    Objectives This study aimed to develop a model for the cross-sectional classification of psychological distress among older adults, using a Bayesian Neural Network (BNN) to examine the relationships among humor expressions, communication skills, physical activity (PA), and social PA experiences (with friends), and their effects on psychological distress risk. Methods A cross-sectional survey was conducted among 5,265 Japanese adults aged 65 to 89 years. The predictor variables included humor expressions, communication skills, and social PA experiences. Depression was assessed using the K6 Psychological Distress Scale (cutoff ≥ 9). A BNN with three hidden layers was constructed with SHapley Additive exPlanations (SHAP) for feature importance identification. Results The BNN model achieved 81.1% accuracy for high-risk detection. This study revealed self-enhancing humor coping and self-control communication skills as the strongest protective factors. Additionally, past PA experiences with friends and present PA experiences alone reduced the risk for K6-assessed psychological distress. Conclusion The BNN model identified positive humor styles, self-controlled communication abilities, past social PA, and present PA status as important predictors of contributors to the model output for classifying K6-assessed psychological distress among older adults, highlighting the potential relevance of social engagement in communication and PA.

    Background Although exercise can help prevent age-related cognitive decline, the mechanisms underlying its effects on cognition, immune function remain unclear. Therefore, this study comprehensively examined the effects of circuit training on cognitive function, and the plasma proteome in middle-aged to older women. Methods Forty-eight women were assigned to either a four-month circuit training program ( n  = 23, exercising more than twice per week) or control group ( n  = 25). Cognitive function was assessed using Stroop, Flanker, N-back, and source memory tasks. Statistical analyses were performed using linear mixed-effects models with Type III analysis of variance. The plasma proteome was measured using Olink technology (1,098 proteins). Proteomic patterns were explored using orthogonal partial least squares discriminant analysis (OPLS-DA). Furthermore, between-group comparisons of protein changes (post-baseline differences) were performed using unpaired t -tests. For functional annotation, significantly altered proteins (identified by volcano plot analysis) were subjected to pathway enrichment analyses. Results Within-group changes in Stroop interference and d-prime were observed in the circuit training intervention group. Furthermore, the proteins significantly elevated in the exercise intervention group were functionally categorized into anti-inflammatory and immunosuppressive pathways (LRRC25, IL10RA), angiogenesis and tissue remodeling (PROK1, TNFRSF12A), and immune cell activation (CD7). However, no protein–cognition associations remained significant after FDR correction. Conclusions This open-label randomized controlled trial provides preliminary evidence that a 4-month circuit training intervention may improve selected cognitive outcomes and alter circulating proteins related to immune, inflammatory, and carbohydrate-binding functions in middle-aged and older women. However, protein–cognition associations did not remain significant after FDR correction. These findings should be interpreted as hypothesis-generating and require confirmation in larger studies with integrated cognitive and molecular analyses.

    Abstract DunedinPACE quantifies the pace of biological aging. No study has examined its association with cardiorespiratory fitness (CRF). Additionally, the physiologically relevant CRF thresholds associated with slow aging remain unclear. The purpose of this study was to investigate the association between CRF and the pace of epigenetic aging, as measured by DunedinPACE, and to identify a CRF threshold indicative of slower biological aging. Here, we analyzed data of 144 older men (aged 65–72 years) enrolled in the WASEDA’S Health Study to examine the association of CRF (VO₂/kg at peak and ventilatory threshold (VT)), physical fitness, anthropometric parameters, and nutritional intake with epigenetic aging. Epigenetic aging was assessed using DunedinPACE derived from blood DNA methylation profiles. We performed Pearson’s and partial correlation analyses adjusted for age, smoking status, and drinking status, followed by receiver operating characteristic (ROC) curve analyses with bootstrapped optimal cutoff determination. In the unadjusted analyses, VO₂/kg at peak ( r  =  − 0.17, p  = 0.041) and VT ( r  =  − 0.17, p  = 0.046) were inversely associated with DunedinPACE, and VO₂/kg at peak showed a significant association even in the adjusted models ( r  =  − 0.16, p  = 0.046). The ROC curve analysis revealed a potential threshold of VO₂/kg at peak (26.2 mL/kg/min) for differentiating individuals with slower biological aging, supported by bootstrap distributions of optimal cutoff points and Youden’s index. This study suggests that higher CRF is associated with a slower pace of epigenetic aging, as measured using DunedinPACE. The identified VO₂peak threshold may provide a biomarker-based fitness target to support healthy aging in older adults.

    Introduction This study aimed to examine the effects of a digital visual art learning intervention on the cognitive and psychological functions of healthy older adults with no prior experience in art. Methods An open-label pilot randomized controlled trial was conducted, with participants (mean age = 69.43 ± 2.70 years) aged 65 to 74 assigned to either an intervention (n = 37) or control (n = 35) group. Cognitive and psychological functions were assessed before and after 20 sessions of digital visual art learning. In each session, participants mainly worked on lectures and prepared tasks related to basic themes in art. In the final stage, each participant engaged in individual creative activities and aimed to complete their works. Results In the Frontal Assessment Battery, which evaluates frontal lobe function, a significant trend was observed in the interaction between group and time (p = 0.062), and although an improvement trend was observed in the intervention group, the change did not reach statistical significance. Conclusion These findings suggest that digital art could potentially enhance executive function in older adults, providing important insights into its applicability within this population.     Abstract <p>Background Japan has one of the foremost aging populations in the world, and from a dementia prevention perspective, effective and easy-to-implement approaches to dementia prevention are becoming increasingly important. Artistic activities have been suggested to improve cognitive function and psychological well-being among older adults. Although visual art therapy has been shown to improve cognitive function in older adults with cognitive impairment, studies targeting healthy older individuals are limited. Furthermore, considering the spread of digital visual art, incorporating digital devices into art engagement for older adults may improve cognitive function. This study protocol aims to clarify the effects of digital visual art learning interventions on the cognitive and psychological functions of healthy older individuals, as well as the relationships between drawing movement data and related variables.Methods Eighty older adults will be randomly assigned to the digital visual art group or the control group (1:1). The digital visual art group will take part in a participatory digital visual art learning program, with a total of 20 sessions, while the control group will continue their usual daily lives. All participants will undergo cognitive and psychological function tests before and after the intervention. The effects of the program on cognitive and psychological functions will be determined by analysis of variance in the results of cognitive and psychological function tests conducted before and after the intervention. To further clarify the mechanism of intervention effects in the digital visual art group, the relationship between the test results before and after the intervention and related indicators, such as drawing motion, will be clarified using the structural equation modeling.Discussion The study aims to clarify the effects of continuous digital visual art learning on the cognitive and psychological functions of healthy older individuals. The characteristics of this study are threefold: emphasis on preventive approaches, utilization of digital technology in art interventions for older adults, and utilization of drawing motion data obtained from digital pens in the analysis. The digital visual art learning developed through this study is expected to enrich the lives of older adults. This study was registered with the UMIN-CTR (UMIN000051427) on June 23, 2023.</p>

    Introduction With the rapid aging of the population worldwide and the prevalence of dementia and mental health problems among older adults, it is important to extend healthy life expectancy by maintaining brain and mental health. Playing musical instruments, which requires the integration of auditory, visual, and somatosensory functions, is considered an effective way to prevent the development of dementia. However, the effectiveness of group (band) music sessions in healthy older adults has not been investigated. Our purpose, therefore, was to investigate the effects of group music sessions on cognitive and psychological functions among healthy older adults. Methods In this open-label randomized controlled trial, participants aged 65–74, who had no musical experience, were randomly assigned to either the intervention or control group. The intervention group received in weekly 90-minute sessions with the instrument for 16 weeks. The control group received no intervention. Results The results showed that the Mini-Mental State Examination (MMSE) total score and the Wechsler Memory Scale Logical Memory Ⅱ (WMS-LM Ⅱ) score improved significantly, and the Vigor–Activity subscale score of the Profile of Mood States 2nd Edition (POMS 2) tended to improve. Discussion These findings indicated that group music sessions have a potentially beneficial effect for maintaining and improving cognitive and psychological functions in healthy older adults.

    Performance monitoring has been conceptualized as the process that initiates remedial actions when we commit errors. In this project, we confirmed the importance of performance monitoring in motor learning and in the control of learned motor behavior by reviewing previous studies. We further tested the validity of this assertion by recording event-related potentials (ERPs) associated with performance monitoring. We focused on previously-reported phenomena in the motor learning area, including the contextual interference effect, beneficial effects of infrequent feedback, implicit learning, motor programming, and timing function. A series of experiments revealed that ERPs recorded during motor learning were modulated by performance monitoring.
