting oxytocin release from hypothalamic neurons. CD38 is also abundantly expressed in astrocytes in the postnatal brain and is important for astroglial development. Here, we demonstrate that the astroglial-expressed CD38 plays an important role in social behavior during development. Selective deletion of CD38 in postnatal astrocytes, but not in adult astrocytes, impairs social memory without any other behavioral abnormalities. Morphological analysis shows that depletion of astroglial CD38 in the postnatal brain interferes with synapse formation in the medial prefrontal cortex (mPFC) and hippocampus. Moreover, astroglial CD38 expression promotes synaptogenesis of excitatory neurons by increasing the level of extracellular SPARCL1 (also known as Hevin), a synaptogenic protein. The release of SPARCL1 from astrocytes is regulated by CD38/cADPR/calcium signaling. These data demonstrate a novel developmental role of astrocytes in neural circuit formation and regulation of social behavior in adults.

    Primary aldosteronism (PA) is considered the most common form of secondary hypertension, which is associated with excessive aldosterone secretion in the adrenal cortex. The cause of excessive aldosterone secretion is the induction of aldosterone synthase gene (CYP11B2) expression by depolarization of adrenocortical cells. In this study, we found that YM750, an Acyl-coenzyme A: cholesterol acyltransferase (ACAT) inhibitor, acts on adrenocortical cells to suppress CYP11B2 gene expression and aldosterone secretion. YM750 inhibited the induction of CYP11B2 gene expression by KCl stimulation, but not by angiotensin II and forskolin stimulation. Interestingly, YM750 did not inhibit KCl-stimulated depolarization via an increase in intracellular calcium ion concentration. Moreover, ACAT1 expression was relatively abundant in the zona glomerulosa (ZG) including these CYP11B2-positive cells. Thus, YM750 suppresses CYP11B2 gene expression by suppressing intracellular signaling activated by depolarization. In addition, ACAT1 was suggested to play an important role in steroidogenesis in the ZG. YM750 suppresses CYP11B2 gene expression and aldosterone secretion in the adrenal cortex, suggesting that it may be a potential therapeutic agent for PA.

    TWIK-related acid-sensitive K+ (TASK) channels are thought to contribute to the resting membrane potential in adrenal cortical (AC) cells. However, the molecular identity of TASK channels in AC cells have not yet been elucidated. Thus, immunocytochemical and molecular biological approaches were employed to investigate the expression and intracellular distribution of TASK1 and TASK3 in mouse AC cells and H295R cells derived from human adrenocortical carcinoma. Immunocytochemical study revealed that immunoreactive materials were mainly located in the cytoplasm for TASK1 and at the cell periphery for TASK3 in mouse AC cells. A similar pattern of localization was observed when GFP-TASK1 and GFP-TASK3 were exogenously expressed in H295R cells. In addition, p11 that is known to suppress the endoplasmic reticulum exit of TASK1 was localized in the cytoplasm in mouse AC and H295R cells, but not in adrenal medullary cells. Proximity ligation assay (PLA) suggested formation of heteromeric TASK1-3 channels that were found predominantly in the cytoplasm and weakly at the cell periphery. A similar distribution was observed following exogenous expression of tandem TASK1-3 channels in H295R cells. When stimulated by angiotensin II, however, tandem TASK1-3 channels were present mainly in the cytoplasm in all H295R cells. In contrast to that in H295R cells, tandem channels were exclusively located at the cell periphery in all non-stimulated and exclusively in the cytoplasm in stimulated PC12 cells, respectively. From these results, we conclude that TASK1 proteins are present mainly in the cytoplasm and minimally at the cell periphery as a heteromeric channel with TASK3, whereas the majority of TASK3 is at the cell periphery as homomeric and heteromeric channels.

    Hepatocyte nuclear factor 4α (HNF4α) has essential roles in controlling the expression of a variety of genes involved in key metabolic pathways, including gluconeogenesis in the liver. The mechanistic and physiological significance of peroxisome proliferator-activated receptor gamma co-activator-1α (PGC-1α) for HNF4α-mediated transcriptional activation models for gluconeogenic genes is well characterized. However, the transcriptional repression of HNF4α for those genes remains to be examined. In this study, we applied novel proteomic techniques to evaluate the interactions of HNF4α, including those with biochemically labile binding proteins. Based upon our experiments, we identified interferon regulatory factor 2 binding protein 2 (IRF2BP2) as a novel HNF4α co-repressor. This interaction could not be detected by conventional immunoprecipitation. IRF2BP2 repressed the transcriptional activity of HNF4α dependent on its E3 ubiquitin ligase activity. Deficiency of the IRF2BP2 gene in HepG2 cells induced gluconeogenic genes comparable to that of forskolin-treated wild-type HepG2 cells. Together, these results suggest that IRF2BP2 represents a novel class of nuclear receptor co-regulator.

    Glucose is not only the energy fuel for most cells, but also the signaling molecule which affects gene expression via carbohydrate response element binding protein (ChREBP), a Mondo family transcription factor. In response to high glucose conditions, ChREBP regulates glycolytic and lipogenic genes by binding to carbohydrate response elements (ChoRE) in the regulatory region of its target genes, thus elucidating the role of ChREBP for converting excessively ingested carbohydrates to fatty acids as an energy storage in lipogenic tissues such as the liver and adipose tissue. While the pathophysiological roles of ChREBP for fatty liver and obesity in these tissues are well known, much of the physiological and pathophysiological roles of ChREBP in other tissues such as the kidney remains unclear despite its high levels of expression in them. This review will thus highlight the roles of ChREBP in the kidney and briefly introduce the latest research results that have been reported so far.

    Carney complex (CNC) is a rare hereditary syndrome that involves endocrine dysfunction and the development of various types of tumors. Chromosome 2p16 and PRKAR1A on chromosome 17 are known susceptibility loci for CNC. Here we report a mother and son with CNC caused by an 8.57-kb deletion involving the transcription start site and non-coding exon 1 of PRKAR1A. The proband is a 28-year-old male with bilateral large-cell calcified Sertoli cell testicular tumors and pituitary adenoma. Comprehensive genomic profiling for cancer mutations using Foundation One CDx failed to detect any mutations in PRKAR1A in DNA from the testicular tumor. Single-nucleotide polymorphism array analysis of the proband's genomic DNA revealed a large deletion in the 5' region of PRKAR1A. Genomic walking further delineated the region an 8.57-kb deletion. A 1.68-kb DNA fragment encompassed by the deleted region showed strong promoter activity in a NanoLuc luciferase reporter assay. The patient's mother, who is suffering from recurrent cardiac myxoma, a critical sign for CNC, carried an identical deletion. The 8.57-kb deleted region is a novel lesion for CNC and will facilitate molecular diagnosis of the disease.

    The androgen receptor (AR) plays an essential role in the development of prostate cancer, and androgen-deprivation therapy is used as a first-line treatment for prostate cancer. However, under androgen-deprivation therapy, castration-resistant prostate cancer inevitably arises, suggesting that the interacting transcriptional coregulators of AR are promising targets for developing novel therapeutics. In this study, we used novel proteomic techniques to evaluate the AR interactome, including biochemically labile binding proteins, which might go undetected by conventional purification methods. Using rapid immunoprecipitation mass spectrometry of endogenous proteins, we identified enhanced at puberty 1 (EAP1) as a novel AR coregulator, whereas its interaction with AR could not be detected under standard biochemical conditions. EAP1 enhanced the transcriptional activity of AR via the E3 ubiquitin ligase activity, and its ubiquitination substrate proteins included AR and HDAC1. Furthermore, in prostate cancer specimens, EAP1 expression was significantly correlated with AR expression as well as a poor prognosis of prostate cancer. Together, these results suggest that EAP1 is a novel AR coregulator that promotes AR activity and potentially plays a role in prostate cancer progression.

    The adrenal cortex produces steroid hormones as adrenocortical hormones in the body, secreting mineralocorticoids, glucocorticoids, and adrenal androgens, which are all considered essential for life. Adrenocortical tumors harbor divergent hormonal activity, frequently with steroid excess, and disrupt homeostasis of the body. Aldosterone-producing adenomas (APAs) cause primary aldosteronism (PA), and cortisol-producing adenomas (CPAs) are the primary cause of Cushing’s syndrome. In addition, adrenocortical carcinoma (ACC) is a highly malignant cancer harboring poor prognosis. Various genetic abnormalities have been reported, which are associated with possible pathogenesis by the alteration of intracellular signaling and activation of transcription factors. In particular, somatic mutations in APAs have been detected in genes encoding membrane proteins, especially ion channels, resulting in hypersecretion of aldosterone due to activation of intracellular calcium signaling. In addition, somatic mutations have been detected in those encoding cAMP-PKA signaling-related factors, resulting in hypersecretion of cortisol due to its driven status in CPAs. In ACC, mutations in tumor suppressor genes and Wnt-β-catenin signaling-related factors have been implicated in its pathogenesis. In this article, we review recent findings on the genetic characteristics and regulation of intracellular signaling and transcription factors in individual tumors.

    Cancer stem cells (CSCs) are believed to cause cancer metastasis and recurrence. BEX2 (brain expressed X-linked gene 2) is a CSC-related gene that is expressed in dormant CSCs in cholangiocarcinoma and induces resistance against chemotherapy. The aim of the present study was to identify small compounds that have activity to inhibit BEX2 expression and result in the attenuation of CSC-related phenotypes. We screened 9600 small chemical compounds in high-throughput screening using cholangiocarcinoma cell line HuCCT1 expressing BEX2 protein fused with NanoLuc, and identified a compound, BMPP (1, 3-Benzenediol, [4-(4-methoxyphenyl)-1H-pyrazol-3-yl]). BMPP was found to exert decreasing effects on BEX2 protein expression and G0 phase population of the tumor cells, and increasing effects on ATP levels and chemotherapeutic sensitivity of the cells. These findings indicate that BMPP is a valuable chemical compound for reducing dormant CSC-related phenotypes. Thus, the identification of BMPP as a potential CSC suppressor provides scope for the development of novel therapeutic modalities for the treatment of cancers with BEX2 overexpressing CSCs.

    Aldosterone is synthesized in the adrenal by the aldosterone synthase CYP11B2. Although the control of CYP11B2 expression is important to maintain the mineral homeostasis, its overexpression induced by the depolarization-induced calcium (Ca2+) signaling activation has been reported to increase the synthesis of aldosterone in primary aldosteronism (PA). The drug against PA focused on the suppression of CYP11B2 expression has not yet been developed, since the molecular mechanism of CYP11B2 transcriptional regulation activated via Ca2+ signaling remains unclear. To address the issue, we attempted to reveal the mechanism of the transcriptional regulation of CYP11B2 using chemical screening. We generated a cell line by inserting Nanoluc gene as a reporter into CYP11B2 locus in H295R adrenocortical cells using the CRSPR/Cas9 system, and established the high-throughput screening system using the cell line. We then identified 9 compounds that inhibited the CYP11B2 expression induced by potassium-mediated depolarization from the validated compound library (3399 compounds). Particularly, tacrolimus, an inhibitor of phosphatase calcineurin, strongly suppressed the CYP11B2 expression even at 10 nM. These results suggest that the system is effective in identifying drugs that suppress the depolarization-induced CYP11B2 expression. Our screening system may therefore be a useful tool for the development of novel medicines against PA.

    Angiotensin II (Ang II) is a well-known peptide that maintains the balance of electrolytes in the higher vertebrates. Ang II stimulation in the adrenal gland induces the synthesis of mineralocorticoids, mainly aldosterone, through the up-regulation of aldosterone synthase (CYP11B2) gene expression. Additionally, it has been reported that Ang II activates multiple signaling pathways such as mitogen-activated protein kinase (MAPK) and Ca2+ signaling. Although Ang II has various effects on the cellular signaling in the adrenal cells, its biological significance, except for the aldosterone synthesis, is still unclear. In this study, we attempted to search the novel target gene(s) of Ang II in the human adrenal H295R cells using a proteomic approach combined with stable isotopic labeling using amino acid in cell culture (SILAC). Interestingly, we found that Ang II stimulation elevated the expression of phosphofructokinase type platelet (PFKP) in both protein and mRNA levels. Moreover, transactivation of PFKP by Ang II was dependent on extracellular-signal-regulated kinase (ERK) 1/2 activation. Finally, we observed that Ang II treatment facilitated glucose uptake in the H295R cells. Taken together, we here identified PFKP as a novel target gene of Ang II, indicating that Ang II not only stimulates steroidogenesis but also affects glucose metabolism.

    <title>Abstract</title> Fatty acid binding protein 7 (FABP7) is an intracellular fatty acid chaperon that is highly expressed in astrocytes, oligodendrocyte-precursor cells, and malignant glioma. Previously, we reported that FABP7 regulates the response to extracellular stimuli by controlling the expression of caveolin-1, an important component of lipid raft. Here, we explored the detailed mechanisms underlying FABP7 regulation of caveolin-1 expression using primary cultured FABP7-KO astrocytes as a model of loss of function and NIH-3T3 cells as a model of gain of function. We discovered that FABP7 interacts with ATP-citrate lyase (ACLY) and is important for acetyl-CoA metabolism in the nucleus. This interaction leads to epigenetic regulation of several genes, including caveolin-1. Our novel findings suggest that FABP7-ACLY modulation of nuclear acetyl-CoA has more influence on histone acetylation than cytoplasmic acetyl-CoA. The changes to histone structure may modify caveolae-related cell activity in astrocytes and tumors, including malignant glioma.

    Background and Objective: Obesity is highly complicated by hypertension and hyperglycemia.In particular, it has been proposed that obesity-related hypertension is caused by adipocyte-derivedfactors that are recognized as undetermined proteins secreted from adipocytes. Adipocyte-derived factorshave been known to be related to aldosterone secretion in the adrenal gland. So far, Wnt proteins,CTRP-1, VLDL, LDL, HDL and leptin have been demonstrated to stimulate aldosterone secretion. Incontrast, it has not yet been clarified whether adipocyte-derived factors also affect adrenal cortisol secretion. Methods and Results: In the present study, we investigated the effect of adipocyte-derived factors oncortisol synthase gene CYP11B1 mRNA expression in vitro study using adrenocortical carcinomaH295R cells and mouse fibroblast 3T3-L1cells. Interestingly, adipocyte-derived factors were demonstratedto have the ability to stimulate CYP11B1 mRNA expression. Conclusion: Since CYP11B1 is well known as a limiting enzyme of cortisol synthesis, our study suggeststhat adipocyte-derived factors may stimulate cortisol secretion, as well as aldosterone secretion.Taken together, adipocyte-derived factors may be the cause of metabolic syndrome due to their stimulatingeffects on aldosterone/cortisol secretion. Therefore, the innovation of novel drugs against themmay possibly be a new approach against metabolic syndrome.

    Glucocorticoids and glucocorticoid receptors (GRs) suppress pituitary pro-opiomelanocortin (Pomc) gene expression. O-linked β-N-acetylglucosamine (O-GlcNAc) modification, mediated by O-GlcNAc transferase (OGT), plays an important role during gene transcription. However, whether OGT is involved in the GR-mediated transrepression that occurs in pituitary corticotroph cells is currently unknown. Here, we report that OGT regulates Pomc expression in the mouse corticotroph cell line AtT-20. The overexpression of OGT has an additive effect on the GR-mediated negative transcription pathway. Both the knockdown of OGT by RNA interference and the use of a chemical OGT inhibitor abolished the repressive effects of Pomc expression induced by GRs. OGT inhibition leads to both the decreased recruitment of GRs and the increased recruitment of RNA polymerase II to the Pomc locus. O-GlcNAc modification is involved in the negative regulation of Pomc transcription in corticotroph cells. OGT may be a promising therapeutic target for the treatment of Cushing's disease.

    CD38 is an enzyme that catalyzes the synthesis of cyclic adenosine diphosphate-ribose from nicotinamide adenine dinucleotide (NAD+). We recently reported that this molecule regulates the maturation and differentiation of glial cells such as astrocytes and oligodendrocytes (OLs) in the developing brain. To analyze its role in the demyelinating situation, we employed cuprizone (CPZ)-induced demyelination model in mice, which is characterized by oligodendrocyte-specific apoptosis, followed by the strong glial activation, demyelination, and repopulation of OLs. By using this model, we found that CD38 was upregulated in both astrocytes and microglia after CPZ administration. Experiments using wild-type and CD38 knockout (KO) mice, together with those using cultured glial cells, revealed that CD38 deficiency did not affect the initial decrease of the number of OLs, while it attenuated CPZ-induced demyelination, and neurodegeneration. Importantly, the clearance of the degraded myelin and oligodendrocyte repopulation were also reduced in CD38 KO mice. Further experiments revealed that these observations were associated with reduced levels of glial activation and inflammatory responses including phagocytosis, most likely through the enhanced level of NAD+ in CD38-deleted condition. Our results suggest that CD38 and NAD+ in the glial cells play a critical role in the demyelination and subsequent oligodendrocyte remodeling through the modulation of glial activity and neuroinflammation.

    A complementary study to our former work is performed about screening of the patient with hyperthyroidism using routine test data. The same calculation is done, however, adopting 17 routine tests in addition to former 14 tests. The result adopting 4 routine tests such as ALP, AST, ALT and T-Cho which thyroid specialist often consider as important to suspect hyperthyroidism, is evaluated. The importance of S-Cr to make screening the patients with hyperthyroidism is stressed.

    The CRF-test is quite useful in evaluating hypothalamic-pituitary-adrenal (HPA) function and used widely in clinical practice. Combined administration of CRF, TRH, LHRH, and GRF has been known to be feasible for assessing anterior pituitary function. Since the rapid ACTH-test is still a necessary adjunct for evaluation of adrenal function, we sought to examine whether simultaneous administration of ACTH had any significant effect on other anterior pituitary function tests, i.e., TRH-, LHRH-, and GRF-tests.<BR>Provocative tests were carried out on 15 healthy volunteers using multiple hormones in the following combinations:(1) ACTH, GRF, TRH, LHRH;(2) GRF, TRH, LHRH;(3) TRH, LHRH; or (4) ACTH alone. After 30 min bed rest in the morning, blood was drawn for measurement of baseline hormone levels, then drugs were administered iv and blood was drawn after 15, 30, 45, 60, 90, and 120 min. Serum levels of anterior pituitary hormones and cortisol were measured by specific enzyme immunoassays.<BR>ACTH administration in combination with GRF, TRH, and LHRH evoked significantly greater GH responses compared with combined administration of GRF, TRH, and LHRH. However, ACTH in combination with three other hormones had no significant effects on TSH, PRL, LH, or FSH responses. GRF administration in combination with TRH and LHRH did not affect significantly TSH, PRL, LH, or FSH responses compared with combined administration of TRH and LHRH. Finally, cortisol response following ACTH administration was not affected significantly by simultaneous administration of GRF, TRH, and LHRH. From these results it is proposed that combined administration of ACTH, TRH, and LHRH may be used as a single test. Combined administration of GRF, TRH, and LHRH may also be used as a single test. However, ACTH administration in combination with GRF may affect GH responses, and therefore, these two agents should be administered separately.

    我々は、当該年度の下記の成果を得た。 1)D532の標的遺伝子の解析:我々は野生型マウス群、1型糖尿病性腎症モデルマウスであるiNOS-トランスジェニック(TG)マウス+Vehicle群、iNOS-TGマウス+D532群の各腎臓から抽出したRNAを用いて、RNA-Sequenceを施行した。その結果、Lpk、Fis1、Kcnb1、Dguok、Slc25a15といったChREBPの標的遺伝子に加えて、Txnipも野生型マウス群に比してiNOS-TGマウス群でその発現が著明に増加していた一方で、iNOS-TGマウス+D532群では発現が顕著に減少していることが明らかとなった。また、GO enrichment analysisでも細胞内シグナル伝達経路やアポトーシス経路の有意な変動が観察された。以上の結果から、TxnipがD532の下流に存在することが確認された。現在、更なるパスウェイ解析を行うことにより、D532作用時の腎臓におけるTxnipの上流遺伝子を検索中である。2)CRISPR-Cas9システムによるChREBP KOマウスの作成:ChREBPのDNA結合ドメインであるヘリックス-ループ-ヘリックス構造(Exon 13~14内)を含むExon 12からExon 14を認識するcrRNAを作製し、マウス受精卵(B6N×B6N)に対してエレクトロポレーション法によりCas9 protein 100 ng/μL、100 ng/μL crRNAs、100 ng/μL tracrRNAを導入した。作製した受精卵は偽妊娠雌マウスの卵管に導入し、変異マウスは胎生19日目に出生した。得られたChREBP KOマウス腎臓でのChREBP欠損はWestern blotによって確認された。

    我々は当該年度に以下の実験を遂行した。 1)糖尿病性腎症(DN)の発症・増悪におけるChREBPの関与・作用機構の解明:A) CRISPR/Cas9システムを用いて作成したChREBPノックアウトマウスからRNAを抽出してRNAシークエンスに供する準備を進めている。B)東北大学医学部倫理委員会にて承認を得たことから、東北大学病院 腎・高血圧・内分泌科の腎生検組織からのRNA抽出・RNAシークエンスに向けて、DNならびに非DN検体の準備を進めている。 2)D532のDN改善効果の機序解明および実用化・臨床応用に向けた構造最適化:A)D532の一端を伸長して末端をビオチン化することにより作成したD532プローブを用いて、アビジン‐ビオチン反応を利用したアフィニティ精製・質量分析にてD532の標的因子の同定を進めた結果、ChREBP自身がD532の標的因子であることが明らかとなった。B)ChIPアッセイの結果、D532がChREBPのDNAへの結合を抑制することが認められた。本結果から、D532が高血糖依存性のChREBP転写活性をDNA結合レベルで抑制することが明らかとなった。C)D532はChREBPの核内移行には影響を及ぼさないことが明らかとなった。D)疎水性であるD532をPMB30Wにてミセル化することにより水溶化が出来ることが明らかとなった。本成果により、今後のD532を用いた非臨床試験・臨床試験に向けて大きく前進できるものと期待される。

    Using a type 1 diabetic nephropathy model (iNOS/RAGE double TG mouse), we aimed to develope a new drug which regulate the expression of mineral corticoid receptor's co-repressor and test the depression effect of diabetic nephropathy and a utility of the combination with the existing MR antagonist. But the creation of the GEMIN4-Luc stable cell line was technically difficult, so we make ChoRE-Luc stable cell line in parallel, it was ssuccessful.Using these cell line, we proceeded high-throughput screening (HTS) using an onset of and finally two hit compounds are obtained. We delivered these compounds to iNOS transgenic (TG) mouse and examined the albumin urine output and compared pharmacological effect with other drugs.

    In this research projecct, we focused on ChREBP, a DNA-binding transcription factor which is believed to be involved in diabetic nephropathy pathogenesis. To understand ChREBP-mediated transcriptional regulation, we tried to identify ChREBP-containing protein complex using renal mesangial cells. In this attempt, we identified several coregulator candidates for ChREBP and, we also identified cross-talk mechanisms between ChREBP and inflammation signals. To developing drugs targeting these factors might be promising strategy for diabetic nephropathy and other metabolic syndromes.

    PA024 dose-dependently suppressed AII-induced CYP11B2 mRNA expression/promoter activity. Additionally, high-dose PA024 significantly decreased aldosterone secretion. The suppression of CYP11B2 promoter activity by PA024 was observed in the deletion mutants analyses using the region from -1521 (full length) to -135 including NBRE-1, Ad4, and Ad5 elements. PA024 also suppressed mRNA expression of transcription factors Nurr1 and NGFIB in a dose-dependent manner. Since PA024-mediated suppression of CYP11B2 promoter activity was rescued by Nurr1 overexpression, decrease of Nurr1 expression may contribute to the suppression. PA024 also inhibited mRNA expression of StAR, HSD3β2, and CYP21A2. Therefore, PA024-mediated suppression of these enzymes may also be involved in the inhibition of aldosterone synthesis. Furthermore, PA024 significantly lowered systemic blood pressure in THM.

    EXTL3 functions as a glycosyltransferase involved in heparan sulfate proteoglycans biosynthesis. We generated pancreatic β-cell specific Extl3 knockout (βExtl3KO) mice. In βExtl3KO mice at 2-8 weeks old, the β-cell proliferation activity was decreased and histological examination of pancreatic islets revealed an atypical organization. βExtl3KO mice at 8 weeks old showed glucose intolerance coupled to impaired insulin secretion upon glucose challenge. These data indicate that heparan sulfate plays important role(s) in β-cell proliferation and the mechanism of insulin secretion.

    膵β細胞の増殖因子として発見されたREG (Regenerating gene)は、REG Iα、 REG Iβ、 REG III、 HIP/PAP、 REG IVの5つの遺伝子ファミリーから構成されている。近年、種々の消化器がんにおいてREG Iα遺伝子が発現していること、さらにはREG Iα遺伝子の発現が患者の予後と負の相関関係を示すことが明らかとされつつある。本研究の目的は、各REGタンパク質に対するモノクローナル抗体を作成し、それらを用いて種々の消化器がんにおける各REGの発現を検討することにより、それらの発現とがん患者の生命予後との関係を明らかにすることである。本年度は、以下の成果が得られた。 1)昨年度に精製まで完了したREG Iα、 REG IβおよびREG IVに対するモノクローナル抗体に加え、本年度ではREG IIIおよびHIP/PAPに対するモノクローナル抗体の精製が終了した。 2)各REG遺伝子を過剰発現させた培養細胞系の上清を精製することにより、非常に純度の高い(〜100%)各REGタンパク質が得られた。 3)2)で得られた各REGタンパク質を抗原に用いて、ウエスタンブロット法を施行した。その結果、1)で得られた各モノクローナル抗体の特異性が確認された。 以上の結果により、各REGタンパク質の発現を特異的に認識することが可能となった。現在、各モノクローナル抗体を用いた免疫組織染色の条件設定を行っている。

    Study on ATRA-resistance caused by mutant PML-RAR α ; In this study, we examined the cellular and molecular RA-response of a human APL cell line, UF-1, established from a patient who had ATRA-resistance and a mutation in the PML-RAR α chimeric protein. For cellular RA-response, differentiation and apoptosis was evaluated in UF-1 cells treated with retinoids. For molecular RA-response, RA-dependent transcriptional activity of PML-RARa chimeric protein and RT-PCR of PML-RAR α, RAR α, RAR β, and RAR γ was determined. In response to ATRA and Am80 at 100 nM or lower doses, neither cellular nor molecular RA-response was detected. By contrast, in response to 1 □M ATRA and Am80, UF-1 showed a full-scaled maturation with apoptosis, whereas molecular RA-response of the mutant PML-RARa was insufficient. These findings suggest that the dose-dependently altered ATRA-response of UF-1 cells may be determined through the interaction between the mutant PML-RARa and other intrinsic nuclear RA receptors.

    In this study period, we have investigated the role of vascular peroxisome proliferator activated-receptor ganma (PPAR-γ) in hypertensive arteriosclerosis in terms of molecular biology, physiology, and clinical science. We previously reported that PPAR-γ activation suppressed thromboxaen (TX) synthase gene. In this study, we have newly observed that PPAR-γ can also suppress TX receptor gene, as well as angiotensin type 1 receptor gene at the transcription level. In the gene transcription mechanism, transcription co-factors (CBP etc) are involved. Additionally, retinoic acid, a ligand of RXR, has also shown to suppress TX receptor gene transctription. In the physiology study, we have demonstrated that PPAR-γ ligand troglitazone dilated biphasically renal glomerular afferent and efferent arterioles, and the later phase dilatation is possibly mediated via a genomic action. PPAR-γ ligand pioglitazone has shown to increase plasma BNP levels in DM-2 patients with mild congestive heart failure. Non-genomic action of aldosterone has shown and thus a novel aldosterone action affecting NaCl metabolim via renal microcirculation has been suggested.

    The hypothalamic-pituitary-adrenal axis is necessary to maintain the internal milieu of living organisms. Norepinephrine (NE) has been shown to stimulate CRH gene expression in the paraventricular nucleus of the hypothalamus (PVN). On the other hand, CRH colocalizes with arginine vasopressin (AVP) in the parvocellular division of the PVN. We investigated the effect of NE on primary transcripts of CRH and AVP in the PVN by microinjection of NE directly into the PVN of conscious rats. NE stimulated CRH gene transcription, whereas it had no significant effect on AVP gene transcription. We reported the same experiments using rats, adrenalectomized and supplemented with low-dose corticosterone. It became clear that glucocorticoid affects CRH- and AVP gene in a different manner, AVP gene being more sensitive to the circulating glucocorticoid in this acute experimental paradigm. Furthermore, TPA stimulated CRH gene expression in a human neuroblastoma cell line, BE(2)-M17, suggesting the physiological role of the PKC pathway in the regulation of CRH gene expression.

    1) We have studied the transcription regulation of rat thromboxane synthase (TXS) gene by peroxisome proliferator-activated receptor γ (PPARγ) in Mφ. The transcription activity of cloned 5'-flanking region (1.6 kb) of rat TXS gene (5'FL-TXS) was examined by luciferase reporter gene assay. TXS mRNA levels and transcription activity of 5'FL-TXS were inhibited by PPARγ. Deletion analysis showed that the element responsible for the PPARγ effect was located in a region containing the NF-E2/AP-1 site (-98/-88), which was indicated to be bound by NF-E2 related factor 2 (Nrf2). Finally, a direct interaction between PPARγ and Nrf2 was confirmed by GST pull-down assay. In conclusion, the Nrf2-binding site (-98/-88) is the major promoter of 5'FL-TXS in Mφ, which can be suppressed by activated PPARγ via a protein-protein interaction with Nrf2. 2)Thiazide-sensitive Na-Cl cotransporter (TSC) is localized exclusively to the renal distal convoluted tubule (DCT) in kidney. We examined the molecular mechanism of this renal TSC expression localization. We generated transgenic rats harboring the transcription regulatory region of TSC fused upstream of the LacZ gene. Immunohistochemical analysis clearly showed that LacZ gene expression was co-localized to cortical distal tubules with TSC, indicating that the 5'FL/rTSC regulates the renal tubule-specific expression of TSC. Transcriptional activity of the rTSC gene was stimulated by overexpression of HFH-3, and electrophoretic mobility-shift assay showed a direct binding of in vitro synthesized HFH-3 to the HFH-3/rTSC. In conclusion, the transcription regulatory region of TSC gene confers the DCT-specific gene expression in the kidney. The DCT-specific transcription factor HFH-3 may be involved in the renal tubule-specific transcription of TSC gene.
