Background Oral mucositis is a frequent adverse effect of chemotherapy that significantly affects patients’ quality of life and treatment continuity. Despite its clinical relevance, effective preventive strategies remain limited. This study aims to compare the efficacy and safety of betamethasone mouthwash with those of sodium gualenate hydrate mouthwash for preventing chemotherapy-induced oral mucositis. Methods This Phase II, multicenter, open-label, randomized controlled trial aims to enroll 296 adult patients (≥18 years) undergoing chemotherapy for solid tumors (excluding head and neck and hematological cancers). Participants will be randomized 1:1 into two arms: a betamethasone mouthwash (intervention) group and a sodium gualenate hydrate mouthwash (control) group. The primary outcome is the prevention of Grade 1 oral mucositis, assessed using the Common Terminology Criteria for Adverse Events v3.0 and v5.0. Secondary outcomes include prevention of Grade 2–3 oral mucositis, the incidence of oral candidiasis, and chemotherapy completion rates. Data will be analyzed using Kaplan–Meier survival curves, log-rank tests, and Cox regression models. Expected Outcomes This study aims to establish betamethasone mouthwash as an effective preventive strategy for potentially enhancing the quality of life of patients with cancer. Trial registration This study was registered with the Japan Registry of Clinical Trials on September 20, 2024 (jRCTs071240060).

Abstract Multinucleated giant cells like osteoclasts (OCs) often contain >10 nuclei. This raises the question of whether nuclei residing in the same cytoplasm are all regulated similarly or whether they are heterogeneous, expressing a different set of genes regulated by nucleus-specific epigenetic mechanisms. We combined imaging of living cells with confocal microscope, picking of intracellular components inside a single cell using the Single Cellome™ System SS2000 (Yokogawa, Japan), allowing next generation sequencing at high resolution for mRNA (iscRNA-seq) and for DNA methylation (iscWGBS; whole genome bisulfite sequencing) of any cellular components including a single OC nucleus. Our results reveal that individual nuclei within the same cell are heterogeneous in terms of gene expression and epigenetic regulation, possibly affecting regional cell function.

One of the important cell lines derived from bone marrow is the MSC. In recent years, markers of MSC isolation have been reported, but they are still heterogeneous cell populations. Therefore, it is difficult to accurately evaluate the MSC characteristics. In this study, scRNA-seq analysis was used to identify seven genetically distinct subpopulations of mouse bone marrow MSCs. ATAC-seq analysis also revealed epigenetic landscapes, genetic similarities and functional characteristics. These results elucidate stem cell niche signals in the bone marrow microenvironment, reconstitute bone marrow in vitro, and are potentially important for subpopulations identified in various clinical applications in the treatment of bone and bone marrow-related disorders. It made it possible to shed light on the role.

The author examined to isolate mesenchymal cell populations by analyzing MSCs at the single cell level. The MSCs were analyzed by scRNA-Seq, and seven specific clusters were confirmed from the gene expression profile. In addition, each cluster were confirmed that they could be separated using FACS from the gene expression profile. Furthermore, author revealed that seven clusters were independent cell populations by RNA-Seq and ATAC-seq analysis. Next, author performed in vitro characterization of these clusters. As a result, aouthor found that some of the clusters had a function closely stem cells. Author aim to identify stem cell-like cell populations by direct comparison with ES cells in the future.

In this study, we will elucidate stemness signals of MSCs induced by hematopoietic stem cells and aim to establish a method to achieve the promotion and maintenance of bone formation by utilizing signal related molecules in recombinant human (rh) BMP-2 induced regenerated bone. In addition, we aim at application to bone regeneration by realizing functional bone regeneration that can maintain bone volume and bone quality for a long time. Eventually, we studied with the aim of contributory to develop oral surgery and regenerative medicine. From the results of in vivo and in vitro studies, we indicated in order to maintain stem cell properties, mobilize and establish bone marrow cells, persistent release of stemness factor is required. Therefore, we suggest that be necessary to study the continuing release of the factor in the future.

We examined the factors associated in immune tolerance by cytokine assay. However, ASC isolated by FACS was remained an issue, as these cells were heterogeneity cell population. Therefore, we verified whether there are cells involved in immune tolerance in the population using single cell RNA-Seq. As a result, ASC isolated by the conventionally method revealed that were consists of ten populations from genetic similarity. Furthermore, through global gene analysis, expression of a genes related in an immune tolerance were upregulated in a specific cell population X. When this cell population was co-cultured with CD3-positive T cells in LPS-supplemented medium, we confirmed differentiation of CD3 positive T cells to CD4/25/Foxp3 positive Treg was significantly increased. In addition, the signal existing between population X and CD3 positive T cells was analyzed by RNA-Seq. As a result, we identified candidate factors associated in the immune tolerance.
