    ABSTRACT Lignans are bioactive polyphenols in sesame oil that contribute to its antioxidant and metabolic health benefits. Their composition changes drastically during roasting and refining, but conventional methods such as HPLC–UV are time‐consuming and require organic solvents. Here, we established a rapid and non‐destructive near‐infrared (NIR) spectroscopic method coupled with partial least squares regression (PLSR) to simultaneously quantify six lignan species—sesamin, sesamolin, episesamin, diasesamin, sesaminol, and sesamol—in sesame oil. A total of 100 samples, including both roasted and refined oils, were analyzed. Reference data were obtained by HPLC–UV, and PLSR models were constructed using second‐derivative NIR spectra. The models achieved excellent predictive accuracy ( R 2  = 0.970–0.996; RMSE = 0.0008%–0.020%), even for trace‐level lignans such as sesamol (0.005%–0.03%) and diasesamin (&lt; 0.04%). The approach successfully distinguished structurally similar lignans and revealed spectral regions (around 7400 cm −1 ) linked to processing‐induced compositional changes. The proposed method provides a powerful, solvent‐free platform for rapid lignan profiling, allowing analysis within minutes without sample pretreatment, and offers strong potential for industrial quality control and process monitoring.

    Monitoring oxidation causes (i.e. radical and/or photo-oxidation) of vegetable oil gives very important information for its quality control. This study aimed to develop a rapid and simple near infrared (NIR) spectroscopy method to explore the oxidation causes in vegetable oil by quantifying triacylglycerol hydroperoxide (TGOOH) positional isomers proceeded by different oxidation causes. First, the concentrations of TGOOH isomers were determined by liquid chromatography-mass spectrometry. Partial least square regression for concentrations of TGOOH isomers (R2: 0.994-0.998) using NIR spectra in the regions of 7500-6000 cm-1 and 5500-4500 cm-1 as explanatory variables. Principal component analysis revealed that those models were created because NIR spectra capture trace changes of the peak around 6980 cm-1(-CH2-) and 5260 cm-1 (H2O) derived by different oxidation causes (i.e. radical and/or photo-oxidation). The application of this method to various kinds of vegetable oil is expected to lead to a simple and rapid quality evaluation of vegetable oil.

    Freezing hydrogels can compromise their network structures and modify their properties as a result of ice crystal formation. Therefore, understanding the internal structure, including ice crystals and the state of chemical components within hydrogels, is essential. In this study, we evaluated the elemental distribution in bio-based hydrogels subjected to freezing-thaw process using X-ray absorption spectroscopy with synchrotron radiation. A bio-based hydrogel, prepared from Alaska pollock, underwent both slow and rapid freezing processes. Tomographic images and linear X-ray absorption coefficient distributions of the rapidly frozen hydrogel displayed a uniform image with a mean absorption coefficient of 2.81 cm-1. Conversely, the slowly frozen sample exhibited distinct contrasts with peaks at 2.516 cm-1 (dark) and 3.691 cm-1 (bright), occupying 28% and 72% of the image, respectively. The mean absorption coefficient of the slowly frozen sample was comparable to that of the rapidly frozen sample, indicating no elemental loss. The elements within the hydrogel were categorized into organic elements, macrominerals, and trace elements. The bright areas in the images were attributed to the concentration of macrominerals. Notably, Cl and Na were the primary contributors to the absorption coefficients among the elements present, signifying salt migration during freezing. These findings suggest that the contrast observed in X-ray computed tomography images after freezing reflects the elemental distribution within the hydrogel and successfully demonstrates element localization due to cryoconcentration.

    In this study, the bubbles in rice flour dough were investigated under a constant temperature. The bubble size distribution is important for the control of food texture. If bubble sizes depend mainly on the inertial force, viscous force, and surface tension, then the normalized mean bubble diameter should be a function of the Reynolds number and Weber number. We obtained experimental data using a hand mixer, and compared the properties of doughs prepared using six rice flours; each flour was prepared through a different milling process. We also added the size effect of the rice flour particles as the Bond number. Furthermore, we performed a dynamic wettability test to estimate the wettability of the rice flour surface. The results of this test were described well by the Washburn equation, and <italic>dc</italic>cos<italic>θ</italic> was calculated as a wettability parameter (where, <italic>dc</italic>: effective diameter of a capillary in a powder bed, cos<italic>θ</italic>: the contact angle). The mean bubble diameter (<italic>dbm</italic>) generated by whipping was expected to be affected by the thickness (<italic>d</italic>) of the rod of the mixer, its movement speed, physical properties of the material, and gravitational acceleration. Therefore, dimensionless mean diameter (<italic>dbm/d</italic>) was expressed by a dimensionless equation. The empirical equation obtained was generally applicable to the variety of materials selected for this study.

    <p>The aim of this study was to clarify the change in the powder properties of rice flour depending on the milling process. Rice flour samples, which have gradual mechanical shock properties, were prepared using different milling methods. Furthermore, the correlation between the starch damage, owing to mechanical shock, and powder properties of rice flour was investigated. The particle size was changed gradually through each milling process; however, the change did not clearly correlate with starch damage. The results of the X-ray diffraction (XRD) pattern of nongelatinized samples showed the typical A-type structure of starch. The crystal structure of starch in rice flour may change to a disorder state with the progress of milling; thus, in this study, instead of crystallinity, we considered the disorder index (DI) calculated from the XRD intensity of samples. Relationship between DI and starch damage was confirmed with R2 = 0.923. Therefore, the mechanical shock caused by the milling process contributes to the crystal state of starch. The parameter qm calculated from the Guggenheim-Anderson-de Boer (GAB) equation of each sample corresponded to the DI. This result suggests that the sorption site of rice flour decreased, and a positive correlation was observed between the parameter K and DI. Thus, the interaction between the rice flour and water molecules weakened because of the mechanical shock. In addition, the use of a SEM image supports the insight that the change in parameter K may reflect the structural change in the solid phase. These results demonstrated that the change in powder properties of rice flour caused by mechanical shock of the milling could evaluate quantitatively.</p>

    <p>Monitoring of water activity at meat surface is important on controlling the quality of aging beef, because the fermentation microorganisms are grow on the meat surface. Although in previous study water activity of beef under drying process was investigated by near infrared (NIR) spectra, spectra data have been obtained from the whole beef cut. Therefore, this study aimed to estimate water activity of the surface of the sample using NIR spectra in the 700-1050 nm. Different thick samples in the 2-10 mm were prepared and the optimization of the measurement conditions was carried out for surface monitoring. Consequently, the optimal measurement condition were determined for monitoring the spectra from the surface of a beef cut. The change in moisture content and water activity about lump sample of beef were investigated for drying at 4℃. A linear decrease in water activity estimated by NIR spectra was observed, and the estimated water activity reached 0.8 after around 22~25 days. These results showed the evaluation potential of dry state at surface of beef by using NIR spectroscopy.</p>

    <p>A cultivation behavior in high density cultivation of mung bean sprouts was investigated from the engineering viewpoint. The effect oxygen concentration on the growth rate was evaluated, because heterotrophic growth and secondary metabolism reactions were related to oxygen. The cart with controllable circuit for gas phase was used for high density cultivation. In a growing test of mung bean in high density and individual cultivations under various oxygen concentrations, courses of fresh weight, and whiteness index of seedling of mung bean sprouts were measured, and specific growth rate based on fresh weight were investigated. In high density cultivation, specific growth rate was faster than in individual cultivation, and differences in growth rate according to growth stage were observed with oxygen concentration. On low oxygen concentration whiteness index of the seedling raised. Therefore, it is assumed that oxygen concentration was so low that secondary metabolism reaction of brown substances in root of seedling was limited. These results suggest that growth and secondary metabolism reactions in high density cultivation could be regulated by controlling oxygen concentration in gas phase. In this study, we proposed that the concept of Controlled Atmosphere (CA) might be applied in plant cultivation as well as CA storage.</p>

    植生の水ストレスは、可視・近赤外(400-1100nm)領域の反射スペクトルを用いているが、この領域を利用した評価は、クロロフィルバンドの変化を介した間接的なものであった。近赤外・赤外領域のスペクトルは、水を直接検出するバンドが存在し、非破壊計測分野では指標として考案されているものがあるが、そのスペクトル挙動自体を詳細に検討した事例は多くないうえ、過剰ストレスに対応した水ストレス指標はまだ開発されていない。そこで、本研究では、過剰水分にも対応する分光的水分評価指標を開発することを目的として、近赤外分光と赤外分光を併用した解析を実施している。本研究の独創的な点は植物葉の主要な成分であるセルロースをキー物質として分光学的手法による検出を目指している点である。2023年は、基礎的な評価として、デンプン系素材の水分活性を変えることで、スペクトル挙動の解析を行い、特に900-100カイザー付近の遠赤外領域を用いた検討を行った、その結果、水分量が増加するにつれて水に由来すると考えられるベースラインの変動が認められた。デンプン系素材に対して、この領域のPLS回帰モデルを構築した結果、水分活性と有意な相関が認められ、水分量そのものに関しては、高水分域で検量線が飽和する可能性が認められた。この結果は、水分量の増加による検量線のさらなる詳細なスペクトル挙動との検討が必要であると考えられるが、遠赤外領域での水分評価の可能性を言及することに成功した。2024年度は、セルロースと水分の関連性を遠赤外領域を中心として、検証し水分過剰/欠乏ストレス構築の可能性について明確化していく予定である。
