    3-Monochloropropanediol esters (3-MCPDEs) are contaminants formed during high-temperature cooking and oil refining. While meat products are a source of 3-MCPDE exposure, related risk assessments remain limited. This study analyzed 3-MCPDE levels in pan-fried marinated meats (pork, chicken, beef, and salmon, with 0-10 % NaCl) using gas chromatography-mass spectrometry (GC-MS) following American Oil Chemists' Society (AOCS) Cd 29a-13 and assessed exposure in the Taiwanese population. Salt addition and higher unsaturation promoted lipid oxidation, increasing 3-MCPDE formation. Strong correlations (R > 0.9) were found between 3-MCPDE contents and total oxidation (totox) values across different meat types. Pork, with the highest fat content, had the highest 3-MCPDE levels, followed by salmon. Exposure assessments showed 3-MCPDE intake remained below the tolerable daily intake (TDI), with pork being the largest contributor. The highest P95 exposure from four meat types with 10 % NaCl reached 41.35 % of the TDI, suggesting potential risk when combined with other dietary sources.

    This study developed water-dispersible γ-oryzanol (WD-OZ) using microemulsion system and assessed their absorption in rats. While γ-oryzanol itself is hardly soluble in water, WD-OZ exhibited high water dispersibility, and γ-oryzanol, along with its metabolites, was detected in rat plasma. These findings provide a solid basis for future application of the microemulsion-based approach to enhance the bioavailability of γ-oryzanol in food.

    Lipid droplets (LDs) are organelles that store and supply lipids based on cellular needs. While mechanisms preventing oxidative damage to membrane phospholipids are established, the vulnerability of LD neutral lipids to peroxidation and protective mechanisms are unknown. Here, we identify LD-localized Ferroptosis Suppressor Protein 1 (FSP1) as a critical regulator that prevents neutral lipid peroxidation by recycling coenzyme Q10 (CoQ10) to its lipophilic antioxidant form. Lipidomics reveal that FSP1 loss leads to the accumulation of oxidized triacylglycerols and cholesteryl esters, and biochemical reconstitution of FSP1 with CoQ10 and NADH suppresses triacylglycerol peroxidation in vitro. Notably, polyunsaturated fatty acid (PUFA)-rich triacylglycerols enhance cancer cell sensitivity to FSP1 loss and inducing PUFA-rich LDs triggers triacylglycerol peroxidation and LD-initiated ferroptosis when FSP1 activity is impaired. These findings uncover the first LD lipid quality control pathway, wherein LD-localized FSP1 maintains neutral lipid integrity to prevent the buildup of oxidized lipids and induction of ferroptosis.

    Conjugated fatty acids (CFAs) have been known for their anti-tumor activity. However, the mechanism of action remains unclear. Here, we identify CFAs as inducers of glutathione peroxidase 4 (GPX4) degradation through chaperone-mediated autophagy (CMA). CFAs, such as (10E,12Z)-octadecadienoic acid and α-eleostearic acid (ESA), induced GPX4 degradation, generation of mitochondrial reactive oxygen species (ROS) and lipid peroxides, and ultimately ferroptosis in cancer cell lines, including HT1080 and A549 cells, which were suppressed by either pharmacological blockade of CMA or genetic deletion of LAMP2A, a crucial molecule for CMA. Mitochondrial ROS were sufficient and necessary for CMA-dependent GPX4 degradation. Oral administration of an ESA-rich oil attenuated xenograft tumor growth of wild-type, but not that of LAMP2A-deficient HT1080 cells, accompanied by increased lipid peroxidation, GPX4 degradation and cell death. Our study establishes mitochondria as the key target of CFAs to trigger lipid peroxidation and GPX4 degradation, providing insight into ferroptosis-based cancer therapy.

    Monitoring oxidation causes (i.e. radical and/or photo-oxidation) of vegetable oil gives very important information for its quality control. This study aimed to develop a rapid and simple near infrared (NIR) spectroscopy method to explore the oxidation causes in vegetable oil by quantifying triacylglycerol hydroperoxide (TGOOH) positional isomers proceeded by different oxidation causes. First, the concentrations of TGOOH isomers were determined by liquid chromatography-mass spectrometry. Partial least square regression for concentrations of TGOOH isomers (R2: 0.994-0.998) using NIR spectra in the regions of 7500-6000 cm-1 and 5500-4500 cm-1 as explanatory variables. Principal component analysis revealed that those models were created because NIR spectra capture trace changes of the peak around 6980 cm-1(-CH2-) and 5260 cm-1 (H2O) derived by different oxidation causes (i.e. radical and/or photo-oxidation). The application of this method to various kinds of vegetable oil is expected to lead to a simple and rapid quality evaluation of vegetable oil.

    Liposome lipid peroxidation induced by cold atmospheric pressure plasma jet (CAPPJ) irradiation was investigated. The formation of thiobarbituric acid reactive substances (TBARS), an indicator of lipid peroxidation final products, as a function of irradiation was observed. Lipid radicals, peroxidation reaction intermediates generated by CAPPJ irradiation, were confirmed by increased NBD-pen fluorescence intensity. Additionally, lipid peroxidation products, liposomal phosphatidylcholine (PC) isomers, were analyzed by LC-MS/MS. Products specific to singlet oxygen (1O2) oxidation, 16:0/10-hydroperoxy-8E,12Z-octadecanoic acid (10-8E,12Z-HpODE) PC and 16:0/12-9E,13E-HpODE PC, were not detected, but radical oxidation specific products 16:0/13-9E,11E-HpODE PC and 16:0/9-10E,12E-HpODE PC were. This suggests that during CAPPJ irradiation, radicals, rather than 1O2, are the primary reactive species of lipid peroxidation. This is also supported by the β-carotene quenching of 1O2 not suppressing TBARS and lipid radical generation. Also, neither TBARS formation nor lipid radical generation were suppressed by SOD, indicating that the superoxide radical (O2 •-) is not responsible for the lipid peroxidation reaction. As the CAPPJ irradiation of water produces large quantities of hydroxyl radical (•OH) and •OH scavengers decreased the amount of TBARS produced by CAPPJ irradiation, it is highly plausible that •OH is the primary species involved in CAPPJ-induced liposome lipid peroxidation.

    The roasting process of sesame oil is expected to alter its internal composition and stability under oxidation condition. Presumably, the effect of roasting may differ with oxidation conditions (i.e., thermal and photo-oxidation), but such studies have not been undertaken. To further evaluate this notion, several type of sesame oils (raw and refined as unroasted oil, and roasted oil) and rapeseed oils as comparison were subjected to thermal oxidation (120℃) and photo-oxidation (50,000 lx) for 5 and 10 hours. The result revealed that the roasting sesame oil exhibited good stability under thermal oxidation, possibly due to the change on antioxidant agents such as sesamol and Maillard products during the roasting process. In contrast, the refined sesame oil (unroasted) demonstrated high stability under photo-oxidation, indicating that the refining process has a more significant impact on the oxidative stability in sesame oil compared to the alterations in its components caused by the roasting process. Taken together, this study is the first to show that the roasting and refining processes of sesame oil alter its internal composition and show different variations in sesame oils' oxidative stability under thermal and photo-oxidation, which holds significance considering its global consumption.

    Determining the content of the nutrient choline in foods and obtaining the required amount from the diet are crucial. One way to measure choline in foods is by converting choline esters to free choline via acid hydrolysis, followed by quantifying the total choline, as adopted by the AOAC method (AOAC-Choline); however, certain choline esters are difficult to hydrolyse. Here, we investigated various acid hydrolysis conditions to establish a reliable method for determining the total choline in foods by detecting free choline using highly sensitive and selective mass spectrometry. Hydrolysis in 0.055 mol/L HCl for 8 h in an autoclave (121 °C) was found to be optimal for the hydrolysis of choline esters in various foods. Twenty-four foods, including grains, seed, vegetables, fruits, mushroom, algae, fish, meats, beverage, processed foods, and egg, were measured. The trends in the total choline content were consistent with previous reports; however, the choline content was 10-20% higher than that measured using AOAC-Choline. Therefore, re-evaluation of the total choline content in foods using our constructed method is recommended. This reassessment will allow for a more reliable determination of choline intake for maintaining health.

  14. 1‐Deoxynojirimycin attenuates pathological markers of Alzheimer's disease in the in vitro model of neuronal insulin resistance

    Abstract Insulin resistance, the hallmark of type 2 diabetes mellitus (T2DM), has emerged as a pathological feature in Alzheimer's disease (AD). Given the shared role of insulin resistance in T2DM and AD, repurposing peripheral insulin sensitizers is a promising strategy to preserve neuronal insulin sensitivity and prevent AD. 1‐Deoxynojirimycin (DNJ), a bioactive iminosugar, exhibited insulin‐sensitizing effects in metabolic tissues and was detected in brain tissue post‐oral intake. However, its impact on brain and neuronal insulin signaling has not been described. Here, we investigated the effect of DNJ treatment on insulin signaling and AD markers in insulin‐resistant human SK‐N‐SH neuroblastoma, a cellular model of neuronal insulin resistance. Our findings show that DNJ increased the expression of insulin signaling genes and the phosphorylation status of key molecules implicated in insulin resistance (Y1146‐pIRβ, S473‐pAKT, S9‐GSK3B) while also elevating the expression of glucose transporters Glut3 and Glut4, resulting in higher glucose uptake upon insulin stimuli. DNJ appeared to mitigate the insulin resistance‐driven increase in phosphorylated tau and Aβ1‐42 levels by promoting insulin‐induced phosphorylation of GSK3B (a major tau kinase) and enhancing mRNA expression of the insulin‐degrading enzyme (IDE) pivotal for insulin and Aβ clearance. Overall, our study unveils probable mechanisms underlying the potential benefits of DNJ for AD, wherein DNJ attenuates tau and amyloid pathologies by reversing neuronal insulin resistance. This provides a scientific basis for expanding the use of DNJ‐containing products for neuroprotective purposes and prompts further research into compounds with similar mechanisms of action.

    The Ryukyu long-furred rat is an endangered species confined to the southernmost three small islands of Japan (Amami-Oshima, Tokunoshima, and Okinawa). Its population is rapidly decreasing because of roadkill, deforestation, and feral animals. To date, its genomic and biological information are poorly understood. In this study, we successfully immortalized Ryukyu long-furred rat cells by expressing a combination of cell cycle regulators, mutant cyclin-dependent kinase 4 (CDK4R24C) and cyclin D1, together with telomerase reverse transcriptase or an oncogenic protein, the Simian Virus large T antigen. The cell cycle distribution, telomerase enzymatic activity, and karyotype of these two immortalized cell lines were analyzed. The karyotype of the former cell line immortalized with cell cycle regulators and telomerase reverse transcriptase retained the nature of the primary cells, while that of the latter cell line immortalized with the Simian Virus large T antigen had many aberrant chromosomes. These immortalized cells would be valuable for studying the genomics and biology of Ryukyu long-furred rats.

    The delivery of curcumin (CUR) using the solid dispersion system (CUR solid dispersions; C-SDs) has been shown to improve CUR bioavailability. However, it is unclear how different particle sizes of C-SDs affect the bioavailability and biological activities of CUR. Hence, we prepared C-SDs in different sizes using food-grade excipients and evaluated their bioavailability and biological activities. By pulverizing large particle sizes of C-SDs using zirconia beads, we successfully prepared C-SDs I-IV (particle size: (I) 120, (II) 447, (III) 987, (IV) 1910 nm). When administrated orally in rats, the bioavailability of CUR was increased with decreasing C-SDs size, most likely by improving its solubility in micelles. When administrated intravenously in rats, blood concentrations of CUR were increased with increasing particle size, suggesting that larger C-SDs presumably control the metabolic conversion of CUR. In RAW264 cells, more CUR was taken up by cells as their sizes reduced, and the more potent their anti-inflammatory activities were, suggesting that smaller C-SDs were taken up through a number of cellular uptake pathways. Altogether, the present study showed an evident effect of C-SDs size on their bioavailability and anti-inflammatory activities-information that serves as a basis for improving the functionality of CUR.

    Pyrroloquinoline quinone (PQQ) is a water-soluble, vitamin-like compound discovered in bacteria as the third redox coenzyme. PQQ is reportedly present especially in natto (fermented soybeans) and tea, and the compound has been reported to have physiological activities such as antioxidant effect and nerve protection. However, the distribution and physiological function of PQQ have been still unclear, probably due to limitation of its analytical method. In this study, we tried to develop a LC-MS/MS method of PQQ. By using a reverse-phase column with an ion-pair reagent (dibutylammonium acetate), PQQ standard could be analyzed at the sensitive order of picogram level. Furthermore, since PQQ could easily react with amino acids including Arg, Gln, Gly, His, Lys, and Phe, a LC-MS/MS method for analysis of PQQ-amino acid adducts was also developed. The developed method could be useful for not only investigation of distribution of PQQ and PQQ-amino acid adducts in nature, but also evaluation of their absorption, metabolism and physiological activities in vivo.

    We recently reported that allergic reaction induced disruption of liver lipid metabolism and that tocotrienol (T3) had a potential to improve allergic reaction. The goal of this study was to examine the influence of T3 administration on liver lipid metabolism in allergia-induced mice. A model of atopic dermatitis was caused in NC/Nga mice treated with picryl chloride to induce allergic reaction. Allergia-induced mice caused lipid accumulation in the liver. On the other hand, T3 administration attenuated the liver lipid accumulation. The expression levels of the genes related to the liver lipid metabolism in the allergic mice was examined using DNA microarray analysis and quantitative RT-PCR. Allergic reaction caused upregulation of Cdkn1a, which is strongly associated with lipid accumulation, and downregulation of Acox1, which is associated with fatty acid β-oxidation, in the liver of allergic mice. In the liver of T3-administered allergic mice, the changes in the expression levels of Cdkn1a and Acox1 were not observed. These results suggest that T3 administration can improve the disturbance of liver lipid metabolism induced by allergic reaction.

    Since the Japanese are known for their long life span, it has been theorized that a Japanese-style diet has potential for health promotion. To identify the health effects of components of the Japanese diet that have changed over the last five decades, we conducted a study to examine their influence on obesity in mice. Based on National Nutrition Survey data, one-week menus of Japanese foods representative of those that used to be consumed in 2005, 1990, 1975 and 1960 were prepared, cooked, and powderized. Each of these diets was given to ICR mice for four weeks. It was found that Japanese food representative of 1975 suppressed lipid accumulation in white adipose tissue and expansion of adipocytes. To examine the underlying mechanisms responsible, we conducted DNA microarray analysis of the liver, which is responsible for lipid energy metabolism, and found that Japanese food representative of 1975 promoted the expression of genes related to high-energy expenditure. We then prepared purified diets mimicking Japanese foods from each period based on their energy balance of protein, fat and carbohydrate, and fed them to mice for four weeks. No significant differences in white adipose tissue weight were evident among the four groups. Overall, our findings suggested that Japanese foods typical of the period around 1975 are more effective than modern Japanese foods for preventing obesity, and that this effect is not dependent on energy balance.

    Japanese food is of interest worldwide as healthy food. However, comparison of Japanese food with European and American food has not been performed using DNA microarray analysis. In this study, we examined differences in gene expression levels in the liver of rats fed "Japanese food" or "American food" using a DNA microarray. Two meals were cooked based on a menu of Japanese food and American food. The cooked meals were prepared to a freeze-dried powder and given to rats for three weeks as test diets. Total RNA was then extracted from rat liver and used in DNA microarray analysis. The expression levels of stress response genes were lower in rats fed Japanese food compared to those fed American food, and expression of genes of the sugar and lipid metabolism system was higher in rats fed Japanese food. Expression of genes associated with cholesterol catabolism increased markedly in rats fed Japanese food, although the ingested lipid content was low, and cholesterol accumulation in rat liver was prevented. Therefore, the results suggest that Japanese food is healthy and profitable compared with American food due to activation of metabolism and reduction of stress.

    This study describes the occurrence of 3-hydroxy-5-hydroperoxy-2-methyl-5,6-dihydropyran-4-one (HMDP) in plasma obtained from normal subjects and patients with type 2 diabetes. We have shown previously that HMDP is a novel hydrophilic hydroperoxide formed in the in vitro Maillard reaction that could be analyzed using ultrasensitive chemiluminescence detection-high-performance liquid chromatography (CL-HPLC). The HMDP concentration was 273+/-227 nmol/l in normal subjects and 656+/-535 nmol/l in patients with diabetes. The HMDP concentration was proportional to the plasma glucose concentration level (r=0.640; P<0.01) but not with the glycated hemoglobin level. To investigate the in vivo effects of HMDP, a range of concentrations of the compound was incubated for different time periods with human serum albumin and lipoproteins. HMDP was found to induce denaturation of these macromolecules by modifying lysine residues and causing cross-linking and polymerization of proteins. In the presence of metal ions such as iron and copper, HMDP induced peroxidative degradation of lipoprotein lipids as measured by an elevation in thiobarbituric acid reactive substances (TBARS) concentration. These results suggested that HMDP is produced as a consequence of both hyperglycemia and increased oxidative stress, and may have the potential to contribute to the pathogenesis of arterial complications in diabetes mellitus.

    Despite an increasing number of nutritional studies on food constituents, there has been some difficulty in assessing their bioavailability in human studies. In many cases, biological evaluation has been limited by a lack of proper analytical methods. In this paper, analytical techniques for glycated lipids (newly suggested to be pathogenetic agents) and food components (e.g., tea catechins), for which micro-analysis has been shown to be difficult, were introduced, and their application to nutritional studies was discussed.

    The methods for evaluating antioxidant property of natural products in in vitro and in vivo lipid peroxidation systems are reviewed. Antioxidant activity of tocopherols, carotenoids, flavonoids and curcuminoids have been demonstrated against lipid peroxidation induced in microsomes, erythrocytes, plasma lipoproteins and in animal trials. Recently, in vivo antioxidant function of natural products was extensively investigated in humans together with explanation of their absorbability and metabolic fate.
