    The Cryptococcus neoformans species complex (CNSC), responsible for cryptococcosis, is controlled by Th1-type immunity, in which IFN-γ-activated macrophages form granulomas that contain infection. Nevertheless, CNSC can evade host immunity and, after the primary infection phase, can shift to a latent infection similar to tuberculosis. Reactivation is thought to occur when immune control fails, but the underlying mechanisms remain poorly understood. FTY720, a functional antagonist of sphingosine-1-phosphate receptors, is primarily used in the treatment of multiple sclerosis. However, there have been reports linking its use to instances of cryptococcosis in patients undergoing treatment. To explore this, we established a novel mouse model using CnT-II mice, which express CD4+ T cell receptors specific for chitin deacetylase 2 (Cda2), a major T cell antigen of CNSC. Following infection with encapsulated CNSC, mice developed persistent pulmonary fungal burdens (10²-10³ CFU) accompanied by granulomatous responses, modeling latent infection. Upon FTY720 administration, mice exhibited increased pulmonary fungal burdens, disrupted granuloma integrity, and reduced IFN-γ and IL-12 production. FTY720 also markedly decreased CD4+ effector memory (Tem) and effector T cells (Teff) in the lungs, with a particularly profound loss of IFN-γ-producing Tem cells. These findings indicate that our model successfully recapitulates latent cryptococcal infection and reactivation, and demonstrate that FTY720 promotes relapse by depleting protective IFN-γ-producing CD4+ Tem cells and impairing Th1-mediated immunity. Given the rising use of FTY720, our study highlights its potential risk in patients with subclinical cryptococcosis and underscores the need for preventive strategies to avert disease progression.

    The inflammatory response after skin injury initiates wound healing by promoting the release of cytokines and growth factors essential for tissue repair. Modern wound dressings promote healing by maintaining a moist, hypoxic environment. ATKPAD is a novel wound dressing that accelerates healing developed by Okamoto Industries, Inc. Composed of reduced starch syrup and a semi-permeable membrane, it was launched in Japan as a Class III medical device in 2024. Transparent ATKPAD allows wound visibility and absorbs exudate in a molecular-weight-dependent manner, with its semi-permeable membrane retaining larger molecules, like cytokines, while letting small molecules pass through osmotic effects rather than specific binding. However, the detailed molecular and cellular mechanisms of ATKPAD remain unclear. In this study, we examined its effect on full-thickness excisional wound healing. We analyzed the in vitro permeability of cytokines and growth factors and evaluated granulation tissue formation, angiogenesis, neutrophil infiltration, and cytokine and growth factor production in vivo. We observed an increase in cytokines and growth factors alongside accelerated wound healing with ATKPAD. Its application enhanced granulation tissue formation and neutrophil infiltration, accompanied by increased expression of Hif1α. These findings suggest that ATKPAD promotes wound healing by creating a hypoxic microenvironment that enhances early-phase inflammation. (Word count: 200).

    Obesity is recognized as a chronic low-grade inflammation that contributes to metabolic disorders. Weight loss (WL) is well known to improve metabolic disorders. However, recent studies have shown that the changes in immune cells associated with obesity exhibit immunological memory even after WL. Obesity impairs skin barrier function and exacerbates inflammatory skin diseases, such as psoriasis. While skin immune cells maintain homeostasis and defense, the impact of obesity on these cells in intact skin is understudied, as research mainly focuses on skin with an inflammatory skin disease model in the context of obesity. The effect of WL on skin immunity is even less clear. Therefore, this study aimed to investigate these discrepancies. Mice were assigned to the lean group (regular diet), the obese group (high-fat diet for 18 weeks), and the WL group (high-fat diet for 9 weeks followed by regular diet for 9 weeks). Following 18 weeks of feeding, mouse skin was excised, and immune cell populations within the skin were analyzed using real-time PCR and flow cytometry. In addition, the effects of WL on psoriasis were examined using an imiquimod-induced psoriasis mouse model. We observed increased expression of RORγt, IL-17A, and CCL20 in intact skin in both the obese and WL groups. In the psoriasis model, disease severity was further exacerbated by WL. Moreover, whereas the Vγ4+Vγ5- γδ T cell population was increased in the lean group, the Vγ4-Vγ5- γδ T cell population remained elevated following WL, similar to levels observed in the obese group. These observations indicated that obesity may imprint a memorized immune response in the skin, which is not abrogated by WL. This study is the first to demonstrate this persistent effect on skin immunity. These findings imply that individuals who have experienced obesity may remain at increased risk for inflammatory skin conditions, even after WL.

    Pneumonia is one of the leading causes of death worldwide, with Streptococcus pneumoniae being a major causative agent of community-acquired pneumonia. In immunocompromised infants and the elderly, this pathogen can invade the bloodstream and cerebrospinal fluid, leading to invasive pneumococcal diseases (IPDs), such as bacteremia and meningitis. Currently, vaccines targeting the capsular polysaccharides of pneumococcus are available to prevent pneumococcal infection. However, these vaccines have limitations, including incomplete coverage of all serotypes. Therefore, there is a need for next-generation vaccines that are independent of serotype variation. In this study, we developed a novel pneumococcal vaccine using pneumococcal surface protein A (PspA), a virulence factor presents on the surface of all pneumococcal strains, as the antigen. PspA was conjugated with iron oxide nanoparticles (IONPs), serving as both a carrier and a potential adjuvant. When administered intratracheally to mice, this vaccine, PspA-IONP, induced the formation of inducible bronchus-associated lymphoid tissue (iBALT) in the lungs and stimulated antibody production both systemically and on mucosal surfaces, thereby conferring resistance to pneumococcal infection. We focused on invariant natural killer T (iNKT) cells due to their role in antibody production by B cells. Notably, mucosal IgA antibodies induced by this vaccine were significantly reduced in iNKT cell-deficient (Jα18KO) mice compared to wild-type mice. The expression of pIgR, which is crucial for IgA secretion, as well as the production of several cytokines involved in pIgR expression and B cell activation, were also significantly decreased in Jα18KO mice. These results suggest that iNKT cells play a central role in promoting IgA secretion by directly or indirectly inducing multiple cytokines. PspA-IONP demonstrates potential as a novel vaccine platform that induces anti-pneumococcal immunity. Furthermore, this platform can be adapted to combat other respiratory infections by modifying the antigen conjugated to the IONPs.

    BACKGROUND: In burn wound healing, inflammation and oxidative stress are major contributors to delayed repair. Damage-associated molecular patterns (DAMPs) derived from injured tissue activate inflammatory signaling pathways through receptors, such as C-type lectin receptors (CLRs). Dectin-1, one of the receptors for DAMPs, contributes to the production of inflammatory cytokines and reactive oxygen species (ROS). The purpose of this study was to investigate the role of Dectin-1 in burn wound healing using Dectin-1-deficient (KO) mice and the antagonist laminarin. METHOD: Full-thickness burn wounds were created on the dorsal skin of wild-type (WT) C57BL/6 mice and Dectin-1KO mice by applying a 90°C soldering iron for 10 s. We analyzed the percentage of wound closure, the area of granulation tissue, angiogenesis, and inflammatory responses. To evaluate the effect of the topical administration of the Dectin-1 antagonist laminarin, we applied laminarin or vehicle at the burned sites daily and examined burn wound healing. RESULTS: The percentages of wound closure, granulation tissue area, and angiogenesis were significantly enhanced in Dectin-1KO mice compared with WT mice. Dectin-1 deficiency also led to reduced production of inflammatory cytokines such as TNF-α and IFN-γ, and the oxidative damage marker 4-HNE. Furthermore, daily topical administration of the antagonist laminarin promoted burn wound healing. CONCLUSIONS: These results suggest that Dectin-1-mediated signaling inhibits burn wound healing by inducing the production of inflammatory cytokines and oxidative stress. Furthermore, topical administration of the Dectin-1 antagonist laminarin may serve as a promising novel treatment for healing after burn.

ATKPAD is unique in its transparency, allowing clear wound visibility, and selectively absorbs wound exudate through the combined effects of osmotic pressure and its semi-permeable membrane. However, the detailed molecular and cellular mechanisms of ATKPAD remain unclear. In this study, we examined its effect on acute wound healing. We analyzed the in vitro permeability of cytokines and growth factors and evaluated granulation tissue formation, angiogenesis, neutrophil infiltration, and cytokine and growth factor production in vivo. We found that cytokines and growth factors were selectively retained by ATKPAD. Its application enhanced granulation tissue formation and neutrophil infiltration, accompanied by increased hypoxia marker expression. These findings suggest that ATKPAD promotes wound healing by creating a hypoxic microenvironment that enhances early-phase inflammation while preserving key bioactive factors at the wound site.</p>

    BACKGROUND: In the process of burn wound healing, promoting healing and suppressing wound progression by enhancing blood flow is considered essential. Various agents that increase blood flow are currently being assessed. Batroxobin (DF-521; Defibrase®) is a denitrogenating agent extracted from Bothrops moojeni, used as a thrombin-like serine protease to improve ischemic conditions. However, it remains unclear how this agent affects the burn wound healing process. In this study, we conducted analyses to define the effects of batroxobin administration on burn wound healing. METHODS: Full-thickness burn wounds were created on the dorsal skin of C57BL/6 mice by applying a 90℃, 5-mm-diameter soldering iron for 10 s. Immediately after wounding, batroxobin (30 batroxobin units/kg/mouse) was administered intraperitoneally daily. As a vehicle control, the same volume of saline was administered. We analyzed wound area, histological findings, blood flow, growth factor and chemokine synthesis, and ischemia-reperfusion related factor expression. RESULTS: We found that the systemic administration of batroxobin prevented burn wound progression, which was accompanied by decreased expression of TNF-α and NOX2 and reduced synthesis of Hif-1α. In addition, this agent promoted wound healing by enhancing blood flow, increasing S100A4-positive fibroblast accumulation, and stimulating the production of growth factors (bFGF, EGF, and PlGF). CONCLUSIONS: These results indicate that the systemic administration of batroxobin prevented burn wound progression and accelerated the healing process by enhancing blood flow. (Word count: 222).

    本年度は急性創傷におけるナノ型乳酸菌の効果の検証と熱傷創モデルの確立を中心に研究を進めた。 急性創傷におけるナノ型乳酸菌の効果に関しては、直接投与の濃度を検証し、さらに初回単回投与での創閉鎖率の改善を認めた。加えて、病理学的解析においても、CD31陽性血管数やαSMA陽性筋繊維芽細胞数の増加を認め、創傷治癒に関わる肉芽の増生を認めた。上皮化に関しても、上皮の進展率の上昇を認め、増生した肉芽上に皮膚が進展し、治癒が促進していることを確認できた。 急性期において炎症の調節に必要なサイトカインに関してELISA法での解析を行ったところ、IL-6やTNF-αの早期での上昇を認めた。加えて、好中球の遊走に関与するCXCL2やCXCL1なども著明な上昇を認めた。これらは、乳酸菌を急性創傷に投与することで創部における炎症が惹起され、創傷治癒過程における炎症期のメカニズムと合致する。炎症を惹起することで創の清浄化が促進する可能性が示唆された。さらに、抗炎症に作用するI L-10も後期にて上昇を認めた。炎症期から増殖期への移行を適切にかつ早期に誘導することで、創傷治癒を早めている可能性がある。今後、フローサイトメトリーで創部に遊走される白血球の分画を解明し、創傷治癒過程の変化を観察する予定である。 熱傷創モデルに関しては、マウス背部に電気ごてで作成した熱傷創の治癒過程及び病理学的変化を解析した。熱傷創が治癒する過程と、温度による変化、保護剤による違いなどを検討している。今後、標準的な治癒過程を明らかにし、ナノ型乳酸菌投与による変化を比較検討していく予定である。

    The wound healing promoting effect of protamine, a substance derived from salmon milt, was examined. The cytokine release and changes in cell number induced by protamine administration in cultured cells were examined in detail. The involvement of cytokine induction not only in mouse macrophages but also in dendritic cell stimulation was suggested, and the lipopolysaccharide in the protamine formulation used may be involved, and the effect was examined in an endotoxin inactivation experiment using polymyxin B. As a result, no induction of inflammation was observed with protamine alone, suggesting the possibility that the substances contained in the product promote healing.

    Chronic wounds are intractable due to the combined effects of biofilm formation and impaired blood flow. However, there is no universal wound care that can simultaneously approach multiple causes. In this study, we aimed to establish universal wound care by elucidating the effect of defibrase, which has already been shown to inhibit neutrophil extracellular trapping, on wound healing. Using acute and chronic wound models, we analyzed the effects of defibrase on wound healing and found that defibrase promotes wound healing by analyzing wound closure rate, re-epithelialization rate, granulation area, and angiogenesis.

    The purpose of our study is to develop an “innovative wound care using heat-killed nano-type lactic acid bacteria (nano-type LAB) through skin-intestine networks”. We analyzed villus length, the Foxp3 positive cells and IL-10 positive cells in small intestine after oral administration of nano-type LAB in immunosuppressive mice. We found that nano-type LAB contributed to increased villus length, Foxp3 positive cells and IL-10 positive cells. Thus, we concluded that heat-killed nano-type LAB may be involved in controlling intestional conditions by anti-inflammatory activity.

    The purpose of our study is to elucidate the crosstalk between bacterial communities and biofilm formation. N-(3-Oxododecanoyl)-L-homoserine lactone (3-oxo-C12-HSL) is synthesized by an autoinducer synthase encoded by the bacterial lasI gene in P. aeruginosa, which regulates the production of virulence factors and biofilm formation in this bacterium. Wounds were created on the backs of Sprague-Dawley (SD) rats and 3-oxo-C12-HSL was administered on to the wound surface. Administration of 3-oxo-C12-HSL significantly promoted wound healing and neutrophil accumulation, and these responses were reversed by treatment with anti-3-oxo-C12-HSL antiserum. These results suggest that 3-oxo-C12-HSL may be involved in biofilm formation through inflammatory response such as neutrophil accumulation.

    C-type lectin receptors (CLRs) recognize microbial polysaccharides. The precise roles of these CLRs in wound healing remain unclear. This study was conducted to determine the roles of Dectin-1 and Dectin-2 in wound healing, with a particular focus on the kinetics of neutrophilic inflammatory response. Wounds were created on the backs of C57BL/6 mice, and the effects of Dectin-1 or Dectin-2 deficiency. We also analyzed wound closure, histological findings, and neutrophilic inflammatory response, including neutrophil extracellular trap formation. We found that Dectin-1 contributed to the acceleration of wound healing by inducing early-phase neutrophil accumulation, whereas Dectin-2 was involved in prolonged neutrophilic responses and neutrophil extracellular trap formation, leading to delayed wound healing. These results suggest that Dectin-1 and Dectin-2 have different roles in wound healing through their different effects on the neutrophilic response.

    The purpose of our study is to develop an “innovative skin wound care using nano-type lactic acid bacteria (heat-killed KH2).” We analyzed percent wound closure and granulation area. We found that heat-killed KH2 contributed to the acceleration of the formation of granulation tissue by inducing tumor necrosis factor-α, interleukin-6, basic fibroblast growth factor, transforming growth factor (TGF)-β1 production. In addition, heat-killed KH2 also improved wound closure, which was accompanied by the increased production of TGF-β1 in diabetic mice. Topical administration of heat-killed KH2 might have therapeutic potential for the treatment of chronic skin wounds in diabetes mellitus. In the present study, we concluded that heat-killed KH2 promoted skin wound-healing through the formation of granulation tissues and the production of inflammatory cytokines and growth factors.

    Interferon (IFN)-γ is mainly secreted by CD4+ T helper 1 (Th1), natural killer (NK) and NKT cells after skin injury. Although IFN-γ is well known regarding its inhibitory effects on collagen synthesis by fibroblasts in vitro,information is limited regarding its role in wound healing in vivo.In the present study, we analyzed how the defect of IFN-γ affects wound healing. Full-thickness wounds were created on the backs of wild type (WT) C57BL/6 and IFN-γ-deficient (KO) mice. We analyzed the percent wound closure, accumulation of leukocytes, and matrix metalloproteinases (MMPs). IFN-γKO mice exhibited significant attenuation in wound closure on Day 10, characteristics that are associated with prolonged neutrophil accumulation. Expression levels of MMP-2 mRNA were significantly greater in IFN-γKO than in WT. Moreover, under neutropenic conditions created with anti-Gr-1 monoclonal antibodies, wound closure in IFN-γKO mice was recovered through low MMP-2 expression levels.

    Bacteria are detected in more than 90% of chronic wounds such as pressure ulcers and diabetic leg ulcers, and Pseudomonas aeruginosa is one of the most frequently detected bacteria. In this study, we analyzed the effect of NKT cell activation on the elimination of P. aeruginosa in wounds. Administration of α-GalCer, a specific activator of NKT cells, resulted in significantly decreased the number of live colonies in the wounded tissues on days 5 and 7 after P. aeruginosa inoculation, and significantly increased the number of neutrophils on day 3. IFN-γ, IL-22, IL-23, IL-12p70, and S100A9 production were significantly higher in α-GalCer-treated group than in vehicle-treated group. These results suggest that iNKT cells promote P. aeruginosa clearance during skin wound healing.

    It is well known about immunocompromised state and neutrophil dysfunction in diabetic foot ulcers. But the detailed mechanism of neutrophil dysfunction remains unclear. In the current study, to identify the possible contribution of NETosis the co-localization between Ly6G (marker of neutrophils) and citrullinated histon (an indicator of NETosis) and elastase activity after α-mannan administration were examined. We discovered that the expression of citrullinated histone was accerated at the wound sites. In addition, elastase activity level was significantly higher in α-mannan-administrated mice than in vehicle-administered mice. We next investigated the effect of sivelestat sodium, a neutrophil elastase inhibitor, on wound closure in WT mice administered α-mannan, and the administration of sivelestat sodium was found to significantly improve the delayed wound closure caused by α-mannan.

    This study was to evaluate the effect of the spine, left lateral decubitus, and right lateral decubitus position on autonomic nervous activity elderly by using spectral analysis heart rate variability (HRV). Forty-five elderly adults were enrolled. The lowest heart rate continued for 10 min, when participants were left lateral decubitus position on compared with the other two positions, The low frequency HRV to high-frequency HRV ratio (LF/HF) for the right lateral decubitus position was significantly lower than that for the other position. The right lateral decubitus position may attenuate sympathetic nerve activity in elder adults.

    Chronic wound infections are considered one of the most severe wound problem. Indeed, some studies reported bacteria are present over 90 % of chronic wounds. Pseudomonas aeruginosa is frequently isolated from chronic wounds and thought to be a cause of delayed wound healing. Previously, we showed that iNKT cell deficiency resulted in a delayed wound healing process. Other group showed that NKT cells enhanced pulmonary clearance of this bacteria. However, the role of NKT cells in the host defense to P. aeruginosa infection at the wound sites remains to be elucidated. In this study, we conducted to elucidate the role of NKT cells in the host defense to P. aeruginosa infection at the wound site. NKT cells promote re-epithelialization and P. aeruginosa clearance during wound healing. This mechanism may be associated with increased levels of IL-17A and β-defensin1.

    In chronic wounds, the inflammatory responses are prolonged with persistent neutrophil infiltration. Recently, we showed that skin wound healing was delayed and the healing process was impaired under conditions lacking invariant natural killer T (iNKT) cells, an innate immune lymphocyte with potent immuno-regulatory activity. In the present study, to identify the possible contribution of NKT cells activation, the effects of α-Galactosylceramide (α-GalCer) administration on wound healing, specific activator of iNKT cells, were examined. Administration of a-GalCer at the wound sites significantly promoted wound closure, granulation tissue formation, angiogenesis compared with vehicle treatment. In addition, the inflammatory cells were rapidly decreased and granulation tissue were enhanced in α-GalCer administered mice.

    The role of endogenous damage-related molecular patterns (DAMPs) in skin wound healing remains unclear. In the present study, to identify the possible contribution of CLRs, such as Dectin-2, the expression of Dectin-2 mRNA was examined. Dectin-2 mRNA expression reached peak levels at 12 hours in WT mice, and expression levels declined thereafter to the basal level. we found that both neutrophils and macrophages expressed Dectin-2, and the proportions of these cells expressing Dectin-2 were 40.1±5.0% and 32.1±2.9%, respectively. In immunohistochemical analysis, DAMPs recognized by Dectin-2 was increased in infiltrating cells at the wound site.

    Macrophage inducible C-type lectin (Mincle) is an activating receptor, expressed in macrophages and dendritic cells, which senses PAMPs and DAMPs. However, the role of Mincle at the wound sites has not been elucidated. In the present study, we addressed a question how defect of Mincle affected to the healing process of wounds. Wounds were created on the backs of wild type (WT) C57BL/6 and Mincle-deficient (KO) mice. We analyzed percent wound closure, height of granulation tissue, expression of Mincle by real-time PCR and immunohistochemistry. The percent wound closure was significantly increased in Mincle KO mice compared to WT mice, which was associated with the increased height of granulation tissues. Mincle was detected at the early phase after wound creation. These results suggest that Mincle may be involved in the regulation of granulation at the wounded tissues, which may lead to delayed wound healing.

    Pseudomonas aeruginosa is frequently isolated from chronic wounds.NKT cells play an important role in various aspects of the regulation immune responses, including protection against bacterial infections. However, it is not fully understood how this lymphocyte is involved in the host defense against P. aeruginosa wound infection. In the present sudy,we elucidated the role of NKT cells in clearance of P. aeruginosa from the wound. NKT cell-deficient mice showed impaired clearance of P. aeruginosa. These results indicate that NKT cells may be involved in the clearance of P. aeruginosa from wounds.

    In particular, parasympathetic nerve activity as well as arterial baroreflex sensitivity reduces with aging process. In elderly patients with coronary artery disease, high parasympathetic nerve activity and low sympathetic nerve activity have been observed in right lateral decubitus position compared with supine and left lateral decubitus positions. However, little research has been conducted to determine sex and age differences in autonomic nervous activity in different positions, i.e., supine, left recumbent, and right recumbent positions. The aim of this study was to evaluate the effect of the left and right recumbent positions on heart rate variability in elderly was the lowest among the three positions regardless of sex. In elderly men, the LF/HF ratio in right recumbent position was significantly lower than that in supine position. Right recumbent position may attenuate cardiac sympathetic nerve activity in elderly adult men.

    Interleukin-17 (IL-17) is a proinflammatory cytokine. The role of this cytokine in the wound healing process is not fully clarified. In this study, IL-17A knockout mice showed promoted wound healing and decreased neutrophil accumulation compared with wild-type mice, and the administration of recombinant IL-17A led to delayed wound closure, accelerated neutrophilic accumulation and improved wound repair with treatment of a neutrophil elastase inhibitor. Neutrophils, macrophages and gamma-delta T cels showed IL-17A expression after wounding in wild-type mice. These results indicated that IL-17A hampared the wound healing process and neutrophilic inflammation caused by IL-17 led to impaired wound healing in skin.

    Low birth weight(LBW) conformed as a risk of high blood pressure in later life.However, a number of studies have shown that Mongolian people have higher prevalence of hypertension compared to other ethnic groups. including the Japanese. The purpose of this study was to identify whether there are differences in the relationship of LBW and the risk of hypertension between Japanese and Mongolian.We measured BP and heart rate variability at resting condition and postal change for both Japanese and Mongolian volunteers with LBW and normal birth weight,respectively. We conclude that Mongolian young adults a higher risk of hypertension compared to the Japanese counterpart.
