Dr. Peng Chen is an Associate Professor at the Graduate School of Dentistry, Tohoku University, Japan. He received his Ph.D. through the Waseda–NIMS Joint Graduate Program in 2013 and subsequently joined the Institute of Biomaterials and Bioengineering at Tokyo Medical and Dental University. In 2023, he moved to Tohoku University. His research focuses on bioactive metallic materials, multiscale surface engineering, antibacterial biomaterials, material–cell interactions, and smart biointerfaces for dental and biomedical implants. Outside of research, he enjoys music, swimming, marathon running, and outdoor activities.

    Titanium and its alloys are essential in orthopedic and dental treatments owing to their high strength, corrosion resistance, and superior biocompatibility compared with those of other metals. However, titanium alloys are bioinert. Previous studies have indicated that alkali treatment (AT) is a straightforward method to create a surface oxidization layer on titanium, thereby improving its bioactivity. Herein, alkali-hot water pretreatment was used to enhance the osteoconductivity of titanium and to identify a simple and efficient means of enhancing the interaction between osteoblasts and implants for clinical applications. Commercial pure titanium plates were ground (CP Ti) and subjected to alkali solution and hot water treatments (AWT). Single-process CP Ti specimens were prepared via either AT or hot water treatment (WT). Network-like structural features were observed in the AT specimens and were further refined and densified in the AWT specimens. Water contact angle testing revealed that the hydrophilicity of the titanium specimen (80° for CP Ti) increased by 19° for the AT specimens but decreased by 59° for the AWT specimens. Mouse preosteoblasts (MC3T3-E1 cells) were used for in vitro evaluation. After 24 h of culturing, the number of attached MC3T3-E1 cells on the AWT specimens was 1.5 times larger than that on the CP Ti specimens, suggesting that the alkali-hot water treatment enhanced the initial cell attachment. Cell proliferation evaluation indicated that fewer cells were detected in the AT and AWT specimens compared with those in the CP Ti or WT specimens. However, osteogenic differentiation evaluation on day 10 revealed a 1.5-fold higher alkaline phosphatase expression in cells cultured on the AWT specimens than in cells cultured on the CP Ti specimens. These findings demonstrate the good cytocompatibility and osteoconductivity of AWT Ti, highlighting its benefits in orthopedics and dental treatments.

    An electrochemical surface treatment was developed to visualize the corrosion-inducing inclusions of Zr in chloride environments. Pure Zr and Zr alloy (Zr–0.5O–0.5C) were evaluated in this study. The electrochemical surface treatment consisted of repeated galvanostatic anodic polarization and potentiostatic cathodic polarization. After the electrochemical surface treatment, only one brittle and non-conductive shell of Zr oxide was observed at the corrosion initiation site on the tested surface. The corrosion-inducing inclusions were found inside the corrosion pit under the optimum polarization conditions. At the corrosion initiation site on pure Zr, the area inside the corrosion pit contained larger amounts of O, C, and Si than the surrounding matrix. In the case of the Zr-0.5O–0.5C alloy, relatively larger inclusions were observed after the treatment. Extremely large amounts of Si, together with O and C, were present in the inclusions. The inclusions that induced localized corrosion on the pure Zr and Zr-0.5O–0.5C specimens were found to be precipitated, involving the specific aggregation of Si. The surface treatment developed in this study is expected to be utilized as a powerful tool to elucidate the localized corrosion mechanism of Zr in chloride environments.

    Recently, biofilm formation caused by bacterial adhesion and colonization has been recognized as the major cause of failure in orthopedic and dental implant surgeries. In this study, a customized micro-arc oxidation (MAO) treatment technique was developed to obtain desirable antibacterial properties on Ti surfaces. The two-step MAO treatment was applied in the fabrication of specimens with Ag and with/without Zn in their surface oxide layer. In order to simulate practical usage, surface analyses and immersion tests were performed to evaluate the incorporation of Ag and Zn into the resulting oxide layer and ion release behavior, respectively. Additionally, the antibacterial properties of the specimens after long-term immersion in physiological saline were evaluated using Gram-negative facultative anaerobic bacteria. The MAO-treated specimens containing Ag and Zn exhibited excellent antibacterial properties against Escherichia coli, which were sustained even after 6 months of immersion in physiological saline to simulate practical usage. Moreover, the Ag ions released from the surface oxide indicate the antibacterial properties of the specimen in the early stage, while the release of the corrosion products of Zn demonstrates its antibacterial properties in the later stage.

    Abstract Surface-modified commercially pure titanium (Cp-Ti) with zirconium (Zr)-based thin film metallic glasses (Zr-TFMGs) and ZrO2 thin films were surgically implanted into the tibiae of rats; the bone formation was analyzed to examine the performance of the coatings as a biomaterial. Zr-TFMGs and ZrO2 thin films were coated on Cp-Ti substrates to monitor the control of assimilation in vitro and in vivo. The microstructural and elemental analyses were carried out for the as deposited thin films by x-ray diffraction (XRD), transmission electron microscopy and x-ray photoelectron spectroscopy. TFMG- and ZrO2-coated Ti specimens were immersed in simulated body fluid (SBF) for a period of 21 days to evaluate the calcium phosphate precipitation in vitro. XRD, x-ray photoelectron spectroscopy and scanning electron microscopy/energy dispersive x-ray spectroscopy were used to quantify the mineralization on the coated Zr-TFMG and ZrO2. In vitro corrosion studies showed that the Zr-based TFMG and ZrO2 coatings sustained in the SBF, exhibited superior corrosion resistance to the bare crystalline Ti substrate. Wettability studies showed TFMG and ZrO2 coatings with a hydrophobic nature, and the TFMG-coated SBF-submerged specimens showed a hydrophilic nature. The in vitro cell viability of MC3T3-E1 cells showed good cell proliferation and low cytotoxicity. The calcification deposits were evaluated by staining with alizarin red S, which showed a lower calcium formation on Zr-TFMG compared to ZrO2. The present work also aims to assess the assimilation behavior of Cp-Ti, Zr-TFMG and ZrO2in vivo by inserting the coated specimen in the femur of rats. After post-implantation of 8 weeks, specimens were examined by micro-CT evaluation. The bone contact ratios as calculated were 72.75%, 15.32% and 38.79%. Consequently, the bone affinity was Cp-Ti wire >ZrO2-coated Ti wire >Zr48Cu36Ag8Al8-coated Ti wire.

    This study investigated the time transient effect of zinc (Zn) in the porous titanium dioxide formed by micro-arc oxidation (MAO) treatment routinely performed for Zn-containing electrolytes. The aim of our analysis was to understand the changes in both the chemical and biological properties of Zn in physiological saline. The morphology of the Zn-incorporated MAO surface did not change, and a small amount of Zn ions were released at early stages of incubation in saline. We observed a decrease in Zn concentration in the oxide layer because its release and chemical state (Zn2+ compound to ZnO) changed over time during incubation in saline. In addition, the antibacterial property of the Zn-incorporated MAO surface developed at late periods after the incubation process over a course of 28 days. Furthermore, osteogenic cells were able to proliferate and were calcified on the specimens with Zn. The changes related to Zn in saline had non-toxic effects on the osteogenic cells. In conclusion, the time transient effect of Zn in a porous titanium dioxide layer was beneficial to realize dual functions, namely the antibacterial property and osteogenic cell compatibility. Our study suggests the importance of the chemical state changes of Zn to control its chemical and biological properties.

    The Co-Cr-Mo alloy is applied for dental casting alloy. This alloy though both mechanical properties, corrosion resistance are excellent, research and development in which in addition, the property is improved is advanced. In this study, mechanical properties and corrosion resistance were examined on the alloy which replaced Mo of Co-33Cr-5Mo-0.3N alloy in W. On The result showed that mechanical properties were improved, as the composition of W increases. And, though on the corrosion resistance, the improvement by addition of W was not observed, the lowering could not be observed either.

    Although zirconia has excellent mechanical properties, it cannot be used as an artificial bone because it does not have bone-binding properties. On the other hand, calcium phosphates have inferior mechanical properties to zirconia, but have bone-binding properties and are therefore used clinically as an artificial bone. In this study, we aimed to develop an innovative zirconia-based artificial bone that combines the advantages of both zirconia and calcium phosphates. The material made by mixing powder made from carboxylate-containing octacalcium phosphate with zirconia and sintering the mixture was found to strongly inhibit the propagation of cracks. In addition, it was demonstrated that the material has strength three times that of conventional calcium phosphate, and the design guidelines for next-generation artificial bones were successfully established.

    The aim of this project, with originality in conceiving a smart surface design that promotes new bone formation around the implant body, the titanium (Ti) surface with different patterned periodic micro/nano-topographies were created by femtosecond laser irradiation. As a result, it was showed that the adhesion protein was well arranged by micron/nano periodic structure (Hybrid) using laser irradiation. In addition, we designed checkerboard surface pattern with micron/nano periodic structure showed the alternative hard tissue compatibility, both osteogenic differentiation and proliferation. Therefore, the checkerboard surface pattern with micron/nano periodic topography we created successfully promoted the regeneration of new bone in vitro and our findings provide a basis for the design of novel biomaterial surfaces that can regulate specific cellular functions with potential clinical applications.

    The aim of this project was to promote the regeneration of new bone around metallic implant materials by designing a nano-scaled topography on titanium (Ti) surface with femtosecond laser irradiation. Ti specimens with paralleled or crossed periodic grooves with multi-scales (micron, nano, or micron/nano hierarchical (hybrid) scales) were fabricated by femtosecond laser scanning. Among designed surface topographies, the hybrid surface topography showed an effective control of cellular adhesion behavior. In addition, our results also showed that a significant promotion of osteogenic differentiation and followed calcification deposition were observed by both human mesenchymal stem cells and mouse preosteoblasts cultured on the hybrid topography. Therefore, the micron/nano hierarchical topography we created successfully promoted the regeneration of new bone in vitro and our findings provide a basis for the design of novel biomaterial surfaces that can regulate specific cellular functions.
