    ダイズ種子の耐湿性向上による湿害軽減はダイズの収量増加のために重要である.本課題では,ゲノム改良の手段を用いず耐湿性種子を生産することを目指し,個体内の着粒位置や粒重などと種子耐湿性との関係を明らかにすることを目的とした. 本年度は,昨年度のサンプルについて種子耐湿性試験を引き続き行った.また栽培試験として,エンレイとミヤギシロメを供試し摘芯処理の影響を検討した.開花直前に主茎を摘芯処理することにより,受光体勢の改善や倒伏の軽減などが期待できる.これまでの本課題の研究により,摘心される主茎上部は耐湿性種子の割合が高い部位であった.開花前に摘芯処理による耐湿性種子の割合の高い部位の喪失とそれに伴う群落環境の変化が種子耐湿性に与える影響を明らかにしようとした. 昨年度のサンプルについて,種子耐湿性試験が終了し,現在解析中である.解析結果の一部として以下のことがわかった.昨年度の栽培試験では,播種期と栽植密度の異なる試験区を設けた.6月播種と7月播種では,1粒重と種子耐湿性との関係が異なった.さらに7月播種では標準の栽植密度と比べ密植により種子耐湿性が高まり,7月播種の密植栽培により耐湿性種子の割合を高められる可能性が示唆された.播種期が異なる生育期間中の気象条件の比較により子実肥大盛期の気温と降水量の影響が示唆された.さらに栽培地の比較により気温と降水量のそれぞれの影響の大きさも推察されたが,今後さらに調査を重ねる必要がある.

    It is known that there is the genetic difference in SCP (the strength to control the priority of spikelets within the panicle for grain filling) in rice. Data from the experiments using the genotype with high sink capacity recently released showed that the early growth and filling of grains at the lower positions within the panicle (inferior grains) often decrease in varieties with strong SCP, but such a decrease dose not occur in varieties with strong SCP. Therefore, in addition to high sink capacity, weak SCP should be proposed as ideal characteristic of grain filling in super high yielding rice genotype which will be developed in the near future.

    There is the genetic difference in determining the priority of spikelets within the panicle for grain filling in rice. Under the low source/sink ratio, the early growth of grains at the lower positions within the panicle (inferior grains) is easy to delay in Sasanishiki and Koshihikari, but is difficult to delay in 92133 (China) and Akinishiki. Consequently, the percentage of grain filling is lower in the former varieties than in the later varieties. The physiological mechanism which accounts for this genetic difference was investigated. Shading after heading lowered the abscisic acid (ABA) levels in the inferior grains in Sasanishiki and Koshihikari, but had no effect in 92133 (China) and Akinishiki. There was no relation between the early growth of inferior grain and its endogenous cytokinin or free sugar levels. It was known that exogenous cytokinin promotes the early grain growth and grain filling. Therefore, it was concluded that the genetic difference in determining the priority of spikelets for grain filling was regulated by the endogenous ABA levels in the grains. Generally, ABA promotes the ethylene evolution. But ABA did not change the ethylene evolution from the panicle, and some chemicals changed the ethylene evolution without the changes of the early grain growth, suggesting that ABA promote the grain growth directly. Not only ABA but also cytokinin promotes the early growth of inferior grains. In this study, exogenous cytokinin or ABA and root cutting promoted the early growth of inferior grains. Endogenous cytokinin levels in inferior grains was increased by increasing the nitrogen level, but was decreased by root cutting. Endogenous ABA level was increased by exogenous cytokinin or ABA and root cutting, and was closely related to the grain growth. These results suggest that cytokinin promotes the grain growth through the increase of the levels of endogenous ABA.

    Physiological mechanisms controlling sink abortion in legume crops were analyzed. 1. Intra-raceme variation in pod-set probability and cytokinin content was investigated. The amount of cytokinins in racemes that were identified by GC-MS and enzyme immunoassay peaked one to two weeks after the first flowering on a raceme, when pod development was initiated. Within individual racemes, the total amount of cytokinins were greater at more basal floral positions, as the probability of pod set. Thus, pod-set probability was significantly associated with the cytokinin contents at different floral positions within individual racemes. 2. Effects of organic materials and gypsum on root formation and cytokinin production in the root system were analyzed. A continuous application of gypsum enhances expansion of root system and nodule formation in soybean. Contents of isopentenyl-adenin in the root exudate tended to be higher in the plants which were applied gypsum and organic materials. 3. Effects of a growth regulator, figaron, on pod formation of legume crops were studied. Figaron application on racemes increased grain yield of soybeen and peanut, mainly due to enhancing abscission of lately opened flowers. The same effects were not observed in aduki bean. These results indicate that development of opened flowers is regulated by hormones, particularly by cytokinins, and that rhizosphere is an important factor affecting the production capacity of cytokinins in the root.

    The purpose of this study is to elucidate the role of phytohormone and carbohydrate in the grain filling of rice. 1. Early growth and dry matter accumulation of grains Shading after the heading decreased the early growth rate and increased the linear dry matter accumulation rate in the grain at the lower spikelet positions. Both the shading before the heading which reduced the carbohydrate content at the heading and elevated CO_2 after the heading had no effects on the early grain growth, but increased the linear dry matter accumulation rate in the grain. These results suggest that lights promote the early grain growth not through the increment of carbohydrate supply, but through phytohormone which concerned with the grain growth and the endosperm development. 2. Diurnal changes of the early grain growth Grains were expanded and cells of maternal tissue were divided during the day at the early developmental stage. The level of abscisic acid was higher in the day than in the night, but the level of cytokinin was higher in the night than in the day. Sugar pool sizes of grains did not show the clear diurnal change. These results suggest that light regulates the early grain growth through the abscisic acid and, abscisic acid acts as a growth promoter. 3. Varietal differences in early grain growth and characteristics of grain filling Under the defoliated condition, there was a varietal difference in the degree of the growth delay of the grains at the lower spikelet positions within panicle. The variety in which the early gram growth is difficult to delay had higher endosperm cell number and final grain weight than the variety in which the growth is easy to delay. It seemed to have to elucidate the varietal difference in the early grain growth, physiologically.

  8. Development of a Water storage-type Deep-irrigation Method for Rice Aimed at Increasing the Environment-protecting Function of Paddy Fields

    We developed a deep-irrigation method to grow rice at a steady high yield in the Tohoku district, while increasing the function of water storage, which is one of the functional benefits of paddy fields in environmental protection. The main objective for the promotion of the water storage function is to delay the runoff into the sea of the river water, which is abundant due to snow-melting water and rainfall in the rainy season, until summer when there is often a shortage of water supply. We designed a water managing system taking into consideration the following points : 1 : To increase the water storage function of the paddy field, the water is made as deep as possible using the present levee (the maximum water depth, 25 - 30 cm). 2 : Considering the steady high yield in the Tohoku district, the water depth at the meiotic stage is made 25 - 30 cm, to protect from low-temperature. 3 : The water depth is increased continuously until mid July when water is abundant, assuming the water supply is steady, and the depth is controlled to have only one peak. 4 : The yield is increased by deep irrigation, referring to the yield-increasing technique of deep-irrigation method used by an outstanding farmer. The growth of rice plants were analyzed. As the result of stratified clip method, it became clear that the deep-irrigation method is put on the rice plant canopy 10 cm. Three years investigations indicated many available results, one of them is that controlled release coated fertilizer is effective in order to increase the yield.

  9. PHYSIOLOGICAL MECHANISM OF STERILITY-TYPE COOL-WEATHER DAMAGE IN RICE PLANTS

    1. Focus of the research on cool-weather damage in rice plants Literatures were reviewed for research on cool-weather damage in rice plants, and the elucidation of the causal sequence from nitrogen application to unfertilization, placing a particular focus on the number of pollen grain in the anther, was pointed out to be highly important for the understanding of the physiological mechanism of the sterility-type cool-weather damage. 2. Varietal difference in the number of pollen grains The number of pollen grains was larger in clutivars and lines with more highly resistant to the cool weather damage. It showed a tendency, however, to be divided into two subgroups within each group of the same ranking of resistance. This fact implies that a genetic factor other than pollen number is relating to the degree of cool-weather damage. 3. Importance of the relative amount of roots The root/leaf-stem-panicle ratio or the root/spikelet ratio showed a high correlation with the fertility of plants which were cooled at the critical stage. This fact implies that the relative amount of roots to aerial parts of the plant or the number of spikelets is relating to the degree of the cool-weather resistance. 4. Variations in the number of pollen grains due to cultivation conditions The number of pollen grains decreased with an application of larger amount of nitrogen fertilizer. An application of larger amount of phosphate fertilizer counteracted to this nitrogen application and recovered decreased number of pollen grains, resulting in the recovery of unfertilization also. The number of pollen grains decreased by a condition of lower solar incidence. 5. Effect of phytohormone Gibberellin or cytokinin or the mixture of them augmented the degree of unfertilization induced by cool temperatures at the critical stage.

    これまでの研究から,イネの最終粒重が大きい品種ほど穀粒生長速度(GGR)は速く,胚乳細胞数,胚乳細胞当たりの乾物蓄積速度も大きくなった.胚乳細胞当たりの乾物蓄積速度は,基質である胚乳中の糖濃度との相関がなかったので,胚乳細胞当たりの乾物蓄積速度と胚乳中のデンプン合成に関連する酵素活性との関係を本研究で調査した. 粒重の異なる7品種を用い,分げつ除去土耕栽培し,出穂期以降昼24℃夜19℃のファイトトロンに移動した.籾殻を透かして穎果が籾殻を埋め尽くした日から,0,2,4,6,8,10日後に強勢な穎果をサンプルした.これらについて,生体重,乾物重,糖濃度,酵素活性を測定し,完熟時のサンプルから胚乳細胞数を算出した.デンプン合成に関連する酵素として,スクロース合成酵素(SuS),ADP-グルコースピロホスホリラーゼ(ADPGPP),可溶性デンプン合成酵素(sStS),不溶性デンプン合成酵素(bStS)の酵素活性を測定した. SuSでは,1粒当たりの活性には有意な品種間差があったが,生体重当たりの活性と胚乳細胞当たりの活性には有意な品種間差がなかった.また胚乳中の糖濃度にも有意な品種間差がなかったことから,胚乳細胞当たりの乾物蓄積速度にSuS活性は影響を与えていないことが示唆された.ADPGPP,sStS,bStSでは,1粒当たりの活性,生体重当たりの活性,胚乳細胞当たりの活性の全てに有意な品種間差があった.生体重当たりの活性とGGR,胚乳細胞当たりの乾物蓄積速度との間では,ADPGPP,sStS,bStSそれぞれに有意な相関があった.しかし胚乳細胞当たりの活性では,胚乳細胞当たりの乾物蓄積速度と有意な相関があったのは,ADPGPPだけであった. 以上,胚乳細胞当たりの活性を検討することにより,ADPGPPが胚乳細胞当たりの乾物蓄積速度に影響を与えている可能性が示唆された.

    本研究では,粒重が大きく異なるイネ5品種,ササニシキ(玄米乾物重20.9mg),アキヒカリ(22.6mg),フクノハナ(25.7mg),房吉(33.5mg),オオチカラ(39.6mg)の強勢な穎果を用いて,穀粒生長速度(Grain Growth Rate : GGR),有効乾物蓄積期間(Effective Filling Period : EFP),最終粒重,胚乳細胞数,胚乳中の糖濃度の関係について解析した. ポットで分げつ除去,土耕栽培した.出穂直前にファイトトロンに移動し,登熟期の温度環境を一定に保った.穎果の直線的な乾物蓄積期間に一定の間隔で数回と完熟時にサンプルを行い,新鮮重,乾物重を測定した.このデータからGGRやEFPを算出した.さらにこのサンプルについて,エタノールで糖の抽出を行い,イオン交換樹脂を通した後,TMS化を行い,ガスクロマトグラフィで糖含量を測定した.胚乳細胞数の測定法には,穀粒の完熟後,横断切片と縦断切片から細胞数を測定する方法を用いた. 最終粒重が大きい品種ほど胚乳細胞数が多く,胚乳細胞当たりの乾物重も多くなった.最終粒重にはGGRもEFPも有意に影響したが,GGRの影響の方が大きくなった.このGGRへは胚乳細胞数と胚乳細胞当たりの乾物蓄積速度の両方が影響していた.胚乳細胞当たりの乾物蓄積速度は,胚乳の糖濃度が十分にある場合には,胚乳細胞の乾物蓄積能力の影響を受け,胚乳の糖濃度が不十分の場合は,胚乳の糖濃度の影響を受けると考えられる.今回の実験では,品種間における胚乳細胞当たりの乾物蓄積速度と,胚乳の糖濃度との間には,正の関係が見られなかった.むしろ,胚乳細胞当たりの乾物蓄積速度が大きいものほど,胚乳の糖濃度が低い傾向がみられた.これは,胚乳の糖濃度十分であることを示しており胚乳細胞当たりの乾物蓄積速度は糖を乾物すなわちデンプンに変換する能力により決定されていることが示唆された。
