    Microbial fuel cells (MFCs) present a promising alternative to traditional activated sludge treatment for livestock wastewater, offering a carbon‐neutral, sustainable approach to wastewater management. Activated sludge treatment requires significant energy input for aeration and produces unpleasant odors. MFCs eliminate the need for energy‐intensive aeration, simultaneously generating energy during wastewater treatment. Platinum‐based electrodes commonly used in the cathode of MFCs pose a significant cost barrier, necessitating advancements in electrode materials for practical, large‐scale application. This study reports on the performance of a continuous methanogen biocathode MFC system engineered to simultaneously treat cattle‐barn wastewater and utilize carbon dioxide without 2‐bromoethanesulfonic acid (BES). Carbon felt treated with nitric acid without BES successfully reduces methane production by 93%. An MFC configuration utilizing nitric acid‐treated carbon felt as the anode and an oak‐derived carbon electrode as the cathode effectively treat wastewater and convert CO2 to methane, yielding a power density of 5.5 mW m−2 and Coulombic efficiency of 7.3%, approximately twice those without nitric acid treatment and surpassing even the performance of the system with BES treatment. This system represents a promising, low‐cost, and environmentally sustainable approach to renewable energy production and livestock wastewater treatment.

    Long-chain fatty acid (LCFA) degradation primarily involves several species of Syntrophomonas and hydrogenotrophic methanogens, constituting the rate-limiting step in anaerobic digestion. It is crucial to augment their abundance to enhance LCFA degradation. Utilizing microbial carriers presents an effective strategy to maintain the microorganisms on the surface and prevent their washout from the digester. In this study, we aimed to identify a suitable microbial carrier with a superior adsorption capacity for LCFA-degrading microorganisms. We tested various polymers, poly(vinyl alcohol) (PVA), polypropylene (PP), polyethylene glycol (PEG), and polyvinylidene chloride (PVDC), adding them to the sludge at the concentration of 28.25 g L-1 and incubating with olive oil. The amplicon sequencing analysis revealed that PVDC retained Syntrophomonas more abundantly than the other polymers. Remarkably, PVDC predominantly adsorbed LCFA-degrading S. sapovorans and S. zehnderi, whereas medium- to short-chain fatty acid-degrading S. wolfei was abundant in the sludge. Moreover, hydrogenotrophic Methanospirillum hungatei was detected at 2.3-9.5 times higher abundance on PVDC compared to the sludge. Further analysis indicated that not only these LCFA-degrading syntrophic microbial communities but also Propionispira and Anaerosinus, which are capable of lipid hydrolysis and glycerol degradation, became dominant on PVDC. Actually, chemical analysis confirmed that adding PVDC promoted the olive oil degradation. These results underscore the potential of PVDC in promoting anaerobic LCFA degradation.

    A single-contig, circular metagenome-assembled genome (cMAG) of Candidatus (Ca.) Patescibacteria was reconstructed from a mesophilic full-scale food waste treatment plant in Japan. The genome is of small size and lacks fundamental biosynthetic pathways. Taxonomic analysis using the Genome Taxonomy Database revealed that this cMAG belonged to the genus JAEZRQ01 (Ca. Parcubacteria).

    The present study aimed to evaluate a semi-wet biocathode composed of oak white charcoal and agarose gel as an alternative to the standard carbon felt biocathodes used in microbial fuel cells (MFCs). The MFC containing the oak white charcoal cathode (Oak-MFC) recorded a higher current value than that of the MFC containing a carbon felt cathode (CF-MFC). The Oak-MFC produced approximately 4.0-fold more electrons in the external circuit and 1.7-fold more methane (CH4) than the CF-MFC. A real-time PCR targeting mcrA showed that the number of methanogens adhering to the oak white charcoal cathode was approximately 15-fold that adhering to the carbon felt cathode. These results suggest that the methanogens attached to the cathode of both MFCs received electrons and CH4 was produced from carbon dioxide (CO2). Furthermore, Oak-MFC performed better than CF-MFC, thereby suggesting that oak white charcoal bound by agarose gel can be used as an alternative methanogen cathode. The propionic acid degradation rate of Oak-MFC was faster than that of CF-MFC suggesting that the cathodic reaction may affect the anodic reaction. The use of oak-derived electrode as a methanogen cathode also could contribute to sustainable forest management and promote regular thinning of oak trees. Further, its use will enable carbon fixation and efficient energy conversion from CO2 to CH4, thus contributing to sustainable energy use.

    Rumen fibrolytic microorganisms have been used to increase the rate of lignocellulosic biomass biodegradation; however, the microbial and isozymatic characteristics of biodegradation remain unclear. Therefore, the present study investigated the relationship between rumen microorganisms and fibrolytic isozymes associated with lignocellulosic biomass hydrolysis. Rice straw, a widely available agricultural byproduct, was ground and used as a substrate. The biodegradation of rice straw powder was performed anaerobically in rumen fluid for 48 h. The results obtained revealed that 31.6 and 23.3% of cellulose and hemicellulose, respectively, were degraded. The total concentration of volatile fatty acids showed a 1.8-fold increase (from 85.4 to 151.6‍ ‍mM) in 48 h, and 1,230.1‍ ‍mL L-1 of CO2 and 523.5‍ ‍mL L-1 of CH4 were produced. The major isozymes identified by zymograms during the first 12‍ ‍h were 51- and 140-kDa carboxymethyl cellulases (CMCases) and 23- and 57-kDa xylanases. The band densities of 37-, 53-, and 58-kDa CMCases and 38-, 44-, and 130-kDa xylanases increased from 24 to 36 h. A microbial ana-lysis indicated that the relative abundances of Prevotella, Fibrobacter, and Bacteroidales RF16 bacteria, Neocallimastix and Cyllamyces fungi, and Dasytricha and Polyplastron protozoa were related to fibrolytic isozyme activity. The present results provide novel insights into the relationships between fibrolytic isozymes and rumen microorganisms during lignocellulose biodegradation.

    Lipid-rich wastes are energy-dense substrates for anaerobic digestion. However, long-chain fatty acids (LCFAs), key intermediates in lipid degradation, inhibit methanogenic activity. In this study, TaqMan-based qPCR assays targeting the 16S rRNA gene of the cardinal LCFA-degrading bacterial species Syntrophomonas palmitatica and S. zehnderi were developed and validated. A trial experiment showed the advantage of species-specific quantification versus genus-specific quantification in assessing bacterial capacity for lipidic waste degradation. These qPCR assays will serve as monitoring tools for estimating the LCFA-degrading capacity of anaerobic digester communities and developing an effective strategy to enrich LCFA-degrading bacteria.

    In forest ecosystems, understanding the relationship between the vertical distribution of fine roots and residual soil nitrogen is essential for clarifying the diversity-productivity -water purification relationship. Vertical distributions of fine-root biomass (FRB) and concentrations of nitrate nitrogen (NO3 -N) in soil water were investigated in a conifer plantation with three thinning intensities (Control, Weak and Intensive), in which hardwood abundance and diversity were low, moderate and high, respectively. Intensive thinning led to the lowest NO3 -N concentration in soil water at all depths (0-100 cm) and highest FRB at shallow depths (0-50 cm). The NO3 -N concentration at a given depth was negatively correlated with total FRB from the surface to the depth at which NO3 -N concentration was measured, especially at shallow depths, indicating that more abundant fine roots led to lower levels of downward NO3 -N leaching. FRB contributed positively to nitrogen content of hardwood leaves. These findings demonstrate that hardwood mixture in conifer plantations resulted in sufficient uptake of NO3 -N from soil by well-developed fine root systems, and translocation to canopy foliage. This study suggests that productivity and water purification can be achieved through hardwood mixture in conifer plantations.

    Treatment with rumen fluid improves methane production from non-degradable lignocellulosic biomass during subsequent methane fermentation; however, the kinetics of xylanases during treatment with rumen fluid remain unclear. This study aimed to identify key xylanases contributing to xylan degradation and their individual activities during xylan treatment with bovine rumen microorganisms. Xylan was treated with bovine rumen fluid at 37°C for 48 h under anaerobic conditions. Total solids were degraded into volatile fatty acids and gases during the first 24 h. Zymography showed that xylanases of 24, 34, 85, 180, and 200 kDa were highly active during the first 24 h. Therefore, these xylanases are considered to be crucial for xylan degradation during treatment with rumen fluid. Metagenomic analysis revealed that the rumen microbial community's structure and metabolic function temporally shifted during xylan biodegradation. Although statistical analyses did not reveal significantly positive correlations between xylanase activities and known xylanolytic bacterial genera, they positively correlated with protozoal (e.g., Entodinium, Diploplastron, and Eudiplodinium) and fungal (e.g., Neocallimastix, Orpinomyces, and Olpidium) genera and unclassified bacteria. Our findings suggest that rumen protozoa, fungi, and unclassified bacteria are associated with key xylanase activities, accelerating xylan biodegradation into volatile fatty acids and gases, during treatment of lignocellulosic biomass with rumen fluid.

    Rumen microorganisms produce various fibrolytic enzymes and degrade lignocellulosic materials into nutrient sources for ruminants; therefore, the characterization of fibrolytic enzymes contributing to the polysaccharide degradation in the rumen microbiota is important for efficient animal production. This study characterized the fibrolytic isozyme activities of a rumen microbiota from four groups of housed cattle (1, breeding Japanese Black; 2, feedlot Japanese Black; 3, lactating Holstein Friesian; 4, dry Holstein Friesian). Rumen fluids in all cattle groups showed similar concentrations of total volatile fatty acids and reducing sugars, whereas acetic acid contents and pH were different among them. Predominant genera were commonly detected in all cattle, although the bacterial compositions were different among cattle groups. Zymograms of whole proteins in rumen fluids showed endoglucanase activities at 55 and 57 kDa and xylanase activity at 44 kDa in all cattle. Meanwhile, several fibrolytic isozyme activities differed among cattle groups and individuals. Treponema, Succinivibrio, Anaeroplasma, Succiniclasticum, Ruminococcus, and Butyrivibrio showed positive correlations with fibrolytic isozyme activities. Further, endoglucanase activity at 68 kDa was positively correlated with pH. This study suggests the characteristics of fibrolytic isozyme activities and their correlations with the rumen microbiota.

    Scum is formed by the adsorption of long-chain fatty acids (LCFAs) onto biomass surface in anaerobic digestion of oily substrates. Since scum is a recalcitrant substrate to be digested, it is disposed via landfilling or incineration, which results in biomass washout and a decrease in methane yield. The microbes contributing to scum degradation are unclear. This study aimed to investigate the cardinal microorganisms in anaerobic scum digestion. We pre-incubated a sludge with scum to enrich scum-degrading microbes. Using this sludge, a 1.3-times higher methane conversion rate (73%) and a faster LCFA degradation compared with control sludge were attained. Then, we analyzed the cardinal scum-degrading microbes in this pre-incubated sludge by changing the initial scum-loading rates. Increased 16S rRNA copy numbers for the syntrophic fatty-acid degrader <italic>Syntrophomonas</italic> and hydrogenotrophic methanogens were observed in scum high-loaded samples. 16S rRNA amplicon sequencing indicated that <italic>Syntrophomonas</italic> was the most abundant genus in all the samples. The amino-acid degrader <italic>Aminobacterium</italic> and hydrolytic genera such as <italic>Defluviitoga</italic> and <italic>Sporanaerobacter</italic> became more dominant as the scum-loading rate increased. Moreover, phylogenic analysis on <italic>Syntrophomonas</italic> revealed that <italic>Syntrophomonas palmitatica</italic>, which is capable of degrading LCFAs, related species became more dominant as the scum-loading rate increased. These results indicate that a variety of microorganisms that degrade LCFAs, proteins, and sugars are involved in effective scum degradation.

    A food waste recycling system was constructed using a small-scale methane fermentation system that incorporates hot spring heat. Local people provide the food waste and reap the benefits of the rich liquid byproduct. Biogas produced from food waste using methane fermentation is being used to boil water for the Ene Caf&eacute; METHANE. Instead of money, customers make payment in food waste to drink a cup of tea. The system has gained a following, now with over 1900 club members participating. Community members collect their organic food waste and utilize the liquid fertilizer (i.e. methane fermentation-digested liquid) generated as a byproduct of the system. The system at Ene Caf&eacute; METHANE is a completely interdependent food recycling system that makes customers happy because they can recycle their food waste while enjoying the company of fellow locals over a cup of tea. It has also been developed as a tourism resource and is creating stronger face-to-face communication within the community. This unique system is introduced here, along with a description of possibilities for its future development.

    The genus Cryptosporidium, which is an obligate intracellular parasite, infects various vertebrates and causes a diarrheal disease known as cryptosporidiosis. Wild rodents are naturally infected with zoonotic Cryptosporidium; thus, they are potential reservoirs of the parasites. Mice are common rodents frequently found in agricultural areas and have many opportunities to contact other wild animals, livestock, and humans. Irrespective of the potential risk, there are few epidemiologic studies of Cryptosporidium in wild mice because of their low economic importance and the difficulty in conducting surveys. Hence, the species and genotypes of Cryptosporidium in wild mice living around various areas remain unclear. We investigated the species and genotype distribution and prevalence of Cryptosporidium in the large Japanese field mouse (Apodemus speciosus) in an agricultural site in Osaki, Miyagi Prefecture, Japan. In total, 15 mice were captured and examined in this study. By microscopic analysis, only one mouse (JFM 3) was determined to be Cryptosporidium-positive, while the parasite were detected in four mice (JFM 3, 6, 10, and 15) by a molecular approach using partial SSU rRNA gene sequences. Based on nucleotide sequence and phylogenetic analysis, the Cryptosporidium isolates were identified as C. ubiquitum (from JFM 10) and C. muris (from JFM 3 and 6). In contrast, the Cryptosporidium in JFM 15 was not identified as a known species or genotype and is therefore proposed as a novel genotype; the Naruko genotype. More molecular data are necessary to elucidate the taxonomic identity of this novel Cryptosporidium genotype. The C. muris Japanese field mouse genotypes showed marked divergence compared to that in a previous report. The large Japanese field mouse might thus represent a reservoir of multiple Cryptosporidium spp.

    Little information is available on the epidemiology of Cryptosporidium in pigs in central Vietnam. The aims of this study were to investigate the prevalence and to characterize the genotype distribution of Cryptosporidium isolates in pigs in this region. A total of 193 pig fecal samples were screened for the presence of Cryptosporidium oocysts using the modified Ziehl-Neelsen staining method, and 28 (overall prevalence 14.5 %) were identified as positive by microscopic observation. Positive samples were further analyzed by polymerase chain reaction amplification and sequencing. Genetic identification based on the 18S ribosomal RNA and 70 kDa heat shock protein genes revealed that pigs in Vietnam are infected with two species/genotypes (Cryptosporidium suis and Cryptosporidium pig genotype II). This study is the first molecular characterization of Cryptosporidium in pigs in Vietnam. The presence of these host-adapted species/genotypes suggests that pigs may not pose a significant public health risk in this area. More extensive studies are necessary to ascertain the zoonotic potential of Cryptosporidium in porcine hosts in Vietnam.

    We conducted a pilot test of a small-scale methane fermentation system using heat from a hot spring, and evaluated its environmental impact. In the pilot test, although both the concentration and quantity of methane gas were influenced by temperature, they increased with increasing microbial growth. Therefore, a system responsive to temperature change could be designed by increasing the diversity of acclimatized microbes. When income and output for 1 year were taken into consideration, the impact of this system was equivalent to or smaller than that of a standard methane fermentation system. Therefore, in this area, it would be both environmentally and economically advantageous to install systems in several places, rather than a standard methane fermentation system in one place.

    The objective of this study is to develop of the small-scale methane fermentation system that hot spring heat is used for warming of a fermentation tub, and construct a new conveyance system of materials that tourists bring leftover food garbage to the reactor. Biogas production from leftover food garbage was investigated under different HRT conditions. In addition, the composition of per capita of leftover food garbage was investigated in ryokan. The results indicated that most suitable HRT was around 10 days. The amount of biogas production from leftover food volume per capita was estimated about 140L.

    In this study, the effects of Fe-type, Ca-type, and low-sodium-level Ca-type artificial zeolites, made from waste coal ash, on methane production and the methanogen communities under ammonium-rich conditions were investigated. Methane production with Fe-type zeolite was higher than that with the other types of zeolites, and was about four times higher than that without zeolite. Acetate was not accumulated during anaerobic digestion of ammonium-rich organic waste with Fe-type zeolite. The methanogen community which were analysed by real-time polymerase chain reaction (PCR) with Fe-type zeolite was almost the same as that with Ca-type zeolite. These results showed that quantities of methanogen have not relation with the volume of methane production. A comparison of methane production with Fe-type zeolite and that with Fe ions or Ca ions in anaerobic digestion under ammonium-rich conditions confirmed that Fe-type zeolite was the most effective in enhancing methane production, Ca ions addition was not enhancing the methane production contrary. Fe-type zeolite is considered to play two roles in anaerobic digestion; namely, supplying iron to the anaerobic microbes, and decreasing the ammonium concentration.

    To investigate the influence of materials released from lake sediment on the flora of phytop1ankton, we cultured three algal species, the green algae Scenedesmus acutus, the cyanobacterium Microcystis aeruginosa and filamentous cyanobacterium Oscillatoria agardhii with water obtained from an elution experiment of lake sediment. The effect of materials released from sediment on the growth of algae clearly varied from species to species of algae. The growth of green algae S. acutus was influenced by nitrogen released from sediment, though nitrogen was not actually supplied from the sediment somuch. The growth of filamentous cyanobacterium O. agardhii was mainly influenced by phosphorus released from the sediment. On the other hand, the growth of cyanobacterium M. aeruginosa was influenced by some materials, which were not nitrogen and phosphorus. From the results of an incubation experiment with adding EDTA and past research reports, it was suggested that these materials were organic complexes of iron, which stability constants was high, and inhibited M. aeurignosa from getting iron. This is a new finding in the relation between algae and lake sediment, and indicates that organic matter as complex-ligand released from sediment is one of the important factors affecting the flora of phytoplankton.

    Effective treatment of livestock feces and urine is necessary to prevent surface and ground water from being polluted. In this study, thermophilic oxic process (TOP) was applied to the treatment of swine waste. The proper environment condition for the thermophilic bacteria was investigated. The results were summarized as follows : It was impossible to treat swine waste by TOP. As for the reason, the calorie of swine waste lacked for the calorie of water evaporation and temperature rising. However, it could be treated by adding food oil as an assistant calorie source. When BOD5 load was 3 kg·m-3·day-1, aeration rate was 100l·m-3·min-1 and 120ml of food oil was added, 86% of carbon was converted to CO2 and 98% of water was evaporated in one cycle. ATP concentration and the number of thermophilic bacteria became the highest values under 55% of average water content in one cycle. And in this experiment 7 hours for the duration time of over 60°C in one cycle was suitable for maintaining 55% of average water content. The temperature condition of over 60°C was also very important to increase microbial activity.

    Effective treatment of livestock feces and urine is required to prevent surface and ground water from being polluted. The purpose of this study is to investigate the treatment characteristics of swine waste by TOP in long-term operation. The results were summarized as follows: Though swine waste could be treated effectively until 60th day, the treatment performance was gradually declined after 60th day. 87% of carbon was converted to CO2 on 45th day. 41% of carbon was converted to CO2 on 85th day. It was recovered by replacing half of cedar chip by new one on 86th day. Under suitable condition of TOP, 90% of total input swine waste (40kg) and food oil (2kg) was removed in 60 days. The number of thermophilic bacteria was maintained about 1011CFU·g-1 during this period. The reason of the decline of treatment performance was considered thatthe activity of thermophilic bacteria come down due to the accumulation of some residue and deterioration of cedar chip.

    In recent years, seaweed farming has gained attention as a blue carbon credit mechanism, and the establishment of a sustainable, economically viable system that also reduces CO2 emissions is essential. This study focuses on the seaweed Ecklonia cava, which is distributed in the coastal areas of Japan from Kyushu to Kanto. We investigated a seedling cultivation system aimed at reducing the CO2 load by generating hydrogen and methane gas through anaerobic fermentation of Ecklonia cava and utilizing the residual liquid from the fermentation process as a nutrient source for further seaweed cultivation. Hydrogen production was confirmed through anaerobic fermentation, indicating that in addition to marine bacteria, Enterobacteriaceae from rumen fluid may play a key role in hydrogen fermentation. Furthermore, 4% of the dissolved organic matter in the digestate was identified as refractory dissolved organic carbon (RDOC), suggesting its potential contribution to long-term carbon sequestration. These findings highlight the potential of Ecklonia cava to be integrated into a sustainable system that not only produces energy but also contributes to CO2 reduction and carbon fixation, supporting the development of blue carbon initiatives.

    <p>Because paper sludge discharged from the waste paper recycling process contains high levels of lignin and ash, it is not converted into biogas effectively during methane fermentation. The aim of this study was to improve the efficiency of biogas production from paper sludge by the pretreatment with rumen fluid. We performed the pretreatment of paper sludge with rumen fluid at 37°C for 6 hours. After pretreatment, methane fermentation was conducted at 35°C for 45 days. Pretreatment with rumen fluid hydrolyzed paper sludge and increased the VFAs concentration during the pretreatment process. Methane production from pretreated paper sludge was 12.1 % higher than that from untreated paper sludge. These results revealed that pretreatment with rumen fluid improved the efficiency of methane production from paper sludge. Next, we investigated the effects of protozoa on methane fermentation of paper sludge. Protozoa was removed from rumen fluid with sodium dodecyl sulfate, and the pretreatment and methane fermentation of paper sludge was conducted. We observed the positive relationship between the number of protozoa and methane gas production. The results indicated that protozoa possessed positive effects on methane production from paper sludge.</p>

    Microbial fuel cells (MFCs) are devices that harvest electrons to release when anaerobic microbes oxidize organic matter. Most MFCs use an expensive metal catalyst. Therefore we proposed using methanogens as the biocatalyst in the cathode. This system is reasonable and may perform power generation and methanation at the same time. In this study, we designed and tested the ability of the MFC that use methanogens as the catalysts.

    Hydrolysis of the lignocellulosic biomass is the rate-limiting step in biogas production. In this study, rapeseed stems were used as a model of plant biomass, and were pretreated with rumen fluid. Rapeseed was solubilised by rumen-fluid treatment, resulting in production of volatile fatty acids. Methane fermentation of 6-h-treated rapeseed was performed; higher methane yield was obtained compared with untreated rapeseed, showing a significant pretreatment effect. Rumen fluid has higher endo- and exo-glucanase activities and non-phenolic lignin unit oxidising capacity, compared to methane sludge. Therefore, lignocellulose components were effectively degraded by rumen-fluid treatment, resulting in increased methane production.

    Marine products industry discharges large amounts of fish waste in Tohoku coastal area. Fish waste can be used for anaerobic digestion because of its high chemical oxygen demand (COD). However, fish waste is protein-rich substrates, and their degradation products (ammonium) can inhibit the process (Chen et al., 2008). To decrease the inhibition, it was investigated that the influences of oyster shell and carbon carrier for anaerobic digestion of fish waste. The results shown that the methane production was 37% increased on anaerobic digestion of fish waste with oyster shell by carbon carrier.

    A small-scale methane fermentation system was developed using a hot spring for warming the reactor. Materials were added directly into the first tank without grinding the leftover food, including the paper bags. The temperature of the fermentation tank was adjusted to 〜55℃ by running hot spring water through a tube coiled around the tanks. Improving the methane production efficiency, it was compared methane production and volatile fatty acid (VFA) concentrations between two operations used different carriers (polyurethane and carbon fiber) in the reactors. Methane produced during the operation with carbon fibers carrier was higher than that with polyurethane carriers. Concentrations of total volatile fatty acid (T-VFA) in the operation with carbon fiber carrier were 1/10 times concentrations in the operation with polyurethane carrier.     Lignocellulosic biomass are potential raw materials for producing several high-value products. The hydrolysis of plant cell wall to monosaccharide and several monomers were considered the important step in lignocellulosic biomass decomposition. Then we tried to develop the effective decomposition method, which used rumen fluid when decomposition of plant cell wall prior to methane production. This method could increase volatile fatty acids production, and methane production from lignocellulosic biomass by effective decomposition. However, this study was examined only the batch experiment. Therefore, we investigated changing of the chemical properties and microbial community structure during continuous pretreatment of lignocellulosic biomass using rumen fluid. During the incubation period, pH was decreased below 6.0 after 3 days. Volatile fatty acids and biogas production was also significantly decreased after 3 days incubation. Microbial communities analyzed by PCR-DGGE method showed that bacterial communities were not changed during the incubation period. These results suggested that hydrolysis and methanogenic processes in rumen pretreatment system were limited by low pH, meanwhile bacterial communities were not changed during the incubation period.

    The purpose of this study was to evaluate the improvement in the efficiency of methane fermentation by filling with a carbon fiber carrier. By this means, accumulation of organic acids such as propionic acid fell and methane production improved. Analysis of the microbial community structure in the reactor, revealed the presence of Anaerobaculum mibile, which participates in organic acid decomposition, and Coprothemobacter proteolyticus, which participates in protein decomposition. Methanothemobacter thermautotrophicus (100% sequence similarity), a hydrogen-assimilating bacterium, was detected. Some studies have found that C. proteolyticus and Methanothermobacter act symbiotically. The adhesion of these microbes to the carbon fiber carrier in the reactor, may have increased the efficiency of methane production.

The influence of oyster shell and carbon carrier for anaerobic digestion of fish waste was investigated. On mesophilic anaerobic digestion of fish waste, it was observed that oyster shell contracted the inhibition and improved methane production. The methane production was increased on anaerobic digestion of fish waste with oyster shell by carbon carrier. When fish content of the substrate was 20% and HRT was 20 days, methane yield was about 0.5 on continuous anaerobic digestion of fish waste with oyster shell.

    To make the robust design of anaerobic digestion system under temperature fluctuation condition, it was investigated that methane production under the different temperature conditions. The temperatures were controlled to 35-65℃ with the fluctuating range, or 55℃. Volume of methane production from the fluctuating range condition was approximately a half of that from no range condition. When the microbial carriers were exchanged between with and no range conditions, then the cumulative methane production during 8 days was investigated under 35-65℃. The methane production from the carrier derived from with range condition were higher than that derived from no range condition.

    It is important to produce bioethanol from cellulosic biomass, which doesn’t compete with food. Ethanol production process from cellulosic biomass is classified by many pretreatment and saccharification methods. This study considers three methods that are using concentrated sulfuric acid hydrolysis (NEDO), dilute sulfuric acid pretreatment and enzyme saccharification (NREL), and hot water and milling pretreatment and enzyme saccharification (AIST). Three processes are designed by process simulator PRO/Ⅱ, and these material and energy balances are calculated. Conversion efficiencies of saccharification and fermentation are sensitive for ethanol production system, so the range of efficiency values is considered. Finally, these processes are evaluated by amount of CO2 emission reduction. NEDO process needs a large amount of thermal energy for sulfuric acid recovery, and AIST process uses a lot of power for disk milling. NREL process achieves the highest CO2 emission reduction. In the case of no byproduct (mainly lignin) use, however, all processes has a potential to be not able to reduce CO2 emission. In order to reduce CO2 emission, it is essential to use byproduct for process energy supply.

    Nowadays, the concept of "biomass town", which postulates sustainable utilization of local biomass resources, is becoming popular in Japan. In this research, a biomass accounting scheme was suggested as a supportive tool of biomass-related activities' evaluation. The scheme summarizes both economic and environmental indices in one table, which allows conducting integrated evaluation of a biomass town performance and presenting the results to citizens and stakeholders in an easy-to-understand way. Moreover, a case study was conducted in order to check the possibility of the scheme's application. Data on the Shobara city biomass town were processed and three biomass accounting tables were created in order to compare past (scenario 1), present (scenario 2), and planned (scenario 3) biomass town. Analysis of the three scenarios showed that present biomass town has lower economic and social performance comparing to past due to unprofitable and small-scale activities; however, its performance was improved in scenario 3 due to introduction of comparatively large scale pellet production.

    Process estimation of ETBE/BTL production process that is containing bioethanol production by organosoluble treatment was examined. Process design and simulation were performed and then heat and material balance were obtained. From the data, it was found that liquid fuel of 42.3 t/d could be produced in 60 t/d plant.

    In this study, a micro anaerobic digestion system with a 30 L capacity was developed. The system was easily constructed using materials purchased from home improvement stores. In addition, stable fermentation was established using familiar, readily available materials such as seed microorganisms. In the laboratory test, although no mixing in the fermenter was carried out, the methane production efficiency was approximately 60% of the theoretical production value for the COD of the added food waste. In the outdoor experiment on an island in the Seto Inland Sea, by covering the whole fermenter with insulation, the fermentation could be maintained with a 200 W heater even during their winter season.

    本研究は、牛の胃液(ルーメン液)からバイオマス分解促進材を開発するものである。ルーメン液は食肉処理場の汚水処理に大きな負荷を与える廃棄物であるが、我々は、このルーメン液は古紙や植物茎葉などのリグノセルロースの分解促進に有効で、メタン発酵の高効率化が可能なことを明らかにし、日・米・欧において特許化した。実用化のための課題は、ルーメン液の微生物機能の保持または促進、および運搬コスト低減のための減量化である。そこで、有効微生物活性を高め、バイオマス分解を促進する希釈、保存、濃縮、乾燥の方法を開発し、食肉処理場の遠隔地および海外においても使用できるバイオマス分解促進材の確立を目指すこととした。

    With the goal of establishing "microorganism printer" that artificially and freely aligns and attaches methanogens to the electrode surface, (1) elucidation of the adhesion mechanism of methanogens, and (2) dispersion of highly attached CNT of methanogens (3) Accumulation methanogen on the electrode for a short time , and manufacture high-performance electrodes. As a result, (1) Methanothermobacter thermautotrophicus had a good adhesion to that COOH and OH groups on the electrode surface at a ratio of 1: 0.65, and (2) had a high affinity with single-layer supergrowth CNTs. In addition, BSA 0.5 mg / L was suitable as the CNT dispersion. (3) At -600 mV (vs. Ag/AgCl), methanogen adhered to 107 copies/electrode in 2 days. A 7.5-fold higher current density was obtained by microbial fuel cell which methanogen adhered twice as densely. As for the electrode orientation, the same methane conversion rate was obtained even with 1/10 of methanogen as compared with the non-alignment.

    After the treatment of crop residues or waste paper by cattle rumen fluid, methane fermentation of these materials was accelerated. Enzymes and microbial communities in the process was investigated. Fiber‐degrading cellulase was diverse in the process; however, specific enzymes of xylanase showed high enzymatic activity continuously. Cellulase activity in the rumen treatment tank was continued for more than one month. At least five types of cellulases having different molecular sizes were existed. Microbes producing or involving these cellulases were unclassified species and not dominant in the microbial community.

    The microbial community structure related to nitrogen circulation by forest management of Japanese cedar plantation forest was investigated. The management was compared in the unmilling area, the weak thinning area, and the intensity thinned area. Each soil was collected and analyzed for the nosZ gene of N2O reductase. As a result, the amount of nos Z gene was higher in the intensity thinned plot than in the other plots. In addition, the species composition was also detected only in the thinning plot, closely related to Bradyrhizobium japonicum USDA 110 strain known as soybean root nodule bacteria. Undergrowth vegetation in the intensity thinned section had legume plants not found in other plots. Therefore, bacteria with high denitrification ability existed due to the influence of undergrowth vegetation in the intensity thinned plot, and as a result, the possibility that nitrogen removal was high was considered.

    The effects of water quality in the Tuul River in Mongolia on the genetic diversity of fish and animal health were investigated. The water in the Tuul River was more polluted downstream. In 2014, very high numbers of Escherichia coli were detected in well water. Pathogenic E. coli was detected in animal feces. Sheep and goats had high carriage rates. Cryptosporidium was detected in cattle feces and river water. In 2015, the numbers of E coli in well water decreased after changing the well structure. Sheep that drank water downstream had a higher rate of pathogenic E. coli infection. The genetic diversity of fish was similar to that of fish in Lake Baikal. Satellite image analysis showed that vegetation was better farther from roads. The regulation of well construction and the use of pastures may make the pasturing environment sustainable.

    In this study, the relationship between thinning intensity and ammonia-oxidizing prokaryotes was investigated. Concentrations of NO_3, NH_4 included in pore water of forest soil were analyzed. Ammonia oxidizing bacteria(AOB) and ammonia oxidizing archaea(AOA) were quantified by real-time PCR. The comparative community compositions of ammonia-oxidizing microbes among thinning intensities were determined using PCR-DGGE. Concentration of NO_3 in pore water of strong thinned forest soil was lowest than that of other forest soils. In June, intensive thinning significantly increased the amoA gene copies of AOB, as compared to unthinned or weakly thinned forests, whereas in the other months this did not differ greatly among thinning intensities. The amoA gene copies ofAOA were lowest in intensively thinned forests in May and July. The ratio of AOA/AOB in intensively thinned forests strongly decreased, as compared to those in unthinned and weakly thinned forests from May to July. These results suggested that ammonia oxidizingmicrobes in strong thinned were effected to nitrogen cycle.

    The influence of bacteriophages on wastewater treatment was investigated, together with the development of methods to analyze bacteriophages. Host DNA was detected in the lysate of phages, and the finding lead to the possibility to identify the lysed host by analyzing DNA in supernatant of wastewater treatment processes. It was estimated that around 1 to several % of bacteria are daily lysed by phages, and it may be possible to use phages to reduce waste sludge. Phage DNA was resistant to digestion by restriction enzymes, suggesting the complex relationship between phages and host. Yet, further compilation of case studies is still needed to know the impact of phages on wastewater treatment.

    本研究は、亜熱帯沿岸域の金属(鉄・マンガン)供給およびその形態におけるマングローブ林の役割を解明することを目的に、炭素電極を用いた電気化学的測定法による鉄・マンガン錯体の検出を行ってきた。その結果、サンプリング時期によって、同地点のサンプルにおいてピーク検出されない結果が得られた。そのため、炭素電極の再検討をおこなった。比較炭素電極にはGC電極、PFC電極を用い、昨年度より使用しているGRC電極との検出値を比較した。その結果、昨年より使用しているGRC電極が、最も感度が高いことが明らかになった。また、マングローブ底泥間隙水に見られる+0.19Vの還元波を詳細に解析した結果、マングローブ底泥由来マンガン錯体と考えられたピークは、マンガン錯体のピークだけではなく、その他の物質のピークも重なっていることが考えられた。このピークは、酸化還元波の可逆性が低いため、金属錯体ではなく有機物である可能性が考えられた。マングローブ由来鉄錯体、マンガン錯体の沿岸海域への供給を明らかにするためには、マングローブ底泥の浸透流を解明することも必要である。また、マンガン錯体等の現場測定を可能にするためには、現場の塩分濃度測定が必要であることが昨年度の結果より示唆されたことから、現場の塩分濃度を連続的にモニタリングする装置を作成し、マングローブ底泥の塩分濃度連続測定法を検討した。この結果、マングローブ底泥深度30cm下の塩分濃度は潮の満干によって大きく変動しており、またマングローブ林のサンプリング位置によって塩分濃度変化が大きく異なることがわかった。これらの結果より、マングローブ底泥由来錯体の沿岸海域への供給は、潮位変動によって、マングローブ底泥の様々な地点から供給される可能性が示唆された。また、マングローブ錯体の検出変化の原因の一つとして、マングローブ底泥における浸透流がマングローブ底泥由来錯体形成に大きく影響していると考えられた。

    2016年3月、宮城県石巻市に移設されている国立競技場の聖火台に、エコ燃料・バイオメタンガスの火が灯った。2020年の東京オリンピック・パラリンピックでの使用を目指し行われた実験、指揮を取ったのは東北大准教授の多田千佳さん。家庭や学校などで出る生ごみを持ってきてもらいガスを精製するなど、地域の人たちの協力を得ながらバイオメタンガス普及の道を探っている。技術的な困難を乗り越え、「生ごみから聖火を」と情熱を傾ける多田さんの活動を追った。

    持続可能な社会構築には、環境負荷軽減が必要である.旅館等の宿泊施設から排出される生ゴミは大きな問題である。宿泊施設からの生ゴミ量は1人当たり一日平均の3倍強の700gと言われる.平成21年の全国宿泊客数延べ人数は約3億人であるため、年間21万トンの排出量になる.これらの処理コスト・処理エネルギー量は大きく、その軽減が切望されている。含水率の高い生ゴミの有効活用法にメタン発酵がある。メタン発酵はエネルギーおよび経済性収支をプラスにするため、一日処理量50t以上の大規模システムが一般的である。しかし、これを温泉地に導入するには、いくつかの課題がある。初期投資が莫大である、加温の消費エネルギーが大きい、原料運搬が非効率、消化液の処理、これらシステムの環境評価が十分でないと言う点である。そこで本研究では温泉地域での生ゴミの資源化のために小規模のメタン発酵システムの導入を検討する.それには消費エネルギーを含む環境負荷を低減することが不可欠であり,これを①加温に必要なエネルギーを温泉の熱を利用すること,②運搬に必要なエネルギーは観光客が観光の一環として生ゴミの運搬に取り組めるようなシステム作り,および③発酵残さであるメタン消化液を積極的に利用することによって達成することを試みる.さらに,その導入から運用まで地域内で協議しながら実規模でおこない,④環境負荷や経済性をモニタリング・評価することで温泉地への小規模メタン発酵システムの導入を核とした低炭素観光(=エネツーリズム)の確立を目指す.

    持続可能な社会システム構築が切望される中、汚水処理施設も例外ではなく、省エネルギー・自然共生に不可欠な生態系機能活用型浄化の一層強化・コントロールが重要である。活性汚泥法は微生物機能を活用した手法であり、真核・原核生物が混在した微生物生態系機能による浄化技術である。活性汚泥法の研究は歴史が長く、生物顕微鏡下で観察可能な原生動物等真核生物による処理性能の指標化研究や個々の原生動物と純粋細菌による捕食被食関係・増殖速度等の知見がある。近年では、遺伝子解析技術の発展に伴い、活性汚泥の細菌群集構造解析報告が多い。しかし、既往研究では個々の真核生物あるいは細菌群集のみに着目しており、活性汚泥での真核生物と細菌群集構造との相互関係性を遺伝子解析技術によって解明した例はほとんどなく、活性汚泥生態系の現実を反映した機能性解析が不十分である。本研究では、上記の点をふまえ、真核生物と原核生物の生物間相互作用に着目し、遺伝子解析技術を活用した相互関係性解明と生態系機能を解明し、真核・原核生物からなる微生物生態系の新規知見を基にした浄化機能の一層強化による持続可能な汚水処理システム構築を目指す。
