    Abstract X-ray Talbot and Talbot–Lau interferometers consisting of transmission gratings are widely used for X-ray phase imaging, which depicts soft materials. This study exploits the use of a pair of concave and convex parabolic gratings instead of a conventional rectangular phase grating to enhance the phase signal optically. To gain insight into the optimal design, signal enhancement is evaluated by directly measuring the self-image formed downstream of the pair. An increase in the differential phase signals is demonstrated as a function of the distance between the pair, and prospects for deploying this concept into a practical phase imaging technique are discussed.

    Developing long bones alter their shape while maintaining uniform cortical thickness via coordinated activity of bone-forming osteoblasts and bone-resorbing osteoclasts at periosteal and endosteal surfaces, a process we designate trans-pairing. Two types of trans-pairing shift cortical bone in opposite orientations: peri-forming trans-pairing (peri-t-p) increases bone marrow space and endo-forming trans-pairing (endo-t-p) decreases it, via paired activity of bone resorption and formation across the cortex. Here, we focused on endo-t-p in growing bones. Analysis of endo-t-p activity in the cortex of mouse fibulae revealed osteoclasts under the periosteum compressed by muscles and expression of RANKL in periosteal cells of the cambium layer. Furthermore, mature osteoblasts were localized on the endosteum, while preosteoblasts were at the periosteum and within cortical canals. X-ray tomographic microscopy revealed the presence of cortical canals more closely associated with endo- than with peri-t-p. Sciatic nerve transection followed by muscle atrophy and unloading induced circumferential endo-t-p with concomitant spread of cortical canals. Such canals likely supply the endosteum with preosteoblasts from the periosteum under endo-t-p, allowing bone shape to change in response to mechanical stress or nerve injury.

    We have developed a new neutron phase imaging system with a Talbot–Lau interferometer for utilization at the CN-3 port of the Kyoto University Reactor. To achieve efficient differential-phase imaging and visibility (dark-field) imaging at this beamline, we adopted a relatively shorter design wavelength of 2.7 Å. By fabricating neutron absorption gratings with thick gadolinium absorbers, we were able to obtain clear moiré fringes with a high visibility of 55% for thermal neutrons. As a demonstration of its imaging capabilities for expanded actual utilization in the medium-sized sources, we observed additively manufactured rods of Inconel 718. Using visibility imaging, we successfully examined variations in the size of defects in the rods caused by hot isostatic pressing process. In addition, we conducted tomography measurements of the rods, which allowed us to reveal the spatial distribution of defects at sub-micrometer scales.

    Abstract In grating-based X-ray Talbot interferometry, the wave nature of X-ray radiation is exploited to generate phase contrast images of objects that do not generate sufficient contrast in conventional X-ray imaging relying on X-ray absorption. The phase sensitivity of this interferometric technique is proportional to the interferometer length and inversely proportional to the period of gratings. However, the limited spatial coherency of X-rays limits the maximum interferometer length, and the ability to obtain smaller-period gratings is limited by the manufacturing process. Here, we propose a new optical configuration that employs a combination of a converging parabolic micro-lens array and a diverging micro-lens array, instead of a binary phase grating. Without changing the grating period or the interferometer length, the phase signal is enhanced because the beam deflection by a sample is amplified through the array of converging-diverging micro-lens pairs. We demonstrate that the differential phase signal detected by our proposed set-up is twice that of a Talbot interferometer, using the same binary absorption grating, and with the same overall inter-grating distance.

    X-ray phase contrast image has higher sensitivity to materials with light elements than the conventional absorption contrast. Phase image measurement technique using diffraction gratings can be used in the laboratory. Furthermore, X-ray phase microscope using the diffraction gratings has been also developed. However, phase measurement with diffraction gratings has a drawback that the spatial resolution is limited to the grating period. To overcome this problem, a scanning system using a triangular phase grating has recently been proposed, which is expected to archive a large field of view and a high spatial resolution. This study aims to investigate the feasibility of the system and a deep understanding of the optical system by simulation.

    The locomotor system is highly bilateral at the macroscopic level. Homochirality of biological molecules is fully compatible with the bilateral body. However, whether and how single-handed cells contribute to the bilateral locomotor system is obscure. Here, exploiting the small number of cells in the swimming tadpole larva of the ascidian Ciona, we analyzed morphology of the tail at cellular and subcellular scales. Quantitative phase-contrast X-ray tomographic microscopy revealed a high-density midline structure ventral to the notochord in the tail. Muscle cell nuclei on each side of the notochord were roughly bilaterally aligned. However, fluorescence microscopy detected left-right asymmetry of myofibril inclination relative to the longitudinal axis of the tail. Zernike phase-contrast X-ray tomographic microscopy revealed the presence of left-handed helices of myofibrils in muscle cells on both sides. Therefore, the locomotor system of ascidian larvae harbors symmetry-breaking left-handed helical cells, while maintaining bilaterally symmetrical cell alignment. These results suggest that bilateral animals can override cellular homochirality to generate the bilateral locomotor systems at the supracellular scale.

    Auditory ossicles in the middle ear and bony labyrinth of the inner ear are highly mineralized in adult mammals. Cellular mechanisms underlying formation of dense bone during development are unknown. Here, we found that osteoblast-like cells synthesizing highly mineralized hearing-related bones produce both type I and type II collagens as the bone matrix, while conventional osteoblasts and chondrocytes primarily produce type I and type II collagens, respectively. Furthermore, these osteoblast-like cells were not labeled in a "conventional osteoblast"-specific green fluorescent protein (GFP) mouse line. Type II collagen-producing osteoblast-like cells were not chondrocytes as they express osteocalcin, localize along alizarin-labeled osteoid, and form osteocyte lacunae and canaliculi, as do conventional osteoblasts. Auditory ossicles and the bony labyrinth exhibit not only higher bone matrix mineralization but a higher degree of apatite orientation than do long bones. Therefore, we conclude that these type II collagen-producing hypermineralizing osteoblasts (termed here auditory osteoblasts) represent a new osteoblast subtype.     Owing to the development of X-ray focusing optics during the past decades, synchrotron-based X-ray microscopy techniques allow the study of specimens with unprecedented spatial resolution, down to 10 nm, using soft and medium X-ray photon energies, though at the expense of the field of view (FOV). One of the approaches to increase the FOV to square millimetres is raster-scanning of the specimen using a single nanoprobe; however, this results in a long data acquisition time. This work employs an array of inclined biconcave parabolic refractive multi-lenses (RMLs), fabricated by deep X-ray lithography and electroplating to generate a large number of long X-ray foci. Since the FOV is limited by the pattern height if a single RML is used by impinging X-rays parallel to the substrate, many RMLs at regular intervals in the orthogonal direction were fabricated by tilted exposure. By inclining the substrate correspondingly to the tilted exposure, 378000 X-ray line foci were generated with a length in the centimetre range and constant intervals in the sub-micrometre range. The capability of this new X-ray focusing device was first confirmed using ray-tracing simulations and then using synchrotron radiation at BL20B2 of SPring-8, Japan. Taking account of the fact that the refractive lens is effective for focusing high-energy X-rays, the experiment was performed with 35 keV X-rays. Next, by scanning a specimen through the line foci, this device was used to perform large FOV pixel super-resolution scanning transmission hard X-ray microscopy (PSR-STHXM) with a 780 ± 40 nm spatial resolution within an FOV of 1.64 cm × 1.64 cm (limited by the detector area) and a total scanning time of 4 min. Biomedical implant abutments fabricated via selective laser melting using Ti–6Al–4V medical alloy were measured by PSR-STHXM, suggesting its unique potential for studying extended and thick specimens. Although the super-resolution function was realized in one dimension in this study, it can be expanded to two dimensions by aligning a pair of presented devices orthogonally.

    X-ray phase computed tomography (CT) is used to observe the inside of light materials. In this paper, we report a new study to develop and test a laboratory assembled X-ray phase CT system that comprises an X-ray Lau interferometer, a rotating Mo anode X-ray tube, and a detector with high spatial resolution. The system has a high spatial resolution lower than 10 μm, which is evaluated by differentiating neighbouring carbon fibres in a polymer composite material. The density resolution is approximately 0.035 g/cm3, which enables to successfully distinguish the high-density polyethylene (HDPE, 0.93 g/cm3) from the ultra-low-density polyethylene (ULDPE, 0.88 g/cm3) in the sample. Moreover, the system can be switched to operate on another mode based on a Talbot–Lau interferometer that provides a wider field of view with a moderate spatial resolution (approximately 100 μm). By analyzing sample images of the biological, this study demonstrates the feasibility and advantages of using hybrid configuration of this X-ray phase CT system.

    Electron microscopy is a powerful tool for visualizing the shapes of sub-nanometer objects. However, Contrast Transfer Function (CTF) principally restricts lower frequency components in the image. To overcome this problem, phase-plate techniques have been proposed and currently Hole Free Phase Plate (HFPP) and Volta Phase Plate (VPP) are widely used especially for biological specimens to retrieve low frequency information of the sample's potential distributions. In this report, we have developed a new phase-contrast scanning transmission electron microscope (STEM) in which a probe beam including side robes is formed with an amplitude Fresnel zone plate (FZP) and the interference patterns produced by the zero and first order diffracted waves generated by the FZP are detected. We name it FZP Phase Contrast STEM (FZP-PC-STEM) hereinafter. The amplitude FZP was manufactured by using focused ion beam (FIB) equipment, and the diffraction data were collected by using diffraction imaging technique. The validity of our proposed optical model was confirmed by comparing experimental and simulated images. Observations of carbon nanotube (CNT) bundles by using this method showed that the contrast of low-spatial-frequency components in the CNT image was significantly enhanced. This method does not, in principle, require the post-image processing used in the diffraction imaging method, and it can be easily introduced into pre-existing equipment without major modifications. The stability and robustness of the FZP inserted in condenser system were also confirmed during long-time operation. We expect that the FZP-PC-STEM will be widely applicable to high-contrast observations of low-Z samples with simple and easy operation.

    A two-dimensional array of compound refractive lenses (2D array of CRLs) designed for hard X-ray imaging with a 3.5 mm2 large field of view is presented. The array of CRLs consists of 2D polymer biconcave parabolic 34 × 34 multi-lenses fabricated via deep X-ray lithography. The developed refractive multi-lens array was applied for sub-pixel resolution scanning transmission X–ray microscopy; a raster scan with only 55 × 55 steps provides a 3.5 megapixel image. The optical element was experimentally characterized at the Diamond Light Source at 34 keV. An array of point foci with a 55 µm period and an average size of ca. 2.1 µm × 3.6 µm was achieved. In comparison with the conventional scanning transmission microscopy using one CRL, sub-pixel resolution scanning transmission hard X-ray microscopy enables a large field of view and short scanning time while keeping the high spatial resolution.

    Under the JST-ERATO project in progress to develop X-ray and neutron phase-imaging methods together, recent achievements have been selected and reviewed after describing the merit and the principle of the phase imaging method. For X-ray phase imaging, recent developments of four-dimensional phase tomography and phase microscopy at SPring-8, Japan are mainly presented. For neutron phase imaging, an approach in combination with the time-of-flight method developed at J-PARC, Japan is described with the description of new Gd grating fabrication.

    X-ray radiographic imaging technique has found applications in various fields. However, it is not enough to get clear X-ray images of samples with low absorbance, such as biological soft tissues. To resolve this problem, we proposed a method using an X-ray Talbot interferometer of X-ray phase imaging. In this X-ray Talbot interferometer, X-ray gratings were required to have a fine, high accuracy, high aspect ratio structure. Then, we have developed and succeeded high aspect ratio X-ray gratings with a pitch of 5.3 μm and a height of 30 μm using X-ray lithography technique. We discuss that the X-ray gratings having a large effective area in order to obtain imaging size of practical use in medical application. In currently study, we have fabricated the X-ray gratings with a large effective area of 60 mm×60 mm. And, we conducted X-ray phase tomography of mouse at the chest and abdominal using X-ray Talbot interferometer. As a result, we successfully observed soft tissues and high density tissues. With the aim of broadening a field of view, we try to fabricate X-ray gratings that have a pitch of below 5.3 μm and larger area of 100 mm square. This result suggests that X-ray Talbot interferometer is a novel and simple method for phase sensitive X-ray radiography.

    Three-dimensional tomographic imaging using x-ray phase information is reported. The contrast in normal x-ray transmission images is generated by the difference in x-ray absorption. Therefore, structures in weakly x-ray absorbing objects are hardly depicted. Because the interaction cross section of x-ray phase shift is about a thousand times larger than that of absorption for low-Z elements, highly sensitive imaging is achieved by detecting the x-ray phase shift. We used an x-ray interferometer to measure interference patterns and phase-shifting interferometry to obtain x-ray phase maps from the interference patterns. Phase-contrast x-ray CT reconstructs three-dimensional images from the x-ray phase maps. This paper introduces the principle of this technique and observation results obtained for cancerous tissues using synchrotron radiation.

    X-ray transmission imaging that creates image contrast from the distribution of the X-ray absorption coefficient is not sensitive to materials consisting of light elements such as hydrogen, carbon, nytrogen, and oxygen. On the other hand, the X-ray phase shift caused by the light elements is substantial enough to be detected even when absorption is almost zero. Hence, phase-contrast X-ray imaging is a promissing technique for observing the structure inside biological soft tissues without the need for staining and without serious radiation exposure. Using fringe scanning X-ray interferometry, the X-ray phase shift caused by an object was measured. Three-dimensional image reconstruction of cancerous tissues using the measured phase shifts was enabled under tomographic configuration phase-contrast X-ray computed tomography (CT) .

    Phase-contrast X-ray computed tomography (CT) enabling the observation of biological soft tissues without contrast enhancement has been developed. The X-ray phase shift caused by an object is measured and input to a standard CT reconstruction algorithm. A thousand times increase in the image sensitivity to soft tissues is achieved compared with the conventional CT using absorption contrast. This is because the X-ray phase shift cross section of light elements is about a thousand times larger than the absorption cross section. The phase shift is detected using an X-ray interferometer and computer analyses of interference patterns.<BR>Experiments were performed using a synchrotron X-ray source. Excellent image sensitivity is demonstrated in the observation of cancerous rabbit liver. The CT images distinguish cancer lesion from normal liver tissue and, moreover, visualize the pathological condition in the lesion. Although the X-ray energy employed and the present observation area size are not suitable for medical applications as they are, phase-contrast X-ray CT is promising for investigating the internal structure of soft tissue which is almost transparent for X-rays. The high sensitivity also provides the advantage of reducing X-ray doses.

    Even for hard X-rays, X-ray phase information allows image formation with higher contrast for such a biological soft tissue, which cannot be sufficiently delineated by conventional absorption-based imaging. To generate projection data from raw fringe pattern images in the process of this CT image reconstruction procedures, frequently 2π-phase jump must be corrected, i.e., the phase unwrapping. However, it is difficult to obtain the satisfactory corrections for noisy fringe pattern images. In previous attempt, regarding the phase unwrapping as a combinatorial optimization, we have proposed a robust phase-unwrapping algorithm for phase-contrast X-ray CT by using genetic algorithm. However, the cost of calculation has the severe problem, because it takes several hours. Here, we have proposed an improvement of GA procedure for shortening of processing time by applying these procedure on divided local area. The validity of this method is demonstrated and discussed experimentally.

    In hard X-ray regions, use of phase information allows image formation with higher contrast for a phase object such as a biological soft tissue, which cannot be sufficiently delineated by conventional absorption-based imaging. To generate projections from raw fringe pattern images in the course of the CT image reconstruction procedures, 2π-phase jump must be corrected, i. e., the phase unwrapping. However, it is difficult to obtain the satisfactory results for low S/N fringe pattern images. In this research, we propose a method to correct the failed unwrapped results for the phase-contrast CT imaging. We investigate the effectiveness for actual data obtained in Spring-8, and point out the left problems.

    X-ray phase imaging has been attracting attention since it can be applied to polymers and biological soft tissues consisting low-Z elements, which are not imaged clearly with conventional X-ray imaging methods. The study performed so far was for static samples during imaging. However, dynamical imaging is desirable to understand the property and function of materials. We studied the methods for high-speed phase imaging and four-dimensional X-ray phase tomography with X-ray Talbot or Talbot-Lau interferometry. More concretely, we developed methods in combination with white synchrotron radiation and storoboscopic methods for periodic phenomena as a breaktrough to dynamical X-ray phase imaging.

    Using the X-ray phase measurement techniques with X-ray transmission grating(s), including X-ray Talbot interferometry, in combination with an X-ray imaging microscope with a Fresnel zone plate, X-ray phase-sensitive imaging microscopy has been developed, enabling high-sensitive and high-resolution observation for weakly absorbing objects, such as polymers and biological tissues. In addition, by repeating the phase measurement at plural projection directions, three-dimensional image reconstruction of the refractive index (X-ray phase tomography) has also been demonstrated.

    Interferometry, which is indispensable in modem science and technology, has been quite successfully developed in the visible light region. In the region of soft X-rays whose wavelength is much shorter than that of visible light, however, interferometry is underdeveloped, and few researches into the metrology taking advantage of wave nature are reported. It is dear that a variety of applications would be opened up when a soft X-ray interferometer is developed. However, the short wavelength of soft X-rays and poor quality of optical elements for soft X-rays have obstructed the development of soft X-ray interferometry. We have experience of constructing and operating interferometers for hard X-rays whose wavelength is further shorter than that of soft X-rays. We proposed this research project as an expansion of our activity into the soft X-ray region. In the hard X-ray region, crystal optical elements are available, and therefore interferometers were operated in spite of the hard-X-ray wavelength (ca. 0.1 nm). Although problems characteristic of soft X-rays, such as that crystals are not available, and that an interferometer has to be constructed in vacuum, are pointed out, we consider that the development of a soft X-ray interferometer can be within our scope basing on our achievement in the hard X-ray region. We constructed a Mach-Zehnder type interferometer for 13-nm synchrotron soft X-rays employing free-standing Ru/SiN films for beam splitters. We selected this configuration because its operation is comparatively easy and a variety of applications are expected. We found that only two accurate tuning stages are required in order to align four optical elements, and fabricated a prototype comprising accurate petitioners driven by piezoelectric actuators. Furthermore, because the plan for alignment is important, we devised a method that depends on the visibility of visible laser. Experiments using soft X-rays were canned out at the Photon Factory, High Energy Accelerator Research Organization. Unfortunately, we have not observed interference yet. We consider that vibration and/or deformation of optical elements would be responsible for current results. We will continue experiments, devising to get rid of the problems. Once interference is observed, we have a plan to add a mechanism for changing optical path lengths and to move on the development of soft X-ray Fourier interferometry to realize innovative energy resolution in the field of soft X-ray spectroscopy.
