    To evaluate the effects of the soluble fiber isomaltodextrin in a protein-biased diet, a 21-day protein diet trial was conducted in rats, with 60% of the calories derived from protein. The results revealed that the high-protein diet alone led to a significant increase in kidney weight. In contrast, the consumption of water with 5% isomaltodextrin dissolved in it, along with a high-protein diet, suppressed this weight gain. To elucidate this mechanism, an analysis of serum urea toxins confirmed that the concentrations of phenyl sulfate were significantly higher with high protein, and significantly lower with isomaltodextrin. The impact of a high-protein on increased phenol in cecal contents, an increased that was mitigated by the isomaltodextrin. This suggests that the inhibitory effect of isomaltodextrin on renal hypertrophy was due to the suppression of urea toxin precursor production by the gut microbiota.

    Abstract Patients with end-stage renal disease have lower fasting plasma glucose and HbA1c levels, with significantly higher insulin levels. For a long time, it has been believed that this higher insulin level in renal failure is due to decreased insulin clearance caused by reduced renal function. However, here we reported that accumulation of the gut microbiota-derived uremic toxin, phenyl sulfate (PS) in the renal failure, increased insulin secretion from the pancreas by enhanced glucose-induced insulin secretion. Other endogenous sulfides compounds which accumulated as in the renal failure also increased glucose-induced insulin secretion form b-cells. In addition, we also found that PS increased insulin resistance through lncRNA expression and Erk1/2 phosphorylation in the adipocytes. To confirm the relationship between PS and glucose metabolism in human, we recruited 2 clinical cohort studies (DKD and CKD), and found that here was a weak negative correlation between PS and HbA1c. Because these trials did not collect fasting insulin level, we alternatively used the urinary C-peptide/creatinine ratio (UCPCR) as an indicator of insulin resistance. We found that PS-induced insulin resistance in patients with eGFR < 60 ml/min/1.73 m2. These data suggest that the accumulation of uremic toxins modulates glucose metabolism and induced insulin resistance in CKD and DKD patients.

    The erythroid growth factor erythropoietin (EPO) is mainly produced by the kidneys in adult mammals and induces expansion of erythroid cells and iron use for hemoglobin synthesis. The liver also produces EPO at a lower level than the kidneys. Renal and hepatic EPO production is fundamentally regulated by hypoxia-inducible transcription factors (HIFs) in a hypoxia/anemia-inducible manner. Recently, small compounds that activate HIFs and EPO production in the kidneys by inhibiting HIF-prolyl hydroxylases (HIF-PHIs) have been launched to treat EPO-deficiency anemia in patients suffering from kidney disease. However, the roles of the liver in the HIF-PHI-mediated induction of erythropoiesis and iron mobilization remain controversial. Here, to elucidate the liver contributions to the therapeutic effects of HIF-PHIs, genetically modified mouse lines lacking renal EPO-production ability were analyzed. In the mutant mice, HIF-PHI administration marginally increased plasma EPO concentrations and peripheral erythrocytes by inducing hepatic EPO production. The effects of HIF-PHIs on the mobilization of stored iron and on the suppression of hepatic hepcidin, an inhibitory molecule for iron release from iron-storage cells, were not observed in the mutant mice. These findings demonstrate that adequate induction of EPO mainly in the kidney is essential for achieving the full therapeutic effects of HIF-PHIs, which include hepcidin suppression. The data also show that HIF-PHIs directly induce the expression of duodenal genes related to dietary iron intake. Additionally, hepatic EPO induction is considered to partially contribute to the erythropoietic effects of HIF-PHIs but to be insufficient to compensate for the abundant EPO induction by the kidneys.

    Sensitive biomarkers can enhance the diagnosis, prognosis, and surveillance of chronic kidney disease (CKD), such as diabetic kidney disease (DKD). Plasma growth differentiation factor 15 (GDF15) levels are a novel biomarker for mitochondria-associated diseases; however, it may not be a useful indicator for CKD as its levels increase with declining renal function. This study explores urinary GDF15's potential as a marker for CKD. The plasma and urinary GDF15 as well as 15 uremic toxins were measured in 103 patients with CKD. The relationship between the urinary GDF15-creatinine ratio and the uremic toxins and other clinical characteristics was investigated. Urinary GDF15-creatinine ratios were less related to renal function and uremic toxin levels compared to plasma GDF15. Additionally, the ratios were significantly higher in patients with CKD patients with diabetes (p = 0.0012) and reduced with statin treatment. In a different retrospective DKD cohort study (U-CARE, n = 342), multiple and logistic regression analyses revealed that the baseline urinary GDF15-creatinine ratios predicted a decline in estimated glomerular filtration rate (eGFR) over 2 years. Compared to the plasma GDF15 level, the urinary GDF15-creatinine ratio is less dependent on renal function and sensitively fluctuates with diabetes and statin treatment. It may serve as a good prognostic marker for renal function decline in patients with DKD similar to the urine albumin-creatinine ratio.

Canagliflozin is a sodium-glucose cotransporter (SGLT) 2 inhibitor, and it also exerts a modest inhibitory effect on SGLT1.     Roxadustat (FG-4592, Rox) is a stabilizer for hypoxia-inducible transcription factors (HIFs), which induce production of the erythroid growth factor erythropoietin, and has been listed by the World Anti-Doping Agency as a prohibited substance for athletes since 2011. Although the detection technologies for Rox and its glucuronide-conjugated metabolite (Rox-Gluc) have been developed exploiting triple quadrupole mass spectrometry (MS/MS), the production of metabolites from Rox in the human body remains to be clarified. Here, we established a protocol for the detection of unknown metabolites in plasma and urine samples from Rox-doping mice by global metabolomics using an ultra high-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UHPLC-QTOF/MS). We identified methylated Rox (Rox-Methyl), a novel metabolite, and Rox-Gluc in mouse urine by principal component analysis and orthogonal partial least squares discriminant analysis based on detected features by UHPLC-QTOF/MS analysis. The estimated pharmacokinetic parameters of Rox-Methyl and Rox-Gluc in mouse plasma showed similar profiles to that of Rox and both compounds showed similar biological activities. Of note, Rox-Methyl showed shorter half-life than Rox-Gluc in vivo, implying the easy escape from anti-doping screen. These results demonstrate that the global metabolomics method introduced in this study will contribute to the identification and detection of HIF-analog doping.

    We have synthesized a novel phthalocyanine dimer, in which two phthalocyanine units are linked directly at the, so-called, alpha-position. The electrochemical and spectroscopic properties of the dimer and the corresponding monomer as a reference have been investigated to clarify the unique electronic structures of the obtained dimer.

    Highly chemo- and regioselective deprotonative alumination of functionalized aromatic and heteroaromatic compounds was realized using a newly designed aluminum ate complex. Direct alumination followed by electrophilic trapping proved to be a powerful tool for the preparation of 1,2- or 1,2,3-multisubstituted aromatic compounds. In addition, the functionalized aromatic aluminate intermediate was found to undergo transition metal-catalyzed C–C bond-forming reactions and oxidation with high regio- and chemoselectivities. A structural study of iBu3Al(TMP)Li and a mechanistic investigation of this novel deprotonation reaction strongly suggested that an unique coordinative and electronic environment of the lithium/aluminum-hetero-bimetallic ate complex allows the facile and selective deprotonation reaction.

    We have developed a new organozincate-mediated anionic polymerization (ZAP) using tBu4ZnLi2 as an initiator. This method possesses great versatility and high chemoselectivity, and multi-functionalized monomers, such as NIPAm, AN, and HEMA, could be polymerized in high yields. Furthermore, the current anionic polymerization by using tBu4ZnLi2 has high compatibility with protic solvents, which accelerate the reactions significantly. The polymer growth was also well-regulated, and the terminal zincate of the polymer end could be modified by electrophilic traps.

    スフィンゴ脂質は、細胞膜形成あるいは脂質メディエーター機能により、生体の恒常性維持に必須の分子群であり、がんや免疫疾患発症に関連することが示唆されている。また、細胞膜に含まれる脂質ラフトや細胞外小胞のエクソソームの主要構成脂質でもあり、スフィンゴ脂質の代表分子であるスフィンゴシン一リン酸(S1P)を介したエクソソーム分泌機構が細胞間シグナル伝達に重要な役割を担うことが判っている。 しかしながら、スフィンゴ脂質変動を細胞膜脂質ラフトレベルで直接観察することは困難であり、機能解析の障壁となっていた。そこで本研究の目的は、スフィンゴ脂質を細胞オルガネラレベルで検出できるMSイメージング法を開発し、脂質ラフトあるいはエクソソーム分泌過程で分布蓄積するスフィンゴ脂質を可視化し、病変部位での変動を捉えることで創薬標的を明らかにすることである。 投影型MSイメージングによるオルガネラレベルでの可視化を実現するためには、各分析条件における高感度化が必須である。R5年度は、主に①投影型MSイメージング分析による細胞オルガネラ由来に含まれる分子の検出、②CyclicIMSによる糖セラミドの分離、③導電性粘着フィルムの高感度化、④Phostag誘導体化試薬の合成および⑤細胞外小胞に含まれるスフィンゴ脂質分析の最適化を実施した。今後開発した条件の再現性を評価し、全ての技術を組み合わせることにより、オルガネラレベルにおけるスフィンゴ脂質の可視化に挑戦する。

    The purpose is to develop a new disease diagnostic method by modified nucleosides profile using LC-MS/MS. Modified nucleosides for standard and its stable isotopes for internal standard were synthesized in this study. As a result, it was possible to prepare a standard curve for modified nucleosides, which could not be quantified before, and to achieve precise quantification in human plasma. The constructed quantitative system was applied to lung cancer patient plasma samples. A logistic regression analysis of the modified nucleoside quantification values showed a superior diagnostic model compared to existing lung cancer biomarkers. This quantitative system for modified nucleosides with high comprehensiveness and high throughput is expected to play an important role in biomarker research and elucidation of pathology.

    Sphingosine monophosphate (S1P) is a bioactive lipid and has been implicated in the pathogenesis of immunological diseases, cancer, and atherosclerosis. S1P and its related molecules expressed in their signaling pathway are considered potential drug targets. However, it has been difficult to visualize the tissue distribution of S1P using conventional analytical methods. In this study, we developed a S1P-specific detection method and a highly sensitive and high-resolution tissue section by MS imaging. We constructed a library of Phostag derivatization reagents by synthesizing novel Phostag derivatization reagents, and succeeded in highly sensitive detection of S1P.

    Enzymatic evaluation methods and enzymatic evaluation methods for compounds that inhibit or increase the enzymatic activity of CD73 or CD39 for the small molecule discovery of novel cancer immunotherapy targeting the PD-1 / PD-L1 independent immunosuppressive pathway. As a result of screening and evaluating 6,080 species of the Tohoku University compound library using the evaluation method using mouse spleen cells, 19 promising seed candidate compounds were obtained. We succeeded in obtaining three compounds selective for CD73 from the enzyme reaction curves of one of the 13 analog compounds.

    Cyclic ADP ribose (cADPR) is the nicotinamide adenine dinucleotide (NAD) major metabolite. cADPR is an important messenger molecule that has been pointed out to be involved in various life phenomena. For example, oxytocin release, insulin secretion, inflammatory response, and even stem cell self-renewal. In this study, we focused on intracellular cADPR, which could not be quantified due to the extremely small amount in the previous metabolome studies, and constructed a highly sensitive and accurate quantification method for cADPR using LC / MS. A stable isotope of cADPR was synthesized as an internal standard and a quantitative system of cADPR was constructed. Sensitivity was increased 1,000 times more than previously reported, and sufficient sensitivity was achieved to quantify intracellular cADPR.

    Cancer immunotherapy with PD-1 antibody has been applied in clinical settings. Because pathological conditions are differences in each cancer patients, it is expected to develop an evaluation for different immune suppression mechanism such as PD-1 and/or PD-L1. In this study, we focus on modified nucleosides and nucleotides that have long been implicated in cancer pathologies, but whose pharmacokinetics and immune function is still unknown in many ways. The quantitative, qualitative, accurate, and comprehensive methods were developed in this study, We also found that nucleic acids such as methylated adenosine suppress T cell activation, suggesting that modified nucleic acids may be involved in cancer immunosuppression. This result indicates the possibility that cancer immunopathology can be grasped and treated based on a new standard different from the past, and it can be expected to contribute to the further development of cancer treatment.

    In this study, we aimed to identify the molecules involved in radioresistance of cancer cells and to elucidate its mechanism. Here, we quantitatively compared the proteomes and the metabolomes of clinically relevant radioresistant (CRR) cell lines and their parent strains. In CRR cells, the chaperones and the proteins involved in oxidative stress were higher than control cell lines. Pathways such as amino acid metabolism and fatty acid metabolism were found to be highly enriched in CRR cells. These molecules can be expected as radiation tolerance markers and/or new targets for cancer therapies.

    Theoretical calculations have been employed to elucidate pi-deficient system Aromatic Nucleophilic Substitution and Meisenheimer Complexes. The computations suggest that the Meisenheimer complexes don't exist as intermediate structures except for the case that it was special. As a result of the reaction pathway analysis, it was found that SNAr proceeds via a transition structure, not via Meisenheimer complexes. Thus, the classical interpretation to determine the reactivity and regioselectivity by the stability of intermediates is denied, and it is thought that an argument is necessary for transition structure and activation energy to clarify reactivity and regioselectivity of SNAr.

    Function, expression, stability and cellular localization of proteins are determined by their three dimensional structure. Therefore, control of the conformation of proteins results on regulation of their function/stability/localization. Abnormal conformation of specific proteins results in various incurable diseases. This research projects resulted in creation of: (1) various nuclear receptor ligands which control the function of the receptors by regulating their conformation, (2) small molecules (pharmacological chaperons) which correct abnormal trafficking of mutated proteins (ex. mutated rhodopsin which causes retinitis pigmentosa, NPC1 which caused Niemann-Pick type 1 disease), and (3) small molecules (SNIPERS) which induce degradation of target proteins in living cells.
