    The subtraction of background components from observed spectra is essential in performing multivariable analysis, frequently applied to Raman spectra of cells. The subtraction procedure, however, is manual and time consuming, especially for big data. Here, we propose an automated method for removing background information from measured spectra of cells, exhibiting the theoretical framework and an experimental application.

    Reactive oxygen species generated in dermal cells of human skin is related to skin disorders or diseases. In this study, Raman analysis effectively clarified the identities of three types of human skin models after the models were stimulated with hydrogen peroxide. With the Caucasian skin model, the major Raman bands underwent large intensity changes within 4-5 days of stimulation. With the Black skin model, the Raman bands remained almost unchanged. The changes in the Asian skin model were unique compared to those in the above two. Eumelanin and pheomelanin are probably the main compounds that differentiate dermal cells in terms of their sensitivity to hydrogen peroxide.

    The use of a hollow fiber as a Raman probe, which gives strong advantage of a free link in space, was confirmed to be a versatile and standard analytical method, since Raman data obtained through a hollow fiber probe assures a sufficient link to conventional Raman data. In this paper, we confirmed that a Raman spectrum given by the hollow fiber probe becomes identical to a Raman spectrum measured by a conventional approach, if one is multiplied by an optimized coefficient. In addition, Raman signal intensity changes were related to various types of curved geometries of the probe. The Raman signal intensity value at a curved geometry of the probe, which is one of the most frequently used positions, became 0.35 compared to the value at the standard position of the probe (straight lined).

    We describe a theoretical framework for a model-based approach to two-dimensional correlation spectroscopy that is generally applicable to any arbitrary model function. The method is based on the correlation between spectral data and a set of model waveforms with a varying correlation index, the global phase angle Θ. When experimental spectral intensity variations are expressed as sinusoidal, exponential, Lorentzian, or quadratic functions, the proposed approach allows us to estimate the quantitative values of the target parameters in those expressions. In addition, this method enables us to assess the sequential order in a series of bands undergoing non-identical intensity changes in a dynamic data set. We present both simulated and experimentally obtained data that illustrate that the deviations from linearity of the absorption band intensity waveforms are clearly detected and can be quantitatively estimated using quadratic functions.

    The effect of hydrothermal treatments (HTT) on merocyanine (MS)–Cd arachidate mixed Langmuir–Blodgett (LB) films has been studied. At temperatures ranging from 30 to 90 °C, the redshifted J-band in the as-deposited state has been found to reorganize under the condition of 100% relative humidity, forming a new phase of further redshifted bands with spectra sharper than those of the as-deposited J-band. The HTT-induced phase closely resembles that induced by the heat treatment without humidity control, while the occurrence of the latter is limited to 60–70 °C. The water content introduced into the LB system during HTT is suggested to play a catalytic role in facilitating the reorganization by its lubricating and/or hydrolyzing effects to soften the film structure.

    In this paper we propose a new method for pattern recognitions of band shifting, overlapping, and broadening using a global phase map derived from generalized two-dimensional correlation spectroscopy (global 2D phase map). To obtain the distinct patterns for band shifting, overlapping, and broadening, original spectra are subjected to scaling so that the spectra after the scaling do not cross one another. After such pretreatment the band shifting creates the tilting of a correlation plane, the band overlapping yields four flat terraces, and the band broadening generates an asymmetric curved plane with respect to the diagonal line in the global 2D phase map. An example of the application of this method has been investigated using two bands due to the CH2 symmetric stretching and CN stretching vibrations in temperature-dependent infrared (IR) transmission spectra of an eleven-layer Langmuir–Blodgett (LB) film of 2-dodecyl-7,7,8,8-tetracyanoquinodimethane (dodecyl-TCNQ). The global 2D phase maps develop the clearly distinct patterns due to the shifting, overlapping, and broadening even if the shifting and broadening bands are overlapped with other bands.

    A spectral analysis based on the concept of a global phase map derived from generalized two-dimensional (2D) synchronous and asynchronous correlation spectra has been applied to actual experimental data to demonstrate its potential. As an application of a global 2D phase map, temperature-dependent infrared transmission spectra from 30 to 80 °C of an eleven-layer Langmuir–Blodgett (LB) film of 2-dodecyl-7,7,8,8-tetracyanoquinodimethane (dodecyl-TCNQ) have been investigated. Although the generalized 2D correlation asynchronous map presents only a very weak signal due to the CH3 asymmetric stretching vibration compared with much stronger signals due to the CH2 stretching vibrations of dodecyl-TCNQ, the global 2D phase map clearly shows the distinct nonlinear response arising from the CH3 asymmetric stretching vibration. The detection of nonlinear responses suggests that the active thermal movement of the alkyl-chain loses a trans-zigzag skeleton plane in the intermediate state above 80 °C between the solid and melt states.

    The concept of a global phase map derived from generalized two-dimensional synchronous and asynchronous correlation spectra is introduced to extract the purely correlational information by intentionally canceling amplitude information. For the practical usage of a global 2D phase map, a filtering method based on the standard deviation spectrum obtained from the time series intensity profiles is proposed to remove the effect of small amplitude noise. A simulation created with the use of three synthesized bands with exponential decay has clearly detected the only difference in the characteristic rate constant of the decay process in the global 2D phase map, although the three bands have different initial values of the peak intensity amplitudes.

    Studies on highly conductive LB films based on ditetradecyldimethylammonium-Au(dmit)2 [2C14-Au(dmit)2] salt have been reviewed. The room temperature conductivity of the 2C14-Au(dmit)2 LB film is as high as 40 S/cm after electro-oxidation. The resistance vs. temperature plot depends on the gap distance for the measurement. A weak metallic behavior is seen from room temperature down to 210-250 K, when the gap distance is 500 μm, while it extends down to 56-106 K, when the gap distance is 5 μm, indicating that the disorders in the film are important limiting factors for the macroscopic resistance. The data of the complex magnetic susceptibility (χ' + iχ") suggest the existence of the Meissner state below 3.9 K. Although some samples show resistance drop on cooling below 4 K, the typical resistance behaviors are of activation type at the lower temperature. We consider that disorders such as grain boundaries and/or defects are important limiting factors that should be solved to improve the electrical conduction toward the realization of global superconductivity.

    An extension of the two-dimensional (2D) correlation analysis scheme for multi-dimensional perturbation is described. A simple computational form is provided to construct synchronous correlation and disrelation maps for the analysis of microscopic imaging data based on two independent perturbation variables. Sets of time-dependent attenuated total reflection infrared (ATR-IR) spectra of water and cellulose mixtures were collected during the evaporation of water from finely ground cellulose. The system exhibits complex behaviors in response to two independent perturbations, i.e., evaporation time and grinding time. Multiple perturbation 2D analysis reveals a specific difference in the rate of evaporation of water molecules when accompanied by crystallinity changes of cellulose. It identifies subtle differences in the volatility of water, which is related to the crystalline structure of cellulose.

    Langmuir–Blodgett (LB) films of dioctyldimethylammonium–Au(dmit)2 [2C8–Au(dmit)2] salt show a relatively high room-temperature conductivity of up to 17 S/cm. The conductivity is as high as $50 \pm 23$ S/cm after electrochemical oxidation. A distinct absorption band is seen at 287 nm and it increases linearly with the number of layers, indicating a reproducible film deposition for up to 20 layers. During the post-treatment, the absorption peak decreases together with some shoulders and a broad band appears in the near-infrared (NIR) region with an isosbestic point at 609 nm, indicating the formation of conduction electrons. The UV/visible absorption spectrum changes markedly in the first 60 min of the post-treatment and becomes almost constant later, which corresponds well with the change in conductivity during the post-treatment.

    Recently it became possible to measure Raman spectra of a single live cell using a standard Raman microscope. We developed a bio-Raman microscope in order to monitor the dynamics of cellular differentiation, proliferation, and apoptosis. To do so, living cells were incubated on the microscope stage. Using the bio-Raman microscope, we defined cellular states in a non-destructive and non-labeling manner. The obtained information of cellular conditions is very important to control cellular destinies aiming tissue engineering. Advanced mathematical methods, developed in this project, were found effective to spectral analysis in a general manner.

    A human breast cancer cell line, MCF-7 differentiates into mammalian grand-like cells upon addition of heregulin to the culture medium. In this study, we traced changes of cellular internal states by means of Raman spectrum measurement, which is a potent method to detect multi-dimensional intracellular dynamics in single cells with minimal disruption. Analysis of the Raman dynamics up to 12 days though the differentiation process has revealed pseud-circular trajectory of the major intracellular components.

    研究代表者の研究目的は、ジテトラデシルジメチルアンモニウム-Au(dmit)_2塩(2C_<14>-Au(dmit)_2塩)に基づくLangmuir-Blodgett(LB)膜の構造に関する情報を主に赤外分光法を用いて抽出し、LB膜が導電相、超伝導相を実現するメカニズムを明らかにすることであった。赤外スペクトルを解析するために、研究代表者が提案した位相角表示一般化二次元相関法を実験系に適用した。平成16年度の研究成果は、以下に要約される。 (1)これまで、LB膜の電気伝導度等の機能測定を継続的に行ってきた。今年度も引き続き、これを行った。低温領域の電気物性についても研究した。 (2)既に作製した温度変化赤外スペクトル測定システムを用いて、LB膜を温度変化させた時の赤外スペクトルを測定し、その構造変化について詳しく研究した。構造に関する情報に基づいて、LB膜に温度処理を施し、その導電性の向上に取り組んだ。 (3)申請者が提案した位相角表示一般化二次元相関分光法の理論体系の整理を行い、拡張した。具体的には、バンドシフト、ブロードニング問題を解決した。拡張した位相角法を実験データに適用した。 (4)研究成果をまとめ、その内容を権威ある雑誌に掲載した(J.Phys.Chem.BとChem.Phys.Lett.に1報ずつ掲載。)研究成果を国内外の学会で報告した。例えば、米国内の分析化学、分光関連学会(The Federation of Analytical Chemistry and Spectroscopy Societies (FACSS))において、招待講演を行うなどした。
