    Somatic plant cells can regenerate shoots and/or roots or adventitious embryonic calluses, which may induce organ formation under certain conditions. Such regenerations occur via dedifferentiation of somatic cells, induction of organs, and their subsequent outgrowth. Despite recent advances in understanding of plant regeneration, many details of shoot induction remain unclear. Here, we artificially induced shoot stem-like green organs (SSOs) in Arabidopsis thaliana roots via simultaneous induction of two transcription factors (TFs), ARABIDOPSIS THALIANA HOMEOBOX PROTEIN 25 (ATHB25, At5g65410) and the B3 family transcription factor REPRODUCTIVE MERISTEM 7 (REM7, At3g18960). The SSOs exhibited negative gravitropism and differentiated vascular bundle phenotypes. The ATHB25/REM7 induced the expression of genes controlling shoot stem characteristics by ectopic expression in roots. Intriguingly, the restoration of root growth was seen in the consecutive and adjacent parts of the SSOs under gene induction conditions. Our findings thus provide insights into the development and regeneration of plant shoot stems.

    Recent progress in plant science has revealed the molecular functions of many genes involved in plant development and environmental responses. However, to understand the fundamental mechanisms in plant biology, it will be essential to reproduce biological processes orchestrated by multiple genes. We have previously produced shoot stem-like greening organs (SSOs) in parts of the root by simultaneous induction of a set of genes specifically expressed in the shoot apical meristem (Hanano et al., 2020). Induction of the SSOs elucidates the fundamental mechanisms underlying plant development. Here, we will report the reconstruction of nitrogen-fixing symbiosis by using the synthetic biological approach. In comparison with the genome sequences of a nitrogen-fixing symbiosis plant, Lotus japonicus (legume), and a non-nitrogen-fixing symbiosis plant, Fragaria vesca (strawberry), we found the candidate sets of the genes involved in the nitrogen-fixing symbiosis. We are now simultaneously inducing these multiple genes, constructed on a binary vector by the PRESSO method (Takita et al., 2013), in F. vesca by using Rhizobium rhizogenes -mediated hairy root transformation.

    Recent advances in plant biology have revealed molecular mechanisms underlying plant development. However, many details of shoot-stem induction remain unclear. Here we took a synthetic biological approach by simultaneously inducing multiple transcription factors expressed around the shoot apical meristem. Co-induction of two transcription factors (TFs), ARABIDOPSIS THALIANA HOMEOBOX PROTEIN 25 (ATHB25, At5g65410) and the B3 family transcription factor REPRODUCTIVE MERISTEM 7 (REM7, At3g18960) artificially formed shoot stem-like green organs (SSOs) in Arabidopsis thaliana roots. The SSOs exhibited negative gravitropism and differentiated vascular bundle phenotypes. The ATHB25/REM7 induced the expression of genes controlling shoot stem characteristics by ectopic expression in roots. Intriguingly, the restoration of root growth was seen in the consecutive and adjacent parts of the SSOs under gene induction conditions.     Differentiation of shoots and roots is not only one of the major events during early development in plants but also a very interesting phenomenon of terrestrial plant evolution. Previous studies have reported many genes that generate and maintain shoot apical meristems (SAMs), but the mechanism of differentiation of shoot and root remains unclear. Here, we artificially converted parts of the root into the organs with shoot stem characteristics (termed as SSOs) via simultaneous induction of two SAM-specific transcription factors, ARABIDOPSIS THALIANA HOMEOBOX PROTEIN 25 (ATHB25) and the B3 family transcription factor REPRODUCTIVE MERISTEM 7 (REM7). In the SSOs, we observed negative gravitropism, the formation of chloroplasts that have photosynthetic activity, differentiation of vascular bundles, induction of expression of genes originally expressed in the shoot organs, and suppression of genes expressed in the roots. The SSOs once formed in the roots sustainably restored the normal root growth. This is the first report to convert the roots into the shoots without apparent dedifferentiation by a synthetic biological approach. The SSO formation will be a valuable model to understand the molecular mechanisms and cell-to-cell communications on the shoot-root differentiation, which has not been elucidated due to the nascent event during embryogenesis.     A genomic DNA fragment of Arabidopsis thaliana (~100 kbp, containing 12 genes) was introduced into a photoreceptor mutant of Marchantia polymorpha L, which diverged 400 million years ago. The transformed Marchantia polymorpha recovered the deficiency of photo reception. The result indicates that the M polymorpha still recognizes the Arabidopsis gene.
