    Abstract The aim of this study was to investigate the bactericidal effect and recovery of biocompatibility of contaminated titanium surfaces using a combination treatment involving silver, copper, or iron ion application along with 400 nm purple-LED light irradiation. Additionally, the study sought to develop a functional calcium phosphate (CaP) coating treatment on titanium surfaces following disinfection, to promote re-osseointegration. A purple-LED emitting light at 400 nm was utilized to irradiate Staphylococcus aureus suspensions and biofilms in the presence of various concentrations of silver, copper, and iron solutions for 1 min. The bactericidal effect and electron spin resonance (ESR) spectrum were subsequently evaluated. Additionally, the hydrophilicity of the titanium surface and cell viability of MC3T3-E1 cells after combination treatment with silver ion was evaluated. Furthermore, a titanium surface coating with CaP gene transfection carrier containing plasmid DNA was developed using an electric current. The activity of hard tissue formation was then evaluated both in vitro and in vivo post-treatment. The bactericidal effect of the combination treatment with silver ions was attributed to the generation of hydroxyl radicals, whereas the effects from iron and copper treatments were not radical-mediated. The silver treatment significantly restored the hydrophilicity and cell affinity of the titanium surface. Moreover, CaP coating applied via an electric current (30 µA for 5 min) enhanced hard tissue formation activity on the titanium surface in both in vitro and in vivo settings. The combination treatment utilizing silver ions and purple-LED irradiation significantly enhanced bactericidal effects by generating high levels of hydroxyl radicals. Additionally, coating the titanium surface with functionalized CaP promoted early osseointegration, suggesting a promising strategy for improving implant outcomes.

    Objectives: This study aimed to evaluate the effectiveness of fully remote practical training for the preparation of removable partial dentures, using the video meeting platform Zoom and the communication app. LINE. Materials and Methods: Remote training was applied for one student using Zoom and LINE at the same time as in-person training for the rest of the class. Consumable and instruments needed were delivered to the student subject to remote training in advance. The student’s work was assessed in front of a web camera, at a pre-determined checkpoint during each step. The actual appliances were then mailed to the instructors and reevaluated. The extent of agreement among the assessors for the video-based and direct evaluations, as well as the extent of agreement between the video-based and direct evaluations were calculated. Results: The introduction of Zoom facilitated mutual communication between the student and the instructors. LINE has proven beneficial for practical training. A fully remote practical training class can be conducted smoothly by assigning a dedicated instructor for remote training. The Κ coefficient for agreement between monitor-based and direct evaluations was 0.704 ± 0.12. The survey results indicated that this practical training style was effective in enhancing knowledge and skills. Conclusions: Fully remote blended learning practical training is feasible using Zoom and LINE; however, the tasks to be conducted must be carefully selected. This study is a case report of a single student; thus, future studies should involve fully remote training of small groups to evaluate its educational effectiveness.

    With dental implant treatment becoming the gold standard, the need for effective bone augmentation prior to implantation has grown. This study aims to evaluate a bone augmentation strategy integrating three key growth factors: bone morphogenetic protein-2 (BMP-2), insulin-like growth factor 1 (IGF-1), and vascular endothelial growth factor (VEGF). Collagen scaffolds incorporating BMP-2, IGF-1, or VEGF were fabricated and categorized into five groups based on their content: scaffold alone; BMP-2 alone (BMP-2); BMP-2 and IGF-1 (BI); BMP-2, IGF-1, and VEGF (BIV); and BMP-2 and IGF-1 with an earlier release of VEGF (BI + V). The prepared scaffolds were surgically implanted into the calvarias of C57BL/6JJcl mice, and hard tissue formation was assessed after 10 and 28 days through histological, tomographic, and biochemical analyses. The combination of BMP-2 and IGF-1 induced a greater volume of hard tissue augmentation compared with that of BMP-2 alone, regardless of VEGF supplementation, and these groups had increased levels of cartilage compared with others. The volume of hard tissue formation was greatest in the BIV group. In contrast, the BI + V group exhibited a hard tissue volume similar to that of the BI group. While VEGF and CD31 levels were highest in the BIV group at 10 days, there was no correlation at the same time point between hard tissue formation and the quantity of M2 macrophages. In conclusion, the simultaneous release of BMP-2, IGF-1, and VEGF proved to be effective in promoting bone augmentation.

    OBJECTIVES: An antimicrobial technique utilizing hydroxyl radicals generated by the photolysis of 3% H2O2 has been developed recently. The present study aimed to evaluate the effect of H2O2 photolysis treatment on tooth demineralization caused by Streptococcus mutans biofilm. MATERIALS AND METHODS: To induce tooth demineralization, S. mutans biofilm was allowed to form on the maxillary first molars collected from Wistar rats via 24-h culturing. The samples were immersed in 3% H2O2 and irradiated with 365-nm LED (H2O2 photolysis treatment). Viable bacterial counts in the biofilm were evaluated immediately after treatment and after an additional 30-h culturing by colony counting. The acidogenicity of the biofilm, re-established 30 h after treatment, was assessed by measuring the pH. The effect of H2O2 photolysis treatment on tooth demineralization was assessed by measuring the depth of the radiolucent layer in micro-CT images. RESULTS: H2O2 photolysis significantly reduced viable bacterial counts in the biofilm to 3.7 log colony forming units (CFU)/sample, while the untreated group had 7.9 log CFU/sample. The pH of the biofilm re-established after treatment (6.6) was higher than that of the untreated group (5.3). In line with the pH measurement, the treatment group had a significantly lower depth of radiolucent layer in dentin than the untreated group. CONCLUSIONS: H2O2 photolysis treatment was effective not only in killing the biofilm-forming S. mutans but also in lowering the acidogenicity of the biofilm. Thus, this technique could inhibit tooth demineralization. CLINICAL RELEVANCE: H2O2 photolysis can be applicable as a new dental caries treatment.

    We performed a tarnish test on titanium and investigated how color difference is affected by the ultrasonic cleaning of the specimens in water and storage environment conditions of a reference specimen. The following results were obtained. 1. Ultrasonic cleaning of titanium specimens in water for 2 minutes after polishing did not affect color difference. 2. Storage environment conditions, especially humidity, affected the color difference of the reference specimens in air. 3. Color difference (ΔE*_<ab>) due to passivation was attributed to ΔL * and Δb*, suggesting that this color difference originates from the difference in the thickness of passivation film.

    The magnetic shield material containing no nickel was developed using SUSXM27 stainless steel with absorption of nitrogen gas. After specimens of SUSXM27 were kept at 1200℃ in nitrogen gas at a pressure of 1 atm for 1-10 hours, X-ray diffraction showed nitrogen gas changed an alpha to a gamma phase of the specimens. Anodic polarization curves of the specimens composed of only the alpha and gamma phase respectively were measured in a 0.9%NaCl solution at 37℃. Although pitting corrosion was observed on both specimens, breakdown potentials of the gamma phase were higher than those of the alpha phase.

    本研究は、リン酸カルシウムナノ粒子を基材とした遺伝子導入剤を、DefensinやMagaininなど様々な抗菌ペプチドでコーティングすることで口腔内細菌に効果的な抗菌性を示し、かつ歯肉結合組織由来細胞に対して高い遺伝子導入効率を示す組み合わせを検討する。次に遺伝子導入剤内部に抗炎症性を示すsiRNAやBMP-2などの硬組織誘導性細胞賦活因子を発現させるplasmid-DNAなどを複数種搭載させ、骨造成能をin vivoで検証する。これらの結果を基に、インプラント周囲炎に適用し、オステオインテグレーション獲得性を評価する。さらに組織の早期治癒のために高周波振動刺激を行い骨形成促進効果について検討するものである。 R4年度は、可視光・UVC照射装置を購入し、感染チタン表面において銀イオン存在下で青色LEDを照射することで殺菌効果に加え、光触媒活性によるチタン表面性状の回復ができるのではないかという仮説のもと、黄色ブドウ球菌を用いてその検証を行った。 インプラント表面を模倣したチタンディスク(サンドブラスト処理、リン酸エッチング処理後、黄色ブドウ球菌を播種、培養しバイオフィルムを作成)上に、濃度の異なる硝酸銀溶液を加え、青色LEDを1分間照射し、殺菌効果をコロニー形成単位の測定により評価した。また、各処理を行った後のチタン表面の接触角を測定して、表面性状の親水性を評価した。さらに各処理後のチタンディスク上に骨芽細胞様細胞(MC3T3E1)を播種し、培養2日後にMTT試験を行い、細胞親和性の評価を行った。 その結果、銀イオンと可視光領域の青色LED照射の併用法は、浮遊状ならびにバイオフィルムを形成した黄色ブドウ球菌の殺菌作用だけでなく、感染チタン表面の親水性を回復する効果が認められた。

    インプラント治療は表面性状の発展に伴い治療の成功率が飛躍的に向上することで、世界的に普及する治療方法となった半面、インプラント周囲炎の報告も増加している。しかし、インプラント周囲炎に対する有効な治療方法はまだないことが歯科治療において大きな問題となっている。インプラント治療では、細菌などで汚染した表面の殺菌療法だけでは不十分で、その後に骨の再付着を誘導するようなシステムの確立が重要となる。そこで、本研究ではラジカル殺菌処置後に抗菌性リン酸カルシウム遺伝子導入ナノ粒子によるコーティング層を構築することで、再感染予防と再オステオインテグレーションの獲得をする新しいインプラント周囲組織再生療法を確立することを目的としている。まず、「リン酸カルシウムコーティング層の作製条件」を検証したところ、照射強度や照射時間は一定量を超えると、リン酸カルシウムの粒子の付着量に変化がなくなり、上限があることが明らかにしてきた。また、殺菌効果については、同じ照射強度、照射時間でも対象となる細菌の濃度には適正濃度があること、その適正濃度は細菌の種類によって異なることが明らかとなった。一方、「CaP コーティング層の表面性状、生体親和性」について解析したところ、菌種によってはリン酸カルシウム自体が細菌の増殖を助長していることが確認された。そこで、新たにこれまでと異なる抗菌ペプチドを付与したリン酸カルシウムナノ粒子を製作し、同細菌に対して殺菌試験を行ったところ、リン酸カルシウム自体による細菌増殖効果が抑制されることが明らかとなった。現在は同リン酸カルシウムナノ粒子の遺伝子導入効率や生体親和性をインプラント周囲に存在する歯周組織関連細胞を用いて検証している。

    The aim of the present study was to evaluate 1) the antimicrobial activity of hydrogen photolysis treatment against multi-species biofilm containing cariogenic bacteria, and 2) reactions of the dental pulp to the treatment. The hydrogen photolysis treatment is a novel antimicrobial technique in which oxidative power of hydroxyl radicals generated by photo-irradiation of hydrogen peroxide is used. The treatment could effectively reduce viable bacteria in the multi-species biofilm as in the case of single-species biofilm. The reactions of the dental pulp were evaluated in an animal study using rats. The treatment generated localized degenerative tissue at pulp horn region, whereas infiltration of inflammatory cells was not observed. The degenerative tissue was repaired within 5 days, and tertiary dentin formation was observed 21 days after treatment.

    歯科インプラントは,欠損補綴における標準的な治療オプションとして歯科臨床において広く普及した.しかしながら,日常臨床においては,欠損部の骨量・骨質が不十分な場合や,骨造成を行った場合,さらには骨粗鬆症や糖尿病など骨治癒不全ケースでは,インプラント埋入後の周囲骨形成が充分になされず,オッセオインテグレーションが獲得されない,もしくは治癒期間が大幅に延長してしまう等の症例が依然として多く存在する.これら理由から治療を断念する患者は,歯科インプラントを希望する患者の実に2割以上に及ぶとされる.このような臨床的課題に対して,薬物による副作用や外科的侵襲を伴わず,安全で,簡便に骨形成とオッセオインテグレーション獲得促進をサポートする支援デバイスの開発は非常に意義深い. 本研究は,歯科インプラント治療支援を目的としたインプラント周囲骨形成・オッセオインテグレーション促進効果をもつ低強度・高周波振動刺激デバイスの開発を目指すものである.アカデミア主導型医療機器開発であり,新規口腔デバイス開発(基礎データ収集,技術の知財化,機器開発),トランスレーショナルリサーチ,非臨床POC(Proof of Concept)の取得という2つのステップから構成される. 昨年度は,ヒト口腔内への応用を想定した小型の刺激デバイスの試作機の設計,口腔内に維持・固定する方法および振動刺激の周波数・強度等の条件の検討を行った.

    The present study aimed to evaluate the potential effects of proanthocyanidin-rich grape seed extract (GSE) on the healing of bone defects as well as implant osseointegration in ovariectomized (OVX) rats, a model of osteoporosis in postmenopausal women. Female Wistar rats were used. The animals underwent OVX operation. One week after OVX surgery, the second surgery was performed to create calvarial bone defects. In addition, one week after the second surgery, implant surgery was performed to install titanium mini-screws in tibiae. During the healing period, GSE or vehicle (water) was daily administered by oral gavage. OVX tended to impair healing of the defects. However, GSE counteracted the OVX-induced impairment in bone healing. Similarly, GSE increased the implant removal torque and bone-to-implant contact compared to those of the water-administered animals. These findings suggest that oral administration of GSE may be beneficial in dental implant therapy in osteoporosis patients.

    Protamine-coated multi-shell calcium phosphate (CaP) was developed as a non-viral vector for tissue regeneration therapy in this study. The antibacterial effects of CaP nanoparticles against Streptococcus mutans and Aggregatibacter actinomycetemcomitans increased with an increase in the protamine dose. In the combination treatment with BMP-2 and IGF-1, the concentration ratio of BMP-2 and IGF-1 is an important factor affecting bone formation activity. During gene transfection treatment, BMP-2 and IGF-1 were released simultaneously after gene transfer; the loaded dose of the plasmid DNA encoding IGF-1 did not impact the BMP-2 or IGF-1 yield or new bone formation ratio in vitro and in vivo. In conclusion, two growth factor-releasing systems were developed using an antibacterial gene transfer vector, and the relationship between the loaded plasmid DNA dose and resultant growth factor yield was determined in vitro and in vivo.

    The aim of this research subject is to study the bacteriostatic mechanism of Ti-Ag alloys. Various titanium alloys were developed and their microstructures and mechanical properties were investigated. The microstructures of Ti-Ag alloys were controlled by the heat treatment. Bacteriostatic action of Ti-Ag alloys whose microstructures were controlled was examined. The bacteriostatic action of massive alpha was superior to that of Ti2Ag. It was found that the bacteriostatic mechanism was different from antibacterial action by Ag ions.

    100nM 1alpha,25-dihydroxy Vitamin D3 enhanced Cementum protein 1 (CEMP1) and F-spondin gene expressions considered as specific cementoblastic markers in human periodontal ligament cells. 1alpha,25-dihydroxy Vitamin D3 also enhanced Osterix, Runx2, Alkaline phosphatase (ALP), Osteocalcin (OCN) and Bone sialoprotein (BSP) gene expressions which are regarded as important factors for osteoblastic and cementoblastic differentiation and mineralization. These results indicated that 1alpha,25-dihydroxy Vitamin D3 regulates periodontal ligament cells differentiation. Furthermore, Cells strongly expressed CEMP1 protein were appeared with long term 1alpha,25-dihydroxy Vitamin D3 stimulation. 100nM 1alpha,25-dihydroxy Vitamin D3 decreased mineralized nodule formation. 100nM 1alpha,25-dihydroxy Vitamin D3, Ascorbic acid, beta-glycerophosphate and dexamethazone enhanced mineralized nodule formation, however CEMP1 protein expression was decreased compared with 1alpha,25-dihydroxy Vitamin D3 alone.

    This study aimed to evaluate the capability of treatment based on hydroxyl radical for peri-implantitis. In the study, sandblasted titanium discs were used as specimens. Firstly, treated specimens were observed to investigate the effect of this treatment on a titanium surface. There were not significant differences between treated discs and untreated discs. Then I analysed proliferation of mouse osteoblastic cells (MC3T3-E1) cultured on the treated discs. Compared with intact discs, biofilm (using Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, Fusobacterium nucleatum and Streptococcus mitis) contamination lowered cell proliferation on the specimen surface,whereas this treatment recovered cell proliferation. These findings suggest that treatment based on hydroxyl radical can recover the degraded biocompatibility of biofilm-contaminated titanium surfaces without damage and can potentially be utilised for peri-implantitis treatment.

    The aim of this study is that the development of new dental pulp protection treatment with a Anti-inflammatory effect using gene silencing and regeneration by the fluoroapatite synthesis. In this study, a gene transfection vector based on Calcium phosphate(CaP), which is composed of bone, enamel and dentin, was prepared and evaluated the gene transfection efficiency and cell viability of CaP with different peptide. For rat dental pulp derived (DP) cells, CaP nanoparticles with octa-ariginine or Protamine induced high transfection efficiency and cell viability. Furthermore, this reduced the TNF-a mRNA expression of the rat DP cells stimulated by LPS. These results suggested that the gene silencing using CaP with siRNA(anti-TNF-a) reduced inflammation. On the other hand, the synthesis of fluoroapatite having vertical crystal arrangement against dental surface. Furthermore, the application of CaP with siRNA(anti-TNA-a)reduced the inflammation in histological observation.

    Aiming to develop functionally gradient bacteriostatic alloys, we prepared experimental Ti-Ag alloys and investigated their machinability. The machinability of Ti-Ag alloys was superior to that of pure titanium. On the other hand, experimental Ti-Nb-Cu ternary alloys were prepared and investigated their properties. The differences in the mechanical properties are attributable to solid-solution strengthening or to precipitation strengthening by the dual-phase (alpha + beta) structure. Additionally, we developed a simplified fixating device for performing intraoral radiography in beagle dogs.

    In this project, in vitro and in vivo tests were performed with an aim of application of radical disinfection technique to treatment for peri-implantitis. In vivo test, where peri-implantitis caused around implants installed in dogs were treated with radical disinfection technique. However, there was no significant difference between the test and control groups. Thus, to find more suitable treatment conditions, in vitro test was additionally performed. The results demonstrated that the radical disinfection technique could recondition the titanium surface that had been previously contaminated by bacteria for subsequent osteoblastic cells. Therefore, future study should focus more on the establishment of treatment condition that enables to acquire re-osseointegration around the implants suffering from peri-implantitis.

    Induction of ectopic hard tissue formation and resin-bonding affinity using αTCP-4META/MMA-TBB resin composits and ascorbic acid/copper chloride or EDC crosslinked nanohydroxyapatite/collagen complex (nHAC) impregnated with BMP-2 were examined in vitro and in vivo in order to improve the clinical outcome of vertical root fracture treatent. As results, a hybrid layer was observed at the interface between resin and bone in both optical microscope and SEM observation, and direct resin-bone contact was observed without connective tissue intervention, when nHAC crosslinked with ascorbic acid/copper chloride was adhered to resin and impregnated with BMP-2 and implanted into connective tissue.

    Fundamental elements of tissue regeneration include cells, growth factors, and a scaffold. This study aimed to fabricate a growth factor releasing biodegradable scaffold for tissue regeneration. We prepared multi-shell calcium phosphate (CaP) nanoparticles functionalised with DNA, polyethyleneimine (PEI), protamine, and octa-arginine (R8), and compared their respective transfection activity and cell viability measures using human mesenchymal stem cells. These experiments demonstrated that the scaffolds were associated with moderate hMSC cell viability and were capable of releasing the BMP-2 protein into hMSCs following gene transfection. In particular, the scaffold loaded with protamine-containing CaP nanoparticles demonstrated the highest cell viability and transfection efficiency in hMSCs; thus, it might be suitable to serve as an efficient growth factor releasing scaffold.

    To improve the bonding treatment of vertically fractured roots, we aimed to the newly cementum formation on a compound of resin and dentin powder, which may improve the binding between resin and cementum. In vitro, cytotoxicity and Micro Tensile Bond Strength (MTBS) test were assessed. It had little cytotoxicity and exhibited higher bond strength. In vivo, in some cases, we observed the newly cementum formation on a compound of resin and dentin powder. This experiment might exhibit a possibility of the newly cementum formation on a resin.
