    Abstract Silk‐fibroin is utilized as a biocompatible and bioresorbable enzyme immobilization matrix and exemplarily demonstrated for a screen‐printed amperometric glucose biosensor. The silk‐fibroin is derived from the silkworm Bombyx mori. The enzyme immobilization matrix consisting of silk‐fibroin, together with glucose oxidase from Aspergillus niger, is applied to a screen‐printed carbon‐based, biocompatible, and biodegradable working electrode on a flexible silk‐fibroin substrate. The biosensor is characterized electrochemically at physiological glucose concentrations in the range from 0.5 to 10 mm. The results are compared to a “conventional” glucose biosensor, also fabricated on a flexible silk‐fibroin substrate, however utilizing a laboratory standard enzyme immobilization matrix based on bovine serum albumin and glutaraldehyde. Furthermore, the influence of pH (pH 5.5 to pH 8.0) and temperature variations (21 to 70 °C) on these two different immobilization matrices are studied.

    A light-addressable potentiometric sensor (LAPS) is a semiconductor-based sensor platform for sensing and imaging of various chemical species. Being a potentiometric sensor, no faradaic current flows through its sensing surface, and no electrochemical reaction takes place in the course of LAPS measurement. In this study, a four-electrode system is proposed, in which a LAPS is combined with the conventional three-electrode electrochemical system. A LAPS is included as the fourth electrode for potentiometric sensing and imaging of the target analyte in the course of an electrochemical reaction taking place on the surface of the working electrode. The integrated system will be useful for analyzing dynamic processes, where both the electrochemical process on the electrode surface and the ion distribution in the solution need to be simultaneously investigated. Different grounding modes of control circuits that can simultaneously conduct potentiostatic/galvanostatic polarization and LAPS measurement are designed, and their functionalities are tested. The interference between polarization and LAPS measurement will also be discussed.

    The artificial olfactory image was proposed by Lundström et al. in 1991 as a new strategy for an electronic nose system which generated a two-dimensional mapping to be interpreted as a fingerprint of the detected gas species. The potential distribution generated by the catalytic metals integrated into a semiconductor field-effect structure was read as a photocurrent signal generated by scanning light pulses. The impact of the proposed technology spread beyond gas sensing, inspiring the development of various imaging modalities based on the light addressing of field-effect structures to obtain spatial maps of pH distribution, ions, molecules, and impedance, and these modalities have been applied in both biological and non-biological systems. These light-addressing technologies have been further developed to realize the position control of a faradaic current on the electrode surface for localized electrochemical reactions and amperometric measurements, as well as the actuation of liquids in microfluidic devices.

    Nowadays, bioelectronic devices are evolving from rigid to flexible materials and substrates, among which thermally-drawn-fiber-based bioelectronics represent promising technologies thanks to their inherent flexibility and seamless integration of multi-functionalities. However, electrochemical sensing within fibers remains a poorly explored area, as it imposes new demands for material properties—both the electrochemical sensitivity and the thermomechanical compatibility with the fiber drawing process. Here, we designed and fabricated microelectrode fibers made of carbon nanotube (CNT)-based hybrid nanocomposites and further evaluated their detailed electrochemical sensing performances. Carbon-black-impregnated polyethylene (CB-CPE) was chosen as the base material, into which CNT was loaded homogeneously in a concentration range of 3.8 to 10 wt%. First, electrical impedance characterization of CNT nanocomposites showed a remarkable decrease of the resistance with the increase in CNT loading ratio, suggesting that CNTs notably increased the effective electrical current pathways inside the composites. In addition, the proof-of-principle performance of fiber-based microelectrodes was characterized for the detection of ferrocenemethanol (FcMeOH) and dopamine (DA), exhibiting an ultra-high sensitivity. Additionally, we further examined the long-term stability of such composite-based electrode in exposure to the aqueous environment, mimicking the in vivo or in vitro settings. Later, we functionalized the surface of the microelectrode fiber with ion-sensitive membranes (ISM) for the selective sensing of Na+ ions. The miniature fiber-based electrochemical sensor developed here holds great potential for standalone point-of-care sensing applications. In the future, taking full advantage of the thermal drawing process, the electrical, optical, chemical, and electrochemical modalities can be all integrated together within a thin strand of fiber. This single fiber can be useful for fundamental multi-mechanistic studies for biological applications and the weaved fibers can be further applied for daily health monitoring as functional textiles.

    Stainless steel plays an important role in industry due to its anti-corrosion characteristic. It is known, however, that local corrosion can damage stainless steel under certain conditions. In this study, we developed a novel measurement system to observe crevice corrosion, which is a local corrosion that occurs inside a narrow gap. In addition to pH imaging inside the crevice, another imaging technique using an infrared light was combined to simultaneously visualize surface roughening of the test piece. According to experimental results, the lowering of local pH propagated inside the crevice, and after that, the surface roughening started and expanded due to propagation of corrosion. The real-time measurement of the pH distribution and the surface roughness can be a powerful tool to investigate the crevice corrosion.

    Recently, robotic studies based on biological materials have been attracting a lot of attention, because of its intrinsic abilities. Biological materials have some interesting capabilities in such a way that tissues as devices can be self-organized, adaptable, self-repaired, and so on. Such significant abilities are based on cellular behavior that reflects mechanical stimuli during the development of the tissues. Under this circumstance, in this paper, we intend to deal with development of a micro-actuator by exploiting mechanical stimulus response of muscle cells. To do so, we carry out the induction of differentiation into myofilaments from a cultured myoblasts C2C12 under the periodic stretching forces for 70 hours. As a result, interestingly, we found that induction of differentiation into myofilament-like calls are promoted under the mechanical stimulus.

    The mechanism to encode and store the information on time is essential to uncover the nature of temporal information processing in the sensory, memory and higher-order areas in the brain. In this article, the results of psychophysical experiments on short-term memory of tone durations in auditory sequence were reported. Subjects were requested to recall the durations of all the tones presented serially (sequence) by a telegraph key. The performance of recall was evaluated by correlation coefficients and the distribution in correlation diagram between presented and recalled tone durations, and the relationship to the serial order and the number of tones in a presented sequence were investigated. It was shown that the performance of the recall of tone durations could be evaluated by cross-correlation, and the performance decreased when the serial order of the target tone in the presented sequence was later or the number of tones in a sequence was larger. It was also shown that there were some types of characteristic distribution on the correlation diagram between presented and recalled tone durations which corresponded to the degree of memory decay. This result indicated that the behavioral process of memory decay of the analogue (non-categorized) information could be observed by the profile of correlation diagram.

    Atomic force microscopy (AFM) has been used for nanotribological studies of silicon surfaces covered by oxide layers of various kinds: chemical oxides prepared by SC 1 (NH4OH/H2O2/H2O) and SC 2 (HCl/H2O2/H2O) treatments, a thermally treated silicon surface and a H-terminated oxide-free silicon surface. Only in the case of SC 1 chemical oxide, scratching of the oxide and ploughing of the silicon by a Si3N4 AFM tip were observed. On the other hand, no wear of the sample was noted on other surfaces. By annealing the SC 1-treated surface in N2 gas above 200oC for 30 min, the oxide layer could not be scratched. By soaking the thermal oxide in KOH, the oxide layer was then scratched. It is suggested that the presence of OH bases is a requirement for the nano-scratching of the oxide layers.

    A light-addressable potentiometric sensor (LAPS) with Poly-L-Omithine with Laminin (PLOL)-coated SiO2 layer as the only insulating material on the bulk Si in SiO2/Si structure, is investigated as a possible biological interface. Comparing with the conventional structure (Si3N4/SiO2/Si) of the LAPS, much steeper photocurrent response was observed for the PLOL-coated SiO2/Si structure, which shows higher surface potential sensitivity and pH resolution. PLOL-coated SiO2 layer is kept soaked with the electrolyte for ten days and no appreciable change in the surface potential sensitivity was noticed. The pH sensitivity of the sensor was investigated in the pH range of pH4 to pH10, and a nearly Nernstian sensitivity was observed.

    A light-addressable potentiometric sensor (LAPS) is a semiconductor-based chemical sensor, in which a measurement site on the sensing surface is defined by illumination. This light addressability can be applied to visualize the spatial distribution of pH or the concentration of a specific chemical species, with potential applications in the fields of chemistry, materials science, biology, and medicine. In this review, the features of this chemical imaging sensor technology are compared with those of other technologies. Instrumentation, principles of operation, and various measurement modes of chemical imaging sensor systems are described. The review discusses and summarizes state-of-the-art technologies, especially with regard to the spatial resolution and measurement speed; for example, a high spatial resolution in a submicron range and a readout speed in the range of several tens of thousands of pixels per second have been achieved with the LAPS. The possibility of combining this technology with microfluidic devices and other potential future developments are discussed.

    A BCI (brain-computer interface) system that detects visual ERPs (event-related potentials) to control a cursor on the computer screen is proposed in this paper. The ERPs induced by the selective attention to one of the eight circles around the cursor which was blinked in random order, was used to control the cursor movement. Information bit rates for five subjects were about 11 to 16 bit/min. Improvement of an information bit rate and increasing a speed of cursor movement are left for further studies.

    To detect the imagined limb from EEG related to the motor imagery for the use in BCI, the spatial filter using neighboring channels was commonly used. But, the spatial filter has been selected mainly by the empirical method on trial-and-error basis. In this study, we recorded the EEG during left hand, right hand and feet motor imagery from five subjects and the ICA was applied to discovering the spatial filter for extracting event-related EEG components to the motor imagery. It was shown that the spatial filter can be generated semi-automatically by applying ICA to recorded multichannel EEG.

    The authors have evaluated the effect of neurofeedback training for subjects on the brain activity to be used in the brain-computer interface (BCI) which detected motor imagery from cerebrum blood flow measured by NIRS (near-infrared spectroscopy). The concentrations of oxy-hemoglobin on motor cortex during motor imagery of a subject's right hand were measured, and were visually fed back to the subject. It was shown that the training enhanced the cerebral blood flow during motor imagery on two out of five subjects. Investigation of more effective training and neurofeedback methods were left for the further study.

    An auditory brain-computer interface (BCI) which detected event-related potential (ERP) elicited by selective attention to one of the tone streams was proposed. Each tone in two kinds of auditory oddball sequences with different frequencies of standard tones was presented alternatively to subjects, and they were perceived by subjects as two kinds of segregated streams. Event-related potentials elicited by two kinds of deviant tones were classified by linear discriminant analysis (LDA) to find which streams subjects paid selective attention. By the experiments to six healthy subjects, it was shown that this system could achieve binary selection of two kinds of tone streams.

    A Brain-Computer Interface (BCI) is a communication system that translates brain activity into commands for controlling a computer or an external device for disabled users like amyotrophic lateral sclerosis (ALS) patients. We developed a BCI system using event-related potentials during selective attention to tone frequencies and a classification accuracy of detecting event-related potentials caused by two kinds of deviant stimulation with different frequencies was evaluated. A classification accuracy of 60〜90% and a classification rate of 3〜4bit/min were obtained respectively for two subjects.

    In this study, we investigated the effect of feedback training to the user to improve the accuracy of command detection on BCI system which detects a command from EEG during motor imagery. During the feedback training session, subjects were visually presented a white bar on LCD whose length was proportional to the EEG band power of the specific frequency range. The effects of feedback training were evaluated in terms of the change of the band power and the accuracy of command detection. It was shown that the feedback training was effective on two subjects out of four.

    A generic platform for realizing online brain-computer interface (BCI) system was developed. The platform is module-based, and has a GUI-based diagram editor for building desired system. The platform can be extended easily, and supports multiple acquisition hardware and operating system. An online BCI system that detects the band power of EEG beta oscillation during hand or foot motor imagery was built on this platform. We confirmed that the system was able to realize an online BCI system. The platform is extendable, and a various system can be realized by adding new modules.

    EEG-based asynchronous brain-computer interface (BCI) system to realize binary switch by using simple thresholding of EEG beta band power was tested to six healthy subjects. EEG signal was measured from a bipolar channel on the vertex of the head, and was band-pass filtered, squared and smoothed on-line to extract the band power of beta oscillation. Subjects were requested to imagine foot movement, and if the band power exceeded a pre-defined threshold value, a command was generated. It was shown that two subjects out of six were able to induce bursts of beta oscillations by foot movement imagery, and binary commands could be detected in higher correct rates by these subjects.

    The human intafacing system to detect the direction and distance of voluntary eye gaze movement from EOG (electrooculogram) was investigated. In this study, a method to detect 8-class directions and 2-class distances of voluntary eye gaze movements from AC-amplified vertical and horizontal EOGs was proposed. This technique could avoid strict eye tracking which required DC amplification of very small signal. It was found that this technique enabled detection of eye gaze movement with high correct rate by AC-amplified EOG. It may be useful to develop robust human inter facing system based on menu selection for severely paralyzed patients.

    Combined with thermal annealing, the locally confined low energy electron-beam from a scanning tunneling microscope (STM) tip can be used for nanofabrication of Si oxide film on Si substrate as nano-lithography mask; the oxide layer within the field emission a-beam exposed area can be decomposed, and then evacuated from the surface at elevated temperatures of about 600°C. Different manometer scale patterns of the oxide windows on Si substrate such as dot window arrays, lines, and circles can be formed. The minimum feature size attained so far is 25nm and the rms edge roughness of line patterns is less than 3.5nm. Mechanism and controllability of the STM nanofabrication processing are discussed.

    本研究は、1枚のセンサで複数の種類のイオンや分子の濃度分布を同時に画像化できる「マルチアナライト化学イメージセンサ」の実現を目的としている。従来、1枚のセンサで測定できる化学種は1つに限られていたたが、本研究では測定したい各化学種に応答する複数の感応膜を1枚のセンサ面上に集積する方法と、それらの応答を高速かつ正確に読み出す機構を開発することによって、目的の達成を目指している。 本年度は昨年度に引き続き、各種イオノフォアを含有する高分子膜をセンサ面上に作製し、感度や測定限界の違いを調べた。センサ面とイオン感応膜の密着性や、得られるスロープ感度の再現性に課題があったが、センサ面と各感応膜の間に画素の大きさに対応する導電性の下地層を設けることによりこれらの課題を解決できる見込みを得た。また、光走査によって多数の感応膜から正確に信号を読み出すための機構についても、昨年度に引き続き改良を進め、100万画素の画像を読み出せるようになった。 実用上問題になると思われる、試料溶液と半導体基板との容量結合による信号電流のロスについて、MIS構造における実験と回路モデルの解析により周波数特性などを調べ、感応膜アレイを有するセンサ構造において高周波動作を可能にする方法について検討を行った。 本システムの生物学的応用のため、センサ面上に設けた多数の培養チャンバに微生物を含む培養液を自動分注できるシステムの開発を行ったほか、植物根の生育をモニターする実験を行った。また、化学イメージセンサと各種電気化学システムを複合するための各種制御回路の開発を行った。

    For realization of a high-definition chemical imaging sensor system, methods to improve the spatial resolution, to shorten the measurement time, and to use a large-area sensing plate were developed. The spatial resolution was improved by using a hybrid illumination that suppressed the lateral diffusion of photocarriers in the semiconductor layer. The measurement time was shortened by parallel readout of data using a plurality of light probes. A distributed-element model was constructed to evaluate the influence of a large-area sensing plate, and the dependence on various parameters such as the frequency, the conductivity of the solution, and the series resistance was investigated. The developed chemical imaging sensor system was applied to visualization of the corrosion process on a metal surface and the metabolism of microorganisms.

    本研究では、微小流体デバイスに組み込み可能な半導体化学センサとして期待される light-addressable potentiometric sensor (LAPS)の空間分解能を向上させる新しい原理の実証と装置の開発を行った。LAPSは、画素構造や配線を形成することなく、光照射位置ごとにpHやイオン濃度の読み出しが行える光アドレス機能を有しており、1枚のセンサチップ上で複数の検体やイオンを測定したり、空間分布を画像化することができる。 LAPSは、半導体センサ裏面の光照射で発生した電子・正孔対が、センサ表面のイオン濃度に応答して形成される空乏層で電荷分離される際に生じる電流を信号として検出する。このとき、面内方向への電荷の拡散によって空間分解能が決まる。本研究では、光をパルス状に照射し、過渡電流を時間分解的に測定することによって、照射点近傍のイオン濃度を高い空間分解能で測定する方法を開発した。前年度までの研究でこの原理の実証が行えたので、最終年度はさまざまなパラメータを変化させて測定条件の最適化を行った。 実験により、パルス照射後の電流信号の積分時間に対する空間分解能の依存性を得ることができた。例として、積分時間2μsおよび10μsにおける空間分解能はそれぞれ110ミクロンおよび240ミクロンであった。積分時間を2μsより短くすることによって空間分解能をさらに向上できることが示されたが、S/N比との間にトレードオフ関係が認められた。実際にこの方法を用いて微細テストパターンのイメージングを行い、高解像度の化学イメージが得られることを確認した。また、pH感度については、従来法と本質的に同じ値を得ることができた。

    In this study, the data acquisition rate of the chemical imaging sensor, which can visualize the two-dimensional distribution of pH or ion concentration in the sample placed on a semiconductor sensor plate, was enhanced to realize a movie recording of chemical species at a maximum frame rate of 1000 fps. The developed system was applied, for example, to dynamic analysis of reaction in a microfluidic channel constructed on the sensing surface and visualization of pH distribution in the vicinity of a corroding metal surface.

    The BCI (brain-computer interface) based on motor imagery is to detect the intension of subjects from their brain activities from sensorimotor cortex during imagining to move their own limbs. This study aimed to investigate the methodology to use neurofeedback training (NF), on which the processed EEG or NIRS (brain blood flow measured by near-infrared spectroscopy) data is presented to subject in real time, for improving the applicability and accuracy on motor-imagery based BCI. It was found that the motor related EEG on mu, beta and gamma band could be extracted by applying local and sparse spatial filters designed by ICA (independent component analysis) to measured EEG data, and such components could be used for neurofeedback training to improve the performance of motor-imagery based BCI.

    The methods to improve user-applicability of BCI (brain-computer interface) using non-invasive measurements of brain activity were proposed and investigated. The methodologies of feature extraction, pattern classification and online neurofeedback were studied by EEG, NIRS (near-infrared spectroscopy) and fMRI measurements. And the BCIs based on event-related potentials (ERPs) with higher usability were proposed and evaluated.

    In this study, a flat-bed-type chemical image scanner was developed, which was capable of visualizing the distribution of specific ions or molecules in the specimen placed on the sensing surface. In the developed system, a large-area sensor was scanned by a plurality of light sources in parallel, which resulted in great reduction of measurement time in comparison to the conventional chemical imaging sensor system. A new measurement mode was proposed, in which the ion concentration was determined by measuring the phase of the photocurrent, and its insensitivity to fluctuation of light intensity and defects in the sensor plate was demonstrated.

    形態変化するDNA分子の設計方法:萩谷は、連続ヘアピンからなる分子マシンに関して、ヘアピンの配列を変化させたときに、3連続ヘアピンから成る分子マシンの挙動がどのように変化するかを調べた。分子マシンの光制御を目指した光機能性超分子の構築:浅沼は、これまでとは逆に「cis-体で二重鎖形成、trans-体で解離」というスイッチングが可能な光応答性DNAの設計と実現に成功した。ヘアピンとバルジによる並行計算:萩谷は、Whiplash PCRの熱力学的な解析と、状態遷移の効率化(Displacement WPCR)を行った。レトロウィルスによる並行計算:陶山は、二つの正帰還と一つの負帰還反応から構成されたオシレータをRTRACの基本反応を用いて構築した。シミュレーションにより発振可能な条件が存在することを確かめた後、実装を進めた。翻訳系による並行計算:横山は、翻訳システムを利用した「オートマトン」を動物細胞(培養細胞)内でも構築することを目標に、非天然型アミノ酸が細胞に与えられた場合にのみ活性化され、サプレッサーtRNAにアミノ酸を結合するような酵素(アミノアシルtRNA合成酵素)を用いることにより、サプレションを制御する機構を構築した。DNA Walker:萩谷は、DNA Walkerの構築に向けて、温度、pH、光の三種類の入力によって駆動する分子マシンに関する予備実験を行った。特に、これらの三種類の入力の独立性について調べた。マイクロチップのための微量液体制御機構の開発:藤井は、液滴操作に必要な周辺技術等の整備を進め、オンデマンド式で液滴の生成・合一の操作が可能にした。また、本技術を用いてDNAとPNAのハイブリダイゼーション反応と電気泳動による反応産物の分離操作をデバイス上で実現した

    As a platform for chemical sensors that can simultaneously detect many chemical species, a measurement system based on the light-addressable potentiometric sensor (LAPS) was developed. 1.Development of thin-film a-Si LAPS The spatial resolution of the LAPS depends on the beam size of the laser beam as a light source, the thickness of the semiconductor layer and the diffusion length of minority carriers. Compared with the LAPS with a bulk Si substrate, the thin-film a-Si LAPS is advantageous for high spatial resolution, realization of large-area sensor and combination with optical sensing methods due to its translucency. It was demonstrated that the fabricated a-Si LAPS worked as a chemical sensor with a sensitivity comparable to that of the conventional LAPS. The spatial resolution of this sensor was higher than 1μm. 2.Micropatterning of biomolecules For the application of LAPS to biosensors and biochips, the micropatterning technology was developed for immobilization of biomolecules on the semiconductor surface. The probe of an atomic force microscope (AFM) was used for local anodic oxidation of the semiconductor surface, and the oxide pattern was employed as a template for immobilization of biomolecules. With this technology, it was possible to fabricate structures such as a line pattern of protein molecules with a thickness narrower than 100nm and a DNA wire structure. 3.Fabrication of sensors with microchannels For highly efficient measurement of analyte with a small valume, a sensor chip with a microchannel was fabricated. The SU-8 walls were fabricated with photolithography, and a glass plate with a conductive film was used as the ceiling of the microchannel. By using a conductive ceiling, the current image of the microchannel became uniform.

    DNAの塩基配列の解読により、生命現象を解析するプロテオミクスや、SNP(単一塩基多型)解析に基づいたオーダーメイド医療の実現など、21世紀のバイオテクノロジーを牽引する研究が活発になりつつある。これらの研究を進展させるためには、生体関連物質の高感度分析・解析技術の開発が必要不可欠であり、その技術開発には,20世紀に成熟した半導体エレクトロニクス技術、特に表面ナノスケール制御・解析技術と,バイオテクノロジーとのインテリジェントな融合が有効である.この融合をわれわれは「表面ナノバイオトロニクス」と名づける。この「表面ナノバイオトロニクス」の目標は,表面という2次元空間を作業空間として、生体情報を物理信号(エレクトロン(電子)やフォトン(光子)など)に高効率に変換し,また,その逆に物理信号を生体系にフィードバックするような、高度なバイオ・インタフェイスシステムを構築することである。国内外を見るに、現在,この種の研究は緒についたばかりである。そこで本研究は、半導体工学や表面科学の研究者と、生命科学、バイオテクノロジー分野の研究者が、有機的な医理工連携を図ることにより、「表面ナノバイオトロニクス」技術の戦略目標を討論することを目的とした。 平成16年(2004年)には、応用物理学会秋季大会シンポジウム「ナノバイオテクノロジーにおける表面・界面の課題」を開催し、本研究のメンバーを中心に講演・討論を行った。さらに、平成16年10月に内外の著名な研究者を招待して国際研究集会「International Workshop on Surface-Biotronics」(東京)を開催し、表面バイオトロニクスに関する研究調査を行い、関連情報の収集に努めた。 以上の調査研究に基づいて、平成17年度発足の特定領域研究「生体階層構造に適合した表面バイオトロニクスの創成」(代表:岩崎裕)を申請した。

    1. Development of multi-ion-imaging technology The conditions for the fabrication of ion-sensitive membranes with ionophores were optimized to implement various ion sensitivities on the sensing surface of the light-addressable potentiometric sensor (LAPS). A photo-patterning process was developed for the integration of ion-sensitive photocurable membranes on a single sensor chip. The distribution of the ion concentration was visualized with a sensor with integrated K^+-and Ca^<2+>-sensitive membranes. 2. Development of Enzyme LAPS For the application of the LAPS and the chemical imaging sensor to biological specimens, enzyme LAPS was developed with urease and butyrylcholinesterase. Various methods for the immobilization of enzyme molecules on the sensor surface were compared in terms of sensitivity and stability. 3. Development of a pen-shaped portable LAPS device To allow more flexible use of LAPS; a pen-shaped portable LAPS device was developed. The control and measurement circuits are built in the device, which can be directly dipped into the solution to be measured. The sensor chip has 4 or 16 measurement points on the surface, each of which can be independently assigned for measurement of different ion species. 4. Application to the microfluidic devices For the application to the detection and measurement of chemical species in the microfluidic devices such as μTAS and Lab-on-a-Chip, a LAPS-based system was combined with a microfluidic channels fabricated with the SU-8/PDMS technology. This system allows label-free measurement of ion concentration at an arbitrary point in the microchannel. The distribution of the ion concentration in the microchannel was visualized.

    本グループでは、電解質溶液またはゲル状試料中におけるpH等の化学的情報の2次元的な空間分布を画像化することができる、化学イメージングセンサの開発を行ってきた。この方法では、センサ表面の電荷によって半導体層中に空乏層幅の分布が生じることを利用して、その静電容量を場所ごとに読み出していくことになる。具体的にはセンサ裏面から変調レーザビームを照射し、静電容量に応じて外部回路に流れる交流光電流を計測する。現在までに、空間分解能10ミクロン、pH分解能0.01、測定速度毎秒100ピクセルを達成している。このうち、空間分解能およびpH分解能については多くのアプリケーションにおいて十分な値であるが、測定速度については1画面の測定に分オーダーの時間を要することから、変化の速い現象の測定に用いることは困難であった。 そこで本研究では化学イメージングセンサの実時間化に向けて装置および測定方式の改良を行った。昨年度は各ピクセルごとの測定に単発の光パルスを用いることで10倍以上の高速化を達成し、また低周波ノイズの影響が低減できることを明らかにした。本年度は化学イメージングセンサを用いて、ゲル中におけるイオンの拡散を可視化し、拡散係数の計測に応用できることを明らかにした。平行平板電極を有する電気化学システムにおいて、電気分解によって生じる電極間のpH分布が時間とともに変化する様子を観測し、pH分布の時間変化から拡散係数を計測した。また、分子量の異なるイオンについて各々の拡散係数を求め、分子量との関係を調べた。

    In this study, the chemical imaging sensor system was further developed to achieve higher performance and multifunctions that will meet the requirements of future applications. As an example, a dynamically changing electrochemical system was visualized and quantitatively analyzed. Visualization of chemical quantities other than pH was realized. A high-speed measurement system was proposed. Firstly, the diffusion process of ions was observed by measuring the distribution of ion concentrations in the proximity of the electrode after electrolysis. The Gaussian distribution of the ion concentration was observed as derived from the Fick's law. The ionic diffusion constants were experimentally determined for various ions. Secoedly, a sensor with a Au dot array pattern was developed for visualization of the redox potential distribution. As an application, the photo response of the porphirin was measured. Finally, a high-speed measurement system was proposed for the chemical imaging sensor. The technologies developed in this study can be combined and applied to chemical imaging of various specimens.

    本年度はナノ構造形成への応用が考えられるステップのバンチングとファセッティングについて調べた。これらの方法により、原子レベルで平坦なステップフリーの広いテラスが得られ、ナノ構造の形成や量子機能の発現に有利であるほか、高さや密度を制御した多段ステップを量子細線の作製に利用したり、核生成サイトとしての応用が考えられる。 Si(111)微斜面における直流電流の通電によるステップのバンチングおよびデバンチングの過程をin situで観察することができる光散乱測定装置を試作した。超高真空中チャンバ内に置いた試料に対して約1240℃でステップダウン方向に通電加熱を行ない回折光を測定した。その結果、平均テラス幅は3〜14ミクロンの広い範囲にわたって時間の1/2乗に比例して増加することがわかった。またテラス幅分布の標準偏差は平均テラス幅の0.24倍であった。次にステップアップ方向に通電を行なったところ回折ピークの位置は変化せず次第にピークが消滅する様子が観察された。デバンチングにおいてはバンチの間隔は変化せずに各バンチが崩壊していることを示している。 また、Si(113)微斜面においてアニールによるファセッティングの様子を超高真空STMで調べた。高温側ではsingle step相となるのに対して、700℃以下ではファセッティングが進行し、テラス幅が時間の1/6乗で増加することを確認した。また、近距離でのステップ間相互作用を考慮したモンテカルロシミュレーションによってこれを再現した。

    DNAコンピューティングは、DNAの分子認識機能を利用することによって、モル当たりアボガドロ数オーダーの超並列演算を可能にする方法として期待されており、特に、NP完全問題のように問題の規模に対して計算量が著しく増大する場合において、従来の計算機に比べて取り扱える問題の規模が大きくなることが期待されている。DNAコンピューティングについては、これまでにいくつかのインプリメンテーションが提案されており、小規模な問題については実際に演算も行なわれているが、必ずしもそのまま大規模な問題に対してスケーラブルではない。 本研究では、まず、組み合わせ問題のブール代数化と、DNA鎖長へのコーディングを用いてナップサック問題を解くアルゴリズムを提案し、実際に小規模な問題について演算を行ない、解の有無を判定することができた。この方法では、まず、DNA断片の全ての組み合わせからなるDNAをLigation反応によって生成した後、PCR反応によって条件を満たす組み合わせのみを増幅し、電気泳動によって解の有無を判定する。また、解となる組み合わせを具体的に知る方法として、大腸菌プラスミドを用いたクローニングを行ない、DNA配列を読み取った。この方法では、複数の解が存在する場合にも各々分離して解が得られるという利点がある。 さらに、DNAコンピューティングにおいて問題の規模を大きくする際に現実的な問題として現われるてくるであろう、分子生物学的プロセスの信頼性の問題に注目し、間違った組み合わせに対してLigation反応が起こる割合を塩基対ごとに調べ、エラー率の評価を行なった。

    本研究では、単電子デバイス構造製作時の結晶成長や加工において問題となるラフニングや、デバイスの長時間安定性にとって問題となるナノ構造やステップ構造の熱的揺らぎについての知見を得ることを目的とし、統計力学に基づいた解析、及びSi表面のSTM観察を行なった。 まず、熱平衡状態における微斜面上の原子ステップの揺らぎの性質をSolid-On-Solid(SOS)モデルに基づいたモンテカルロシミュレーションで調べた。SAステップ及びSBステップの2種類のステップが交互に存在するSi(001)表面に対して、熱平衡状態のシミュレーション像と実際の表面のSTM像におけるステップのゆらぎを比較することにより、結合エネルギーをおよそε(SA)=0.15eV、ε(SB)=0.046eVと見積もった。 熱平衡状態以外では、MBE成長時における表面のカイネティックラフニングと成長条件の関係や、Si(113)表面におけるステップバンチングの時間依存性について調べた。MBE成長のモンテカルロシミュレーションでは表面マイグレーション距離に応じてバリスティックデポジション→3次元成長→レイヤーバイレイヤーモード→ステップフローモードと変化する様子を観察した。 また、実際にナノ構造の揺らぎが電気的特性に与える影響を調べるための準備として、AFM探針を用いたSi表面やAl細線の酸化加工を行ない、幅20nm長さ1μmの細線を作製した。

    本研究では、シリコン窒化膜で覆われたシリコン基板をセンサ面とし、同センサ面上の2次元電位の変化とその分布を顕微的に高速にイメージングする事が可能な装置を新たに開発し、その動作を確認した。さらにこのセンサ面上に人工的に培養した動物の神経細胞を伝達する活動電位を測定することを試み、予備的な結果を得た。 センサ面上で神経細胞を培養し、センサ面上に張り付いた神経細胞の活動電位を測定する場合には、(1)神経培養液の電位と半導体基板電位との差により直流バイアス電位差がセンサ面上に均一に印加されている。(2)神経に活動電位が生じた場合には、神経細胞の内部の電位が外部の電位=培養液の電位に対しパルス的に正に振れ、細胞膜と絶縁膜を介して容量結合により半導体基板のバンドの曲がり方が変化し、交流光電流の振幅が変調されることになる。 本装置の動作を確認するため、センサ面上に金属電極ドットを設け、半導体基板との間に適当な直流バイアスを印加し、パルス電位を重畳し変調光を照射した時の交流光電流の振幅を測定し、交流光電流の振幅がパルス電位により変調されていることを確認した。実際の神経細胞において活動電位の変化を位置敏感に測定できるこを確認するためには、比較的大きな単一又は少数の神経細胞を培養し、その活動電位をガラス電極と本センサで同時に測定し、それらの結果を比較する必要がある。 この目的のために、電極マニピュレータをシステムに組み込んだほか、アンプや培養装置など神経細胞を用いた実験を行なう環境を整備した。また、東海大学の榊原教授に御指導いただき、ウミウシの神経細胞をセンサ基板上に張り付くように培養できる条件を確立し、今後はこの細胞を用いた活動電位測定の実験を行なう予定である。

    A novel chemical imaging microscope has been developed with the aid of the present grant. The chemical imaging microscope is based on the principle of the light-addressable potentiometric sensor (LAPS). The sensing surface of a LAPS device in contact with the electrolyte is made of insulating Si _3N_4 film deposited on semiconducting Si substrate. The two-dimensional chemical information in the electrolyte is mapped into the variation of the width of depletion layr in Si substrate. The electrical impedance of the depletion layr is measured as a function of position, in the form of photocurrent induced by scanning-laser-beam that illuminates the rear surface of the Si substrate. The spatial resolution, sensitivity and measurement speed have been investigated and improved to be 10 micron meter, 0.01pH and 100 pixels/sec, respectively. The microscope has been verified to have capability of visualization of two-dimensional bio/chemical information in electrolytic specimens ; distribution of pH of microorganism colonies, specific molecules and redox potential.It could be also used for imaging rapidly changing electrical-potential such as active neuron potential. To demonstrate its applicability to faster processes, we measured the chemical waves in Belousov-Zhabotinsky reaction. Propagation of chemical waves was observed as variation of redox potential either at a fixed point or on a line along the propagating direction. Such measurement would be useful for microscopic analysis of reaction-diffusion and pattern-formation phenomena in redox reaction systems.

  21. Development of an Ultra-Clean Closed- Multi-Wet Processing-System and Study of Si/Liquid lnterfaces

    Ultra clean and precisely controlled wet processes such as ultra pure water cleaning and wet chemical etching processes of ULSI are required for further progresses of semiconductor technology. In this study, as the first step to study new wet treating procedures and apparatus which satisfy such high quality demands, we have newly developed an ultra-clean closed multi-wet process system and confirmed the improved multi-wet treatment. Main results are described as follows. A flow system which is capable of RCA cleaning has been developed. Better solution replacement characteristic than conventional over-flow system and superior wet treatment characteristics have been achieved. For example, in etching uniformity of oxide layr on 4 inch wafer, average etching rate was 50.5 A/min.for 10 minute etching by HF at 17゚C and uniformity of etching rate in the surface was 1.15 A/min.by rms, less than ? B12.3%. Electrochemical STM study was performed for observation of interface between Si and solution in wet processes. At first, step structures were observed on HF treated Si (111) surfaces and the rms roughness of the surface in 50x50 nm^2 area was 0.60 A.Secondly, the double layr steps of a height of 3.2 A were observed on NH_4F treated Si (111) surface and the rms value of the surface was 0.38 A,which is flatter than the HF treated surface in agreement with previous reports. Surface roughening in mesoscopic scale by surface processes such as chemical etching of Si was studied with STM and AFM experimentally and scaling characteristics of surface roughness, especially self-affine roughness index and characteristic correlation length were measured. Further study to make clear the superiority of newly developed wet processes is in progress.
