    During sexual reproduction in the ciliate Tetrahymena thermophila, meiosis occurs in the germline micronucleus, resulting in the formation of four haploid micronuclei. Of these, only one is selected to evade autophagy, and subsequently migrates to the membrane junction with the partner cell for reciprocal pronuclear exchange. We previously demonstrated that the transmembrane protein Semi1 is essential for this nuclear migration. Semi1 is specifically expressed in mating cells and localizes to the periphery of the selected nucleus. Loss of Semi1 disrupts nuclear attachment to the junction, leading to infertility. However, the mechanism by which Semi1 positions the nucleus at the junction remains unclear. Here, we report that the Semi1-interacting protein, Semi2, is also necessary for proper nuclear positioning. Deletion of Semi2 results in the same nuclear mislocalization phenotype and infertility observed in Semi1 mutant cells. Semi2 colocalizes with Semi1, but in the absence of Semi1, Semi2 fails to exhibit perinuclear localization. The selected nucleus anchors to microtubules prior to migration, a process dependent on both Semi1 and Semi2. We propose a model in which Semi1 recruits Semi2 to the selected nucleus, facilitating the interaction between the nucleus and microtubules required for proper nuclear positioning at the membrane junction.

    Long-chain fatty acid (LCFA) degradation primarily involves several species of Syntrophomonas and hydrogenotrophic methanogens, constituting the rate-limiting step in anaerobic digestion. It is crucial to augment their abundance to enhance LCFA degradation. Utilizing microbial carriers presents an effective strategy to maintain the microorganisms on the surface and prevent their washout from the digester. In this study, we aimed to identify a suitable microbial carrier with a superior adsorption capacity for LCFA-degrading microorganisms. We tested various polymers, poly(vinyl alcohol) (PVA), polypropylene (PP), polyethylene glycol (PEG), and polyvinylidene chloride (PVDC), adding them to the sludge at the concentration of 28.25 g L-1 and incubating with olive oil. The amplicon sequencing analysis revealed that PVDC retained Syntrophomonas more abundantly than the other polymers. Remarkably, PVDC predominantly adsorbed LCFA-degrading S. sapovorans and S. zehnderi, whereas medium- to short-chain fatty acid-degrading S. wolfei was abundant in the sludge. Moreover, hydrogenotrophic Methanospirillum hungatei was detected at 2.3-9.5 times higher abundance on PVDC compared to the sludge. Further analysis indicated that not only these LCFA-degrading syntrophic microbial communities but also Propionispira and Anaerosinus, which are capable of lipid hydrolysis and glycerol degradation, became dominant on PVDC. Actually, chemical analysis confirmed that adding PVDC promoted the olive oil degradation. These results underscore the potential of PVDC in promoting anaerobic LCFA degradation.

    Ecological dynamics is driven by complex ecological networks. Computational capabilities of artificial networks have been exploited for machine learning purposes, yet whether an ecological network possesses a computational capability and whether/how we can use it remain unclear. Here, we developed two new computational/empirical frameworks based on reservoir computing and show that ecological dynamics can be used as a computational resource. In silico ecological reservoir computing (ERC) reconstructs ecological dynamics from empirical time series and uses simulated system responses for information processing, which can predict near future of chaotic dynamics and emulate nonlinear dynamics. The real-time ERC uses real population dynamics of a unicellular organism, Tetrahymena thermophila . The temperature of the medium is an input signal and population dynamics is used as a computational resource. Intriguingly, the real-time ecological reservoir has necessary conditions for computing (e.g. synchronized dynamics in response to the same input sequences) and can make near-future predictions of empirical time series, showing the first empirical evidence that population-level phenomenon is capable of real-time computations. Our finding that ecological dynamics possess computational capability poses new research questions for computational science and ecology: how can we efficiently use it and how is it actually used, evolved and maintained in an ecosystem?

    During sexual reproduction/conjugation of the ciliate Tetrahymena thermophila, the germinal micronucleus undergoes meiosis resulting in four haploid micronuclei (hMICs). All hMICs undergo post-meiotic DNA double-strand break (PM-DSB) formation, cleaving their genome. DNA lesions are subsequently repaired in only one ‘selected’ hMIC, which eventually produces gametic pronuclei. DNA repair in the selected hMIC involves chromatin remodeling by switching from the heterochromatic to the euchromatic state of its genome. Here, we demonstrate that, among the 15 Tetrahymena Snf2 family proteins, a core of the ATP-dependent chromatin remodeling complex in Tetrahymena, the germline nucleus specific Iswi in Tetrahymena IswiGTt and Rad5Tt is crucial for the generation of gametic pronuclei. In either gene knockout, the selected hMIC which shows euchromatin markers such as lysine-acetylated histone H3 does not appear, but all hMICs in which markers for DNA lesions persist are degraded, indicating that both IswiGTt and Rad5Tt have important roles in repairing PM-DSB DNA lesions and remodeling chromatin for the euchromatic state in the selected hMIC.

    Little information is available on the epidemiology of Cryptosporidium in pigs in central Vietnam. The aims of this study were to investigate the prevalence and to characterize the genotype distribution of Cryptosporidium isolates in pigs in this region. A total of 193 pig fecal samples were screened for the presence of Cryptosporidium oocysts using the modified Ziehl-Neelsen staining method, and 28 (overall prevalence 14.5 %) were identified as positive by microscopic observation. Positive samples were further analyzed by polymerase chain reaction amplification and sequencing. Genetic identification based on the 18S ribosomal RNA and 70 kDa heat shock protein genes revealed that pigs in Vietnam are infected with two species/genotypes (Cryptosporidium suis and Cryptosporidium pig genotype II). This study is the first molecular characterization of Cryptosporidium in pigs in Vietnam. The presence of these host-adapted species/genotypes suggests that pigs may not pose a significant public health risk in this area. More extensive studies are necessary to ascertain the zoonotic potential of Cryptosporidium in porcine hosts in Vietnam.

    ABSTRACT Spaceflight is important to explore the universe. However, astronauts face many health challenges in space including the increased risk of infection. During spaceflight, the microgravity weakens immunity, which causes opportunistic infections with pathogens such as herpesvirus. However, we have no information about the effects of microgravity on infections caused by other pathogens with a latent stage in their life cycle. In addition, there are no reports on host cell responses or the mechanisms that modulate changes under microgravity conditions. Due to the high cost and effort required for space study in actual microgravity, simulated microgravity plays an important role in mimicking conditions that allow researchers to study risks during spaceflights. Toxoplasma gondii is one of the top five important economic pathogens; it causes toxoplasmosis, may reactivate from the latent phase in immunocompromised individuals, and there is no active drug or vaccine. In this study, we observed the effect of simulated microgravity on T. gondii and on host cells. Simulated microgravity using a 3D clinostat induced host cell proliferation and caused aggregated cells to form three dimensional multicellular spheroids in Vero cells and HFF. Rapid egress and growth of tachyzoites during the acute phase of type II ME49 infection were observed under simulated microgravity.

    BACKGROUND: Serine/arginine-rich splicing factor 1 (SR1), an orthologue of hypothetical RNA-binding protein (HRB1) in yeast, is essential to the asexual development of Plasmodium falciparum and P. berghei. However, their interacting proteins in malaria parasites remain unclear. METHODS: To identify SR1-interacting proteins in malaria parasites, transgenic Pb ANKA expressing green fluorescent protein-fused PbSR1 (PBANKA_1232100) was generated and performed immunoprecipitation coupled to mass spectrometry (IP-MS) using the transgenic parasites. To investigate the developmental stage at which PbSR1 and RhopH2 are expressed, transgenic parasites co-expressing the fusion proteins PbSR1::GFP and RhopH2::mCherry were generated and western blot analysis and live-cell fluorescence imaging were performed. RESULTS: The fluorescence signal of PbSR1::GFP was stronger in nuclei than the cytoplasm of malaria parasites. IP-MS of the transgenic parasites indicated that PbSR1 interacts with nuclear proteins, including RNA-binding protein and small nuclear ribonucleoprotein, and cytoplasmic proteins, such as RhopH1 (or Clag3), RhopH2, and RhopH3. Live-cell fluorescence imaging showed that co-localization of PbSR1 and RhopH2 in the cytoplasm was observed in trophozoites and gametocytes but not mature schizonts and merozoites. From these results, trophozoites, immature schizonts and gametocytes are candidate stages at which cytoplasmic PbSR1 interacts with RhopH2. CONCLUSIONS: PbSR1 interacts with nuclear proteins and rhoptry proteins.

    Abstract Anaerobic digestion of lipid-rich wastes holds significant potential for enhanced biomethane production, due to the high energy density of lipids. However, to fully harness this potential, a deeper understanding of lipolytic microorganisms is essential, as key microbial players involved in lipid hydrolysis remain largely unidentified. In this study, we employed an integrated approach combining zymography, metaproteomics, and metagenomics to identify the lipolytic microorganisms from anaerobic digester sludge. This activity-based strategy identified a novel lipase distantly related to known lipases. Besides, although this lipase originates from a mesophilic environment, it exhibited unexpected extremophilic-like properties, with maximal activity at 97.5 °C and pH 11. We further reconstructed a metagenome-assembled genome encoding this lipase and demonstrated that it likely represents a novel genus closely related to Candidatus Scatomorpha. Metabolic reconstruction suggested that this bacterium hydrolyzes extracellular lipids and utilizes the resulting hydrolysate, glycerol, to produce lactate and ethanol. Habitat analysis revealed that this bacterium is specifically detected in anaerobic digesters, particularly those processing lipid-rich waste. These findings highlight the pivotal role of this bacterium in anaerobic lipid degradation.

    Toxoplasma gondii is an intracellular parasitic protozoan with a high infection rate in mammals, including humans, and birds. There is no effective vaccine, and treatment relies on antiparasitic drugs. However, existing antiprotozoal drugs have strong side effects and other problems; therefore, new treatment approaches are needed. Metal nanoparticles have attracted increased interest in the biomedical community in recent years because of their extremely high surface area to volume ratio and their unique reactivity that could be exploited for medicinal purposes. Previously, we confirmed the anti-Toxoplasma effects of gold, silver, and platinum nanoparticles, in a growth inhibition test. Here, we asked whether the anti-Toxoplasma effect could be confirmed with less expensive metal nanoparticles, specifically iron oxide nanoparticles (goethite and hematite). To improve the selective action of the nanoparticles, we modified the surface with l-tryptophan as our previous findings showed that the bio-modification of nanoparticles enhances their selectivity against T. gondii. Fourier-Transform Infrared Spectroscopy (FTIR) analysis confirmed the successful coating of the iron oxide nanoparticles with l-tryptophan. Subsequently, cytotoxicity and growth inhibition assays were performed. L-tryptophan-modified nanoparticles showed superior anti-Toxoplasma action compared to their naked nanoparticle counterparts. L-tryptophan enhanced the selective toxicity of the iron oxide nanoparticles toward T. gondii. The bio-modified nanoparticles did not exhibit detectable host cell toxicity in the effective anti-Toxoplasma doses. To elucidate whether reactive oxygen species contribute to the anti-Toxoplasma action of the bio-modified nanoparticles, we added Trolox antioxidant to the assay medium and found that Trolox appreciably reduced the nanoparticle-induced growth inhibition.

    Toxoplasma, an important intracellular parasite of humans and animals, causes life-threatening toxoplasmosis in immunocompromised individuals. Although Toxoplasma secretory proteins during acute infection (tachyzoite, which divides rapidly and causes inflammation) have been extensively characterized, those involved in chronic infection (bradyzoite, which divides slowly and is surrounded by a cyst wall) remain uncertain. Regulation of the cyst wall is essential to the parasite life cycle, and polysaccharides, such as chitin, in the cyst wall are necessary to sustain latent infection. Toxoplasma secretory proteins during the bradyzoite stage may have important roles in regulating the cyst wall via polysaccharides. Here, we focused on characterizing the hypothetical T. gondii chitinase, chitinase-like protein 1 (TgCLP1). We found that the chitinase-like domain containing TgCLP1 is partially present in the bradyzoite microneme and confirmed, albeit partially, its previous identification in the tachyzoite microneme. Furthermore, although parasites lacking TgCLP1 could convert from tachyzoites to bradyzoites and make an intact cyst wall, they failed to convert from bradyzoites to tachyzoites, indicating that TgCLP1 is necessary for bradyzoite reactivation. Taken together, our findings deepen our understanding of the molecular basis of recrudescence and could contribute to the development of novel strategies for the control of toxoplasmosis.

    Plasmodium falciparum parasites are the primary cause of malaria across Africa. The problem of drug resistance to malaria is ever growing and novel therapeutic strategies need to be developed, particularly those targeting the parasite and also the host or host-pathogen interaction. Previous studies have shown that the development of cerebral malaria (CM) is related to dysregulation of the immune system in a murine malaria model of experimental cerebral malaria. It involves a complex interaction of events and interferon-gamma seems to be the unifying factor. Therefore, the antiplasmodial activity targeting the parasite and immunomodulatory strategies that reduce overall host inflammation, with IFN-γ in focus, could delay CM onset and prove beneficial in malaria infection therapy. Phyllanthus niruri is used to treat fever and other symptoms of malaria in Nigeria. Its modes of action as an anti-malarial remedy have not been exhaustively investigated. This study therefore examined the aqueous extract of P. niruri (PE) for its antiplasmodial activity in vitro using the Plasmodium falciparum HB3 strain. Furthermore, in vivo murine malaria model using the Plasmodium berghei ANKA strain was used to investigate its anti-malarial effects. We showed that PE has multiple anti-malarial effects, including anti-parasitic and host immunomodulatory activities. Co-culture of P. falciparum with PE and some of its phytoconstituents drastically reduced parasite number. PE also decreased parasitemia, and increased the survival of infected mice. We also observed that the integrity of the blood-brain barrier was maintained in the PE-treated mice. The results confirmed that PE showed moderate antiplasmodial activity. In vivo murine malaria model using P. berghei ANKA for experimental cerebral malaria revealed that PE suppressed parasite growth, and modulate the production of interferon-gamma. The findings demonstrate that PE affects malaria progression, targeting parasites and host cells.

    Cryptosporidium parvum is an apicomplexan parasite that causes severe zoonotic diarrhea in humans and calves. Since there are no effective treatments or vaccines for infants or immunocompromised patients, it is important to understand the molecular mechanisms of the parasite-host interaction for novel drug discovery. Mitogen-activated protein kinase (MAP kinase) is a key host factor in interactions between host and various pathogens, including parasites. Although the function of conventional MAP kinases against parasite infection has been investigated, that of atypical MAP kinases remains largely unknown. Therefore, we focused on one of the atypical MAP kinases, MAPK4, and its effect on C. parvum infection in human intestinal cells. Here, we report that MAPK4-deficient intestinal cells showed a significant reduction in C. parvum infection. We also show that host MAPK4 has a role in host cell survival from C. parvum infection. In addition, we show that C. parvum requires host MAPK4 for its successful invasion and asexual reproduction. Taken together, our data suggest that MAPK4 is an important host factor contributing to C. parvum infection in human intestinal cells.

    Cryptosporidium spp. are gastrointestinal opportunistic protozoan parasites that infect humans, domestic animals, and wild animals all over the world. Cryptosporidiosis is the second leading infectious diarrheal disease in infants less than 5 years old. Cryptosporidiosis is a common zoonotic disease associated with diarrhea in infants and immunocompromised individuals. Consequently, cryptosporidiosis is considered a serious economic, veterinary, and medical concern. The treatment options for cryptosporidiosis are limited. To address this problem, we screened a natural product library containing 87 compounds of Traditional Chinese Medicines for anti-Cryptosporidium compounds that could serve as novel drug leads and therapeutic targets against C. parvum. To examine the anti-Cryptosporidium activity and half-maximal inhibitory doses (EC50) of these compounds, we performed in vitro assays (Cryptosporidium growth inhibition assay and host cell viability assay) and in vivo experiments in mice. In these assays, the C. parvum HNJ-1 strain was used. Four of the 87 compounds (alisol-A, alisol-B, atropine sulfate, and bufotalin) showed strong anti-Cryptosporidium activity in vitro (EC50 values = 122.9±6.7, 79.58±13.8, 253.5±30.3, and 63.43±18.7 nM, respectively), and minimum host cell cytotoxicity (cell survival > 95%). Furthermore, atropine sulfate (200 mg/kg) and bufotalin (0.1 mg/kg) also showed in vivo inhibitory effects. Our findings demonstrate that atropine sulfate and bufotalin are effective against C. parvum infection both in vitro and in vivo. These compounds may, therefore, represent promising novel anti-Cryptosporidium drug leads for future medications against cryptosporidiosis.

    <italic>Toxoplasma gondii</italic> chronically infects the brain as latent cysts containing bradyzoites and causes various effects in the host. Recently, the molecular mechanisms of cyst formation in the mouse brain have been elucidated, but those in the human brain remain largely unknown. Here, we show that abnormal glutamine metabolism caused by both interferon-γ (IFN-γ) stimulation and <italic>T. gondii</italic> infection induce cyst formation in human neuroblastoma cells regardless of the anti-<italic>T. gondii</italic> host factor nitric oxide (NO) level or Indoleamine 2,3-dioxygenase-1 (IDO1) expression. IFN-γ stimulation promoted intracellular glutamine degradation in human neuronal cells. Additionally, <italic>T. gondii</italic> infection inhibited the mRNA expression of the host glutamine transporters SLC38A1 and SLC38A2. These dual effects led to glutamine starvation and triggered <italic>T. gondii</italic> stage conversion in human neuronal cells. Furthermore, these mechanisms are conserved in human iPSC-derived glutamatergic neurons. Taken together, our data suggest that glutamine starvation in host cells is an important trigger of <italic>T. gondii</italic> stage conversion in human neurons.

    Treatment with rumen fluid improves methane production from non-degradable lignocellulosic biomass during subsequent methane fermentation; however, the kinetics of xylanases during treatment with rumen fluid remain unclear. This study aimed to identify key xylanases contributing to xylan degradation and their individual activities during xylan treatment with bovine rumen microorganisms. Xylan was treated with bovine rumen fluid at 37°C for 48 h under anaerobic conditions. Total solids were degraded into volatile fatty acids and gases during the first 24 h. Zymography showed that xylanases of 24, 34, 85, 180, and 200 kDa were highly active during the first 24 h. Therefore, these xylanases are considered to be crucial for xylan degradation during treatment with rumen fluid. Metagenomic analysis revealed that the rumen microbial community's structure and metabolic function temporally shifted during xylan biodegradation. Although statistical analyses did not reveal significantly positive correlations between xylanase activities and known xylanolytic bacterial genera, they positively correlated with protozoal (e.g., Entodinium, Diploplastron, and Eudiplodinium) and fungal (e.g., Neocallimastix, Orpinomyces, and Olpidium) genera and unclassified bacteria. Our findings suggest that rumen protozoa, fungi, and unclassified bacteria are associated with key xylanase activities, accelerating xylan biodegradation into volatile fatty acids and gases, during treatment of lignocellulosic biomass with rumen fluid.

    Scum is formed by the adsorption of long-chain fatty acids (LCFAs) onto biomass surface in anaerobic digestion of oily substrates. Since scum is a recalcitrant substrate to be digested, it is disposed via landfilling or incineration, which results in biomass washout and a decrease in methane yield. The microbes contributing to scum degradation are unclear. This study aimed to investigate the cardinal microorganisms in anaerobic scum digestion. We pre-incubated a sludge with scum to enrich scum-degrading microbes. Using this sludge, a 1.3-times higher methane conversion rate (73%) and a faster LCFA degradation compared with control sludge were attained. Then, we analyzed the cardinal scum-degrading microbes in this pre-incubated sludge by changing the initial scum-loading rates. Increased 16S rRNA copy numbers for the syntrophic fatty-acid degrader <italic>Syntrophomonas</italic> and hydrogenotrophic methanogens were observed in scum high-loaded samples. 16S rRNA amplicon sequencing indicated that <italic>Syntrophomonas</italic> was the most abundant genus in all the samples. The amino-acid degrader <italic>Aminobacterium</italic> and hydrolytic genera such as <italic>Defluviitoga</italic> and <italic>Sporanaerobacter</italic> became more dominant as the scum-loading rate increased. Moreover, phylogenic analysis on <italic>Syntrophomonas</italic> revealed that <italic>Syntrophomonas palmitatica</italic>, which is capable of degrading LCFAs, related species became more dominant as the scum-loading rate increased. These results indicate that a variety of microorganisms that degrade LCFAs, proteins, and sugars are involved in effective scum degradation.

    Treatment with rumen microorganisms improves the methane fermentation of undegradable lignocellulosic biomass; however, the role of endoglucanase in lignocellulose digestion remains unclear. This study was conducted to investigate endoglucanases contributing to cellulose degradation during treatment with rumen microorganisms, using carboxymethyl cellulose (CMC) as a substrate. The rate of CMC degradation increased for the first 24 h of treatment. Zymogram analysis revealed that endoglucanases of 52 and 53 kDa exhibited high enzyme activity for the first 12 h, whereas endoglucanases of 42, 50, and 101 kDa exhibited high enzyme activities from 12 to 24 h. This indicates that the activities of these five endoglucanases shifted and contributed to efficient CMC degradation. Metagenomic analysis revealed that the relative abundances of <italic>Selenomonas</italic>, <italic>Eudiplodinium</italic>, and <italic>Metadinium</italic> decreased after 12 h, which was positively correlated with the 52- and 53-kDa endoglucanases. Additionally, the relative abundances of <italic>Porphyromonas</italic>, <italic>Didinium</italic>, unclassified Bacteroidetes, <italic>Clostridiales</italic> family XI, <italic>Lachnospiraceae</italic> and <italic>Sphingobacteriaceae</italic> increased for the first 24 h, which was positively correlated with endoglucanases of 42, 50, and 101 kDa. This study suggests that uncharacterized and non-dominant microorganisms produce and/or contribute to activity of 40, 50, 52, 53, and 101 kDa endoglucanases, enhancing CMC degradation during treatment with rumen microorganisms.

    We had developed a new pretreatment system using cow rumen fluid to improve the methane production from lignocellulosic substrates. However, the pretreatment conditions differ from the in-situ rumen environment, therefore different microbes may be involved in plant cell wall decomposition. In the current study, shotgun metagenomic analysis using MiSeq platform was performed to elucidate the bacteria which produce cellulase and hemicellulase in this pretreatment system. The rumen fluid which contained waste paper pieces (0.1% w/v) were incubated at 37°C during 120 h. The fluid samples were collected from the reactor at each time-point and analyzed for chemical properties. Rumen microbial DNA was extracted from 0-h and 60-h samples and subjected to shotgun-metagenomic analysis. After pretreatment, approximately half of cellulose and hemicellulose contents of the waste paper were decomposed and some volatile fatty acids were accumulated. Clostridia (e.g., Ruminococcus and Clostridium) were the predominant bacteria before and after 60-h pretreatment, and their relative abundance was increased during pretreatment. However, Prevotella and Fibrobacter, one of the most dominant bacteria in-situ rumen fluid, were observed less than 3% before incubation and they were decreased after pretreatment. Genes encoding cellulase and hemicellulase were mainly found in Ruminococcus, Clostridium, and Caldicellulosiruptor. Calicellulosiruptor, which had not been previously identified as the predominant genus in lignocellulose decomposition in in-situ rumen conditions, might be considered as the main fibrolytic bacterium in this system. Thus, this study demonstrated that the composition of fibrolytic bacteria in this system was greatly different from those in the in-situ rumen.

    Sulfur-oxidizing bacterial diversity at the surface of cattle manure was characterized throughout the composting process using a sulfur oxidation gene (soxB) clone library approach. In the mesophilic phase, clones related to the genera Hydrogenophaga and Hydrogenophilus were characteristically detected. In the thermophilic phase, clones related to the genera Hydrogenophaga and Thiohalobacter were predominant. In the cooling phase, the predominant soxB sequences were related to the genus Pseudaminobacter and a new sulfur-oxidizing bacterium belonging to the class Alphaproteobacteria. The present study showed changes in the community composition of sulfur-oxidizing bacteria at the surface of compost throughout the composting process.

    Cryptosporidium spp. are enteric protozoan parasites that infect a wide range of hosts including humans, and domestic and wild animals. The aim of this study was to molecularly characterize the Cryptosporidium spp. found in calf faeces in Japan. A total of 80 pre-weaned beef and dairy calves' diarrhoeic faecal specimens were collected from nine different prefectures in Japan. A nested polymerase chain reaction targeting the small subunit 18S rRNA and GP60 genes were used to detect the Cryptosporidium genotypes and subtypes. 83.8% (67 out of 80) of the specimens were positive for Cryptosporidium spp.; Cryptosporidium was found in both beef and dairy calves. Cryptosporidium parvum was the predominant species, detected in 77.5% (31/40) of beef calves and 80% (32/40) of dairy calves. Cryptosporidium bovis was also detected, 5.0% (2/40) of dairy calves, and C. ryanae was also found 2.5% (1/40) of dairy calves. One mixed-species infection, 2.5% (1/40) was detected in a beef calf having C. parvum, and C. ryanae. We detected the most common subtype of C. parvum (i.e., IIaA15G2R1), as well as other subtypes (i.e., IIaA14G3R1, IIaA14G2R1, and IIaA13G1R1) that have not previously been detected in calves in Japan. Our results demonstrate the widespread diversity of Cryptosporidium infection in calves in Japan.

    Various support carriers are used for high-density retention of methanogenic archaea in anaerobic wastewater treatment systems. Although the physicochemical properties of carrier materials and microorganisms influence the adhesion of methanogenic archaea, details about the underlying mechanism remain poorly characterized. We applied seven types of chemical surface modifications to carbon felts to clarify the adhesion properties of <italic>Methanothermobacter thermautotrophicus</italic>, a representative thermophilic hydrogenotrophic methanogen. The relationship between carrier surface properties and methanogen adhesion was evaluated. <italic>M. thermautotrophicus</italic> adhesion was significantly increased up to 2.6 times in comparison with control on carbon felts treated with NaOH, HCl, H2SO4, or Na2HPO4. Treated carbon felts showed a lower water contact angle, but no correlation between the carrier surface contact angle and methanogen adhesion was observed. On the other hand, at the surface of the carrier that showed improved adhesion of methanogens, the ratio of -COOH : -OH was 1 : 0.65. Such a ratio was not observed with treated carriers for which methanogen adhesion was not improved. Therefore, in the adhesion of <italic>M. thermautotrophicus</italic>, the functional group abundance was important as well as physical surface properties such as the hydrophobicity. Hydrogenotrophic methanogens are involved in active methanation during the startup of anaerobic digestion. Additionally, these methanogenic archaea function as methanogenic cathode catalysts. Therefore, anaerobic digestion performance will greatly improve by controlling the adhesion of hydrogenotrophic methanogens such as <italic>M. thermautotrophicus</italic>.

    The genus Cryptosporidium, which is an obligate intracellular parasite, infects various vertebrates and causes a diarrheal disease known as cryptosporidiosis. Wild rodents are naturally infected with zoonotic Cryptosporidium; thus, they are potential reservoirs of the parasites. Mice are common rodents frequently found in agricultural areas and have many opportunities to contact other wild animals, livestock, and humans. Irrespective of the potential risk, there are few epidemiologic studies of Cryptosporidium in wild mice because of their low economic importance and the difficulty in conducting surveys. Hence, the species and genotypes of Cryptosporidium in wild mice living around various areas remain unclear. We investigated the species and genotype distribution and prevalence of Cryptosporidium in the large Japanese field mouse (Apodemus speciosus) in an agricultural site in Osaki, Miyagi Prefecture, Japan. In total, 15 mice were captured and examined in this study. By microscopic analysis, only one mouse (JFM 3) was determined to be Cryptosporidium-positive, while the parasite were detected in four mice (JFM 3, 6, 10, and 15) by a molecular approach using partial SSU rRNA gene sequences. Based on nucleotide sequence and phylogenetic analysis, the Cryptosporidium isolates were identified as C. ubiquitum (from JFM 10) and C. muris (from JFM 3 and 6). In contrast, the Cryptosporidium in JFM 15 was not identified as a known species or genotype and is therefore proposed as a novel genotype; the Naruko genotype. More molecular data are necessary to elucidate the taxonomic identity of this novel Cryptosporidium genotype. The C. muris Japanese field mouse genotypes showed marked divergence compared to that in a previous report. The large Japanese field mouse might thus represent a reservoir of multiple Cryptosporidium spp.

    ABSTRACT Plasmodium falciparumparasites are the major cause of malaria across Africa. Due to the appearance of multi-drug resistant parasites, new antimalarial drugs are needed. The medicinal plantPhyllanthus niruriis being used to treat fever and other symptoms of malaria in Nigeria; however, little is known about its antimalarial mechanisms. Here, we show that aqueous extract ofP. niruri(PE) has multiple antimalarial effects, including anti-parasitic and host immunomodulatory activity. We found that co-culture ofP. falciparumwith PE drastically reduced parasite number, but PE did not inhibit parasite development or rupture; rather, it blocked erythrocytes invasion. In addition, we identified Astragalin as one of the antimalarial compounds which are contained in PE. Moreover, we found that PE suppresses the inflammatory activity and apoptosis of immune cells (T cells) and astrocytes and neurons in the central nervous system (CNS). Furthermore, we confirmed that oral administration of PE to mice suppressed parasite growth, excessive inflammation, CNS dysfunction, and the development of experimental cerebral malaria in anin vivomurine malaria model. Our findings demonstrate that PE has multiple effects on malaria progression, targeting both parasite and host cells.

    <p>Because paper sludge discharged from the waste paper recycling process contains high levels of lignin and ash, it is not converted into biogas effectively during methane fermentation. The aim of this study was to improve the efficiency of biogas production from paper sludge by the pretreatment with rumen fluid. We performed the pretreatment of paper sludge with rumen fluid at 37°C for 6 hours. After pretreatment, methane fermentation was conducted at 35°C for 45 days. Pretreatment with rumen fluid hydrolyzed paper sludge and increased the VFAs concentration during the pretreatment process. Methane production from pretreated paper sludge was 12.1 % higher than that from untreated paper sludge. These results revealed that pretreatment with rumen fluid improved the efficiency of methane production from paper sludge. Next, we investigated the effects of protozoa on methane fermentation of paper sludge. Protozoa was removed from rumen fluid with sodium dodecyl sulfate, and the pretreatment and methane fermentation of paper sludge was conducted. We observed the positive relationship between the number of protozoa and methane gas production. The results indicated that protozoa possessed positive effects on methane production from paper sludge.</p>

    After the treatment of crop residues or waste paper by cattle rumen fluid, methane fermentation of these materials was accelerated. Enzymes and microbial communities in the process was investigated. Fiber‐degrading cellulase was diverse in the process; however, specific enzymes of xylanase showed high enzymatic activity continuously. Cellulase activity in the rumen treatment tank was continued for more than one month. At least five types of cellulases having different molecular sizes were existed. Microbes producing or involving these cellulases were unclassified species and not dominant in the microbial community.

    In the ciliate Tetrahymena, the gamete-nuclei formation involves chromatin remodeling induced by a novel DNA double-strand break at the post-meiosis stage. Chromatin remodeling in the pronucleus during the gamete-nuclei formation is an evolutionarily conserved phenomenon. This occurs not only in ciliate but also in animals such as humans. However, the molecular mechanism of this phenomenon has been obscure. This study focused on the chromatin remodelers involved in the euchromatin formation for the gamete-nuclear formation and the changes in the epigenomic state associated with the gamete-nuclear formation. We identified some proteins involved in euchromatin formation and unclosed some parts in the system that controls it.

    In this study, molecular mechanisms and biological roles of a novel formation of post-meiotic DNA double-strand breaks (DSB) in the haploid micronuclei (pronuclei) of Tetrahymena thermophila were analyzed. After completion of meiosis, the TopII, and Spo11 mediated DSB formation in all the four pronuclei. Soon thereafter, DNA repair including histone replacement took place in the pronucleus that was adjacent to the mating-junction. This nucleus then began an additional mitosis as the selected pronucleus to produce gametic nuclei, while the other three unselected nuclei with persistent DSB eventually disappeared from the cytoplasm. Defective DSB formation in mutant strains abolishes not only nuclear selection but also histone replacement. These results suggest that the post-meiotic DSB is likely an intrinsic developmental program of the pronucleus and potentially contributes to chromatin remodeling to produce gametic nuclei capable of forming a zygotic nucleus.

    Up to the present time, protozoan fauna in Japan has not been fully documented. Therefore, we, more than 20 protozoan specialists, have conducted a systematic survey on the Japanese protozoans by 1) collecting original bibliographic records 2) listing up species names and collection points, and 3) extracting reliable list of literature, in order to construct a comprehensive inventory database of Protozoan fauna of Japan. We have also conducted a nation -wide field survey of free-living and parasitic protozoans at more than 100 locations in Japan, including Abashiri area in Hokkaido and Minami Daito Island in Okinawa prefecture. The result of the survey has been presented on line at http://sites.google.com/site/protistsurvey/.

    本研究では、畑や水田、森林、草原、河川など様々な環境がある広大な東北大学農学研究科附属複合生態フィールド教育研究センター(以下FSC)に、主な複数のスタディサイトを儲ける。これにより、ほぼ同一の気候条件の異なる環境で生息する原生生物の生態構造をとらえられ、本研究の主要な目的である土壌周辺を含む生態系での原生生物の流れを詳しく解析できる。また、原生生物の探索は、環境DNAのライブラリによる分子生物学的手法を基軸に行い、その類縁解析は、最新で再精密の分子系統解析を用いる。また、原生生物の生理活性は、mRNAによる環境RNA解析を用いる。ここでは、DNA複製酵素など分裂と関わる分子の遺伝子、あるいはHSPなどの恒常的発現遺伝子などの遺伝子量と、またそれらの系統解析により、生態系で活発な原生生物種を同定できる。

  2. 排熱活用小型メタン発酵による分散型エネルギー生産と地域循環システムの構築

    水産地域で排出される有機性廃棄物は水分を多く含むため、焼却に多大なエネルギーを要する。このような廃棄物の有効処理法としてメタン発酵がある。メタン発酵は嫌気性微生物の発酵によるメタンガス生産であるが、微生物活性を維持するために、反応槽を加温する必要がある。 これまでメタン発酵は、投入エネルギーと生産エネルギーの収支をプラスにするため、大規模システムが当然とされた。しかし、初期投資が数十億円と莫大であり、原料となる有機性廃棄物の調達コストが高い、原料不足が問題となっている。さらに、維持費も大きく、建設しても持続的維持が不可能になる課題があった。東北地方の沿岸域はリアス式海岸で、それぞれの港を含む水産地域は、小さな集落でまとまっているため、従来の大規模システムの導入は不適である。 本研究では、これまでメタン発酵で課題となっていた加温の投入エネルギーを水産加工業からの(煮蛸やしらすをゆでる時、乾燥,冷蔵冷凍時に無駄にされる)排熱を活用し、メタン発酵装置を温め、これまで不可能とされていたメタン発酵槽の小型化・省エネルギー化を可能にし、より安価で簡便にメタン発酵装置を小規模地域に導入し、地域内で水産地域由来の廃棄物系バイオマス処理をかねた分散型エネルギー生産を可能にすることを目的としている。 加えて、地域からの廃棄物を見える形で循環化し、町からの海への環境負荷を省コストに減らし、災害に強く、海を守りながら、低炭素型の持続可能な生活を可能にする町づくりを目的としている。 これにより、水産地域では、これまで廃棄物処理に係った処理コストを削減し、自立的エネルギー生産を可能になるだけでなく、いざというときの緊急電源および廃棄物処理システムを常備した町づくりにするものである。

    生活や生産で排出された有機性廃棄物の新たな処理方法として、微生物を利用した省エネルギーの処理が確立しつつあり、また生成品であるコンポストや消化液には肥料としての価値が期待されている。これら生成品について多くの研究が行われ、肥料として一定の評価が与えられた。だが、これら生成資材は莫大な微生物を含有し、投入による土壌生態系の撹乱は不可避と想定される。しかし生態系に対する影響、特に土壌の原生生物へ注目した研究はない。 本研究は、コンポストや消化液を肥料成分とした視点でなく莫大な微生物の集合体と見なし、使用に伴う土壌生態系の変化を、特に原生生物を中心に観る。解析は、生息する原生生物を直接観察する手法、土壌のDNAやRNA抽出による網羅的解析、また構築モデル系によるメタボローム解析など、新旧の生物学的手法を用いて行い、微生物資材の与えるミクロな生態系への影響を評価する。
