    ABSTRACT Cyclic diaryliodonium salts are widely employed as linchpins for constructing functionalized biaryls and as precursors to arynes under basic conditions. However, their synthetically practical engagement with strongly nucleophilic organometallic reagents has remained largely unexplored. Here we disclose a reaction between these salts and aryl Grignard reagents that delivers iodine‐capped teraryls with predominant meta ‐selectivity. In contrast to ligand coupling at iodine(III) or direct base‐induced aryne formation, the present transformation is initiated by the formation of a cyclic triaryliodane, which serves as both an aryne precursor and a dynamic aryl reservoir within a self‐propagating carbomagnesiation sequence. Spectroscopic analyses and control experiments support the involvement of the triaryliodane and reversible aryl exchange with aryl Grignard reagents, while trapping experiments intercept both aryne and terarylmagnesium intermediates. The reaction tolerates sterically and electronically diverse aryl Grignard reagents with consistent meta ‐selectivity, whereas strongly perturbing substituents on the iodonium salts can modulate the regioselectivity. The resulting iodo‐teraryls retain a versatile C─I bond amenable to further π‐extension or recyclization into cyclic iodonium architectures.

    Cyclic diaryliodonium salts are widely employed as linchpins for the synthesis of functionalized biaryls through ring-opening reactions and as precursors to arynes under basic conditions. Here we uncover a mechanistically distinctive reactivity mode in which aryne generation and arylation are merged into a single, self-propagating sequence mediated by aryl Grignard reagents. The transformation proceeds through a transient cyclic triaryliodane intermediate that undergoes aryne formation and subsequent carbomagnesiation in a self-sustaining cycle. The mechanism, involving key aryne and arylmagnesium intermediates, is corroborated by trapping experiments with both arynophiles and electrophiles. This reaction proceeds with high meta-selectivity for unsubstituted and remotely substituted substrates, while substitution proximal to the aryne moiety can alter the outcome. The process efficiently furnishes iodine-capped teraryl frameworks in high yields across a broad range of aryl Grignard reagents. The transformable C–I bond further renders the products versatile scaffolds for rapid elaboration into complex aromatic architectures.

    Abstract Axially chiral bulky arylbenziodoxol(on)e derivatives were synthesized, among which the one based on a benzoic acid skeleton exhibited markedly lower atropisomeric stability than the previously reported bis(trifluoromethyl) analogs. Experimental and computational studies suggest that the higher basicity of the benziodoxolone oxygen renders it susceptible to protic species, leading to conformational lability.

    Abstract γ‐Amino alcohols are essential motifs in bioactive compounds and chiral catalysts, yet the synthesis of their conformationally constrained variants remains challenging due to the lack of suitable methodologies. Here, we report a formal cyclopropylation of imines with cyclopropanols, enabling the construction of previously inaccessible cyclopropane‐embedded γ‐amino alcohols. This transformation leverages the unique reactivity of enolized zinc homoenolates, which effectively act as a β‐hydroxycyclopropyl anions and engage imines through a sequence of Mannich addition and ring closure. The key to this reactivity lies in the use of bulky N‐heterocyclic carbene (NHC) ligands, which promote efficient coupling with N‐sulfonyl aldimines as well as chiral N‐sulfinyl trifluoromethyl‐ketimines while ensuring excellent diastereocontrol over three contiguous stereocenters. Furthermore, the resulting γ‐amino alcohols can be transformed into β‐ or γ‐aminofunctionalized ketones via homoenolate or β‐keto radical intermediates, offering versatile platforms for downstream derivatization.

    A stereoselective hydroxyallylation reaction of cyclopropenes with cyclopropanols is achieved under zinc-mediated conditions, affording densely functionalized cyclopropanes with excellent diastereocontrol over three contiguous stereocenters within and outside the cyclopropane ring. A racemic variant of the reaction is synergistically promoted by catalytic N-heterocyclic carbene (NHC) and organic base (DBU), whereas chiral amino alcohol-derived bifunctional NHC enables a catalytic enantioselective variant. Mechanistically, the reaction features transient generation of enolized zinc homoenolate via ring-opening of zinc cyclopropoxide and enolization of the resulting homoenolate, followed by its addition to the cyclopropene as a prochiral allylzinc nucleophile.

    We report here on the facile synthesis of amino- and alkoxy-λ3-iodanes supported by a benziodoxole (BX) template and their use as arynophiles. The amino- and alkoxy-BX derivatives can be readily synthesized by reacting the respective amines or alcohols with chlorobenziodoxole in the presence of a suitable base. Unlike previously known nitrogen- and oxygen-bound iodane compounds, which have primarily been employed as electrophilic group transfer agents or oxidants, the present amino- and alkoxy-BX reagents manifest themselves as nucleophilic amino and alkoxy transfer agents toward arynes. This reactivity leads to the aryne insertion into the N–I(III) or O–I(III) bond to afford ortho-amino- and ortho-alkoxy-arylbenziodoxoles, iodane compounds nontrivial to procure by existing methods. The BX group in these insertion products exhibits excellent leaving group ability, enabling diverse downstream transformations.

    Organic iodine(III) compounds represent the most widely used hypervalent halogen compounds in organic synthesis, where they usually perform the role of an oxidant or electrophile to functionalize electron-rich or nucleophilic organic compounds. In contrast to this typical reaction mode and the nature of organohalogen compounds in general, we discovered a novel reactivity of organoiodine(III) compounds that behave as organometallic-like nucleophiles toward arynes. Possessing diverse transferable ligands and being supported by a cyclic spectator ligand, the organoiodine(III) compounds undergo addition across the electrophilic C–C triple bond of arynes while retaining the trivalency of the iodine center. This carboiodanation reaction can forge a variety of aryl–alkynyl, aryl–alkenyl, and aryl–(hetero)aryl bonds along with the concurrent formation of an aryl–iodine(III) bond under mild conditions. The newly formed aryl–iodine(III) bond serves as a versatile linchpin for downstream transformations, particularly as an electrophilic reaction site. The amphoteric nature of the iodine(III) group as a metalloid and a leaving group in this synthetic sequence enables the flexible and expedient synthesis of extended π-conjugated molecules and privileged biarylphosphine ligands, where all the iodine(III)-containing compounds can be handled as air- and thermally stable materials.

    The development of chelation-assisted C-H functionalization reactions has long been driven by noble transition metal catalysts, despite the obvious economic benefits of using earth-abundant transition metals. In this context, seminal reports in the literature suggest the potential of cobalt complexes as catalysts for C-H functionalization. This account summarizes our studies on the development of a series of cobalt-based catalysts for chelation-assisted C-H functionalization reactions such as addition of C-H bonds across carbon-carbon multiple bonds (hydroarylation), and coupling between C-H bonds and organic electrophiles. These studies have demonstrated that cobalt catalysts not only serve as mild and cost-effective alternatives to noble metal catalysts for existing C-H functionalizations, but also enable hitherto unknown or difficult transformations, including branched-selective hydroarylation of styrenes and ortho-alkylation using unactivated secondary alkyl halides.

    C-H結合の直接的かつサイト選択的な官能基化は有機分子の構造的複雑化や多様化を迅速に行う手法として理想的であり，複雑分子の合成や医薬品のリード化合物探索を加速する．本研究では，精密に分子設計された遷移金属と典型金属ルイス酸の複合触媒を用い，医薬品分子において重要な部分構造をなす含窒素複素環について，高度な反応サイト制御を伴ったC-H官能基化手法を創出することを目指している．本年度は，コバルトとルイス酸が空間的に近接した複合触媒の設計に基づくC-H結合活性化反応の探索を中心に検討を行った．具体的には，第三級ホスフィンと第二級ホスフィンオキシドが近接配置された二官能性配位子を種々合成し，それにコバルト触媒とアルミニウムルイス酸を組み合わせた触媒を用いて，複素環C-H結合のアルキンによるアルケニル化反応を検討した．その結果，二官能性配位子はピリジンやピリドンなどの複素環について，ルイス塩基性の窒素または酸素近傍のC-H結合に対して選択的にアルケニル化反応を促進することを見出した．この選択性は，以前に見出した，個別のコバルト触媒とアルミニウム触媒を用いた反応におけるサイト選択性と全く異なるものであり，複合触媒の分子設計の有効性を実証するものであると考えられる．さらに，以前の触媒系ではサイト選択的な結合活性化ができず複雑な混合物を与えたイミダゾピリジン類の反応においても，窒素近傍のC-H結合が選択的かつ効率よく変換されることが分かった．現在，より高活性な触媒の探索を目指して，配位子のリン上の置換基や骨格の修飾を鋭意検討しているところである．また，別のコバルト・ルイス酸複合触媒として，コバルト－二座ホスフィン触媒と亜鉛ルイス酸触媒からなる触媒系が，ニトリル化合物の炭素－シアノ（C-C）結合活性化を経るアルキンのカルボシアノ化およびヒドロシアノ化反応を促進することを明らかにした．

    本研究では,安価で豊富な鉄触媒による炭素-水素結合の切断を経る新規高効率炭素-炭素結合生成反応を開発し,革新的有機合成プロセスとして確立することを目的としている.今年度は,鉄触媒とグリニャール反応剤による脂肪族アミン類のα位炭素-水素結合の切断を経る炭素-炭素結合生成反応を開発した.すなわち,窒素原子上に2-ヨードベンジル基を有する脂肪族3級アミンに対して,触媒量の鉄(III)アセチルアセトナートなどの鉄塩の存在下,グリニャール反応剤を作用させると,ヨードベンジル基の脱ヨウ素化を伴ってアミンのα位にグリニャール反応剤の有機基が効率的に導入されることを見出した.本反応は環状および鎖状アミンに適用でき,また,アリール基・アルケニル基・アルキル基など種々の有機基の導入に用いることができた.重水素ラベル化実験による反応機構の検討の結果,本反応は次のような経路で進むことが示唆された.すなわち,低原子価鉄活性種からヨードベンジル基への一電子移動により生成したアリールラジカルが分子内での1,5-水素移動によりαアミノラジカルへと転位し,これが鉄を介してグリニャール反応剤とカップリングすることにより生成物を与える.α位に置換基を有する脂肪族アミン類は生理活性物質に多く見られる構造であり,その効率的合成法の開発は有機合成上の重要な課題である.しかしながら,単純なアミンのα位炭素-水素結合を直接的に炭素-炭素結合へと変換する手法は少ないうえに,アミンの適用範囲および導入できる有機基の範囲は狭いものであった.今回開発した鉄触媒反応は,こうした化合物を合成するうえで有用な手法となるものと期待される.

    本研究では,不活性炭素-ハロゲンおよび擬ハロゲン結合の活性化における遷移金属と典型金属の協奏作用機構に関する研究と,それに基づく新規触媒系の開発を行っている.今年度は,ニッケル触媒によるフェノール誘導体とグリニャール反応剤のクロスカップリング反応およびフッ化アリールと有機亜鉛反応剤のクロスカップリング反応の開発を行った.まず,ニッケル錯体とヒドロキシホスフィン配位子からなる触媒が.アリールカルバメートやアリールホスフェートなどのフェノール誘導体とグリニャール反応剤のクロスカップリング反応を著しく加速することを見出した.これらのフェノール誘導体は,現在クロスカップリング反応に広く用いられているトリフラートに比べて安価かつ安定であるにもかかわらず,反応性に乏しいため利用例は数少なかった.我々の触媒は,これらの反応基質の有効利用に新たな道を開くものと言える.つづいて,ニッケル触媒によるフッ化アリールと有機亜鉛反応剤のクロスカップリング反応が.新たに開発した3座配位子であるヒドロキシビスホスフィンによって促進されることを見出した.本触媒系の適用範囲は広く,モノフルオロアレーンからパーフルオロアレーンに至るまで種々のフルオロアレーンに適用でき,また種々のアリール基,アルキル基を有する亜鉛反応剤を用いることができた.炭素-フッ素結合は最も強力な化学結合のひとつであり,その活性化は有機金属化学・分子触媒化学における重要な基礎的課題であったが.我々の知る限り,モノフルオロアレーンからパーフルオロアレーンまで適用可能な触媒系はこれが初めてである.また,部分的にフッ素化された芳香環は医農薬や液晶などの材料に含まれる重要な部分構造であり,今回開発した触媒はこうした分子を効率的に合成する上でも有用な手法となることが期待される.

    本研究では開殻遷移金属錯体を活性種とする触媒的有機合成反応に関する分子理論的検討とその合成化学的展開研究を行っている.具体的には鉄およびその周辺の遷移金属を触媒とする炭素-炭素結合生成反応を開発するとともに,その反応機構に関する分子理論的検討を行っている.今年度は,鉄触媒と有機亜鉛およびマグネシウム反応剤を用いた芳香族炭素-水素結合の直接変換反応の開発を行った.具体的には,前年度に開発した鉄触媒とアリール亜鉛反応剤によるアリールピリジンおよびアリールイミンの直接アリール化反応を,より実用的なものとするべく検討を行った.第一の課題は,酸化剤として用いていた高価なハロゲン化物である.これをより安価な酸化剤,望ましくは理想的な酸化剤である酸素に置き換えるべく検討を行った.その結果,鉄触媒直接アリール化反応は,酸素の供給量を制御することで効率的に進行することを見出した.第二の課題は,アリール亜鉛反応剤の調製において,大量の亜鉛塩とグリニャール反応剤を必要とし,原子効率が著しく低い点である.そこで,グリニャール反応剤のみを用いた直接アリール化反応を検討した.その結果,配位子,溶媒,グリニャール反応剤の滴下方法などを制御することにより,定量的な直接アリール化を実現した.特筆すべきことに,グリニャール反応剤を用いた反応は亜鉛反応剤のそれに比べて著しく速く,0度という温和な条件下,数分で完結するというものであった.以上得られた知見をさらに展開していくことにより,鉄錯体の新たな反応性が明らかになることが期待される.

    The present research aimed at mechanistic understanding-driven design of organic synthetic reactions. Thus, we developed highly efficient, stereoselective and functional group-tolerant carbon-carbon bond formation reactions, taking advantage of the reactivity of group 8-13 transition metals and the understanding of their multimetallic cooperation. Fully utilizing element diversity and multimetallic cooperation, we developed synthetic methods of high efficiency, high selectivity and sustainability, and applied these methods for the creation of novelπ-electronic materials for organoelectronic devices.

    本研究では,求核性およびルイス酸性という基本的な触媒機能を原子・分子レベルでの設計により複合化することで,両機能の協奏的発現を可能とし,従来の単一機能触媒を凌駕する高性能触媒を創出することを目的としている.本年度は,炭素-ハロゲン結合および炭素-酸素結合の活性化について反応機構および合成化学検討を行い,以下にまとめる成果を挙げることができた. (1)NiおよびPd触媒クロスカップリング反応におけるハロゲン化アリールの活性化機構:ハロゲン化アリールの低原子価遷移金属への酸化的付加は,触媒的クロスカップリング反応に含まれる重要な素反応であり,これまで錯体化学的な検討が精力的に行われてきた.一方で,実際の触媒反応の条件下における反応機構研究はほとんど行われていない.今回,ニッケルおよびパラジウム触媒によるハロゲン化アリールとグリニャール試薬のカップリング(熊田カップリング)における酸化的付加の機構について,速度論的同位体効果(KIE)を用いて検討した.その結果,酸化的付加の段階に金属と配位子の及ぼす影響を明らかにし,炭素-ハロゲン結合切断の遷移状態に関する知見を得た. (2)フェノール誘導体の高効率熊田カップリング反応:芳香族トリフラートは,反応性の高い擬ハロゲン化物としてクロスカップリング反応に汎用されるが,加水分解に弱く,かつ高価であるなどの欠点がある.そのため,トリフラートをより安価かつ安定なフェノール誘導体に置き換える試みが精力的に行われている.今回,ヒドロキシホスフィン配位子が芳香族カルバメートおよびホスフェートの熊田カップリングを著しく促進することを見出した.カルバメート,ホスフェートいずれの場合においても,以前報告されたNi(acac)_2のみを用いる条件に比べて触媒活性及び選択性の面で格段の向上が実現できた.

    近年,環境に対する配慮から有機溶媒の代わりに水を溶媒として用いる有機合成反応の開発が注目を集めている.また,単なる溶媒としてではなく,極性,水素結合,疎水性相互作用といった水の特質を活かした反応系の構築は,化学的にも興味深い課題である.以上の背景および,我々がこれまで行ってきた有機銅化合物の反応化学に関する理論及び実験研究を基盤として,本研究では水中での遷移金属触媒反応の機構の解明と,それに基づく新規触媒系の合理設計を目的とした.以下に本年度の研究成果をまとめる. (1)銅触媒によるアセチレン二量化の機構:銅触媒による水中でのアセチレン二量化反応の機構について理論的検討を行い,銅ビニリデン錯体を活性種とする新しい反応機構を見出した.触媒サイクル全般を通して,水分子は反応促進に重要な寄与をしていることが分かった.計算によって得られた活性化パラメータは,既知の実験データと概ね良好な一致を示した.副生成物として得られるアセトアルデヒドの生成機構についても,ビニリデン錯体の水和を経る反応経路を明らかにすることができた. (2)有機銅試薬によアシル求電子剤の置換反応の機構:有機銅試薬によるチオエステルおよび酸塩化物の置換反応の機構について理論及び実験的検討を行った.密度汎関数計算による反応経路の検討の結果,チオエステルと酸塩化物の反応は全く異なる機構で進行することが明らかとなった.チオエステルの反応が,銅のC-S結合に対する挿入を経て進行するのに対し,酸塩化物の反応では,C-C1結合は銅と相互作用することなく脱離し,リチウムカチオンによって捕捉される.また,実際の置換反応について12C/13C速度論的同位体効果(KIE)の測定を行い,計算結果とよく一致することを明らかにした.

    本研究課題では,遷移金属と典型金属の複合触媒反応系として典型的な,d10遷移金属によるハロゲン化アリール・アルケニルと有機典型金属試薬によるクロスカップリング反応に焦点を当て,理論的及び実験的検討を行っている.本年度は,初年度及び次年度の基礎的な反応機構研究で得られた知見をもとに,クロスカップリング反応の合理的設計を目的として研究を行った. 反応機構研究で得られた基礎的知見とは次の通りである.従来,Cu(I),Ni(O),Pd(O)などのd10金属に対するハロゲン化アリール・アルケニルの酸化的付加は,金属が炭素ハロゲン結合に挿入する,「3中心型遷移状態」を経て進行するとされてきた.ここで筆者は,リチウム塩やマグネシウム塩などのルイス酸性を有する典型金属化合物が共存する場合,酸化的付加の反応経路としてd10遷移金属と典型金属ルイス酸が同時かつ協働的に関与する「脱離型遷移状態」が存在することを新たに発見した. 今年度は,上記の「遷移金属と典型金属による協働的なsp2炭素ハロゲン結合活性化」という新規概念をもとに反応設計開発を行った.ここで取り組んだのは,炭素フッ素結合の活性化を経る炭素炭素結合生成反応である.具体的には,ニッケル触媒による,活性化されていないフッ化アリールとグリニャール試薬のクロスカップリング反応の検討を行った.種々のホスフィン配位子の検討を行った結果,リン原子近傍にヒドロキシ基などのプロトン性基を有する配位子がフッ化アリールとアリールグリニャール試薬のクロスカップリング反応に高活性を示すことを見出した.本反応は特筆すべきことに,sp2炭素ヘテロ元素(酸素,硫黄)結合共存下において炭素フッ素結合活性化を選択的に起こすことが分かった.
