    Abstract Taurine, biosynthesized from methionine or cysteine in the liver, plays a crucial regulatory role in bile acid conjugation, antioxidant effects, and glucose and cholesterol metabolism. This may influence the metabolic changes associated with fat accumulation in beef cattle. However, the physiological role of taurine in this species has not been fully elucidated. In this study, we explored the physiological role of taurine in Japanese Black steers (Bos taurus) in different phases during the fattening period. To examine the correlation among plasma taurine concentrations, various physiological parameters, and genes related to taurine synthesis in the liver, we used biopsied liver tissues, blood samples, and rumen fluids collected from 21 steers at three different stages, i.e., early (T1; 13 months of age), middle (T2; 20 months of age), and late (T3; 28 months of age) phases. Additionally, to investigate the regulatory mechanisms underlying the expression profile of taurine synthesis genes, primary bovine hepatocytes obtained from 4-week-old Holstein calves were treated with palmitate, oleate, acetate, propionate, or β-hydroxybutyrate (BHBA). Plasma taurine and cholesterol concentrations significantly (P < 0.001) increased in the T2 phase, which is potentially attributable to increased energy intake and assimilation induced by increased intake of concentrated feed. Cysteine sulfinic acid decarboxylase (CSAD) expression significantly increased (P < 0.01) in T2 than in other phases. The expression levels of cysteine dioxygenase type 1 (CDO1) and cholesterol 7 alpha-hydroxylase (CYP7A1) were significantly higher (P < 0.001) in T2 than in T3; moreover, the CDO1/glutamate-cysteine ligase catalytic subunit (GCLC) ratio was higher (P < 0.05) in T2 than in T1. Plasma taurine concentrations were positively correlated with plasma methionine (r = 0.51; P < 0.05) and total cholesterol (r = 0.56; P < 0.05) concentrations at T2. Relative CDO1 mRNA expression was upregulated in cultured bovine hepatocytes treated with oleate and propionate, whereas it was downregulated upon acetate treatment. These findings indicate that the increase in plasma taurine concentrations in the T2 phase is associated with changes in lipid and methionine metabolism in Japanese Black steers.

    Abstract In Japan, Japanese Black cattle, known for their exceptional meat quality owing to their abundant intramuscular fat, undergo a unique three-stage feeding system with varying concentrate ratios. There is limited research on physiological and rumen microbial changes in Japanese Black cattle during these stages. Therefore, this study aimed to examine Japanese Black steers in these three stages: early (T1, 12–14 months), middle (T2, 15–22 months), and late (T3, 23–30 months). The rumen bacteria of 21 cattle per phase was analyzed using 16S rRNA gene sequencing. Rumen bacterial diversity was significantly higher in T1, with a distinct distribution, than in T2 and T3. Specific phyla and genera were exclusive to each stage, reflecting the shifts in feed composition. Certain genera dominated each stage: T1 had Flexilinea, Streptococcus, Butyrivibrio, Selenomonas, and Kandleria; T2 had Bifidobacterium, Shuttleworthia, and Sharpea; and T3 had Acetitomaculum, Mycoplasma, Atopobium, and Howardella. Correlation analysis revealed significant associations between certain microbial populations and physiological parameters. These findings indicate that changes in energy content and feed composition are associated with physiological and ruminal alterations. This study may guide strategies to improve rumen health and productivity in Japanese Black cattle by modifying diets to specific fattening stages.

Sections cover livestock methane emissions, the potential of AI technology, data collection issues, methane’s significance in carbon credit schemes, and current research and innovation. The review emphasizes the critical role of accurate measurement and estimation methods for effective climate change mitigation and reducing methane emissions from livestock operations. Overall, it provides a comprehensive overview of methane emissions in the livestock industry by synthesizing existing research and literature, aiming to improve knowledge and methods for mitigating climate change. Livestock-generated methane, especially from cattle, is highlighted as a crucial factor in climate change, and the review underscores the importance of integrating precise measurement and estimation techniques for effective mitigation.

    High grain feeding or weaning, which could compromise the rumen epithelium by increasing ruminal short-chain fatty acid (SCFA) concentrations with pH reduction, is associated with high levels of ruminal toll-like receptor 5 (TLR5). This study aimed to determine the role of TLR5 in the rumen epithelium. Immunohistochemistry revealed that TLR5 was localized in cells on the basal side (i.e., basal and spinous layers) rather than in the granular layer in the rumen epithelium, where tight junctions are most potent, in pre- and post-weaning calves (n = 9). Primary bovine rumen epithelial cells (BRECs) obtained from Holstein cows (n = 3) were cultured to investigate the factors that upregulate TLR5; however, SCFA, low pH (pH 5.6), BHBA, L-lactate, D-lactate, and LPS did not upregulate TLR5 gene expression in BREC. Primary BREC treated with flagellin (TLR5 ligand) had higher expression of interleukin-1β (IL-1β) (P < 0.05) than BREC treated with vehicle. In addition, BREC treated with IL-1β had higher expression of antimicrobial peptides and C-X-C motif chemokine ligand 8 than BREC treated with vehicle (P < 0.05). These results suggest that ruminal TLR5 may recognize epithelial disruption via flagellin and mediate the immune response via IL-1β during high-grain feeding or weaning.

  7. Effect of residual methane emission on physiological characteristics and carcass performance in Japanese Black cattle

    Abstract This study investigated the physiological characteristics and carcass performance associated with residual methane emissions (RME), and the effects of bull differences on CH4‐related traits in Japanese Black cattle. Enteric methane (CH4) emissions from 156 Japanese Black cattle (111 heifers and 45 steers) were measured during early fattening using the sniffer method. Various physiological parameters were investigated to clarify the physiological traits between the high, middle, and low RME groups. CH4‐related traits were examined to determine whether bull differences affected progeny CH4 emissions. Ruminal butyrate and NH3 concentrations were significantly higher in the high‐RME group than in the low‐RME group, whereas the propionate content was significantly higher in the low‐RME group. Blood urea nitrogen, β‐hydroxybutyric acid, and insulin concentrations were significantly higher, and blood amino acids were lower in the high‐RME group than in the other groups. No significant differences were observed in the carcass traits and beef fat composition between RME groups. CH4‐related traits were significantly different among bull herds. Our results show that CH4‐related traits are heritable, wherein bull differences affect progeny CH4 production capability, and that the above‐mentioned rumen fermentations and blood metabolites could be used to evaluate enteric methanogenesis in Japanese Black cattle.

    This study investigated the relationship between rumen fermentation, stress-related hormones, and behavior in sheep with the aim of providing insights for improving animal welfare and feed management practices. Eight lambs were assigned to either a high concentration or control group. Blood samples were collected for hormone analysis, and an open field test was conducted to observe behavioral stress responses. The results showed that diet composition may affect the behavior of ruminants in response to stressors and novel situations, as evidenced by the higher number of escape attempts in the high-concentration group. In addition, analyses of individual volatile fatty acids (VFAs) showed a significant positive correlation between the acetic acid/propionic acid ratio and sniffing behavior of the novel object (p &lt; 0.05, ρ = −0.414). These findings have important implications for animal welfare and feed management practices. Overall, this study provides insights into the potential impact of diet composition on the behavior of ruminants in response to stressors and novel situations, highlighting the importance of improving animal welfare through feed management practices. Further research is needed to fully elucidate the mechanisms underlying the complex relationship between rumen fermentation, stress-related hormones, and behavior in ruminant animals.

    Angiopoietin-like protein 3 (ANGPTL3) is expressed predominantly in the liver and plays a major role in regulating the circulating triglyceride and lipoprotein fraction concentrations by inhibiting lipoprotein lipase (LPL) activity. Given these physiological roles, ANGPTL3 may play an important role in metabolic changes related to fat accumulation during the fattening period in Japanese Black. This study aimed to reveal the physiological roles of hepatic ANGPTL3 in Japanese Black steers (Bos taurus) during the fattening period and investigate the regulatory effects of hepatic ANGPTL3. To investigate the gene expression and protein localization of ANGPTL3, 18 tissue samples were collected from tree male Holstein bull calves aged 7 weeks. Biopsied liver tissues and blood samples were collected from 21 Japanese Black steers during the early (T1; 13 months of age), middle (T2; 20 months), and late fattening phases (T3; 28 months). Relative mRNA expression, blood metabolite concentrations, hormone concentrations, growth, and carcass traits were analyzed. To identify the regulatory factors of hepatic ANGPTL3, primary bovine hepatocytes collected by 2 Holstein calves aged 7 weeks were incubated with insulin, palmitate, oleate, propionate, acetate, or beta-hydroxybutyric acid (BHBA). The ANGPTL3 gene was most highly expressed in the liver, with minor expression in the renal cortex, lungs, reticulum, and jejunum in Holstein bull calves. In Japanese Black steers, relative ANGPTL3 mRNA expressions were less as fattening progressed, and blood triglyceride, total cholesterol, and non-esterified fatty acid (NEFA) concentrations increased. Relative ANGPTL8 and Liver X receptor alpha (LXRα) mRNA expressions decreased in late and middle fattening phases, respectively. Furthermore, relative ANGTPL3 mRNA expression was positively correlated with ANGPTL8 (r = 0.650; P < 0.01) and ANGPTL4 (r = 0.540; P < 0.05) in T3 and T1, respectively, and LXRα showed no correlation with ANGPTL3. Relative ANGTPL3 mRNA expression was negatively correlated with total cholesterol (r = -0.434; P < 0.05) and triglyceride (r = -0.645; P < 0.01) concentrations in T3 and T1, respectively; There was no significant correlation between ANGTPL3 and carcass traits. Relative ANGTPL3 mRNA expression in cultured bovine hepatocytes was downregulated in oleate treatment. Together, these findings suggest that ANGPTL3 downregulation in late fattening phases is associated with the changes in lipid metabolism.

    This study aimed to identify the growth performance and blood factors associated with carcass weight in Japanese Black calves based on 675 performance tests and field carcass records. We measured the body weight, withers height, and chest girth at the start of fattening age (approximately 8–10 months) and analyzed eight blood factors, including vitamins and metabolites. Single- and two-trait animal models were used to estimate the heritability and genetic correlations. The heritability estimates for growth performance were moderate to high (ranging from 0.48 to 0.74), and those for blood metabolites were low to moderate (ranging from 0.19 to 0.51). Estimates for genetic correlations of carcass or body weight with body weight, withers height, and chest girth were high (ranging from 0.42 to 0.80). The body weight and withers height at 8 months of age are possibly closely related to the final carcass weight. The blood metabolites associated with body weight were vitamin E in steers (castrated males) and β-carotene in heifers. Our findings indicate that body measurements and blood metabolites measured during the growing period could be used to determine the nutritional and physiological status of cattle as well as predict carcass weight.

    Abstract In this study, using enteric methane emissions, we investigated the metabolic characteristics of Japanese Black cattle. Their methane emissions were measured at early (age 13 months), middle (20 months), and late fattening phases (28 months). Cattle with the highest and lowest methane emissions were selected based on the residual methane emission values, and their liver transcriptome, blood metabolites, hormones, and rumen fermentation characteristics were analyzed. Blood β-hydroxybutyric acid and insulin levels were high, whereas blood amino acid levels were low in cattle with high methane emissions. Further, propionate and butyrate levels differed depending on the enteric methane emissions. Hepatic genes, such as SERPINI2, SLC7A5, ATP6, and RRAD, which were related to amino acid transport and glucose metabolism, were upregulated or downregulated during the late fattening phase. The above mentioned metabolites and liver transcriptomes could be used to evaluate enteric methanogenesis in Japanese Black cattle.

    We investigated the physiological changes during the fattening period and production characteristics in Japanese Black steers bred and raised using the typical feeding system in Japan. Here, 21 Japanese Black steers aged 12 months were used, with experimental period divided into early (12-14 months of age), middle (15-22 months), and late fattening phases (23-30 months). The liver transcriptome, blood metabolites, hormones, and rumen fermentation characteristics were analyzed. Blood triglyceride and non-esterified fatty acid concentrations increased, whereas blood ketone levels decreased, with fattening phases. Blood insulin increased with fattening phases and was positively correlated with carcass weight and marbling in late fattening phases. Rumen fermentation characteristics showed high propionate levels and low butyrate levels in late fattening phases, likely due to increased energy intake. Genes related to glucose metabolism, such as SESN3, INSR, LEPR, and FOXO3, were down-regulated in late fattening phases. Genes related to lipid metabolism, such as FABP4, were up-regulated, whereas FADS1 and FADS2 were down-regulated. These findings suggest that the physiological changes resulted from changes in the energy content and composition of diets. Liver metabolism changed with changes in fat metabolism. Insulin was strongly associated with physiological changes and productivity in Japanese Black cattle.

    This study aimed to understand the mechanisms underlying the effects of maternal undernutrition (MUN) on liver growth and metabolism in Japanese Black fetal calves (8.5 months in utero) using an approach that integrates metabolomics and transcriptomics. Dams were fed 60% (low-nutrition; LN) or 120% (high-nutrition; HN) of their overall nutritional requirements during gestation. We found that MUN markedly decreased the body and liver weights of the fetuses; metabolomic analysis revealed that aspartate, glycerol, alanine, gluconate 6-phosphate, and ophthalmate levels were decreased, whereas UDP-glucose, UDP-glucuronate, octanoate, and 2-hydroxybutyrate levels were decreased in the LN fetal liver (p ≤ 0.05). According to metabolite set enrichment analysis, the highly different metabolites were associated with metabolisms including the arginine and proline metabolism, nucleotide and sugar metabolism, propanoate metabolism, glutamate metabolism, porphyrin metabolism, and urea cycle. Transcriptomic and qPCR analyses revealed that MUN upregulated QRFPR and downregulated genes associated with the glucose homeostasis (G6PC, PCK1, DPP4), ketogenesis (HMGCS2), glucuronidation (UGT1A1, UGT1A6, UGT2A1), lipid metabolism (ANGPTL4, APOA5, FADS2), cholesterol and steroid homeostasis (FDPS, HSD11B1, HSD17B6), and urea cycle (CPS1, ASS1, ASL, ARG2). These metabolic pathways were extracted as relevant terms in subsequent gene ontology/pathway analyses. Collectively, these results indicate that the citrate cycle was maintained at the expense of activities of the energy metabolism, glucuronidation, steroid hormone homeostasis, and urea cycle in the liver of MUN fetuses.

    We hypothesized that maternal low or high nutrition would give unique effects to morphological and molecular dynamics in adipose tissue of fetus of fatty breed Wagyu (Japanese Black) cattle which produce highly marbled beef. This study aimed to determine the effects of maternal energy intake in Wagyu cows, during gestation on fetal adipose tissue development, histochemical properties, and gene and microRNA (miRNA) expression. Cows were allocated to one of two nutritional energy groups: 120% (HIGH) or 60% nutritional requirements of (LOW). Fetuses (n = 6 per treatment) were removed from pregnant cows by cesarean section at fetal age 260 ± 8 days and euthanized. Subcutaneous adipose tissue (SAT), thoracic cavity visceral adipose tissue (TVAT), and perirenal adipose tissue (PAT) were collected for analysis. In histochemical analysis, in SAT and PAT, HIGH fetuses had greater diameter of adipocytes than LOW fetuses (P&amp;lt;0.05). Only in SAT, LOW fetuses had more Leptin (LEP) mRNA and tended to have more Peroxisome Proliferator-Activated Receptor gamma (PPARG) CCAAT-enhancer-binding proteins alpha (CEBPA) and Glucose transporter (GLUT) 4 mRNA(P&amp;lt;0.10). In all SAT, TVAT, and PAT, LOW fetuses had higher levels of the brown adipose tissue (BAT) biomarkers Uncoupling Protein (UCP) 1 and PPARG coactivator (PGC) 1α mRNA than HIGH fetuses (P&amp;lt;0.08). Meanwhile, in the other adipose tissue, LOW fetuses had lower PPARG, CEBPA, and Zinc Finger Protein (ZFP) 423 (in TVAT and PAT), FASN (in TVAT), LEP and GLUT4 mRNA (in PAT; P&amp;lt;0.10). In particular, in TVAT and PAT, LOW fetuses exhibited lower expression of WAT biomarkers (PPARG and ZFP423). Differential expression of various miRNAs related to adipogenesis between the LOW and HIGH fetuses was detected in an adipose tissue-specific manner (P&amp;lt;0.10). Based on adipose tissue-specific effects of maternal nutrition, these findings suggested that poor maternal nutrition in Wagyu cattle increased BAT development in SAT, TVAT and PAT, while elevated maternal nutrition stimulated fetal SAT development compared with that of TVAT and PAT.

    In this study, we examined genetic parameters for feed efficiency, growth, and carcass traits in Japanese Shorthorn cattle, based on 714 performance tests and 15,790 field carcass records. Feed efficiency traits, including residual feed intake (RFI) and residual body weight gain (RG), were calculated. Single-trait and two-trait animal models were used to estimate heritability and genetic correlations. Heritability estimates for feed efficiency traits were found to be low to moderate (ranging from 0.03 to 0.36); notably, heritability was moderate for RG and low for RFI. Estimates for genetic correlations between feed efficiency traits and average daily gain (DG) were favorably moderate to high (absolute values of 0.43-0.85), and those with daily feed intake were low (absolute values of 0.00-0.32). We also estimated a high genetic correlation between RG and DG. The backfat thickness (BF) of bull calves showed favorable or no genetic correlation estimates with feed efficiency and growth traits, whereas RG and BF showed favorable or no genetic correlation estimates with carcass traits. Our findings indicate that genetic improvements in both feed utilization ability and carcass traits could be achieved by utilizing RG and BF in Japanese Shorthorn cattle.

    Epithelial barrier function in the mammary gland acts as a forefront of the defense mechanism against mastitis, which is widespread and a major disorder in dairy production. Chemerin is a chemoattractant protein with potent antimicrobial ability, but its role in the mammary gland remains unelucidated. The aim of this study was to determine the function of chemerin in mammary epithelial tissue of dairy cows in lactation or dry-off periods. Mammary epithelial cells produced chemerin protein, and secreted chemerin was detected in milk samples. Chemerin treatment promoted the proliferation of cultured bovine mammary epithelial cells and protected the integrity of the epithelial cell layer from hydrogen peroxide (H2O2)-induced damage. Meanwhile, chemerin levels were higher in mammary tissue with mastitis. Tumor necrosis factor alpha (TNF-α) strongly upregulated the expression of the chemerin-coding gene (RARRES2) in mammary epithelial cells. Therefore, chemerin was suggested to support mammary epithelial cell growth and epithelial barrier function and to be regulated by inflammatory stimuli. Our results may indicate chemerin as a novel therapeutic target for diseases in the bovine mammary gland.

    To elucidate the mechanisms underlying maternal undernutrition (MUN)-induced fetal skeletal muscle growth impairment in cattle, the longissimus thoracis muscle of Japanese Black fetal calves at 8.5 months in utero was analyzed by an integrative approach with metabolomics and transcriptomics. The pregnant cows were fed on 60% (low-nutrition, LN) or 120% (high-nutrition, HN) of their overall nutritional requirement during gestation. MUN markedly decreased the bodyweight and muscle weight of the fetus. The levels of amino acids (AAs) and arginine-related metabolites including glutamine, GABA, and putrescine were higher in the LN group than those in the HN group. Metabolite set enrichment analysis revealed that the highly different metabolites were associated with the metabolic pathways of pyrimidine, glutathione, and AAs such as arginine and glutamate, suggesting that MUN resulted in AA accumulation rather than protein accumulation. The mRNA expression levels of energy metabolism-associated genes, such as PRKAA1, ANGPTL4, APLNR, CPT1B, NOS2, NOS3, UCP2, and glycolytic genes were lower in the LN group than in the HN group. The gene ontology/pathway analysis revealed that the downregulated genes in the LN group were associated with glucose metabolism, angiogenesis, HIF-1 signaling, PI3K-Akt signaling, pentose phosphate, and insulin signaling pathways. Thus, MUN altered the levels of AAs and expression of genes associated with energy expenditure, glucose homeostasis, and angiogenesis in the fetal muscle.

    Levels of alpha-tocopherol (α-Toc) decline gradually in blood throughout prepartum, reaching lowest levels (hypovitaminosis E) around calving. Despite numerous reports about the disease risk in hypovitaminosis E and the effect of α-Toc supplementation on the health of transition dairy cows, its risk and supplemental effects are controversial. Here, we present some novel data about the disease risk of hypovitaminosis E and the effects of α-Toc supplementation in transition dairy cows. These data strongly demonstrate that hypovitaminosis E is a risk factor for the occurrence of peripartum disease. Furthermore, a study on the effectiveness of using serum vitamin levels as biomarkers to predict disease in dairy cows was reported, and a rapid field test for measuring vitamin levels was developed. By contrast, evidence for how hypovitaminosis E occurred during the transition period was scarce until the 2010s. Pioneering studies conducted with humans and rodents have identified and characterised some α-Toc-related proteins, molecular players involved in α-Toc regulation followed by a study in ruminants from the 2010s. Based on recent literature, the six physiological factors: (1) the decline in α-Toc intake from the close-up period; (2) changes in the digestive and absorptive functions of α-Toc; (3) the decline in plasma high-density lipoprotein as an α-Toc carrier; (4) increasing oxidative stress and consumption of α-Toc; (5) decreasing hepatic α-Toc transfer to circulation; and (6) increasing mammary α-Toc transfer from blood to colostrum, may be involved in α-Toc deficiency during the transition period. However, the mechanisms and pathways are poorly understood, and further studies are needed to understand the physiological role of α-Toc-related molecules in cattle. Understanding the molecular mechanisms underlying hypovitaminosis E will contribute to the prevention of peripartum disease and high performance in dairy cows.

    The Japanese Shorthorn is a Japanese Wagyu breed maintained at a small population size. We assessed the degree of inbreeding and genetic diversity among Japanese Shorthorn cattle using pedigree analysis. We analyzed the pedigree records of registered Japanese Shorthorn born between 1980 and 2018, after evaluating the pedigree completeness. The average of the actual inbreeding coefficients increased at the same rates annually from approximately 1.5% in 1980 to 4.2% in 2018 and was higher than the expected inbreeding coefficients over time. The effective population size based on the individual coancestry rate largely decreased from 127.8 in 1980 to 82.6 in 1999, and then remained almost constant at approximately 90. Three effective numbers of ancestors decreased over time until 1995, then remained almost constant. In particular, the effective number of founder genomes (Nge ) decreased from 43.8 in 1980 to 11.9 in 2018. The index of genetic diversity based on Nge decreased from 0.99 in 1980 to 0.96 in 2018 due to genetic drift in non-founder generations. Changes in inbreeding and genetic diversity parameters were similar between Japanese Shorthorn and other Japanese Wagyu breeds, but the magnitude of the changes was lower in the Japanese Shorthorn.

    This study reports findings from the pathological examination of the forestomach of an 11-month-old Japanese Black steer with severely retarded growth (41% of expected weight) and chronic ruminal tympany. The ruminal papillae were weakly formed (0.3-0.5 cm long) and unevenly distributed. The cellulae and cristae reticuli were underdeveloped; the cristae were 0.4-0.7 cm in height and milky white. The keratinized layer in the stratified squamous epithelium was thickened. Ruminal pH was 5.25, and ruminal volatile fatty acid concentration was 11.7 mM. The steer's severely retarded growth was considered to be caused by malnutrition due to developmental and functional failure of the forestomach.

    Bovine leukemia virus (BLV) is a global problem that results in significant economic losses to the livestock industry. We developed three virus strains by inserting the HiBiT reporter tag from NanoLuc luciferase (NLuc) into limited sites within BLV molecular clones. Initial analysis for site selection of the tag insertion revealed a permissible site immediately downstream of the viral envelope gene. Therefore, NLuc activity could be used to measure virus copy numbers in the supernatant and the levels of cell infection. Productivity and growth kinetics of the reporter virus were similar to those of the wild-type strain; therefore, the reporter virus can be used to characterize the replication of chimeric viruses as well as responses to the antiviral drug, amprenavir. Collectively, our results suggest that the BLV reporter virus with a HiBiT tag insertion is a highly versatile system for various purposes such as evaluating virus replication and antiviral drugs.

    This study was conducted to investigate the effect of HS on HSPs gene expression in bovine PBMCs of beef calves in in vitro and in vivo models. In the in vitro experiment, blood samples were collected from the jugular vein of five beef calves (age: 174.2 ± 5.20 days, BW: 145.2 ± 5.21 kg). In the in vivo experiment, sixteen Korean native male beef calves (age: 169.6 ± 4.60 days, BW: 136.9 ± 6.23 kg) were exposed to ambient temperature for seven days (22 to 24 °C, relative humidity 60%; temperature–humidity index (THI) = 68 to 70) and subsequently to the temperature and humidity corresponding to the target THI level for 21 days (HS). For PBMC isolation, blood samples were collected every three days. In the in vitro model, the cell viability was significantly decreased in HS groups compared with the control group (p = 0.015). The expression of HSP70 (p = 0.022), HSP90 (p = 0.003) and HSPB1 (p = 0.026) genes was increased in the HS group in in vitro model. In the in vivo experiment, the HSP70 gene expression was increased after sudden exposure to HS conditions (severe THI levels; THI = 88 to 90), whereas HSP90 and HSPB1 showed no differences among the THI groups (p &gt; 0.05). However, in the severe THI group, the HSP70 gene expression returned to normal range after six days of continuous HS. In conclusion, the HSP70 gene plays a pivotal role in protecting cells from damage and is sensitive to HS in immune cells compared with other HSP genes in in vitro and in vivo models. In addition, the in vivo models suggest that calves exhibit active physiological mechanisms of adaptation to HS after six days of continuous exposure by regulating the HSP70 gene expression.

    This study was conducted to evaluate the effects of supplementation of Wakame seaweed stalks on the immunity and intestinal microflora of pigs. Three separate experiments were performed: Relatively young (start at 20-30 kg; Experiments 1 and 2) and fattening period (70 kg; Experiment 3). All pigs (including the control group) were fed the same commercial feed, free from antibiotic additives, but in the feed for the treatment groups, 1% seaweed powder was added. There were no group differences observed in daily weight gain and feed intake in Experiments 1 and 2 between groups; however, daily weight gain was significantly higher in the treatment group compared to the control group in Experiment 3. The percentage of peripheral blood natural killer cells of the treatment group was significantly higher than that of the control group in all experiments. Although addition of seaweed changed the gene expression of cytokine and toll-like receptors of the small intestinal Peyer's patches slightly, seaweed seems to alter intestinal microflora preferentially, for instance, there was an increase in Lactobacillus and a decrease of Escherichia coli observed. These results suggest that Wakame seaweed can be used as supplement for pig feed to improve the gut health and immunity of pigs.

    This study aimed to characterize the relationship between the growth of rumen papillae in calves and the mRNA expression of insulin-like growth factor-binding proteins (IGFBPs) in the rumen papillae. The length of rumen papillae, the mRNA expression of IGFBPs in rumen papillae by quantitative real-time PCR, and the presence of insulin-like growth factors I and II (IGF-I and II) by immunohistochemistry (IHC) were analyzed in nine Holstein calves divided into three groups: suckling (2 weeks, n = 3), milk-continued (8 weeks, n = 3), and weaned (8 weeks, n = 3). The length of rumen papillae was greater (p < 0.01) in weaned calves than in suckling and milk-continued calves, whereas the expressions of IGFBP2, IGFBP3, and IGFBP6 genes were lower (p < 0.05) in the rumen papillae of weaned calves than in milk-continued calves. Thus, rumen papillae length and IGFBP2, 3, and 6 expressions were negatively correlated. The IHC analysis showed that IGF-I and IGF-II were present in the rumen epithelium of calves. These results suggested that the growth of rumen papillae after weaning is associated with the induction of IGFs by the low levels of IGFBP2, IGFBP3, and IGFBP6.

    The aims of the present study were to identify the differences between two mouse lines (high (H)- and low (L)-oxygen consumption) in terms of mitochondrial respiratory activity when GMP (glutamate, malate, and pyruvate) and succinic acid are used as substrates and to examine the relationship between mitochondrial respiration activity and feed efficiency in both lines. The average daily feed intake, feed conversion ratio (FCR), and residual feed intake (RFI) were significantly higher in the H than the L line. The correlation between FCR and RFI was significant (r = 0.60, p < 0.05). RFI was effective as an indicator of feed efficiency. When succinic acid was used as a substrate, mitochondrial respiration states 2-4, ACR, and proton leak were significantly higher in the H than the L line. When GMP was used as a substrate, respiration states 3 and 4 in the H line were significantly higher than those in the L line, and there were significant positive correlations between FCR and RFI and mitochondrial respiration states 2-4. The results indicated that selection for high or low OC changed the basal metabolic rates estimated from liver mitochondrial respiration activity and feed efficiency.

    The aim of this study was to evaluate butyrate supplementation of antibiotic-free milk replacer and starter on growth performance in male Holstein calves. Twenty-nine calves were divided into two groups. Group C (n = 13) was fed antibiotic-free milk replacer without supplementation, and Group B (n = 16) was fed antibiotic-free milk replacer supplemented with butyrate (1.6 % DM of Gustor BP70® ). Starter in Group B contained 0.3 % DM of Gustor BP70® . The intake of milk replacer was lower in group B than in C (p = 0.07 for the treatment x week interaction). Body weight (BW) and heart girth (HG) in group B was higher than in C during the experimental period (p = 0.07 and 0.01 for the treatment × week interaction, respectively). The duration of the weaning period in group B was shorter than in group C (p = 0.02). β-hydroxybutyrate (BHBA) was higher in group B than in C (p = 0.04). Insulin like growth factor-1 (IGF-1) concentrations tended to be higher in group B than in C (p = 0.07 for treatment × week interaction). Our results show that butyrate supplementation in antibiotic-free milk replacer and starter exerted positive effects on growth performance in suckling calves.

    Acute physiological adaptation of lipid metabolism during the postpartum transition period of cows facilitates peripheral metabolic regulation. Hepatokines, which are hormones secreted from hepatocytes, are presumed to play a critical role in systemic metabolic regulation. Angiopoietin-like protein 8 (ANGPTL8) has been identified as a novel hepatokine associated with circulating triglyceride concentrations in mice and humans. However, regulation of ANGPTL8 and its physiological effects is still unknown in cattle. The present study aimed to reveal changes in ANGPTL8 expression and secretion during the periparturient period, and to investigate its regulatory effect on adipocytes and mammary epithelial cells. In the peripartum period, liver ANGPTL8 mRNA expression was lesser on the day of parturition and 1 wk postpartum than it was 1 wk before parturition (P < 0.05). Moreover, plasma ANGPTL8 concentrations decreased on the day of parturition as compared with that 1 wk before parturition (P < 0.05). In addition, ANGPTL8 expression in cultured bovine hepatocytes was downregulated after oleate and palmitate treatment but upregulated after insulin treatment (P < 0.05). ANGPTL8 decreased hormone-sensitive lipase (HSL) expression in differentiated adipocytes and cluster of differentiation 36 (CD36), fatty acid synthase (FAS), acetyl-coa carboxylase (ACC), and stearoyl-coa desaturase (SCD) in cultured bovine mammary epithelial cells (P < 0.05). These data suggest that hepatic ANGPTL8 production was downregulated postpartum when the cows experienced a negative energy balance. This downregulation was associated with increased concentrations of NEFA and decreased concentrations of insulin in lactating cows, and it facilitated lipid mobilization from adipose tissue to the mammary glands. We speculate that ANGPTL8 might have beneficial effects in reverting or improving the physiological adaptation and pathological processes of lipid metabolism during the peripartum period.

    The length and density of rumen papillae starts to increase during weaning and growth of ruminants. This significant development increases the intraruminal surface area and the efficiency of VFA (acetate, propionate, butyrate, etc.) uptake. Thus, it is important to investigate the factors controlling the growth and development of rumen papillae during weaning. This study aimed to compare the transcriptomes of rumen papillae in suckling and weaned calves. Total RNA was extracted from the rumen papillae of 10 male Japanese Black calves (5 suckling calves, 5 wk old; 5 weaned calves, 15 wk old) and used in RNA-sequencing. Transcript abundance was estimated and differentially expressed genes were identified and these data were then used in Ingenuity Pathway Analysis (IPA) to predict the major canonical pathways and upstream regulators. Among the 871 differentially expressed genes screened by IPA, 466 genes were upregulated and 405 were downregulated in the weaned group. Canonical pathway analysis showed that "atherosclerosis" was the most significant pathway, and "tretinoin," a derivative of vitamin A, was predicted as the most active upstream regulator during weaning. Analyses also predicted IgG, lipopolysaccharides, and tumor-necrosis factor-α as regulators of the microbe-epithelium interaction that activates rumen-related immune responses. The functional category and the up-regulators found in this study provide a valuable resource for studying new candidate genes related to the proliferation and development of rumen papillae from suckling to weaning Japanese Black calves.

    Economic loss of agricultural and forestry resources has overwhelmingly increased in recent decades due to feeding damage of Japanese Deer (Cervus nippon centralis) in Japan. Utilization of a captured deer is becoming critical issue in relation to a management of the deer population. In this research, sensory research was carried out in order to investigate the consumer palatability for the deer meat processed by a sweetener (Trehalose and Sucrose) or seaweed (Wakame seaweed and Kombu). The deer meat was investigated by panel test which consisted of 98 ordinary consumers. Sensory research was carried out using a questionnaire about taste, aroma, toughness, greasiness, juiciness and aggregate score of the meat, where each question was made of five choices. The deer meat processed by Wakame was evaluated significantly higher than the deer meat of no processing in all evaluation items. Interaction between the sex and the preference for deer meat was significant in taste score.

    Sensory research was carried out in order to investigate the consumer palatability ofgoat meat. One of traditional Okinawa dish, soup of goat meat was investigated by panel test. The sensoryresearch was conducted by 123 ordinary consumers (male: 83, female: 40). The survey research was carriedout based on a questionnaire about taste, aroma, toughness, and greasiness of the meat, where each questionwas made of five choices. Factors related to profile of the panelists were sex, age, place of origin and preferenceof the panelists were investigated. In toughness and greasiness, female panelists tended to evaluatethe meat more tough and greasy than male panelists (P < 0.05). In toughness, the panelists in age of 21-60class tended to evaluate the meat more tough than panelists of under 20 (P < 0.05). In aroma and aggregatescore, local panelists tended to evaluate the meat stronger smell than visitors (P < 0.05). This survey studyreveals preference of the panelists for goat soup is the most important factor to make favor/disfavor decision.This tendency was especially significant in evaluation of aroma, where more local panelists tend to favor thearoma of goat soup.

    Isolated adipocytes were used to evaluate the lipogenic activity in subcutaneous adipose tissue of Holstein steers during various fattening periods. During the fattening period, both the mean diameter of adipocytes and the lipogenesis increased continually. There was a positive correlation between the mean diameter of adipocytes and the lipogenesis from glucose (r =0.74, P<0.05) or from acetate (r=0.81, P<0.05). Further, the relationship between the size of adipocyte and the lipogenic activity was investigated. It can be done through measuring the incorporation rate of substrates into lipids in each fraction of adipocytes which were separated by meshes. It was shown that both the incorporation rate of glucose or acetate into lipids and the relative contribution ratio of incorporation rate of acetate to glucose in the adipocytes taken from finishing steers, were significantly larger (P<0.05) than those from the early fattening steers. In the adipocytes which were taken from early fattening steers, the incorporation rate of glucose or acetate into lipids became larger with increment of the mean diameter from fraction 1 (33μm) to fraction 6 (139μm) (both: r=0.72, P<0.05). In the adipocytes which were taken from finishing fattening steers, the incorporation rate of glucose into lipids increased with the increment of the mean diameter from fraction 1 (36μm) to fraction 6 (164μm) (r=0.79, P<0.05), while the incorporation rate of acetate into lipids increased only with increment of the mean diameter from fraction 1 (36μm) to fraction 4 (116μm), then decreased with the increment of the mean diameter from fraction 4( 116μm) to those very large adipocytes in fraction 6 (164μm).

    The fatty acid composition and the diameter of subcutaneous adipocytes from 13 Holstein steers at different fattening stages were investigated. The mean diameter of adipocytes from individual steers was positively (P<0.01) correlated with the percentage of myristoleic acid (14:1)(r=0.91), palmitoleic acid (16:1)(r=0.83) or oleic acid (18:1)(r=0.82), and negatively(P<0.01)correlated with the percentage of stearic acid (18:0)(r=-0.89). The isolated adipocytes from the same finishing steer (n=5)were then separated by a 50μm mesh. The large adipocytes which remained on the mesh (118μm mean diameter)contained more (P<0.01) myristoleic acid (14:1)palmitoleic acid (16:1) and oleic acid (18:1), and less (P<0.01) palmitic acid(16:0), stearic acid (18:0) than the small adipocytes which were filtered through (29μm mean diameter). It is suggested that the growth of adipocyte is one of the important determinants of fatty acid composition in subcutaneous adipocytes of Holstein steers throughout the fattening period.

    2021年は、以下の解析を実施し、雄体内における精子の品質制御メカニズムの解析技術の開発を進めた。まず、精子の前駆細胞の膜上に時期特異的に蛍光標識を入れるためのCreERマウスの作製を進めた。既報のRNAシークエンス解析のデータセットから分化細胞で特異的に発現する遺伝子を1つ選択し、その発現を免疫組織化学染色で確認した後、同遺伝子のプロモータの下流にCreERを導入したベクターを作製後、培養下の胚性幹(ES)細胞内での相同組換えを行った。得られた組換えES細胞を正常型の胚盤胞にインジェクションし、現在までに毛への高い寄与が確認されたキメラを2系統作成し、F1を得るための交配試験を実施している。キメラマウスについて、生殖系列へも高い寄与が予想されるが、系統数が少なく、うまくF1が得られない場合に備え、追加のキメラ作製も検討中である。次に、Creによる組換えによって特異的に精子の前駆細胞を蛍光標識できるレポータマウスを検討した。具体的には、生殖細胞の細胞膜に蛍光タンパク質を取り込める既報のレポーターマウスを第一候補として導入した。上述のCreERマウスおよびレポーターマウスを掛け合わせることで、2022年に精子品質制御機構解析のモデル動物を開発する。なお、今後の解析に必要なイメージング技術の改良および内分泌影響の予備検討も実施した。以上の研究は、雄体内における精子の品質のばらつきの発生要因解析に資する技術基盤であり、この利活用を通じて野生動物や産業動物の雄繁殖能制御に資する成果が得られることが期待される。

    This study was to clarify the interaction between the immune response, homeostasis, and the host immune system that occur in rumen epithelium. The results are as follows:(1) CCL5 elevated after weaning is thought to induce monocyte migration and differentiation into macrophages, and the CCL5-monocyte / macrophage axis may play an important role in maintaining homeostasis in rumen epithelium. (2) In the rumen epithelium, IL-1β contributes to the maintenance of homeostasis when SCFA, LPS, flagellin, etc. increase in the rumen due to weaning and the environment by the environment changes in rumen, and in addition it is suggested to contribute to the repair of damage by promoting cell proliferation.

    This study was aimed to identify the genes associated with the development of the rumen epithelium between suckling and weaned Japanse Black cattle. Digital differential display analysis in silico can be useful for screening candidate genes related to the rumen development, and 3-hydroxy-3-methylglutaryl-CoA synthase 2 (HMGCS2), aldo-keto reductase family 1, member C1-like (AKR1C1), and fatty acid binding protein 3 (FABP3) showed significant changes in the levels of gene expression in the rumen between suckling and weaned calves. In addition, RNA seq analysis that 871 genes in rumen papillae of suckling and weaned calves by RNA seq analysis were changed. IPA analysis showed that the production of ketone bodies were increased and PPAR was activated after weaning.

    This study was aimed to elucidate the physiological function of chemerin in the metabolism transitional period of weaning and lactating dairy cattle. Protein expression of Chemerin in the liver was reduced after weaning, but chemerin regulated the gluconeogenesis by propionic acid in the liver without the regulation of insulin secretion in the weaning period. Chemerin gene expression and secretion was reduced in liver of dairy cows after delivery, but this change is directly related with the regulation hepatic gluconeogenesis. This results suggest that the expression and secretion of chemerin in cattle dynamically changes the metabolism transition and revealed to be involved in the regulation of lipid metabolism or glucose metabolism.

    This study was to conduct the fattening evaluation by characterizing the fat accumulation and endocrine regulation in single nucleotide polymorphism of bovine growth hormone (bGH). (1) GH gene polymorphism changed the oleic acid composition in the intramuscular fat by regulating the SCD-1. (2) Response to GH stimulation was different depending on the GH genotype. (3)Insulin has an action to suppress the secretion of GH acts directly on the anterior pituitary, GH secretion is increased by changes in stress and NEFA concentrations caused by the sharp reduction in blood glucose levels in vivo. These results indicate that endocrine regulations such as insulin may different by GH polymorphism of Japanese Black Cattle, affecting the meat quality and production due to changes in fat accumulation.

    In this study, we applied the induced pluripotent stem (iPS) cell technology to the livestock science. We focused on the porcine, one of the major livestock, and established the porcine derived iPS cell. We obtained porcine derived iPS cells with the expression of six reprogramming factors, including Oct3/4, Klf4, Sox2, c-Myc. The porcine derived iPS cell had different nature from that of human, and it showed the growth dependency to the LIF (leucocyte migration inhibition factor), and low molecular inhibitors. Interestingly, porcine derived iPS did not cause the massive apoptosis even after the enzymatic digestion into the single cell, which nature is different from human. Furthermore, the re-activation of X chromosome was observed in female derived porcine iPS cells. We succeeded the high quality porcine derived iPS cell, which is close to the nature of early embryonic stage.

    The aim of this study was to investigate 1) the expression of OXTR in adipose tissues and possible role in regulating lipid metabolism, and 2) the expression of OXTR during the differentiation of mammary epithelial cells. These results suggest 1) that up-regulation of OXTR during fat accumulation and adipocyte development may intensify the catabolic effects by oxytocin in adipose tissue to cause lipolysis, 2) OXTR expressionwas regulated by adiponectin and lactogenic hormones in bovine mammary epithelial cells.

    This study was to investigate the molecular mechanism of fat accumulation and adipogenesis by DNA chip using adipose tissues from 10- and 24-month-old Japanese Black Cattle. Several candidate genes were separated and identified from global gene expression analysis. Adipogenin, chemerin and mPARM-1 genes were confirmed to relate to lipid metabolism and adipocyte differentiation in bovine adipocytes. These results suggested that several genes were involved with adipogenesis and fat accumulation in bovine.

    We investigated the relationship between GH gene SNP and beef production in Japanese Black cattle. (1) C-type allele was for fat production in high-fat meat although A-type allele was profitable for milk production. (2) A-type allele was profitable for carcass weight, dairy gain, BMS and so on, while C-type allele was profitable for unsaturated fatty acid production in meat. (3) A-type allele showed a significant relationship with greater gene expression of SCD and FASN, while C-type allele showed that with lower SCD expression. (4) GHRH-induced GH release in C-type animals was smallest among all types. (5) The effects on meet production was greater for heifers than for bulls.

    我々は脂肪組織で発現する新規アデイポカイン(Adipokine)としてマウスケメリン(Chemerin)とその受容体を報告している。平成20年度は、ウシケメリンとその受容体の遺伝子をクローニングし、塩基配列の解析を行った。GenomeNetを用いたBlastan解析を行ったところ、ウシケメリンとケメリン受容体と思われる遺伝子配列が得られた。これらの配列を基に、特異的プライマーを作成し、ウシケメリンとケメリン受容体全長を決定した。黒毛和種牛の組織からtotal RNAを抽出し、ウシケメリン遺伝子分離と遺伝子発現に供した。ウシケメリンcDNAは3個のExonからなり、ORFは489bpであった。得られた配列はヒトとマウスの塩基配列との相同性は、各々79%と69%であり、アミノ酸配列の相同性は、各々74%と60%であった。ケメリン受容体のORFは1089bpであり、ヒトとマウスの塩基配列と84%と79%の相同性を示した。各組織ごとにおけるケメリンの遺伝子発現量については、肝臓と脂肪組織で高い発現が見られたが、ケメリン受容体は脂肪組織で高い発現量を示したものの、各組織において差は見られなかった。ウシ分化脂肪細胞において、ケメリンとケメリン受容体の遺伝子発現量は脂肪細胞分化に伴い発現量は上昇した。また、分化脂肪細胞でのTNF-αの処理はケメリンとケメリン受容体の遺伝子発現量を増大させた。以上のことから、ウシ脂肪組織でも同様に脂肪細胞由来のアデイポカインが存在し、脂質代謝系に影響を与える可能性が示唆された。

    In order to build the system of good beef production of Japanese Black Cattle, understanding of fat accumulation among each adipose tissue is important. Therefore, the candidate genes related with fat depots have investigated in Japanese Black Cattle. (1) The gene of CDC42, prefoldin-5, decorin, phosphate carrier, 12S ribosomal RNA gene, and Kbtbd2 was identified by the analysis of the gene expression profile between subcutaneous and visceral adipose tissue using the differential display RT-PCR method. Expression patterns of these genes in Japanese Black Cattle were different with those of a single stomach animal like a pig or a mouse. (2) GPCR43, Adipogenin and GPCR120 were highly expressed in adipose tissue and up-regulated during adipocyte differentiation. GPCR43, Adipogenin, GPCR120, and Claudin-6 were important genes on adipocyte differentiation and adipose tissue development (4) Bovine Adipogenin isoform cDNA received alternatives splicing as a result of base sequence analysis, and it was confirmed that the base sequence is missing as compared with Adipogenin. Bovine isoform cDNA was highly expressed in adipose tissue and up-regulated during the adipocyte differentiation of bovine. This was similar with the results of the Adipogenin gene. A possibility that the isoform of an Adipogenin gene was involved with adipose tissue developments was suggested. (5) Introducing an Adipogenin gene into a C2C12 cell showed the fat accumulation in cells without the changes of genes related with adipogenesis.

    The aim of this project was the production and reserve of endangered, rare and valuable birds using the technique of germ cell transfer. We developed the protocol to obtain a large amount of PGCs from gonads of chicken and quail embryos using PGC specific monoclonal antibodies (anti-SSEA1, 2C9 and QCR1) and magnetism activated cell sorting (MACS). MACS treatment greatly increased the population ratio of avian PGCs in gonadal cells, resulting improved gonadal migration in recipient embryos. Blastodermal and primordial germ cells were transferred to the recipient avian embryos. Treated chimeras were hatched and the donor-derived cells were tracked using species-specific DNA markers. Donor derived genome was observed using PCR and Immunohistochemistry in chimeric quail which introduced blastodermal cells of chicken and blue-breasted quail embryos.

    成長ホルモンの分泌は、これまで視床下部ホルモンであるGHRH(促進)とソマトスタチン(抑制)によって制御されていると考えられていた。一方、GHS受容体(GHS-R)と呼ばれるオーファン受容体を介した制御機構も存在することが報告され、新たな内因性成長ホルモン分泌刺激ホルモンとして胃分泌物質、グレリンが発見された。しかし、グレリンの脂肪細胞分化・形成(アディポジェネシス)、脂肪細胞における作用機構およびグレリン受容体の発現機序についてはまだ不明である。したがって、我々は、脂肪細胞におけるグレリンとGHS-Rの関係を明らかにするために、めん羊およびラット脂肪細胞の生成過程に対する成長ホルモン刺激受容体アンタゴニストである[D-Lys-3]-GHRP-6の作用を調査した。[D-Lys-3]-GHRP-6はめん羊およびラット脂肪組織由来の前駆脂肪細胞の脂肪細胞分化を有意に抑制した。また、脂肪細胞分化のマーカーであるPPAR-γ2の発現量は[D-Lys-3]-GHRP-6を10日間添加することによって減少した。逆に、グレリンは脂肪細胞分化過程においてGPDH酵素活性を増加させ、PPAR-γ2の発現量を上向き調節することにより、脂肪細胞分化を促進した。また、めん羊およびラット脂肪組織から単離した脂肪細胞において、グレリンによる脂質分化抑制能は[D-Lys-3]-GHRP-6添加によって減弱された。我々の結果はGHS-Rを介する脂肪細胞分化・形成過程が、[D-Lys-3]-GHRP-6によって抑制されることを明らかにしたものである。

    本研究は脳下垂体におけるレプチンの成長ホルモン分泌抑制に対する分子生物学的解析を行うことを目的としている。脳下垂体のGH細胞を分離、初代培養を施行し、以下の実験に供する。培養細胞に一定の時間と間隔でレプチンの投与後、total RNAを抽出、GH放出因子とグレリン受容体(成長ホルモン刺激物質受容体)mRNAレベルを測定した。レプチンをヒツジの脳下垂体に短期間(1時間)投与した時には成長ホルモンとGHRHによる成長ホルモン分泌に影響がなかった。長期間(24時間)投与すると10^<-7>-M GHRHによる成長ホルモン分泌を抑制した。レプチンを24、48、72時間投与すると、GHやGH放出因子受容体のmRNAレベルが用量依存的に減少した。しかし、グレリン受容体は24と48時間投与した時には変化がなかったが、72時間投与すると有意に増加した。さらに、レプチンを3日間投与するとGHRHによるGH分泌能は減少したが、GHRP-2によるGH分泌能はやや増加した。しかし、GHRHとGHRP-2を同時に投与した時にはレプチンによるGH分泌能には変化がなかった。以上の結果は2001年Endocrinology 142巻の5167ページに掲載された。 さらに、GH放出因子受容体とグレリン受容体の相互作用をAntisense oligonucleotideを用いて分子レベルで検討し、GH放出因子受容体はGHRP-2によって制御される結果を得て、2002年度Endocrinology143巻の1964ページに発表した。また、この研究費の一部分は、脂肪細胞形成においてグレリン受容体の機能解析に用いられ、グレリン受容体は脂肪細胞分化と伴い増加する結果を得て2002年Animal Science Journal(73巻の305ページ掲載)に発表した。

    食肉の肉質と美味しさは多数の要因によって決定されるが、「脂肪交雑」と「脂肪の質」などによって主に評価される。したがって、家畜の産肉性と生産性の向上には、体内での筋肉内脂肪、皮下脂肪、内臓脂肪組織における脂肪細胞の形成と蓄積過程に関わる新規調節因子の解明は重要である。本研究ではこれまで脂肪細胞由来の脂肪蓄積に関連する因子について同定、解析および生産性との関連を調査した。

  2. 国際競争力強化に向けた黒毛和種短期肥育技術の開発

    肥育開始月齢を8か月齢280kgとし、早期肥育に向く子牛育成方法を開発する。その後の肥育期間を18か月とし26か月齢で出荷できるようにCPやTDNを調整した管理を行い、この間の栄養状態を把握する。対照として慣行肥育を行い、生産された牛肉の解析を行い、差異について検討する。第2回目以降の肥育においては、1回目のデータを検討しプログラムの修正を行うとともに、明らかとなった基礎的知見をもとにビタミンなどの補給も試みる。

    反芻動物のルーメンは、ヒトやマウスなどの単胃動物と異なり、飼料の発酵に重要な役割を果たしている器官である。特に、仔牛の離乳前後における適切な栄養・飼養管理によるルーメン発達を促すことは重要であり、丈夫なルーメンづくりとは、その容積とともに筋肉層と上皮層(第一胃乳頭・絨毛)の両方を発育させることである。本研究は次世代シーケンサーを用いてウシのルーメン絨毛組織の形成・発達に関連する候補遺伝子を選別し、多様な飼養条件(エピジェネティクス制御)で得られたウシのルーメン絨毛組織において遺伝子の機能と解析を行うものである。

    ブタの皮下脂肪厚は枝肉価格を決定する最も重要な因子であり、価格に直結する性質である。ブタの皮下脂肪厚は遺伝的な要因と飼料等の環境要因の両者によって決定されているが、環境要因が精密に制御される近代養豚においては、遺伝的な要因の重要性がより高まっている。また消費者の要求は多様化しており、薄脂肪だけではなく味の良い厚脂肪の豚肉を好む方向も現れてきている。皮下脂肪厚の育種改良には長い期間が必要であり、従来型の育種技術では多様化した消費動向に柔軟に対応することは困難である。本課題では、消費者の要望に添った形質のブタを自由に生産する技術を開発するため、ブタの皮下脂肪厚を制御している遺伝子を特定する。
