    Phospholipase A2 (PLA2) catalyzes the hydrolysis of the sn-2 acyl ester linkage in phospholipid, producing lysophospholipid and fatty acid in the presence of Ca2+. The hydrolysis mediated by PLA2 has attracted much interest in various fields, such as pharmacy and biotechnology. It is recognized that PLA2 cannot hydrolyze phospholipid monolayers at high surface coverage. However, the origin of different PLA2 activities is not fully understood yet. The present study investigated the interaction between DPPC (16:0 PC) monolayer and PLA2 using heterodyne-detected sum frequency generation spectroscopy, which is interface-specific spectroscopy and highly sensitive to molecular symmetry based on a second-order nonlinear optical process. It was revealed that PLA2 adsorbs to the DPPC monolayer on the aqueous solution surface only when the surface coverage is low. The adsorption at the low surface coverage significantly changes the interfacial structures of PLA2 and the hydration, which are stabilized by the presence of Ca2+. Therefore, the restriction of the hydrolysis of phospholipid monolayers at high surface coverage can be rationalized by the inhabitation of the PLA2 adsorption. The present study deepens our molecular-level understanding of the hydrolysis of phospholipids by PLA2.

    Photosynthetic organisms have developed a regulation mechanism called state transition (ST) to rapidly adjust the excitation balance between the two photosystems by light-harvesting complex II (LHCII) movement. Though many researchers have assumed coupling of the dynamic transformations of the thylakoid membrane with ST, evidence of that remains elusive. To clarify the above-mentioned coupling in a model organism Chlamydomonas , here we used two advanced microscope techniques, the excitation-spectral microscope (ESM) developed recently by us and the superresolution imaging based on structured-illumination microscopy (SIM). The ESM observation revealed ST-dependent spectral changes upon repeated ST inductions. Surprisingly, it clarified a less significant ST occurrence in the region surrounding the pyrenoid, which is a subcellular compartment specialized for the carbon-fixation reaction, than that in the other domains. Further, we found a species dependence of this phenomenon: 137c strain showed the significant intracellular inhomogeneity of ST occurrence, whereas 4A+ strain hardly did. On the other hand, the SIM observation resolved partially irreversible fine thylakoid transformations caused by the ST-inducing illumination. This fine, irreversible thylakoid transformation was also observed in the STT7 kinase-lacking mutant. This result revealed that the fine thylakoid transformation is not induced solely by the LHCII phosphorylation, suggesting the highly susceptible nature of the thylakoid ultrastructure to the photosynthetic light reactions.

    <title>Abstract</title> Photosynthetic organisms adjust to fluctuating natural light under physiological ambient conditions through flexible light-harvesting ability of light-harvesting complex II (LHCII). A process called state transition is an efficient regulation mechanism to balance the excitations between photosystem II (PSII) and photosystem I (PSI) by shuttling mobile LHCII between them. However, in situ observation of the migration of LHCII in vivo remains limited. In this study, we investigated the in vivo reversible changes in the intracellular distribution of the chlorophyll (Chl) fluorescence during the light-induced state transitions in Chlamydomonas reinhardtii. The newly developed noninvasive excitation-spectral microscope provided powerful spectral information about excitation-energy transfer between Chl-a and Chl-b. The excitation spectra were detected through the fluorescence emission in the 700–750-nm spectral range, where PSII makes the main contribution, though PSI still makes a non-negligible contribution at room temperature. The technique is sensitive to the Chl-b spectral component specifically bound to LHCII. Using a PSI-specific 685-nm component also provided visualization of the local relative concentration of PSI within a chloroplast at room temperature. The decrease in the relative intensity of the Chl-b band in state 2 was more conspicuous in the PSII-rich region than in the PSI-rich region, reflecting the dissociation of LHCII from PSII. We observed intracellular redistributions of the Chl-b-related light-harvesting abilities within a chloroplast during the state transitions. This observation implies the association of the state transitions with the morphological changes in the thylakoid membrane.

    Blending an optimum amount of friction modifiers into lubricant is one of the important measures to reduce fuel consumption induced by the frictional loss for automobiles. However, the agents containing metal or phosphorus compounds can cause catalyst poisoning and clogging in the filters of the exhaust gas refining system. Thus, development of metal-free and phosphorus-free agents with a long-term stability under real working condition is highly desirable. In the present work, the friction coefficients on the metal surface in lubricants containing different friction modifiers were investigated by the reciprocating friction test in detail. A statistical method based on the quantitative structure-property relationship (QSPR) analysis has been employed to correlate the chemical nature of additives with their effect on the friction reduction behaviors. An empirical equation relating the molecular structure and its friction reduction efficiency has been proposed as a standard for a good friction modifier. Furthermore, a surface-sensitive vibration spectroscopy, sum frequency generation (SFG) spectroscopy, has been used to analyze the molecular structures of the lubricants adsorbed on the metal surface. The SFG observation suggested that a certain relationship between the adsorption structure and friction reduction effect of these agents.

    A novel scanning probe technique was developed by combining an atomic force microscope (AFM) and a quartz crystal resonator (QCR). The resonant frequency of the QCR increased when the AFM tip was placed in contact with the QCR surface and increased further when the force applied to the QCR surface increased. The frequency increase depended on the material with which the tip was in contact and seemed to reflect the local elasticity of the sample. Imaging of the material distribution on a surface was demonstrated for a pattern of polymer and gold by mapping the frequency shift of the QCR as a function of the horizontal position of the AFM tip while scanning the AFM tip over the QCR surface in constant force mode.

    A highly surface-sensitive vibrational spectroscopy, sum frequency generation (SFG), has been employed to investigate the interfacial structure of the Langmuir-Blodgett (LB) thin films on solid substrates. The SFG measurements demonstrate that the molecular structures at the surface layer of the even-numbered LB films of stearic acid considerably change both in air and in solution when Cd2+ cations are present. A flip-over model is proposed to explain the Cd2+-induced surface reorganization process.
