The oxidative modification of low-density lipoprotein (LDL) is involved in the generation of lipid peroxidation-derived electrophilic aldehydes, which covalently react with apolipoprotein B-100 to form various adducts. Although the oxidative modification of LDL is critical in the pathogenesis of atherosclerosis, the structural properties of adducts in oxidized LDL (oxLDL) remain unclear. Thus, we aimed to characterize oxLDL using the adductome approach, a comprehensive mass spectrometry-based analysis designed to detect specific product ions from positively ionized oxidized phosphatidylcholine (oxPC) adducts. The amounts of several adducts, including four major oxPC-lysine (oxPC-Lys) adducts, prominently increased when LDL was oxidized with Cu2+. Analysis of the synthetic adduct candidates through mass spectrometry revealed two oxPC-Lys Schiff base adducts and two novel oxPC-Lys amide-type adducts as the major lysine adducts in oxLDL. The oxPC-Lys Schiff base adducts could be transformed into stable amide-type adducts during LDL oxidation. The amounts of oxPC-Lys adducts were significantly higher in the sera of hyperlipidemic mice than in the sera of control mice. The recognition of oxPC-Lys adducts by macrophages suggested that these adducts were involved in the phagocytosis of oxLDL by macrophages. These findings provide insights into the structural properties of lipoproteins modified under oxidative stress and their biological implications. Moreover, this study is the first to identify and quantify oxPC-Lys amide-type adducts in vitro and in vivo.

γ-Aminobutyric acid (GABA)-rich fermented rice bran (GFRB) is a multi-component fermented food matrix with potential metabolic benefits, but its effects on high-fat diet (HFD)-induced liver injury and associated gut microbial and metabolic changes remain unclear. In this study, we characterized the metabolite profile of GFRB and evaluated its effects in rats with HFD-induced liver injury. Male Sprague-Dawley rats were fed a control diet or HFD with or without GABA or low- or high-dose GFRB supplementation for 12 weeks. Gut microbiota, fecal short-chain fatty acids, and fecal metabolomic profiles were further analyzed as exploratory outcomes. GFRB contained markedly increased GABA and was enriched in amino acid- and fermentation-related metabolites, including glutamine, ornithine, citrulline, lactate, and succinate. HFD feeding induced a relatively mild non-alcoholic fatty liver disease-like phenotype characterized by hepatic histopathological alterations, including fatty changes and inflammation. GFRB supplementation attenuated these hepatic alterations, as reflected by a reduced NAFLD activity score in the low-dose GFRB group and decreased hepatic interleukin-6 levels in the high-dose GFRB group. Exploratory analyses also identified alterations in gut microbiota composition and fecal metabolite profiles, particularly in amino acid- and nitrogen-related metabolic pathways. Overall, these findings suggest that GFRB may attenuate early HFD-induced liver injury, potentially through modulation of hepatic inflammation. The accompanying gut microbiota and metabolomic changes should be considered exploratory observations that warrant further mechanistic investigation.

While short-term antibiotic exposure has been reported to exacerbate the development of allergies, the effects of long-term administration remain less understood. This study addressed the influences of prolonged antibiotic administration in a murine model of food allergy. Mice received a broad-spectrum antibiotic cocktail (ABX) for six weeks from pre-sensitization through allergen challenge. ABX reduced allergic reactions, serum IgE levels, and intestinal mast cell counts. Production of Th2 cytokine, but not of Th1 and Th17 cytokines, was lowered in allergen-stimulated splenocytes of ABX-treated allergic mice, whereas the levels of these cytokines in their intestines decreased. ABX did not significantly alter the levels of regulatory cytokine IL-10 in intestines and regulatory T cells in spleens. ABX reduced gut bacterial amount, though Lactobacillus, Lactococcus, and Streptococcus remained detectable. Collectively, the long-term ABX suppresses allergic reaction by modulating Th2-mediated events dominantly, providing insights into the complex role of antibiotics in allergy models.

Pyrroloquinoline quinone (PQQ), an o-quinone-type nutrient, has been shown to exert diverse beneficial effects on the biochemical and physiological processes of mammals. However, the molecular mechanisms underlying these effects remain incompletely understood. Here, through screening of metabolite-sensing G protein-coupled receptors (GPCRs)─which respond to metabolites produced by gut microbiota or derived from nutrients─we found that PQQ selectively activates G-protein-coupled receptor 35 (GPR35) and characterized the molecular basis of its ligand recognition. Using a transforming growth factor α shedding assay, we demonstrated that PQQ selectively activated GPR35, a class A rhodopsin-like GPCR expressed in various tissues, including adipose tissue and the gastrointestinal tract. PQQ also promoted β-arrestin 1 recruitment to the plasma membrane, further supporting its role as a GPR35 agonist. Direct binding of PQQ to GPR35 was demonstrated using a clickable photoaffinity probe derived from PQQ. Molecular docking simulation and site-directed mutagenesis revealed that the 2-carboxylic acid moiety of PQQ forms critical hydrogen bonds with Arg100, Tyr101, and Arg151 of GPR35. Additionally, Phe163 appears to contribute to π-H interaction with PQQ. These findings indicate that PQQ functions as a food-derived agonist of GPR35 and provide new insights into the molecular mechanisms underlying the potential beneficial effects of PQQ.

G protein-coupled receptors (GPCRs) are the largest superfamily of cell surface receptors. They regulate critical physiological events and serve as potential therapeutic targets. G protein-coupled receptor 35 (GPR35), a class A rhodopsin-like GPCR expressed in various tissues, including adipose tissue and the gastrointestinal tract, has roles in diverse functions, including antioxidant, anticarcinogenic, and anti-inflammatory effects. Although many endogenous and synthetic GPR35 agonists have been identified, the understanding of food-derived agonists is limited. In this study, we discovered pelargonidin as a newly identified food-derived GPR35 agonist through a systematic screening approach. We evaluated 28 dietary phytochemicals using a transforming growth factor α (TGFα) shedding assay to evaluate GPR35 activation, and found that cyanidin, a common 3-hydroxyanthocyanidin present in various red fruits and vegetables, induced GPR35 activation. Among a series of 3-hydroxyanthocyanidins tested, pelargonidin, characterized by its monohydroxylated B-ring, exhibited the most potent agonistic activity. Mutational studies demonstrated that the hydrogen bond between the 3-hydroxy group in the C-ring of pelargonidin and Asn169, as well as the hydrophobic interaction between the A-ring of pelargonidin and Phe163, is crucial for GPR35 activation. Furthermore, pelargonidin inhibited the production of interleukin-8, a pro-inflammatory cytokine, by activating endogenous GPR35 in Caco-2 cells. These findings suggest that GPR35 may serve as a potential receptor for dietary anthocyanidins, such as pelargonidin, and provide new insights into the molecular mechanisms underlying the potential chemopreventive effects of anthocyanidins.

The chalcone skeleton is a fundamental structural motif in numerous natural and synthetic compounds, playing a crucial role in various biological activities, including anticancer and anti-inflammatory effects. Mallotus philippinensis is a remarkable source of chalcones, recognized for its diverse chemical structures and significant bioactivity. The presence of complex dimeric chalcones in this species is both characteristic and rare. However, to the best of our knowledge, only five such complex dimeric chalcones have been identified to date. Here, we report six novel complex chalcones, runachalcones A-F (1-6), which significantly expand the chemical space of dimeric chalcones and highlight the untapped structural diversity within M. philippinensis fruits. Their planar structures were elucidated using a combination of NMR spectroscopy and HR-ESIMS, while their absolute configurations were determined through J-based configurational analysis, ROESY spectra, and quantum mechanical calculations of ECD. In an assay using a macrophage line RAW 264.7, only runachalcone F suppressed lipopolysaccharide (LPS)-induced expression of inducible nitric oxide synthase (iNOS) protein and the production of nitric oxide, suggesting that the compound possesses potential anti-inflammatory properties.

Although herbs and spices have been used in traditional medicine for more than a century owing to their health benefits, the associated underlying mechanism is still not clear. Since the G protein-coupled receptor 35 (GPR35) has been linked to exert various antioxidant and anti-inflammatory effects, we screened 19 different herbs and spices for possible GPR35 agonist(s) to understand the GPR35-dependent functions on herbs and spices. Among the screened extracts, the ethyl acetate extract of thyme exhibited a remarkable GPR35 agonistic activity. Activity-guided separations allowed us to identify two polyphenolic phytochemicals, eriodictyol and thymonin, acting as GPR35 agonists. Both eriodictyol and thymonin showed a potent and specific agonist activity toward GPR35 with half maximal effective concentration values of 5.48 and 8.41 μM, respectively. These findings indicate that these phytochemicals may have beneficial health effects upon GPR35 activation.

Obesity is now recognized as an epidemic, requiring rapid intervention. We previously demonstrated that vanilloids from the African spice known as Grains of Paradise (GOP) exhibit strong anti-obesity effects. Here, the molecular mechanism behind the obesity prevention property of the GOP extract was investigated by employing molecular networking-based lipid profiling and 16S rRNA sequencing. Administration of either GOP extract or two of its components, 6-paradol and 6-gingerol, reversed the unbalanced gut microbiota composition induced by a high-fat diet (HFD) with a decrease in Firmicutes / Bacteroidetes ratio and increase in genera Bifidobacterium and Akkermansia. Moreover, GOP extract ameliorated abnormal fatty acid metabolism in gut bacteria. Furthermore, lipid molecular networking revealed differences in lipid composition, such as triglycerides and bile acids, in the feces. These results suggest that 6-paradol and 6-gingerol can restore an unhealthy gut environment and prevent lipid absorption, a possible consequence of the reconstruction of the gut microbiota.

SCOPE: Grains of Paradise (GOP), the seeds of Aframomum melegueta, has anti-obesity effects. However, the mechanisms underlying the effects remain unclear. METHODS AND RESULTS: We set up to study the anti-obesity impact and homeostatic effects of 6-paradol, a major vanilloid found in GOP, and investigated the physiological outputs and the lipometabolism-related gene in fat and liver in high-fat-induced obese mice with a comparison with structurally similar vanilloids (6-gingerol and 6-shogaol). The vanilloids were synthesized in adequate quantities for performing animal experiments and orally administered to six-week-old male mice over two weeks. We found that 6-paradol decreased body weight gain and visceral and subcutaneous fats in two weeks, whereas 6-gingerol and 6-shogaol had no effect. Additionally, 6-paradol suppressed the hepatic cholesterol and triglyceride and significantly decreased the gene expression related to fatty acid synthesis, lipid transportation, and adipocyte differentiation in both liver and adipose tissue. Moreover, phosphorylation of AMP-activated protein kinase (AMPK) that greatly contributes to lipometabolism was promoted by 6-gingerol but not 6-paradol. CONCLUSION: These results suggest that 6-paradol regulates several obesity-related genes in an AMPK-independent manner. Therefore, it could be the principal active vanilloid in GOP giving it anti-obesity properties with a different mechanism. This article is protected by copyright. All rights reserved.

In order to identify a novel whitening agent, the methanol extract of S. polyanthum leaf was focused on by the screening test using nine Indonesian medicinal plants for the inhibition of melanogenesis and tyrosinase activity in B16 melanoma cells. Three novel compounds [(1) 1-(2,3,5-trihydroxy-4-methylphenyl)hexane-1-one, (2) 1-(2,3,5-trihydroxy methylphenyl)octane-1-one, and (3) (4E)-1-(2,3,5-trihydroxy-4-methylphenyl)decan-1-one and one known compound [(4) 1-(2,3,5-trihydroxy-4-methylphenyl)decan-1-one were isolated from the methanol extract. Our study demonstrated that S. polyanthum leaf methanol extract at 25-200 μg/mL decreased extracellular melanin formation ca. 20-80%, with high cell viability. Compounds 1-4 were found to be active in melanogenesis and tyrosinase inhibition. Compound 3 was the most active against tyrosinase activity (83.98 μM), particularly when L-tyrosine was the substrate. Compounds 1-4 significantly diminished extracellular melanin formation in B16 melanoma cells (> 80%), with high cell viability. Thus, our study suggested that compounds 1-4 isolated from the methanol extract of S. polyanthum leaf play important roles in decreasing extracellular melanogenesis and inhibiting tyrosinase.
