    Mobile radiography is useful for triaging patients during disasters. However, in the face of large-scale disasters, medical equipment, including X-ray systems, may be restricted because of power outages. Therefore, a novel mobile radiography system that can be used in any disaster was developed, and its basic performance was evaluated. This system is designed for use during power outages and can be recharged using both household and solar power sources. The reproducibility of the X-ray output of the system (irradiation dose, tube voltage, radiation quality and irradiation time) was excellent, confirming that radiography could be performed consistently. The entrance surface dose and scattered radiation during chest radiography were measured using a chest phantom. Both results were acceptable, verifying that the system is sufficiently safe from the viewpoint of radiation protection. This novel mobile radiology system is easy to carry and can generate solar power, making it useful during a power outage in the case of a large-scale disaster or when examinations need to be performed outside a hospital, such as at a first-aid station.

    The International Commission on Radiological Protection has reduced the occupational eye lens dose limit to 20 mSv/year. Interventional radiology (IR) physicians are frequently exposed to high levels of scattered X-rays, and radiation protection tools, including lead glasses, are used for minimizing exposure. However, the effectiveness of lead eyewear for radiation workers who wear prescription glasses (e.g. over-glasses-type eyewear) has not been comprehensively investigated. This study evaluated the shielding effectiveness of over-glasses-type eyewear worn over plastic prescription glasses through phantom experiments. Furthermore, the dose reduction provided by the plastic lenses and front frame of the prescription glasses (excluding lead) was assessed. The over-glasses-type eyewear demonstrated high shielding effectiveness, with average shielding rates of 0.72-0.74 and 0.74-0.80 for the left and right eyes, respectively, against radiation incident from the lower left oblique, as typically encountered in clinical settings. In contrast, the plastic lenses of prescription glasses provided <9% dose reduction when it irradiated with 80- and 120-kV X-rays, indicating minimal contribution to eye protection. The plastic front frame offered a 14% reduction at 80 kV and 11% at 120 kV. These findings suggest that the front frame of the prescription glasses contributes to photon attenuation and improves overall shielding. However, the improper fit of over-glasses-type eyewear may lead to gaps, compromising its shielding effectiveness. To minimize gaps and optimize protection, IR physicians should select over-glasses-type eyewear that fits securely over prescription glasses. Additionally, combining protective tools is essential for effective eye protection.

    Fluoroscopy-guided bronchoscopy is an essential tool for diagnosing and treating lung diseases, particularly lung cancer. However, prolonged fluoroscopic exposure raises concerns regarding radiation-induced lens injuries in physicians, such as radiation cataracts. In response to the International Commission on Radiological Protection lowering the occupational lens dose limit to an average of 20 mSv/year over 5 years, there is an increasing need for effective lens protection during such procedures. This study has aimed to develop a novel lens protection device specifically designed for bronchoscopy physicians and evaluate its protective performance through a phantom study. The device consisted of a 0.175 mm lead (Pb) sheet positioned on the left side of the physician's head, secured with headgear to improve stability and comfort during prolonged use. A phantom study was conducted using a trunk phantom to simulate a patient and a head phantom to simulate a physician. The scattered radiation doses were measured at 15 locations on the phantom head using a radiophotoluminescence glass dosimeter, both with and without a protective device. The device demonstrated a protective effect of more than 80% for the left eye across all tested angles, whereas the right eye showed protection ranging from approximately 40% to 75% depending on the angle. This novel lens protection device has the potential to significantly reduce scattered radiation to the left eye while minimizing vision obstruction and discomfort. This offers a practical solution for radiation protection during bronchoscopy and may be applicable to other interventional procedures requiring fluoroscopic guidance.

    The purpose of this study was to estimate the internal dose of radiation in Japanese macaques (aka Nihonzaru or snow monkey) due to the Fukushima nuclear power plant accident. Images of a male Japanese macaque weighing ~10 kg were acquired using a multi-slice computed tomography (CT) scan with a 64-row segment detector. The CT images were used to create voxel phantoms of the bones, bone marrow, brain, eyes, heart, lungs, stomach, liver, spleen, pancreas, kidneys, intestines, bladder, testes, thyroid and miscellaneous tissue. The Particle and Heavy Ion Transport System (PHITS) Monte Carlo code was used to calculate the internal exposure rate conversion factors for 134Cs, 137Cs and 131I isotopes for the created voxel phantoms with a statistical precision higher than 1%. The PHITS-calculated energy deposits were compared with those for rhesus monkeys. The results showed that the fractions of energy deposits for β-radiation in different organs were almost identical between the two species. For γ-radiation, there was excellent agreement in the self-absorption rate with the approximate curve of the Japanese macaque, with an average deviation of 2%. The maximum deviation of 12% was for the kidney, which has two organs, so the error with the approximate curve is slightly larger due to the energy loss created between organs.

    Although interventional radiology (IVR) is preferred over surgical procedures because it is less invasive, it results in increased radiation exposure due to long fluoroscopy times and the need for frequent imaging. Nurses engaged in cardiac IVR receive the highest lens radiation doses among medical workers, after physicians. Hence, it is important to measure the lens exposure of IVR nurses accurately. Very few studies have evaluated IVR nurse lens doses using direct dosimeters. This study was conducted using direct eye dosimeters to determine the occupational eye dose of nurses engaged in cardiac IVR, and to identify simple and accurate methods to evaluate the lens dose received by nurses. Over 6 months, in a catheterization laboratory, we measured the occupational dose to the eyes (3 mm dose equivalent) and neck (0.07 mm dose equivalent) of nurses on the right and left sides. We investigated the relationship between lens and neck doses, and found a significant correlation. Hence, it may be possible to estimate the lens dose from the neck badge dose. We also evaluated the appropriate position (left or right) of eye dosimeters for IVR nurses. Although there was little difference between the mean doses to the right and left eyes, that to the right eye was slightly higher. In addition, we investigated whether it is possible to estimate doses received by IVR nurses from patient dose parameters. There were significant correlations between the measured doses to the neck and lens, and the patient dose parameters (fluoroscopy time and air kerma), implying that these parameters could be used to estimate the lens dose. However, it may be difficult to determine the lens dose of IVR nurses accurately from neck badges or patient dose parameters because of variation in the behaviors of nurses and the procedure type. Therefore, neck doses and patient dose parameters do not correlate well with the radiation eye doses of individual IVR nurses measured by personal eye dosimeters. For IVR nurses with higher eye doses, more accurate measurement of the radiation doses is required. We recommend that a lens dosimeter be worn near the eyes to measure the lens dose to IVR nurses accurately, especially those exposed to relatively high doses.

    Abstract The signal intensities of CO2− radicals in teeth can be utilised as an individual indicator of the cumulative external dose for animals. To accurately determine the external dose, it is desirable to analyse the CO2− radical intensity and improve its detection limit. We recently reported a dose–response in the range of 0–200 mGy and estimated the absorbed dose for seven wild Japanese macaques captured in/around the related areas to the Fukushima Daiichi Nuclear Power Plant accident. Herein, for further improvement of this method, we examined the electron spin resonance spectra of the teeth of these seven and an additional four macaques captured in Fukushima by applying two spectrum-decomposition algorithms.

    The International Commission on Radiological Protection has lowered the annual equivalent eye-lens dose to 20 mSv. Although occupational exposure can be high in nuclear medicine (NM) departments, few studies have been conducted regarding eye-lens exposure among NM staff. This study aimed to estimate the annual lens doses of staff in an NM department and identify factors contributing to lens exposure. Four nurses and six radiographers performing positron emission tomography (PET) examinations and four radiographers performing radioisotope (RI) examinations (excluding PET) were recruited for this study. A lens dosimeter was attached near the left eye to measure the 3-mm-dose equivalent; a personal dosimeter was attached to the left side of the neck to measure the 1-cm- and 70-µm-dose equivalents. Measurements were acquired over six months, and the cumulative lens dose was doubled to derive the annual dose. Correlations between the lens and personal-dosimeter doses, between the lens dose and the numbers of procedures, and between the lens dose and the amounts of PET drugs (radiopharmaceuticals) injected were examined. Wilcoxon’s signed-rank test was used to compare lens and personal-dosimeter doses. The estimated annual doses were 0.93 ± 0.13 mSv for PET nurses, 0.71 ± 0.41 mSv for PET radiographers, and 1.10 ± 0.53 mSv for RI radiographers. For PET nurses, but not for PET or RI radiographers, there was a positive correlation between the numbers of procedures and lens doses and between amounts injected and lens doses. There was a significant difference between the lens and personal-dosimeter doses of PET nurses. The use of protective measures, such as shielding, should prevent NM staff from receiving lens doses &gt; 20 mSv/year. However, depending on the height of the protective shield, PET nurses may be unable to assess the lens dose accurately using personal dosimeters.

    Mobile radiography allows for the diagnostic imaging of patients who cannot move to the X-ray examination room. Therefore, mobile X-ray equipment is useful for patients who have difficulty with movement. However, staff are exposed to scattered radiation from the patient, and they can receive potentially harmful radiation doses during radiography. We estimated occupational exposure during mobile radiography using phantom measurements. Scattered radiation distribution during mobile radiography was investigated using a radiation survey meter. The efficacy of radiation-reducing methods for mobile radiography was also evaluated. The dose decreased as the distance from the X-ray center increased. When the distance was more than 150 cm, the dose decreased to less than 1 μSv. It is extremely important for radiological technologists (RTs) to maintain a sufficient distance from the patient to reduce radiation exposure. The spatial dose at eye-lens height increases when the bed height is high, and when the RT is short in stature and abdominal imaging is performed. Maintaining sufficient distance from the patient is also particularly effective in limiting radiation exposure of the eye lens. Our results suggest that the doses of radiation received by staff during mobile radiography are not significant when appropriate radiation protection is used. To reduce exposure, it is important to maintain a sufficient distance from the patient. Therefore, RTs should bear this is mind during mobile radiography.

    PET検査に従事する看護師4人の水晶体被ばく線量を評価した。水晶体用線量計DOSIRISとガラスバッジによる水晶体被ばく線量、従事した検査数、PET薬剤の投与量を調査した。その結果、左眼水晶体近傍にDOSIRISを装着して測定した3mm線量当量から算出した年間水晶体等価線量は0.93±0.13mSv、ガラスバッジでは0.66±0.17mSvであり、DOSIRISとガラスバッジの線量に十分な相関はなかったが、DOSIRISおよびガラスバッジの線量と、看護師の従事件数、薬剤投与量にはそれぞれ相関がみられた。本研究では年間のPET検査従事件数が推定568件であったことより、看護師が線量限度の20mSv/年を超過する可能性は低いと考えられるが、従事件数や薬剤投与量によっては水晶体の線量が高くなることが予想され、看護師のPET検査従事件数を減らすことが有効な水晶被ばく低減策といえる。

    Radiation protection/evaluation during interventional radiology (IVR) poses a very important problem. Although IVR physicians should wear protective aprons, the IVR physician may not tolerate wearing one for long procedures because protective aprons are generally heavy. In fact, orthopedic problems are increasingly reported in IVR physicians due to the strain of wearing heavy protective aprons during IVR. In recent years, non-Pb protective aprons (lighter weight, composite materials) have been developed. Although non-Pb protective aprons are more expensive than Pb protective aprons, the former aprons weigh less. However, whether the protective performance of non-Pb aprons is sufficient in the IVR clinical setting is unclear. This study compared the ability of non-Pb and Pb protective aprons (0.25- and 0.35-mm Pb-equivalents) to protect physicians from scatter radiation in a clinical setting (IVR, cardiac catheterizations, including percutaneous coronary intervention) using an electric personal dosimeter (EPD). For radiation measurements, physicians wore EPDs: One inside a personal protective apron at the chest, and one outside a personal protective apron at the chest. Physician comfort levels in each apron during procedures were also evaluated. As a result, performance (both the shielding effect (98.5%) and comfort (good)) of the non-Pb 0.35-mm-Pb-equivalent protective apron was good in the clinical setting. The radiation-shielding effects of the non-Pb 0.35-mm and Pb 0.35-mm-Pb-equivalent protective aprons were very similar. Therefore, non-Pb 0.35-mm Pb-equivalent protective aprons may be more suitable for providing radiation protection for IVR physicians because the shielding effect and comfort are both good in the clinical IVR setting. As non-Pb protective aprons are nontoxic and weigh less than Pb protective aprons, non-Pb protective aprons will be the preferred type for radiation protection of IVR staff, especially physicians.

    Since the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, we have established an archive system of livestock and wild animals from the surrounding ex-evacuation zone. Wildlife within the alert zone have been exposed to low-dose-rate (LDR) radiation for a long continuous time. In this study, we analysed the morphological characteristics of the testes and in vitro fertilization (IVF) capacity of cryopreserved sperm of racoons from the ex-evacuation zone of the FDNPP accident. The radioactivity of caesium-137 (137 Cs) was measured by gamma-ray spectrometry, and the measured radioactivity concentration was 300-6,630 Bq/kg in the Fukushima raccoons. Notably, normal spermatogenesis was observed in the seminiferous tubules of the testes, with the germinal epithelium composed of a spermatogenic cell lineage with no evident ultrastructural alterations; freeze-thawing sperm penetration ability was confirmed using the interspecific zona pellucida-free mouse oocytes IVF assays. This study revealed that the chronic and LDR radiation exposure associated with the FDNPP accident had no adverse effect on the reproductive characteristics and functions of male raccoons.

    福島原発事故によっておきた環境汚染による被ばくは、これまで知見の少ない領域である。我々は野生動物を用いて生体への影響調査を行っている。行動履歴の不明な野生動物の被ばく線量を推定するために、被ばく量に依存して歯の中に生成、蓄積されるラジカルを電子スピン共鳴(ESR)測定にて測定する手法に着目した。現在、動物の低線量被ばくに適用させるため改良した新手法を開発しており、現段階での測定精度と実際の推定線量被ばく線量について報告する。

    To monitor radiocesium activity in skeletal muscle of live cattle, the animals were given radiocesium-contaminated feed continuously, then switched to contamination-free feed after radiocecium concentration in peripheral blood (PB) reached plateau. Radioactivity in skeletal muscles of neck and rump was measured by attaching the probe of a NaI survey meter closely on the body surface just above the muscle of the live cattle (external measurement). We validated the strong positive correlation between the value of the external measurement and radiocesium activity concentration of dissected muscle (r = 0.89, P < 0.001 for neck; r = 0.80, P < 0.001 for rump). Accumulation of radiocesium both in muscle and PB was proportional to the total amount of radiocesium cattle ingested. However, radioactivity concentration in PB was constant in the cattle that had continuously ingested radiocesium, lower than 2.0 × 105 Bq in total within 67 days from the beginning of radiocesium intake. In addition, the ratio of radiocesium activity in muscle to that in PB was lower during the time when radiocontaminated feed was ingested than that of contamination-free feed ingestion. Using the correlation of radioactivity between muscle and PB, we confirmed that a majority of the cattle in the ex-evacuation zone of the Fukushima Daiichi nuclear power plant accident, from 167 to 365 days after the accident occurred, were in the declining period of radiocesium intake.

    MicroRNA (miRNA) is a non-coding RNA involved in regulating both cancer gene promotion and suppression. We investigated the role of miRNA in inducing radiation resistance in cancer cell lines using clinically relevant radioresistant (CRR) cells. Analysis using miRNA arrays and qPCR revealed that miR-7-5p is highly expressed in all examined CRR cells. Additionally, CRR cells lose their radioresistance when daily irradiation is interrupted for over 6 months. MiR-7-5p expression is reduced in these cells, and treating CRR cells with a miR-7-5p inhibitor leads to a loss of resistance to irradiation. Conversely, overexpression of miR-7-5p in CRR cells using a miR-7-5p mimic shows further resistance to radiation. Overexpression of miR-7-5p in parent cells also results in resistance to radiation. These results indicate that miR-7-5p may control radioresistance in various cancer cells at the clinically relevant dose of irradiation. Furthermore, miR-7-5p downregulates mitoferrin and reduces Fe2+, which influences ferroptosis. Our findings have great potential not only for examining radiation resistance prior to treatment but also for providing new therapeutic agents for treatment-resistant cancers.

    Several populations of wild Japanese macaques (Macaca fuscata) inhabit the area around Fukushima Daiichi Nuclear Power Plant (FNPP). To measure and control the size of these populations, macaques are captured annually. Between May 2013 and December 2014, we performed a haematological analysis of Japanese macaques captured within a 40-km radius of FNPP, the location of a nuclear disaster two years post-accident. The dose-rate of radiocaesium was estimated using the ERICA Tool. The median internal dose-rate was 7.6 μGy/day (ranging from 1.8 to 219 μGy/day) and the external dose-rate was 13.9 μGy/day (ranging from 6.7 to 35.1 μGy/day). We performed multiple regression analyses to estimate the dose-rate effects on haematological values in peripheral blood and bone marrow. The white blood cell and platelet counts showed an inverse correlation with the internal dose-rate in mature macaques. Furthermore, the myeloid cell, megakaryocyte, and haematopoietic cell counts were inversely correlated and the occupancy of adipose tissue was positively correlated with internal dose-rate in femoral bone marrow of mature macaques. These relationships suggest that persistent whole body exposure to low-dose-rate radiation affects haematopoiesis in Japanese macaques.

    Purpose To understand the molecular mechanisms underlying cancer cell radioresistance, clinically relevant radioresistant (CRR) cells that continue to proliferate during exposure to 2 Gy/day X-rays for more than 30 days were established. A modified high-density survival assay for anticancer drug screening revealed that CRR cells were resistant to an antimicrotubule agent, docetaxel (DTX). The involvement of reactive oxygen species (ROS) from mitochondria (mtROS) in the cross-resistance to X-rays and DTX was studied.<br /> Methods and Materials Sensitivity to anticancer agents was determined by a modified high-density cell survival or water-soluble tetrazolium salt assay. DTX-induced mtROS generation was determined by MitoSOX red staining. JC-1 staining was used to visualize mitochondrial membrane potential. DTX-induced DNA double-strand breaks were determined by γ-H2AX staining. To obtain mitochondrial DNA-lacking (ρ0) cells, the cells were cultured for 3 to 4 weeks in medium containing ethidium bromide.<br /> Results Treatment with DTX increased mtROS in parental cells but not in CRR cells. DTX induced DNA double-strand breaks in parental cells. The mitochondrial membrane potential of CRR cells was lower in CRR cells than in parental cells. Depletion of mtDNA induced DTX resistance in parental cells. Treatment with dimethyl sulfoxide also induced DTX resistance in parental cells.<br /> Conclusions The mitochondrial dysfunction observed in CRR cells contributes to X-ray and DTX cross-resistance. The activation of oxidative phosphorylation in CRR cells may represent an effective approach to overcome radioresistant cancers. In general, the overexpression of β-tubulin or multidrug efflux pumps is thought to be involved in DTX resistance. In the present study, we discovered another DTX resistant mechanism by investigating CRR cells.

    東京電力福島第一原子力発電所事故により福島県内では多くの野生動物が被災した。我々は福島県内で被災した野生ニホンザルに着目し、2013年より現在までに300頭以上から様々な臓器、筋肉などのサンプリングを行っている。採取した18の主要臓器と抹消血の放射性セシウム濃度を測定した結果、体内における放射性セシウム濃度は骨格筋が最も高く、甲状腺が最も低いことが明らかとなった。筋肉中放射性セシウム濃度と抹消血球像との間に明確な相関はみられなかったが、成獣の骨髄中顆粒球系細胞は筋肉中放射性セシウム濃度と負の相関を示し、脂肪細胞割合は筋肉中放射性セシウム濃度と正の相関を示した。これらの結果は、低線量慢性被ばくによる造血機能の低下を示唆するが、骨髄細胞の解析結果が放射線による影響であるかを結論付けるためには今後も解析を継続して行う必要がある

    放射線被ばくによる健康影響は,放射線により誘発されるDNA障害がその原因となって起こる.一方で,細胞には,こうしたDNA障害を修復する能力が普段から備わっており,生成されたDNA損傷を随時修復して排除している.しかしながら,被ばく線量に直線的に依存してDNA障害の数が増加すると,修復の能力を超えたり,あるいは誤った修復をする可能性が出てくる.最近,特に,100mGy(Sv)程度の低線量放射線被ばくによる健康影響について議論される機会が増えているが,疫学調査ではその技術的限界から,科学的に明確な結論を得ることが困難で,このことが,東京電力福島第一原発事故後の放射線に対する人々の不安にもつながっている.そこで,100mGyを下回るような低線量被ばくによるDNA障害の誘発と修復について,科学的に証明された事実を取得することを目的に本研究を計画した.

    水晶体の職業被ばくが生じるインターベンショナル・ラジオロジー(IVR)に従事する医療スタッフの水晶体被ばく線量に関するデータをある程度収集することができた。 野生ニホンザルから採取した水晶体中の酸化ストレス解析方法の改良に取り組んだ。本解析では試料中のマロンジアルデヒド(MDA)とチオバルビツール酸の付加体を形成させて波長480から700 nmまでの範囲を1nmずつ吸光度を測定して得られるスペクトルを解析した。535 nm付近でのみ示される吸収のピークとその他の波長範囲の水平なベースラインの2成分が得られるので、535nmのピーク高から検量線を用いてMDA濃度が定量できる。しかしながら、組織試料解析では、ベースライン部分に起伏が生じて平坦なベースラインが得られないため、535nmのピーク高を正確に取得できずにMDAを評価できない課題が生じた。試料調整前に血液を取り除くことで解析可能なスペクトルが得られるようになった。また、血液除去過程が不十分であるとMDA濃度の正確性に影響を及ぼすことも分かった。本年度は組織試料を用いてMDA濃度を解析するためのプロトコールを確立し、従来法では定量が困難であった個体試料の評価できるようになったこと、従来よりも誤差範囲を小さくできるために低線量率被ばくによるわずかな変化を評価できる解析手法を確立した。 p53が機能するヒト水晶体上皮細胞の樹立するために、白内障手術時に切除される前のう片に接着しているヒト水晶体上皮細胞の初代培養を行った。研究分担者が開発した不死化ベクターは、従来の不死化ベクターと異なり遺伝子の特異的な領域に不死化遺伝子を選択的に導入することが可能であるため、水晶体上皮細胞の特徴が保持された不死化ヒト水晶体上皮細胞株(iHLEC-NY1)を作製することに成功した。

    放射線晩発障害の発症リスクを被ばく後の早期に予測できれば、予防的介入(治療)も可能となり、晩発障害の発症を抑えることが可能となるが、晩発障害のほとんどの疾患は酸化ストレス(活性酸素種(ROS)の増加や抗酸化能の低下によって引き起こされる状態のこと)との関連が明らかになっている。本研究では、抗酸化能を指標とした晩発障害リスク予測の妥当性を明らかにすることを目的としている。初年度の研究実績については、 ①低線量低線量率照射装置を用いてマウスに慢性照射(33 μGy/min~12.5 mGy/hr; 1~60日間;総線量0.1~5 Gy)を行い、照射後からマウスが死亡するまで経時的に採血し、抗酸化能関連指標(血液抗酸化能(i-STrap法による測定)、グルタチオン量など)や酸化ストレスの原因となる慢性炎症関連サイトカイン(IL-6、TNF、IL-1β、MCP-1など)の測定を行うことを目標にしており、現在マウス飼育室と照射の準備を完了し、順次購入されたマウスに照射を行い飼育観察を続けている。 ②福島県で害獣として処分された野生ニホンザルから採血し、抗酸化能関連指標(血液抗酸化能、グルタチオン量など)や酸化ストレスの原因となる慢性炎症関連サイトカイン(IL-6、TNF、IL-1β、MCP-1など)の解析を行い、生息地や体内の放射線量からサルの被ばく線量を割り出し、各指標と被ばく線量、年齢、性別などの関係を明らかにすることを目標にしており、現在冬場に捕獲された野生ニホンザルからの血液のストックを行っている。

    本研究は低濃度で持続的なトリチウム曝露による細胞影響を明らかにすることで、トリチウム影響に関して不足している科学的知見を集積するために実施した。特に内部被ばくの長期化と関連する有機結合型トリチウム(OBT)による影響について、トリチウム水による影響との比較を行なっている。本年度は特に、従来よりも低濃度トリチウム処理を行なった際の細胞内取り込みについて検討を行なった。 細胞内への取り込みについては、培地中にトリチウムを添加して24時間後以降から継日的に培地中と細胞中のトリチウムを液体シンチレーションカウンタで測定した。まず、従来よりも低い濃度(7.4 kBq/ml)のトリチウム水を処理したところ、トリチウム添加24時間後までに増加していた細胞内トリチウムは、その後、添加7日後までに著しい変化はみられなかった。OBTとしてトリチウムグルコースやトリチウムチミジンをトリチウム水と同じ濃度で処理したところ、それぞれ添加後2日目と6日目に細胞内トリチウム濃度が最大となった。トリチウム水や今回使用したOBTは添加量の9割以上が培地内に残っており、細胞内への取り込みはトリチウム水とトリチウムグルコースで1%以下、トリチウムチミジンで4 %程度であることがわかった。また、細胞核と細胞質の分画を抽出してそれぞれの分画中のトリチウムを測定すると、トリチウム水では細胞核と細胞質にほぼ同程度のトリチウムが分布していた一方で、OBTではその化学形態に依存して細胞内に局在していることが示された。従来のOBTを用いた細胞影響では処理濃度で細胞影響を比較することが多かったが、本年度得られた結果をもとに線量評価を行なうことで、トリチウム水とOBTによる生物影響を同一の指標を用いて評価するための基礎的知見として利用することが期待される。

    We here investigated if low dose /low dose rate radiation exposure elicits delayed instability as high dose radiation exposure did. We established the immortalized cells from femoral muscle of wild Japanese Macaque living in ex-evacuation zone. Compared with the immortalized cells established from the muscle of control Japanese Macaque, delayed effects shown as induction of DNA double-strand breaks or global methylation did not appear even in the cells made from exposed Japanese Macaque. Based on the system we here established, investigation using tissue other than muscle is necessary to comprehensively discuss radiation effect among different organs.

    We analyzed the role of mitochondria (mt) on the phenotype expression of CRR cells that continue to proliferate under fractionated irradiation of 2 Gy/day x-rays. Reactive oxygen species (ROS) derived from mt was detected in the parent cells after acute irradiation with 10 Gy, but was not detected in CRR cells, and the mitDNA copy number and ATP amount were decreased. Using ethidium bromide, mtDNA deleted ρ0 cells could be established from parental ells but was impossible from CRR cells. Only one ρ0 cells could be established from HepG2-CRR to which maintenance irradiation paused. Expression of the antioxidant enzyme group in ρ0 cells showed no specific tendency compared to the parental cells but was rather sensitive to H202. It was suggested that the influx of H 2O2 is accelerated by the decrease in cell membrane potential of ρ0 cells.

    We have constructed an archive system of organs affected by the Fukushima Daiichi Nuclear Power Plant accident and obtained the following results. Two years after the accident, almost only radioactive cesium was detected in all organs examined. Oxidative stress related molecules in cattle plasma were correlated with internal dose-rate. DNA double strand breaks in cattle lymphocytes were higher than non-affected controls. Large Japanese field mice were significantly exposed to both gamma- and beta-rays. The turnover of their spermatogenesis was accelerated. However, no abnormalities was detected in mature sperms including cattle. Bone marrow of wild Japanese macaques tended to be hypoplastic, but no abnormality was found in peripheral blood cells. Although no significant changes have been observed, internal dose rate in monkeys is still high. Therefore, it is necessary to continue detailed study to know biological effects of long-term exposure to low dose-rate radiation.

    Recently, the cancer stem cell (CSC) is suggested to be resistant against therapy and recurrence. The fraction of CSCs was higher in clinically relevant radioresistant cells than in their parental cells. However, a long-term low dose fractionated radiation resulted in radioresistance of parental cells but did not increase CSCs fraction. We therefore conclude that the CSC phenotype is not involved in the acquisition of radioresistance of cancer cells.

    We investigated the inducible radioresistant mechanism that cancer cells are able to proliferate under 2 Gy/day of fractionated radiation exposure, so called clinically-relevant radioresistant phenotype (CRR). For CRR formation, we revealed that pre-exposure with less than 2 Gy/day of fractionated dose and stepwise dose-escalation was critically required prior to 2 Gy/day of fractionated exposure. Pre-exposure with lower fractionated dose brought radioresistant phenotype (acquired radioresistance, ARR) and dose-escalation gradually enhanced radioresisntace. We also found that the nature of cancer stem cell or epithelial-mesenchymal transition, which is generally known as the factors contributing acquired radioresistance, was not involved in CRR formation. We also found less DNA damage, especially DNA double strand breaks, in radioresistant cells and that suppression of DNA-damage response might be involved in the mechanism of ARR and/or CRR formation.

    DNA double-strand breaks trigger activation of signal transduction pathways including induction of cell death. Recently, it is suggested that the amplification of DNA damage signaling could be related to the effective cell death induction. In this study, we revealed that the amplification of DNA-damage signaling was suppressed in tumor cell lines.We also demonstrated cell cycle phase-dependency in the amplification of DNA-damage signaling.

    Live-cell imaging of mammary carcinoma cell line revealed prolonged cell cycle arrest as a predominant cell death mode induced by ionizing radiation. Regardless cell cycle phase that exposed to ionizing radiation, prolonged cell cycle arrest was induced in G1 phase. Particularly, direct cell cycle progression from G2 to G1 phase without mitosis was observed by G2-irradiation in presence of p53, suggesting that radiochemotherapy combined with taxane-treatment increases the treatment-sensitivity for p53-depleted carcinoma cells.
