    Physical stimulation, which is a key factor affecting the metabolism of osteoblasts and their precursor cells, plays an important role in bone remodeling; however, the role of micro-vibrations in osteoblast differentiation is unclear. In the present study, we determined the effects of frequency-regulated repeated micro-vibration (FRMV) on cell proliferation and established a method to induce osteoblast differentiation through FRMV using the mouse pre-osteoblast-like cell line MC3T3-E1, which is widely used in bone metabolism research. The results indicated that FRMV significantly influenced the proliferation of MC3T3-E1 cells in a normal growth medium. FRMV at 42.2 Hz significantly promoted proliferation, whereas FRMV at 92.1 Hz showed no effect on the proliferation rate. Moreover, FRMV at 42.2 Hz significantly increased alkaline phosphatase (ALP) enzyme activity and ALP gene expression in MC3T3-E1 cells. Treatment with LDN193189, a bone morphogenetic protein (BMP) signaling inhibitor, revealed that the FRMV-induced upregulation in ALP enzyme activity and ALP gene expression were significantly suppressed in MC3T3-E1 cells. The results suggest that the FRMV protocol developed in the present study induces osteoblast differentiation through the BMP signaling pathway. Thus, FRMV may contribute to the development of effective bone regeneration technologies.

    Abstract Overactivation of poly (ADP-ribose) polymerase-1 (PARP-1) triggers a noncanonical form of programmed cell death (PCD) called parthanatos, yet the mechanisms of its induction are not fully understood. We have recently demonstrated that the aggresome-like induced structures (ALIS) composed of the autophagy receptor SQSTM1/p62 and K48-linked polyubiquitinated proteins (p62-based ALIS) mediate parthanatos. In this study, we identified the D1 dopamine receptor agonist YM435 as a unique parthanatos inhibitor that acts as the disaggregating agent for the p62-based ALIS. We found that YM435 structurally reduces aggregability of the ALIS, and then increases its hydrophilicity and liquidity, which prevents parthanatos. Moreover, dopamine and L-DOPA, a dopamine precursor, also prevented parthanatos by reducing the aggregability of the ALIS. Together, these observations suggest that aggregability of the p62-based ALIS determines the sensitivity to parthanatos, and the pharmacological properties of YM435 that reduces the aggregability may be suitable for therapeutic drugs for parthanatos-related diseases such as neurodegenerative diseases.

    PC12 cells, which are derived from rat adrenal pheochromocytoma cells, are widely used for the study of neuronal differentiation. NGF induces neuronal differentiation in PC12 cells by activating intracellular pathways via the TrkA receptor, which results in elongated neurites and neuron-like characteristics. Moreover, the differentiation requires both the ERK1/2 and p38 MAPK pathways. In addition to NGF, BMPs can also induce neuronal differentiation in PC12 cells. BMPs are part of the TGF-β cytokine superfamily and activate signaling pathways such as p38 MAPK and Smad. However, the brief lifespan of NGF and BMPs may limit their effectiveness in living organisms. Although PC12 cells are used to study the effects of various physical stimuli on neuronal differentiation, the development of new methods and an understanding of the molecular mechanisms are ongoing. In this comprehensive review, we discuss the induction of neuronal differentiation in PC12 cells without relying on NGF, which is already established for electrical, electromagnetic, and thermal stimulation but poses a challenge for mechanical, ultrasound, and light stimulation. Furthermore, the mechanisms underlying neuronal differentiation induced by physical stimuli remain largely unknown. Elucidating these mechanisms holds promise for developing new methods for neural regeneration and advancing neuroregenerative medical technologies using neural stem cells.

    The Gly16Arg polymorphism results in a G to C nucleotide mutation in the human beta 2-adrenergic receptor (ADRB2) gene and has a relationship with obesity; however, this substitution's effects on food preferences are unclear. Therefore, we determined this relationship among healthy young adults (mean age, 23.4; n = 52). To evaluate food preferences, four categories of food (sweet, salty, sour, and bitter) along with high-fat foods were evaluated using a self-reporting questionnaire. Male (n = 26) and female subjects (n = 26) were genotyped for the polymorphism and further divided into three groups (two homozygous groups, GG, CC; and a heterozygous group, GC). Preference for sour foods in the GG group was higher compared with that in the CC group in females (p < 0.05). When sweet foods were classified into low- and high-fat subgroups, preference for high-fat sweet foods in the GG group was higher than that for low-fat sweet foods in all subjects (p < 0.05). The degree of preference for high-fat foods in the GG group was higher than other groups for males (p < 0.05). These results suggest that ADRB2 polymorphism is associated with food preference. Understanding the relationship of ADRB2 substitution to food preference will be valuable for designing individualized anti-obesity strategies.

    Neuritogenesis is the process underling nervous system regeneration; however, optimal extracellular signals that can promote neuronal regenerative activities require further investigation. Previously, we developed a novel method for inducing neuronal differentiation in rat PC12 cells using temperature-controlled repeated thermal stimulation (TRTS) with a heating plate. Based on neurogenic sensitivity to TRTS, PC12 cells were classified as either hyper- or hyposensitive. In this study, we aimed to investigate the mechanism of hyposensitivity by establishing two PC12-derived subclones according to TRTS sensitivity during differentiation: PC12-P1F1, a hypersensitive subclone, and PC12-P1D10, a hyposensitive subclone. To characterize these subclones, cell size and neuritogenesis were evaluated in subclones treated with nerve growth factor (NGF), bone morphogenetic protein (BMP), or various TRTS. No significant differences in cell size were observed among the parental cells and subclones. BMP4- or TRTS-induced neuritogenesis was increased in PC12-P1F1 cells compared to that in the parental cells, while no neuritogenesis was observed in PC12-P1D10 cells. In contrast, NGF-induced neuritogenesis was observed in all three cell lines. Furthermore, a BMP inhibitor, LDN-193189, considerably inhibited TRTS-induced neuritogenesis. These results suggest that the BMP pathway might be required for TRTS-induced neuritogenesis, demonstrating the useful aspects of these novel subclones for TRTS research.

    Osteoblasts are extremely important for bone remodeling processes. Previously, we found a method to induce neurite outgrowth of rat PC12 cells in a temperature-controlled repeated thermal stimulation (TRTS)-dependent manner using a heating plate that is capable of controlling the medium temperature. However, few studies have examined the effect of the TRTS on the osteoblastic differentiation. The present study investigated the effects of the TRTS on the osteoblastic differentiation in murine pre-osteoblastic MC3T3-E1 cells. We clarified the effects of the TRTS on growth rates, alkaline phosphatase (ALP) activity and alizarin red staining (as a marker of osteoblast maturation). These results suggest that TRTS can induce the osteoblastic differentiation in the cells and might offer an effective and novel technique for bone regeneration.

    Neurite formation is a phenomenon that enables functional formation and recovery of the nervous system. Previously, we found a method to induce neurite outgrowth of rat PC12 cells in a temperature-controlled repeated thermal stimulation (TRTS)-dependent manner using a heating plate capable of controlling the medium temperature. However, there is still room for improvement in this method, which induces neuronal differentiation. The purpose of this study is to investigate the versatility of TRTS, the molecular mechanism of differentiation induction, and to improve the differentiation rate. Analysis using signal inhibitors and next-generation sequencing suggested the signaling molecules and signaling pathways that are involved in TRTS-induced neurite formation and in improving the neuronal differentiation rate.

    Dental implants are widely used as highly predictive treatments, however, there is no fundamental solution to peri-implantitis. Therefore, in this research, we aimed to construct a completely new comprehensive implant recovery system for peri-implantitis by selecting the site and time and applying radicals using the latest engineering technology. In this study, we first developed visible light responsive titanium oxide and atmospheric pressure plasma jet irradiation equipment that are most suitable for the implant recovery systems. Using these, we were able to acquire the basic technology to construct an implant recovery system that can be reliably and safely restored without removal in the oral cavity.

    The aim of this study was to construct dental implant recovery system in the oral cavity due to a completely new approach based on three pillars; ① powerful sterilization and infected tissue removal, ② re-activation treatment of the implant surface, ③ tissue regeneration, as a method of obtaining a functional and morphological recovery in the oral cavity without removing the dental implant with peri-implant inflammation. As research results, by application of the latest technology of low-temperature plasma jet, the base of the dental implant recovery system to promote good tissue regeneration was constructed by carrying out the safe sterilization and surface re-activation treatment in the oral cavity for the implant which has lost its biological activity.

    Previously, we developed a novel method for inducing neuritogenesis (NG) using a pulsed electromagnetic field (PEMF) with an exciting coil in rat-derived PC12 cells, which are a model of neuronal differentiation. However, the mechanism that enhances NG is still unknown. In the present study, we examined the mechanism of NG due to PEMF, the effect of Joule heating on NG, and the possibility of PEMF affecting the direction of NG. We demonstrated that the bone morphogenetic protein (BMP) pathway is essential in the process of PEMF-induced NG and that the Joule heat generated by the coil partially contributes to the PEMF-mediated induction of neuronal differentiation. We also found that it is possible to efficiently differentiate the cells into neuronal cells by thermal stimulation alone and that it is difficult to regulate the direction of NG using a mono-directional and parallel magnetic field with a custom-made exciting coil as used in the present study.

    The aim of this study was to establish a new evaluation system for osteoconductivity by focusing on the protein adsorption properties to biomaterials. First, we revealed the difference in the three-dimensional conformational status of the protein on the surface of such osteoconductive biomaterials as hydroxyapatite, as well as the amount of adsorption, concerning the adsorption behavior of serum proteins. Next, by revealing the correlation between the proliferative capacity of mouse pre-osteoblastic cell line MC3T3-E1 and protein adsorption properties on the material, the possibility of a new evaluation system for osteoconductivity was investigated.

    In this study, local bone formation effects of CNP hormone was examined to develop a new clinical treatment of inhabitation of bone resorption on residual ridge. The bone defect of 8.8 mm diameter to expose the dura mater was made in rat cranium. Hydrothermal hydroxyapatite containing saline solution and CNP hormone was prepared and implanted in the defect area. The new bone was formed from peripheral bone and dura mater. The area of newly formed bone increased to 4 weeks after implantation. No significant differences in new bone area showed between saline and CNP groups. The diffusion velocity of CNP hormone in local area should be examined to understand of the in vivo local effect of CNP hormone.

    The aims of this study were to investigate the mechanical-stress-induced gene expression patterns by gene profiling using microarray analysis in the cells of periodontal tissue, and to select the functionally effective molecules to regulate the bone remodeling, and then to establish the transfer method of selected gene into the periodontal tissue. The expression pattern analysis suggested that both properties common among cells and cell-specific properties affect the remodeling mechanism of periodontal tissues in response to mechanical stress. Moreover several signal pathways were identified as candidates. A non-viral gene delivery approach with nano/microbubbles and ultrasound were established for targeting soft tissues.

    The purpose of this study was to examine the possibility of three-dimensional craniofacial form control by the growth control of the suture using i nternalized d evice made of Ni-free Ti-based s hape m emory a lloy from the bi omedical engineering aspect. The animal experiment model which loads mechanical force on the suture of a growth phase rat cranium was produced. Biological and m orphometric al evaluation for the reaction of the suture against the mechanical stress was done in this study. As a result, it was suggested that three-dimensional form control of craniofacial regions could be performed by the device using a new shape memory alloy.

    JNKはストレス応答や細胞分化等の制御因子として機能するキナーゼであるが、初期発生や分化におけるJNKの核外での制御機構は不明な点が多い。一方、JNK結合タンパク質には、JNK結合モチーフが高度に保存されていることから、本研究において、ヒトタンパク質アミノ酸配列ライブラリーを活用した網羅的In silicoスクリーニングを行った結果、私は有力な細胞核外JNK結合候補タンパク質群を得た。 その中で、初期発生や硬組織新生に重要なサイトカイン、BMPの細胞膜受容体の一つ、BMPRII(BMP receptor type II)が、その特異的なC末端領域において、JNKと結合することを見出した。BMPRIIは、JNK以外に、その上流キナーゼのMIKK4やASK1とも会合し、これらは、BMP刺激に応じ空間的制御を受け、BMPRII上でASK1/JNK経路が形成されることを見出した。ASK1により活性化されたJNKは、BMPRIIのスレオニン977(T977)を直接リン酸化した。また、ASK1/JNKシグナル下流で、この部位のリン酸化依存的にプロテアゾーム経路によるBMPRIIのタンパク分解が促進されたが、T977A点変異体では促進されなかった。これらの結果は、BMPRIIがASK/JNK経路の足場タンパク質として機能することを示しており、かつJNKによるBMPRII T977のリン酸化が、半減期を含むBMPRIIの機能制御に関与することを示している。 更に、ニワトリ胚初期発生系で、Smad経路のドミナントネガティブBMPRII変異体に対するT977E点変異体の強制発現が、ドミナントネガティブBMPRII変異体とは異なり、肩甲骨の発生を阻害したことから、初期発生において、このBMPRIIのT977のリン酸化レベルの調節が、non-Smadシグナルの調節に関与している事を示唆している。
