    Translocation of a daughter vesicle from a mother vesicle through a pore is experimentally studied by many groups using a model system of self-reproducing vesicles. However, the theoretical formulation of the problem is not fully understood. In the present study, we present a theoretical formulation of the process based on our previous work [P. Khunpetch et al., Phys. Fluids 33, 077103 (2021)]. In our previous work, we considered the daughter vesicle as a rigid body. In the present work, however, we allow the daughter vesicle to deform during the expulsion process. We thus derive the free energy constituting of the elastic moduli of both the mother and daughter vesicles, and of pressure-driven contribution. The minimum energy path of the translocation is searched by using the string method. With use of experimentally reasonable values of the elastic moduli, our improved model successfully reproduced the progress of the birthing process where there is no free energy barrier between the initial and the final states. The equations of motion of the daughter vesicle have been derived within the framework of the Onsager principle. We found that the translocation time of the daughter vesicle can be reduced when the pressure inside the mother vesicle increases, or the initial size of the daughter vesicle decreases.

    Abstract A “synthetic minimal cell” is considered here as a cell-like artificial vesicle reproduction system in which a chemical and physico-chemical transformation network is regulated by information polymers. Here we synthesise such a minimal cell consisting of three units: energy production, information polymer synthesis, and vesicle reproduction. Supplied ingredients are converted to energy currencies which trigger the synthesis of an information polymer, where the vesicle membrane plays the role of a template. The information polymer promotes membrane growth. By tuning the membrane composition and permeability to osmolytes, the growing vesicles show recursive reproduction over several generations. Our “synthetic minimal cell” greatly simplifies the scheme of contemporary living cells while keeping their essence. The chemical pathways and the vesicle reproduction pathways are well described by kinetic equations and by applying the membrane elasticity model, respectively. This study provides new insights to better understand the differences and similarities between non-living forms of matter and life.

    生体膜における代謝や信号伝達などの生体機能はタンパク質の膜内流動によって制御されている。このため、モデル生体膜(ベシクル)を用いた膜の流動特性の解明が行われてきたが、理論モデルにおける測定可能な膜粘度範囲の制限などから、その流動性の全景については未だ明らかになっていなかった。最近、研究代表者はベシクル膜面上にマイクロインジェクションにより局所的な力を加えることで引き起こされる膜流動パターンを、球面上での流体力学理論[Henle and Levine, Phys. Rev. E (2010)]と比較することにより膜粘度を計測する手法を開発した。この手法を用いて生体膜の主要構成成分である3種類の脂質分子(飽和リン脂質DPPC, 不飽和リン脂質DOPC, コレステロール CHOL)から成るベシクルの流動パターンを、相分離によってできるドメインの動きによって可視化し、組成と膜粘度の関係を測定した。その結果、膜粘度は組成により3桁に渡る広範囲で変化することが明らかになった[Sakuma et al., Biophys. J. (2020)]。 本研究では、モデル生体膜の非対称性などの条件を段階的に生体膜に近づけ、最終的には生体膜の粘度測定を行うことで流動特性を支配する要因の解明を目指す。2020年度は糖鎖に見立てた高分子(polyethyrene glycol:PEG)でベシクル表面を装飾した場合には装飾しないベシクルと比較して膜粘度が1桁程度高くなることを発見した。2021年度はベシクルを用いたボトムアップの立場からのアプローチに対して, 実際の生物の細胞を使ったトップダウンの立場から, 線虫の初期胚にベシクルと同様にマイクロインジェクションによって水流を与え, 膜流動の様子を観察した. この結果, 細胞骨格と細胞膜の間に強固な相互作用がはたらいていることがわかってきた.

    Membrane traffic which is fundamental cell function shows membrane deformations such as adhesion, fusion or pore formation. The purpose of this study is to understand membrane deformations associated with cell functions from soft-matter physics point of view. Membrane deformations of lipid vesicle were reproduced by chemical stimuli using micro-injection method, and were compared with membrane elasticity model. As a result, it is revealed that membrane interactions or membrane tension can be controlled by microinjection of electrolytes to vesicles containing anionic lipid, which realize to membrane deformations relevant to cell functions(e.g. adhesion, fusion, pore formation and so on). In addition, membrane flow induced by microinjection was found in this study, which led to development of an unique technique to measure membrane viscosity for wide range of membrane compositions.

    This research aims to construct minimal cell having essence of biological functions by using non-biological substances, so-called soft materials, whereby we reveal physics underlying the life. We focused on molecular assembly systems composed of synthesizing information polymer encoding vesicle membrane and growth-division vesicle coupled with the synthesis of information polymer. We succeeded to demonstrate the reproduction of vesicles coupled with an enzymatic polymerization reaction (synthesis polyaniline: PANI-ES) occurring on the surface of vesicles composed of AOT and cholesterol. An important characteristic of this reproduction system is that the AOT/cholesterol vesicles encourage the synthesis of PANI-ES and PANI-ES promotes the growth of AOT vesicles. This mutual promotion kinetics captures an essential feature of minimal cells.

    The purpose of this study is to create the molecular assembly (artificial cell) having the essential functions of cellular life (heredity using information molecules, metabolism, self-reproduction of container) and the self-propelling ability. We will reveal the physical background of the cellular life through the artificial cells. 1) We have succeeded to create two types of artificial cells. Both of them demonstrate the vesicle reproduction coupled with the information molecules. One of them shows a primitive model cell cycle comprising four discrete phases (ingestion, replication, maturity and division), i.e., repeatable cell division. Both artificial cell has a potential of heredity and evolvability through the information molecules. 2)We also succeeded to create self-propelled tubular vesicles triggered by chemical stimuli, where the migration is coupled with the wavy motion of the tube. These findings shed light on the way from the molecular assembly to the cellular life.

    To understand the response of molecular assemblies against chemical stimuli is essential for approaching cell dynamics, such as cell deformation and division, from soft matter physics point of view. In this study, I focused on the vesicle dynamics relevant to cell functions induced by chemical stimuli, especially pore formation that plays an important role in membrane traffic. I succeed to reproduce pore formation on the vesicle by injecting hydroxide ions to phospholipid vesicles containing anionic lipids. The interaction between anionic lipid and hydroxide ion decreases cross section area of lipids, which produces the surface tension and then pore formation. This chemical control of pore formation is a first step to reveal biological function non-equilibrium soft matter physics point of view.

    in this research we have investigated vesicle dynamics triggered by chemical stimuli, which might be the first step of the evolution from vesicles to cellular life. By applying chemical stimuli, vesicles show unique dynamics, such as migration, deformations, relevant to biological functions. We revealed that chemical modifications of the surface tension and molecular cross section are responsible for the observed vesicle dynamics. In addition we succeeded to synthesize a new artificial lipid precursor molecule, which is hydrolyzed to produce the membrane molecule at the membrane surface. The membrane molecules are incorporated into the vesicle membrane and causes the vesicle growth.The growing vesicle produces offspring vesicles by division. The vesicle division process was investigated by a molecular dynamics simulation. the simulation revealed that the coupling of molecular shape and vesicle shape plays an important role in the vesicle division.
