    Rhizostomeae (Scyphozoa) jellyfishes are widespread in neritic waters and include species of commercial importance in Asia. This group comprises jellyfish taxa that host endosymbiotic dinoflagellates of the family Symbiodiniaceae, which provide autotrophic benefits. Despite their value, limited molecular data for Japanese rhizostome taxa has hinder accurate taxonomic classification and interpretation of novel traits. This study combines molecular methods to provide the most complete understanding of molecular phylogenetic relations of Rhizostomeae jellyfishes while assessing the number of Symbiodiniaceae taxa that can be hosted in each species at the medusa level through a new method developed herein for tandem amplification of symbionts and host, validated with microscopy. We also evaluate which rhizostomes produce cassiosomes and whether Symbiodiniaceae are found in the core. Phylogenetic analysis of host mitochondrial (16S rRNA, COI) and nuclear (28S) gene regions of 18 medusae from five genera revealed: (1) Mastigias in Japanese waters corresponds to M. albipunctata ; (2) Cassiopea from Kagoshima likely represents an undescribed species, though Cassiopea xamachana may have been introduced; (3) Two cepheid species - Cephea cephea and Netrostoma setouchianum - occur in Japan; (4) Rhopilema esculentum , a commonly harvested species, is endemic to western Japan. Symbiotic Symbiodiniaceae ITS2 analysis identified three dominant genera ( Symbiodinium , Cladocopium , and Durusdinium ). More than one genus among these was found to be hosted in samples of the genera Mastigias and Cassiopea , indicating plasticity in symbiont association at both the taxon and individual medusa level. Microscopy confirmed cassiosome production exclusively in species examined of the suborder Kolpophorae: Cassiopea sp., N. setouchianum , and M. albipunctata , though absent in a juvenile M. albipunctata sample. Conversely, R. esculentum hosts Symbiodiniaceae but appears to lack the ability to produce cassiosomes. Overall, findings support the distinctive evolution of Symbiodiniaceae–Rhizostomeae symbiosis, the monophyly of the suborder Kolpophorae, and the synapomorphy of cassiosome production in Kolpophorae with onset likely influenced by developmental stage. Broader taxon sampling, especially within Dactyliophorae, will provide further clues on the functional evolution and cellular organization underlying photoendosymbiosis and cassiosome production in these medusozoans.

    The endoparasitic dinoflagellates belonging to the genus Amoebophrya can infect a broad range of free-living marine dinoflagellates, including harmful/toxic species. The parasite kills its host; the high prevalence of the parasite has been suggested to be a significant factor for the termination of dinoflagellate blooms in marine systems. The issues involved in culturing host-parasite systems have greatly restricted further research on Amoebophrya biology. Here, we established the culture of a novel strain of Amoebophrya sp. ex Alexandrium catenella (Group I) from Osaka Bay, Japan, and studied its genetic diversity, host specificity, and prevalence in the field. Genetic analysis established that the strain we isolated was a novel culture strain infecting A. catenella. Among the host species tested, the Amoebophrya sp. could infect the genera Alexandrium and Prorocentrum in culture, and the infection was also confirmed in the genus Tripos in a field sample. A maximum prevalence of 73% was recorded during the Alexandrium bloom period in Osaka Bay, after which the host cell density rapidly declined. Our results indicated that the existence of the parasite had a significant effect on the dynamics of A. catenella, especially on the termination of the blooms.

    We analyzed cryptophyte nucleomorph 18S rRNA gene sequences retained in natural Myrionecta rubra cells and plastid 16S rRNA gene and psbA sequences retained in natural cells of several Dinophysis species collected from Japanese coastal waters. A total of 715 nucleomorph sequences obtained from 134 M. rubra cells and 564 plastid 16S rRNA gene and 355 psbA sequences from 71 Dinophysis cells were determined. Almost all sequences in M. rubra and Dinophysis spp. were identical to those of Teleaulax amphioxeia, suggesting that M. rubra in Japanese coastal waters preferentially ingest T. amphioxeia. The remaining sequences were closely related to those of Geminigera cryophila and Teleaulax acuta. Interestingly, 37 plastid 16S rRNA gene sequences, which were different from T. amphioxeia and amplified from Dinophysis acuminata and Dinophysis norvegica cells, were identical to the sequence of a D. acuminata cell found in the Greenland Sea, suggesting that a widely distributed and unknown cryptophyte species is also preyed upon by M. rubra and subsequently sequestered by Dinophysis. To confirm the reliability of molecular identification of the cryptophyte Teleaulax species detected from M. rubra and Dinophysis cells, the nucleomorph and plastid genes of Teleaulax species isolated from seawaters were also analyzed. Of 19 isolates, 16 and 3 clonal strains were identified as T. amphioxeia and T. acuta, respectively, and no sequence variation was confirmed within species. T. amphioxeia is probably the primary source of prey for M. rubra in Japanese coastal waters. An unknown cryptophyte may serve as an additional source, depending on localities and seasons.

    Gene analyses in the food vacuoles of the natural cells of D. acuminata collected from an enclosed port located at Ise Bay, central Japan, were pioneered in this study. From the gene evidences, M. rubrum was revealed as prey item. Water sampling was also conducted weekly or biweekly at the port from March 2012 to July 2015. The water samples were preserved with Bouin’s solution and both organisms (D. acuminate and M. rubrum) were enumerated under a microscope. The temporal changes in the abundances of both organisms often displayed the dynamics in which D. acuminata was flourished after the increase of M. rubrum and vice versa, indicating their prey-predator relationship in nature. In this study, prey organisms and prey-predator relationship for the toxic dinoflagellate D. rotundata were also revealed in nature.

    Totally 516 natural bivalve larvae collected from Matsushima Bay were isolated and identified by molecular methods. A multiplex PCR assay has been developed that enables simultaneous detection of seven bivalve larvae species on the basis of differences in the lengths of the PCR products. The gut contents were analyzed to clarify the potential food sources of the adult oyster. Using universal primer and gene cloning, sequences of suspended phytoplankton, such as diatom Thalassiosira spp. and dinoflagellate Scrippsiella trochoidea, were mainly detected with other sequences of Bolidophyceae, sea squirts, red algae, ciliates, and copepoda.

    This study showed the ecological relationship among Dinophysis, ciliate, and cryptophyte in Japanese coastal areas. Many Dinophysis species ingested the ciliate Myrionecta rubra, and further the ciliate preferentially ingested the cryptophyte Teleaulax amphioxeia. We also revealed that Dinophysis mitra retained more than 100 kinds of plastids robbed from algal prey.

    We examined characteristics of growth, nutritional physiology and lipophilic toxin production of the genus Dinophysis and effects of environmental conditions on them. Characteristics of cellular toxin contents and toxin composition of natural Dinophysis spp. collected in various Japanese coastal waters were clarified. In laboratory experiments growth of Dinophysis spp. depends on specific prey source, the ciliate Myrionecta rubra, feeding on the Cryptophyceae Teleaulax amphioxeia, and was influenced by temperature and light intensity. Toxin production also varied in response to these environmental factors and growth phase. Parts of toxin components were excreted into surrounding seawater with progress of the growth. Cellular toxin contents were occasionally largely different among local strains of Dinophysis species cultivated under the same conditions, suggesting effects of genetic characteristics on the toxin production.
