    Abstract We developed a label-free method for visualizing intracellular temperature distribution using Raman image analysis combined with deep learning. Analysis of the entire region of the O-H stretching band of water using deep learning achieves an accuracy of ∼0.2 °C, allowing intracellular temperature mapping without any pretreatments such as fluorescent labeling. Experiments using photoexcitation of carbon nanotubes attached to living cells demonstrate that this method can obtain transient changes in the temperature distribution within a cell.

    Abstract Liquid–liquid phase separation is a phenomenon that features the formation of liquid droplets containing concentrated solutes. The droplets of neurodegeneration-associated proteins are prone to generate aggregates and cause diseases. To uncover the aggregation process from the droplets, it is necessary to analyze the protein structure with keeping the droplet state in a label-free manner, but there was no suitable method. In this study, we observed the structural changes of ataxin-3, a protein associated with Machado–Joseph disease, inside the droplets, using autofluorescence lifetime microscopy. Each droplet showed autofluorescence due to tryptophan (Trp) residues, and its lifetime increased with time, reflecting structural changes toward aggregation. We used Trp mutants to reveal the structural changes around each Trp and showed that the structural change consists of several steps on different timescales. We demonstrated that the present method visualizes the protein dynamics inside a droplet in a label-free manner. Further investigations revealed that the aggregate structure formed in the droplets differs from that formed in dispersed solutions and that a polyglutamine repeat extension in ataxin-3 hardly modulates the aggregation dynamics in the droplets. These findings highlight that the droplet environment facilitates unique protein dynamics different from those in solutions.

    Liquid-liquid phase separation (LLPS) is an important phenomenon in biology, and it is desirable to develop quantitative methods to analyze protein droplets generated by LLPS. This study quantified the change in protein concentration in a droplet in label-free and single-droplet conditions using Raman imaging and the Raman band of water as an intensity standard. Small changes in the protein concentration with variations in pH and salt concentration were observed, and it was shown that the concentration in the droplet decreases as the conditions become less favorable for droplet formation. The effect of exposure to 1,6-hexanediol was also examined, and this additive was found to decrease the protein concentration in the droplet. A model can be proposed in which the addition of 1,6-hexanediol reduces the protein concentration in the droplet, and the droplet disappears when the concentration falls below a certain threshold value.

    The increasing availability of X-ray free-electron lasers (XFELs) has catalyzed the development of single-object structural determination and of structural dynamics tracking in real-time. Disentangling the molecular-level reactions triggered by the interaction with an XFEL pulse is a fundamental step towards developing such applications. Here we report real-time observations of XFEL-induced electronic decay via short-lived transient electronic states in the diiodomethane molecule, using a femtosecond near-infrared probe laser. We determine the lifetimes of the transient states populated during the XFEL-induced Auger cascades and find that multiply charged iodine ions are issued from short-lived (∼20 fs) transient states, whereas the singly charged ones originate from significantly longer-lived states (∼100 fs). We identify the mechanisms behind these different time scales: contrary to the short-lived transient states which relax by molecular Auger decay, the long-lived ones decay by an interatomic Coulombic decay between two iodine atoms, during the molecular fragmentation.

    2021年度の研究では,2020年度に構築した構造化照明ラマン顕微鏡を用いて,超解像ラマンイメージの取得を行った。まず,試料として直径20 nm程度,長さ数10 μm程度のカーボンナノチューブのバンドルファイバーを対象として,2700 cm-1付近のG’バンドを用いてラマンイメージを取得した。励起波長は532 nmの可視光を用い,対物レンズはNAが1.49の油浸対物レンズを用いた。広視野照明光を用いて取得したラマンイメージでは,200 nm以上に観測されていたカーボンナノチューブが,構造化照明光を用いて複数枚の画像を取得し,フーリエ空間で結合することによって得られたラマンイメージでは,回折限界よりも小さい140 nm程度に観測された。このことから,超解像ラマン顕微鏡の構築に成功したと結論づけた。構造化照明を励起光として取得した各ラマンイメージの露光時間は5秒であり,照明光の構造の位相・向きを変えながら9枚の画像を取得し,フーリエ空間上で結合することで1枚の超解像画像を取得したことから,1枚の超解像ラマンイメージの測定時間は1分以下であり,1分程度の時間分解能で,ラベルフリー超解像観測に成功したと言える。さらに,ウィナーフィルターなどのフィルターを用いることで,よりシグナル/ノイズ比の高い超解像ラマンイメージが取得できることを確認した。また,生細胞を対象として,広視野ラマン顕微鏡を用いてC-H伸縮振動バンドを観測することで,脂肪滴や細胞骨格などの細胞内構造が可視化できることを確認した。

    2021年度の研究では,細胞に外部からストレスを与えた際の細胞内夾雑環境の変化を追跡した。外部からのストレスとして,亜ヒ酸を培地中に添加することによって,酸化ストレスを与えた。亜ヒ酸添加後の細胞を固定化し,G3BP1を免疫染色することで,細胞内にストレス顆粒ができていることを確認した。同様に亜ヒ酸処理した生細胞について,多共焦点ラマン顕微鏡を用いてラマンイメージングを行った。その結果,特にC-H伸縮振動バンドの強度が高い点において,タンパク質に由来するAmide Iバンドや核酸に由来するプリン環のバンドの強度が高くなっていることがわかった。これらはストレス顆粒の主要な構成成分であり,このようなスペクトルは酸化ストレスを与えていないコントロール細胞では見られなかったことから,得られたラマンスペクトルはストレス顆粒のスペクトルと考え,生細胞内のストレス顆粒のラベルフリー観測に成功したと結論した。ストレス顆粒と帰属した領域以外でも,コントロール細胞と比較して,細胞質全体のC-H伸縮振動バンドの強度が高くなっていることがわかった。この結果は,細胞質全体で生体分子の濃度が高くなっていることを示唆しており,酸化ストレスとそれに伴うストレス顆粒の生成に伴って細胞内の夾雑環境が変化していることを示している。緩衝溶液中の液滴形成においても,夾雑度の大きさによって液滴内タンパク質の濃度が変化するなど,夾雑環境によって液液相分離が示唆されていることから,酸化ストレスに伴う細胞内夾雑環境の変化がストレス顆粒にどのように影響を与えているかについて,考察した。

    We have determined three-dimensional structure of a metallo-DNA with uninterrupted one-dimensional (1D) metal-array. In this crystal structure, the DNA duplexes containing a 1D metal-array further formed a pseudo-continuous duplex according to the crystal symmetry. This means that 1D metal-array is continuously formed from one edge of the crystal to another edge. We succeeded in creating a 1D metal-array of a visible length. It should be also mentioned that none could obtain such a long metallo-DNA until now, although many researchers have made a lot of trials all over the world. From now on, using this crystal, we were able to examine electrical properties like a conductivity of a 1D metal-array in the long metallo-DNA. In conclusion, we were able to report truly long molecular nanowire made of metallo-DNA molecule, for the first time.

    By combining a high-repetition-rate nanosecond pulsed laser as a light source, and a high-speed camera as a detector, we constructed a fast structured illumination microscope. A laser pulse was split into two beam lines by using a grating and the structured illumination pattern was obtained on a sample as the interference pattern of two or three beams. The introduction of two Pockels' cells into the beam line(s) enables us to modulate the phase of the illumination pattern electrically within few hundreds microseconds. By the high-speed camera synchronized with the Pockels' cells, we obtained fluorescence images illuminated with different-phase pattern. As the result, we succeeded in obtaining optical sectioned images of a fluorescent stained living cell every 3 ms, and super-resolution images within 5 ms.

    The cross section of hyper Raman scattering is 30 orders of magnitude smaller than that of normal Raman scattering and 10 orders of magnitude smaller than that of two-photon fluorescence, which means it is very difficult to observe the hyper Raman process. It was shown that hyper Raman spectra can be measured when molecules are sandwiched between silver nanoparticles inducing the gap-mode plasmon with megahertz high-repetition pulsed laser excitation. In addition, it was found that a thin film prepared from toluene solution of lithium-encapsulated fullerene on a gold substrate showed high electro-conductivity and fullerene cages in the film were negatively charged from tip-enhanced Raman spectra.

    We have constructed the systems to clarify the response mechanism of biomolecules to the application of an external electric field. The constructed IR electroabsorption system was optimized to observe the electric field effect of molecules in solution state with a very small change of a magnitude of 10-6. We have measured the electric field effects on the IR absorption spectra of fatty acids in solution state and observed the broadening of the spectra in the C = O stretching and C - H stretching bands region, which is due to the so-called Stark shift. The anharmonicity of each vibration mode can be discussed based on the magnitude of the observed broadening.

    28年度には、レーザー誘起相分離過程に現れる相分離構造の光学活性について、特にアミノ酸などの添加物の効果を調べた。また、レーザー誘起相分離過程にある溶液を反応場として、アントラセンカルボン酸の光二量化反応を誘起し、その生成物の光学活性及び異性体比を調べることで、その不斉合成反応への反応場としての性質について調べた。 イソブトキシエタノール-水混合溶液のレーザー誘起相分離過程では光学活性は観測されなかったが、溶質分子として不斉炭素を持つアルギニンやアミノブタノールを加えた溶液では、レーザー照射後100 μ秒程度から光学活性が現れ、500 μ秒程度で最大となった。時間分解光散乱法によって相サイズを調べた結果から、100 μ秒程度かけて相や界面が形成され、その後は形成した相が徐々にそのサイズを成長させる様に相分離過程が進むことがわかった。このことから、光学活性が観測された時間はそれぞれの相や界面が形成した時間であることがわかった。また、溶質分子をトリプトファンやイソロイシンとした場合には、過渡的な光学活性はほとんど観測されなかった。これは、相分離後のそれぞれの相へのアミノ酸分子の溶解度の差や相分離温度の変化が原因であると考えられる。 アントラセンカルボン酸の光二量化反応は、近赤外光レーザーパルス照射によって相分離を誘起したのち、一定の遅延時間をおいて紫外光パルスを照射することによって、相分離過程にある溶液中での二量化反応を誘起した。溶液中に不斉炭素を含まない時には生成物の光学活性は現れなかったが、溶質分子としてアミノブタノールを添加することによって、生成物のCDスペクトルが変化し、光学活性が現れた。CDスペクトルは赤外光パルスと紫外光パルスの遅延時間によって変化したことから、相分離構造によって、反応場としての性質が変化し、生成物の光学活性に影響を与えることがわかった。

    The formation and growth mechanism and the optical property of gold nano-square plates synthesized via laser-induced photo-reduction gold ions in dynamic phase separating media were investigated. Phase separation was initiated by a nanosecond IR pulse, and after certain delay time, a UV pulse was introduced to induce photo-reduction in phase separating media, leading the formation of gold nanostructures. Based on the irradiation time and delay time dependence of the size distribution of gold nano-square plates synthesized, we concluded that the growth process of nano-squares by repetitive irradiation of IR and UV pulses was governed by the size of nanophases during phase separation. Optical properties of gold nano-square plates were also investigated. By taking SEM images, dark field images and scattering spectra of the same nanostructures, the size and orientation dependence of optical property was determined.

    The binary liquid mixture of 2-butoxyethanol (2BE) and water, which has its lower critical soluble temperature, has been studied using Raman spectroscopy. The concentration dependence of Raman spectra implies that micelle-like micro-phases forms even at room temperature when the mole fraction of 2BE is 0.01-0.1. Photo-induced phase separation and phase mixing of water and 2BE, in which spiropyran was dissolved as a photo-responsive solute, was investigated. It was found that the phase separation temperature of the merocyanine (MC) form solution was higher than that of the spiropyran (SP) form solution.
