  1. <i>Plasmodiophora brassicae</i> affects host gene expression by secreting the transcription factor-type effector PbZFE1

    Abstract The protist pathogen Plasmodiophora brassicae hijacks the metabolism and development of host cruciferous plants and induces clubroot formation, but little is known about its regulatory mechanisms. Previously, the Pnit2int2 sequence, a sequence around the second intron of the nitrilase gene (BrNIT2) involved in auxin biosynthesis in Brassica rapa ssp. pekinensis, was identified as a specific promoter activated during clubroot formation. In this study, we hypothesized that analysis of the transcriptional regulation of Pnit2int2 could reveal how P. brassicae affects the host gene regulatory system during clubroot development. By yeast one-hybrid screening, the pathogen zinc finger protein PbZFE1 was identified to specifically bind to Pnit2int2. Specific binding of PbZFE1 to Pnit2int2 was also confirmed by electrophoretic mobility shift assay. The binding site of PbZFE1 is essential for promoter activity of Pnit2int2 in clubbed roots of transgenic Arabidopsis thaliana (Pnit2int2-2::GUS), indicating that PbZFE1 is secreted from P. brassicae and functions within plant cells. Ectopic expression of PbZEF1 in A. thaliana delayed growth and flowering time, suggesting that PbZFE1 has significant impacts on host development and metabolic systems. Thus, P. brassicae appears to secrete PbZFE1 into host cells as a transcription factor-type effector during pathogenesis.

  2. High ultraviolet-B sensitivity due to lower CPD photolyase activity is needed for biotic stress response to the rice blast fungus, Magnaporthe oryzae

    Sensitivity to ultraviolet-B (UVB, 280–315 nm) radiation varies widely among rice (Oryza sativa) cultivars due to differences in the activity of cyclobutane pyrimidines dimer (CPD) photolyase. Interestingly, cultivars with high UVB sensitivity and low CPD photolyase activity have been domesticated in tropical areas with high UVB radiation. Here, we investigated how differences in CPD photolyase activity affect plant resistance to the rice blast fungus, Magnaporthe oryzae, which is one of the other major stresses. We used Asian and African rice cultivars and transgenic lines with different CPD photolyase activities to evaluate the interaction effects of CPD photolyase activity on resistance to M. oryzae. In UVB-resistant rice plants overexpressing CPD photolyase, 12 h of low-dose UVB (0.4 W m−2) pretreatment enhanced sensitivity to M. oryzae. In contrast, UVB-sensitive rice (transgenic rice with antisense CPD photolyase, A-S; and rice cultivars with low CPD photolyase activity) showed resistance to M. oryzae. Several defense-related genes were upregulated in UVB-sensitive rice compared to UVB-resistant rice. UVB-pretreated A-S plants showed decreased multicellular infection and robust accumulation of reactive oxygen species. High UVB-induced CPD accumulation promoted defense responses and cross-protection mechanisms against rice blast disease. This may indicate a trade-off between high UVB sensitivity and biotic stress tolerance in tropical rice cultivars. Graphical Abstract: [Figure not available: see fulltext.].

  3. Isolation of Burkholderia jumbo phages and their utilization as biocontrol agents to suppress rice seedling rot disease

    Jumbo bacteriophages, which have genomic DNA larger than 200 k-bp, are attracting attention as potential biocontrol agents to suppress bacterial diseases in cultivated crops because they generally have a broader host range of phytopathogenic bacteria compared with ordinary bacteriophages. Thus, the identification of new jumbo phages from environmental and agricultural resources is required to develop effective biological control materials. In a survey of phages targeting the phytopathogenic bacterium Burkholderia glumae, bacteriophages FLC8, FLC9, and FLC10 were isolated from fallen leaf compost. All three phages had an icosahedral head and tail structure. FLC8 and FLC9 had 225,545-bp and 321,833-bp genomic DNA, respectively, indicating that they could be classified as jumbo phages, whereas FLC10 had 32,867-bp genomic DNA, indicating that it was an ordinary phage. All three phages clearly suppressed rice seedling rot disease caused by B. glumae: the control values were 77.1 of FLC8, 82.1 of FLC9 and 33.3 of FLC10, respectively. FLC8 and FLC9 could infect five B. glumae and six B. plantarii strains, which were all strains using analysis of phage host range, while FLC10 had a narrow host range. Evaluation of the preservation condition of both jumbo phages revealed that similar to 90% infectivity of them was maintained in phage buffer (pH 7.5) at 4 degrees C for similar to 30 days.

    Reactive nitrogen species (RNS) play an important role in plant immunity as signaling factors. We previously developed a plasma technology to partially convert air molecules into dinitrogen pentoxide (N<sub>2</sub>O<sub>5</sub>), an RNS whose physiological action is poorly understood. To reveal the function of N<sub>2</sub>O<sub>5</sub> gas in plant immunity, Arabidopsis thaliana was exposed to plasma-generated N<sub>2</sub>O<sub>5</sub> gas once (20 s) per day for 3 days, and inoculated with Botrytis cinerea, Pseudomonas syringae pv. tomato DC3000 (Pst), or cucumber mosaic virus strain yellow (CMV(Y)) at 24 h after the final N<sub>2</sub>O<sub>5</sub> gas exposure. Lesion size with B. cinerea infection was significantly (P &lt; 0.05) reduced by exposure to N<sub>2</sub>O<sub>5</sub> gas. Propagation of CMV(Y) was suppressed in plants exposed to N<sub>2</sub>O<sub>5</sub> gas compared with plants exposed to the air control. However, proliferation of Pst in the N<sub>2</sub>O<sub>5</sub>-gas-exposed plants was almost the same as in the air control plants. These results suggested that N<sub>2</sub>O<sub>5</sub> gas exposure could control plant disease depending on the type of pathogen. Furthermore, changes in gene expression at 24 h after the final N<sub>2</sub>O<sub>5</sub> gas exposure were analyzed by RNA-Seq. Based on the gene ontology analysis, jasmonic acid and ethylene signaling pathways were activated by exposure of Arabidopsis plants to N<sub>2</sub>O<sub>5</sub> gas. A time course experiment with qRT-PCR revealed that the mRNA expression of the transcription factor genes, WRKY25, WRKY26, WRKY33, and genes for tryptophan metabolic enzymes, CYP71A12, CYP71A13, PEN2, and PAD3, was transiently induced by exposure to N<sub>2</sub>O<sub>5</sub> gas once for 20 s peaking at 1–3 h post-exposure. However, the expression of PDF1.2 was enhanced beginning from 6 h after exposure and its high expression was maintained until 24–48 h later. Thus, enhanced tryptophan metabolism leading to the synthesis of antimicrobial substances such as camalexin and antimicrobial peptides might have contributed to the N<sub>2</sub>O<sub>5</sub>-gas-induced disease resistance.

    Nitrogen is a very common element, comprising approximately 78% of Earth's atmosphere, and is an important component of various electronic devices while also being essential for life. However, it is challenging to directly utilize dinitrogen because of the highly stable triple bond in this molecule. The present review examines the use of non-equilibrium plasmas to generate controlled electron impacts as a means of generating reactive nitrogen species (RNS) with high internal energy values and extremely short lifetimes. These species include ground state nitrogen atoms, excited nitrogen atoms, etc. RNS can subsequently react with oxygen and/or hydrogen to generate new highly reactive compounds and can also be used to control various cell functions and create new functional materials. Herein, plasma-processing methods intended to provide RNS serving as short-lived precursors for a range of applications are examined in detail.

  7. Cucumber Mosaic Virus Infection in Arabidopsis: A Conditional Mutualistic Symbiont? 査読有り

    A cucumber mosaic virus isolate, named Ho [CMV(Ho)], was isolated from a symptomless Arabidopsis halleri field sample containing low virus titers. An analysis of CMV(Ho) RNA molecules indicated that the virus isolate, besides the usual cucumovirus tripartite RNA genome, additionally contained defective RNA3 molecules and a satellite RNA. To study the underlying mechanism of the persistent CMV(Ho) infection in perennial A. halleri, infectious cDNA clones were generated for all its genetic elements. CMV, which consists of synthetic transcripts from the infectious tripartite RNA genomes, and designated CMV(Ho)tr, multiplied in A. halleri and annual Arabidopsis thaliana Col-0 to a similar level as the virulent strain CMV(Y), but did not induce any symptoms in them. The response of Col-0 to a series of reassortant CMVs between CMV(Ho)tr and CMV(Y) suggested that the establishment of an asymptomatic phenotype of CMV(Ho) infection was due to the 2b gene of CMV RNA2, but not due to the presence of the defective RNA3 and satellite RNA. The accumulation of CMV(Ho) 2b protein tagged with the FLAG epitope (2b.Ho-FLAG) in 2b.Ho-FLAG-transformed Col-0 did not induce any symptoms, suggesting a 2b-dependent persistency of CMV(Ho)tr infection in Arabidopsis. The 2b protein interacted with Argonaute 4, which is known to regulate the cytosine methylation levels of host genomic DNA. Whole genomic bisulfite sequencing analysis of CMV(Ho)tr- and mock-inoculated Col-0 revealed that cytosine hypomethylation in the promoter regions of 82 genes, including two genes encoding transcriptional regulators (DOF1.7 and CBP1), was induced in response to CMV(Ho)tr infection. Moreover, the increased levels of hypomethylation in the promoter region of both genes, during CMV(Ho)tr infection, were correlated with the up- or down-regulation of their expression. Taken altogether, the results indicate that during persistent CMV(Ho) infection in Arabidopsis, host gene expression may be epigenetically modulated resulting from a 2b-mediated cytosine hypomethylation of host genomic DNA.

  8. Plant death caused by inefficient induction of antiviral R-gene-mediated resistance may function as a suicidal population resistance mechanism 査読有り

    Land plant genomes carry tens to hundreds of Resistance (R) genes to combat pathogens. The induction of antiviral R-gene-mediated resistance often results in a hypersensitive response (HR), which is characterized by virus containment in the initially infected tissues and programmed cell death (PCD) of the infected cells. Alternatively, systemic HR (SHR) is sometimes observed in certain R gene–virus combinations, such that the virus systemically infects the plant and PCD induction follows the spread of infection, resulting in systemic plant death. SHR has been suggested to be the result of inefficient resistance induction; however, no quantitative comparison has been performed to support this hypothesis. In this study, we report that the average number of viral genomes that establish cell infection decreased by 28.7% and 12.7% upon HR induction by wild-type cucumber mosaic virus and SHR induction by a single-amino acid variant, respectively. These results suggest that a small decrease in the level of resistance induction can change an HR to an SHR. Although SHR appears to be a failure of resistance at the individual level, our simulations imply that suicidal individual death in SHR may function as an antiviral mechanism at the population level, by protecting neighboring uninfected kin plants.

  9. Isolation and Characterization of a Novel Jumbo Phage from Leaf Litter Compost and Its Suppressive Effect on Rice Seedling Rot Diseases 査読有り

    Jumbo phages have DNA genomes larger than 200 kbp in large virions composed of an icosahedral head, tail, and other adsorption structures, and they are known to be abundant biological substances in nature. In this study, phages in leaf litter compost were screened for their potential to suppress rice seedling rot disease caused by the bacterium Burkholderia glumae, and a novel phage was identified in a filtrate-enriched suspension of leaf litter compost. The phage particles consisted of a rigid tailed icosahedral head and contained a DNA genome of 227,105 bp. The phage could lyse five strains of B. glumae and six strains of Burkholderia plantarii. The phage was named jumbo Burkholderia phage FLC6. Proteomic tree analysis revealed that phage FLC6 belongs to the same clade as two jumbo Ralstonia phages, namely RSF1 and RSL2, which are members of the genus Chiangmaivirus (family: Myoviridae; order: Caudovirales). Interestingly, FLC6 could also lyse two strains of Ralstonia pseudosolanacearum, the causal agent of bacterial wilt, suggesting that FLC6 has a broad host range that may make it especially advantageous as a bio-control agent for several bacterial diseases in economically important crops. The novel jumbo phage FLC6 may enable leaf litter compost to suppress several bacterial diseases and may itself be useful for controlling plant diseases in crop cultivation.

  10. Priming for enhanced ARGONAUTE2 activation accompanies induced resistance to cucumber mosaic virus in Arabidopsis thaliana 査読有り

    Systemic acquired resistance (SAR) is a broad-spectrum disease resistance response that can be induced upon infection from pathogens or by chemical treatment, such as with benzo-(1,2,3)-thiadiazole-7-carbothioic acidS-methyl ester (BTH). SAR involves priming for more robust activation of defence genes upon pathogen attack. Whether priming for SAR would involve components of RNA silencing remained unknown. Here, we show that upon leaf infiltration of water, BTH-primedArabidopsis thalianaplants accumulate higher amounts of mRNA ofARGONAUTE (AGO)2andAGO3, key components of RNA silencing. The enhancedAGO2expression is associated with prior-to-activation trimethylation of lysine 4 in histone H3 and acetylation of histone H3 in theAGO2promoter and with induced resistance to the yellow strain of cucumber mosaic virus (CMV[Y]). The results suggest that primingA. thalianafor enhanced defence involves modification of histones in theAGO2promoter that conditionAGO2for enhanced activation, associated with resistance to CMV(Y). Consistently, the fold-reduction in CMV(Y) coat protein accumulation by BTH pretreatment was lower inago2than in wild type, pointing to reduced capacity ofago2to activate BTH-induced CMV(Y) resistance. A role ofAGO2in pathogen-induced SAR is suggested by the enhanced activation ofAGO2after infiltrating systemic leaves of plants expressing a localized hypersensitive response upon CMV(Y) infection. In addition, local inoculation of SAR-inducingPseudomonas syringaepv.maculicolacauses systemic priming for enhancedAGO2expression. Together our results indicate that defence priming targets theAGO2component of RNA silencing whose enhanced expression is likely to contribute to SAR.

  11. Complete genomic sequence of a novel phytopathogenic Burkholderia phage isolated from fallen leaf compost 査読有り

    In contrast to most Burkholderia species, which affect humans or animals, Burkholderia glumae is a bacterial pathogen of plants that causes panicle blight disease in rice seedlings, resulting in serious damage to rice cultivation. Attempts to combat this disease would benefit from research involving a phage known to attack this type of bacterium. Some Burkholderia phages have been isolated from soil or bacterial species in the order Burkholderiales, but so far there has been no report of a complete genome nucleotide sequence of a phage of B. glumae. In this study, a novel phage, FLC5, of the phytopathogen B. glumae was isolated from leaf compost, and its complete genome nucleotide sequence was determined. The genome consists of a 32,090-bp circular DNA element and exhibits a phylogenetic relationship to members of the genus Peduovirus, with closest similarity to B. multivorans phage KS14. In addition to B. glumae, FLC5 was also able to lyse B. plantarii, a pathogen causing rice bacterial damping-off disease. This is the first report of isolation of a P2-like phage from phytopathogenic Burkholderia, determination of its complete genomic sequence, and the finding of its potential to infect two Burkholderia species: B. glumae and B. plantarii.

  12. Evaluation of plant stress due to plasma-generated reactive oxygen and nitrogen species using electrolyte leakage 査読有り

    The aim of this paper is to highlight the importance of plant-stress measurement, allowing the quantitative evaluation of plant stress due to plasma-generated gas-phase reactive oxygen and nitrogen species (RONS). It is found that electrolyte leakage from Arabidopsis thaliana leaves, detectable within one hour, tends to reflect gas-phase RONS exposure but does not correlate well with subsequently observed wilting. Ion chromatograph analysis of the leaked electrolyte indicates that selective leakage of potassium ions (K+) accounts for nearly 80% of the measured leaked ions. This selective and rapid K+ leakage due to the RONS exposure stress can be interpreted as part of the plant's response, and is potentially involved in subsequent plasma-induced phenomena but can hardly be explained by physical damage. Thus, electrolyte leakage as a plant stress response can be a useful RONS stress measure, particularly for plasma-based agricultural applications.

  13. Single Amino Acid Substitutions in the Cucumber Mosaic Virus 1a Protein Induce Necrotic Cell Death in Virus-Inoculated Leaves without Affecting Virus Multiplication 査読有り

    When Arabidopsis thaliana ecotype Col-0 was inoculated with a series of reassortant viruses created by exchanging viral genomic RNAs between two strains of cucumber mosaic virus (CMV), CMV(Y), and CMV(H), cell death developed in the leaves inoculated with reassortant CMV carrying CMV(H) RNA1 encoding 1a protein, but not in noninoculated upper leaves. In general, cell death in virus-infected plants is a critical event for virus survival because virus multiplication is completely dependent on host cell metabolism. However, interestingly, this observed cell death did not affect either virus multiplication in the inoculated leaves or systemic spread to noninoculated upper leaves. Furthermore, the global gene expression pattern of the reassortant CMV-inoculated leaves undergoing cell death was clearly different from that in hypersensitive response (HR) cell death, which is coupled with resistance to CMV. These results indicated that the observed cell death does not appear to be HR cell death but rather necrotic cell death unrelated to CMV resistance. Interestingly, induction of this necrotic cell death depended on single amino acid substitutions in the N-terminal region surrounding the methyltransferase domain of the 1a protein. Thus, development of necrotic cell death might not be induced by non-specific damage as a result of virus multiplication, but by a virus protein-associated mechanism. The finding of CMV 1a protein-mediated induction of necrotic cell death in A. thaliana, which is not associated with virus resistance and HR cell death, has the potential to provide a new pathosystem to study the role of cell death in virus-host plant interactions.

    Low-temperature atmospheric-pressure air plasma is a source of charged and neutral gas species. In this study, N-carrying tobacco plants were inoculated with plasma irradiated and non-irradiated tobacco mosaic virus (TMV) solution, resulting in necrotic local lesions on non-irradiated, but not on irradiated, TMV-inoculated leaves. Virus particles were disrupted by plasma irradiation in an exposure-dependent manner, but the viral coat protein subunit was not. TMV RNA was also fragmented in a time-dependent manner. These results indicate that plasma irradiation of TMV can collapse viral particles to the subunit level, degrading TMV RNA and thereby leading to a loss of infectivity.

    The outbreak of rice plant diseases can be effectively suppressed in organic farming systems. However, the mechanisms of disease suppression by organic farming systems are not well understood. When Burkholderia-infected rice seeds were sown and cultivated on nine organic-farmed soils which were supplied by nine independent organic rice farmers or standardized commercial conventional soils, the emergence of bacterial seedling diseases was suppressed to equivalent degrees in nine organic-farmed soils, whereas the diseases occurred in two commercial conventional soils. In any organic or commercial conventional soil sown with healthy rice seeds as a control, the diseases did not appear. Upon physicochemical analysis of the nine organic-farmed soils, component common to these organic-farmed soils seemed to not be directly associated with disease-suppressive activity. However, microbiome analyses indicated that the bacterial population in these nine organic-farmed soils was more diverse than those in commercial conventional soils. Intriguingly, the diverse bacterial population structures of organic-farmed soils were preserved after irrigating and sowing rice seeds, but that of commercial conventional soils was clearly changed by them. Thus, organic-farmed soils seem to maintain robust bacterial populations despite the irrigation and seedling growth. Indeed, pathogenic Burkholderia in infected rice seeds also did not proliferate in the seedling grown on organic-farmed soils. Taken together, the common feature of organic-farmed soils might be the correlation between bacterial seedling disease-suppressive activity and higher robustness of the diversified microbiome.

    In Arabidopsis thaliana, RESISTANCE TO CMV(Y) (RCY1), which encodes a CC-NB-LRR class protein, confers the hypersensitive response (HR) to a yellow strain of cucumber mosaic virus [CMV(Y)]. A. thaliana ecotype Col-0 overexpressing RCY1 transgenes (Col::pRCY1-HA#13) shows extreme resistance (ER) to CMV(Y). To identify novel regulatory factors involved in high RCY1 expression and ER, suppressor of RCY1-mediated resistance to CMV(Y) (src) mutants were screened from Col::pRCY1-HA#13 mutagenized with ethyl methanesulfonate. The amounts of RCY1 transcript and RCY1 protein in five src mutant lines was significantly lower than in Col::pRCY1-HA#13. In src mutants, virus systemically spread without formation of necrotic local lesions in the inoculated leaves. Genetic analysis of progenies of crosses between the src mutant and Col::pRCY1-HA#13 suggested that paramutation might have occurred in these src mutants. Indeed, cytosines in the promoters but not in other genomic regions of the RCY1 transgenes are hypermethylated in these src mutant lines. Moreover, decreased RCY1 expression was correlated with increased methylation of the RCY1 promoter region. Treatment with 5-azacytidine decreased methylation in the promoter region and partially restored RCY1 expression. Taken together, SRC may be involved in RCY1-conferred resistance to CMV(Y) through the regulation of cytosine methylation of the RCY1 promoter. (C) 2017 Elsevier Ltd. All rights reserved.

    The control of seedborne rice seedling diseases in the seed beds is important to avoid epidemics in rice nurseries and paddies, which may result in severe yield loss. Recently, irradiation with plasma containing electrons, creating positive or negative ions and neutral species, has been shown to have an antimicrobial effect, probably via generation of reactive oxygen species. This study examines whether two seedborne rice seedling diseases, bakanae disease caused by the fungal pathogen Fusarium fujikuroi, and bacterial seedling blight caused by Burkholderia plantarii, are suppressed by irradiation of infected rice seeds with atmospheric plasma. Seed germination and seedling growth were not inhibited in plasma-treated healthy seeds. When F. fujikuroi-infected rice seeds were irradiated with plasma after being immersed in sterile distilled water, bakanae disease severity index and the percentage of plants with symptoms were reduced to 18.1% and 7.8% of non-irradiated control, respectively, depending on the duration of plasma irradiation. The bacterial seedling blight disease index was also reduced by plasma irradiation in vacuum-inoculated seeds to 38.6% of the non-irradiated control, and in infected seeds harvested from spray-inoculated heads of rice plants to 40.1% of the control. Therefore, plasma irradiation seems to be effective in controlling two independent seedborne rice seedling diseases.

    Magnaporthe oryzae, the fungus causing rice blast disease, should contend with host innate immunity to develop invasive hyphae (IH) within living host cells. However, molecular strategies to establish the biotrophic interactions are largely unknown. Here, we report the biological function of a M. oryzae-specific gene, Required-for-Focal-BIC-Formation 1 (RBF1). RBF1 expression was induced in appressoria and IH only when the fungus was inoculated to living plant tissues. Long-term successive imaging of live cell fluorescence revealed that the expression of RBF1 was upregulated each time the fungus crossed a host cell wall. Like other symplastic effector proteins of the rice blast fungus, Rbf1 accumulated in the biotrophic interfacial complex (BIC) and was translocated into the rice cytoplasm. RBF1-knockout mutants (Delta rbf1) were severely deficient in their virulence to rice leaves, but were capable of proliferating in abscisic acid-treated or salicylic acid-deficient rice plants. In rice leaves, Delta rbf1 inoculation caused necrosis and induced defense-related gene expression, which led to a higher level of diterpenoid phytoalexin accumulation than the wild-type fungus did. Delta rbf1 showed unusual differentiation of IH and dispersal of the normally BIC-focused effectors around the short primary hypha and the first bulbous cell. In the Delta rbf1-invaded cells, symplastic effectors were still translocated into rice cells but with a lower efficiency. These data indicate that RBF1 is a virulence gene essential for the focal BIC formation, which is critical for the rice blast fungus to suppress host immune responses.

    Organic farming systems can effectively control the pathogens that cause plant diseases. When rice seedlings inoculated with Burkholderia glumae at germination were cultivated on each of three independent organic soils supplied by organic growers, or on conventional soil as a control, development of seedling rot symptoms was significantly suppressed on all organic soils, but not on conventional soil. This disease-suppressive activity of the organic soils was also observed for rice seedling damping-off disease caused by Burkholderia plantarii. Interestingly, such disease suppression was completely compromised on organic soils sterilized at 121 °C, indicating biological activity included in organic soils seems to be associated with the suppression of rice seedling diseases. To explore the putative biological factor(s) conferring the disease-suppressive activity of organic soils, we isolated bacteria from the filtrates of organic soil suspensions. Among these filtrates, two bacterial isolates could suppress development of bacterial seedling rot symptoms, whereas Escherichia coli as a control could not. The 16S rDNA and rpoD nucleotide fragment sequences of two bacterial isolates conferring the observed disease-suppressive activity were highly homologous to those of Pseudomonas sp. Furthermore, phylogenetic analysis of these nucleotide sequences suggested that two isolates placed in the P. fluorescens group. These results reveal that certain Pseudomonas sp. that inhabit organic soils likely play a role in the suppression of rice seedling diseases in organic farming systems.

    The suppressive potentials of Bacillus and Paenibacillus strains isolated from the tomato phyllosphere were investigated to obtain new biocontrol candidates against Fusarium crown and root rot of tomato. The suppressive activities of 20 bacterial strains belonging to these genera were examined using seedlings and potted tomato plants, and two Paenibacillus strains (12HD2 and 42NP7) were selected as biocontrol candidates against the disease. These two strains suppressed the disease in the field experiment. Scanning electron microscopy revealed that the treated bacterial cells colonized the root surface, and when the roots of the seedlings were treated with strain 42NP7 cells, the cell population was maintained on the roots for at least for 4 weeks. Although the bacterial strains had no direct antifungal activity against the causal pathogen in vitro, an increase was observed in the antifungal activities of acetone extracts from tomato roots treated with the cells of both bacterial strains. Furthermore, RT-PCR analysis verified that the expression of defense-related genes was induced in both the roots and leaves of seedlings treated with the bacterial cells. Thus, the root-colonized cells of the two Paenibacillus strains were considered to induce resistance in tomato plants, which resulted in the suppression of the disease.

    The accumulation of RCY1 protein, which is encoded by RESISTANCE TO CMV(Y) (RCY1), a CC-NB-LRR class R-gene, is tightly correlated with the strength of the resistance to a yellow strain of Cucumber mosaic virus [CMV(Y)] in Arabidopsis thaliana. In order to enhance resistance to CMV by overexpression of RCY1, A. thaliana was transformed with intron-less RCY1 cDNA construct under the control of strong CaMV35S promoter. Remarkably, a relative amount of RCY1 protein accumulation in the transformants was much lower than that in plants expressing genomic RCY1 under the control of its native promoter. To identify a regulatory element of RCY1 that could cause such differential levels of RCY1 accumulation, a series of RCY1 cDNA and genomic RCY1 constructs were transiently expressed in Nicotiana benthamiana leaves by the Agrobacterium-mediated infiltration method. Comparative analysis of the level of RCY1 accumulation in the leaf tissues transiently expressing each construct indicated that the intron located in the RCY1-coding region of genomic RCY1, but not the native RCY1 genomic promoter or the 5'- and 3'-untranslated regions of RCY1, was indispensable for high level RCY1 accumulation. The increased levels of RCY1 accelerated plant disease defense reactions. Interestingly, such intron-mediated enhancement of RCY1 accumulation depended neither on the abundance of the RCY1 transcript nor on the RCY1 specific-intron sequence. Taken together, intron-mediated RCY1 expression seems to play a key role in the expression of complete resistance to CMV(Y) by maintaining RCY1 accumulation at high levels.

    The gene RESISTANCE TO CMV(Y) (RCY1) encodes a coiled-coil (CC) nucleotide-binding (NB) leucine-rich repeat (LRR)-class resistance protein that confers resistance to a yellow strain of Cucumber mosaic virus [CMV(Y)] with developing the hypersensitive response (HR) cell death in virus-inoculated leaves of Arabidopsis thaliana. To understand the RCY1-mediated virus defense response, host protein that interacts with the CC and NB domains of RCY1 was isolated in a yeast two-hybrid system using the CC-NB domain fragment as bait. The WRKY70 transcription factor could reproducibly bind to the CC-NB domain fragment. This interaction between WRKY70 and the CC-NB domains was confirmed in a co-immunopurification experiment, suggesting that WRKY70 may function in RCY1-conferred resistance to CMV(Y). In CMV(Y)-inoculated leaves of wild-type plants carrying RCY1, virus multiplication was suppressed, and the level of WRKY70 and Pathogenesis-Related 1 (PR-I) transcripts clearly increased. On the other hand, in CMV(Y)-inoculated leaves of the RCY1-transformed wrky70-1 mutant lacking WRKY70 transcripts, virus coat protein accumulation increased significantly, and the induction of PR-1 expression in response to CMV(Y) was partially compromised. However, there was no significant difference in the degree of HR cell death between the wrky70-1 and wild-type plants carrying RCY1. These results indicate that WRKY70 may play a critical role in suppressing virus multiplication, but not in elaborating HR cell death, by directly interacting with the CC-NB domains of RCY1.     Key message Activation of SA-dependent signaling pathway and suppression of JA-dependent signaling pathway seem to play key roles in B. thuringiensis -induced resistance to R. solanacearum in tomato plants. Bacillus thuringiensis, a well-known and effective bio-insecticide, has attracted considerable attention as a potential biological control agent for the suppression of plant diseases. Treatment of tomato roots with a filter-sterilized cell-free filtrate (CF) of B. thuringiensis systemically suppresses bacterial wilt caused by Ralstonia solanacearum through systemic activation of the plant defense system. Comparative analysis of the expression of the Pathogenesis-Related 1(P6) gene, a marker for induced resistance to pathogens, in various tissues of tomato plants treated with CF on their roots suggested that the B. thuringiensis-induced defense system was activated in the leaf, stem, and main root tissues, but not in the lateral root tissue. At the same time, the growth of R. solanacearum was significantly suppressed in the CF-treated main roots but not in the CF-treated lateral roots. This distinct activation of the defense reaction and suppression of R. solanacearum were reflected by the differences in the transcriptional profiles of the main and lateral tissues in response to the CF. In CF-treated main roots, but not CF-treated lateral roots, the expression of several salicylic acid (SA)-responsive defense-related genes was specifically induced, whereas jasmonic acid (JA)-related gene expression was either down-regulated or not induced in response to the CF. On the other hand, genes encoding ethylene (ET)-related proteins were induced equally in both the main and lateral root tissues. Taken together, the co-activation of SA-dependent signaling pathway with ET-dependent signaling pathway and suppression of JA-dependent signaling pathway may play key roles in B. thuringiensis-induced resistance to R. solanacearum in tomato.

    Two photomorphogenic mutants of rice, coleoptile photomorphogenesis 2 (cpm2) and hebiba, were found to be defective in the gene encoding allene oxide cyclase (OsAOC) by map-based cloning and complementation assays. Examination of the enzymatic activity of recombinant GST-OsAOC indicated that OsAOC is a functional enzyme that is involved in the biosynthesis of jasmonic acid and related compounds. The level of jasmonate was extremely low in both mutants, in agreement with the fact that rice has only one gene encoding allene oxide cyclase. Several flower-related mutant phenotypes were observed, including morphological abnormalities of the flower and early flowering. We used these mutants to investigate the function of jasmonate in the defence response to the blast fungus Magnaporthe oryzae. Inoculation assays with fungal spores revealed that both mutants are more susceptible than wild-type to an incompatible strain of M. oryzae, in such a way that hyphal growth was enhanced in mutant tissues. The level of jasmonate isoleucine, a bioactive form of jasmonate, increased in response to blast infection. Furthermore, blast-induced accumulation of phytoalexins, especially that of the flavonoid sakuranetin, was found to be severely impaired in cpm2 and hebiba. Together, the present study demonstrates that, in rice, jasmonate mediates the defence response against blast fungus.

    Heading Chinese cabbage (Brassica rapa ssp. pekinensis) has been recalcitrant to regeneration and transformation. In particular, there are few reports concerning Japanese cultivars. We evaluated the factors that inhibit Agrobacterium-mediated transformation in heading Chinese cabbage. Ten, we investigated the effects of selectable markers using heading Chinese cabbage cv. Chihiri 70 and compared the effects with those in broccoli cv. Ryokurei (B. oleracea var. italica) as a control for an easily transformable Brassica species. To utilize a selectable marker derived from a host plant cisgene, we cloned a genomic DNA fragment containing regulatory and coding sequences of the acetolactate synthase (ALS) gene from Chinese cabbage and mutagenized it to a herbicide resistant form. After transformation of Chinese cabbage and broccoli with this construct, transgenic plants were efficiently selected with the herbicide bispyribac sodium salt and screened by DNA gel blot analysis. Te average transformation frequency of Chinese cabbage was 1.2±0.2%, which was similar to those in reports using antibiotic selectable markers and was lower than for broccoli (13.9±2.0%). Furthermore, the escape rate was restricted at a low level (about 35-50% lower than hygromycin selection), which is an advantage on practical transformation. We confrmed transgene inheritance and herbicide resistance of potted plants in the T1 generation. This report is the first to describe a selection system for the transformation of a Brassica crop that uses a herbicide-tolerant selectable marker derived from a cisgene. © 2013 The Japanese Society for Plant Cell and Molecular Biology.

    RCY1, which encodes a coiled coil nucleotide-binding site leucine-rich repeat (LRR) class R protein, confers the hypersensitive response (HR) to a yellow strain of Cucumber mosaic virus (CMV[Y]) in Arabidopsis thaliana. Nicotiana benthamiana transformed with hemagglutinin (HA) epitope-tagged RCY1 (RCY1-HA) also exhibited a defense response accompanied by HR cell death and induction of defense-related gene expression in response to CMV(Y). Following transient expression of RCY1-HA by agroinfiltration, the defense reaction was induced in N. benthamiana leaves infected with CMV(Y) but not in virulent CMV(B2)-infected N. benthamiana leaves transiently expressing RCY1-HA or CMV(Y)-infected N. benthamiana leaves transiently expressing HA-tagged RPP8 (RPP8-HA), which is allelic to RCY1. This result suggests that Arabidopsis RCY1-conferred resistance to CMV(Y) could be reproduced in N. benthamiana leaves in a gene-for-gene manner. Expression of a series of chimeric constructs between RCY1-HA and RPP8-HA in CMV(Y)-infected N. benthamiana indicated that induction of defense responses to CMV(Y) is regulated by the LRR domain of RCY1. Interestingly, in CMV(Y)infected N. benthamiana manifesting the defense response, the levels of both RCY1 and chimeric proteins harboring the RCY1 LRR domain were significantly reduced. Taken together, these data indicate that the RCY1-conferred resistance response to CMV(Y) is regulated by an LRR domain-mediated interaction with CMV(Y) and seems to be tightly associated with the degradation of RCY1 in response to CMV(Y).

    Virus multiplication is dependent on host metabolism. Thus, cell death at the primary site of virus infection seems to be critical for defense against virus infection. However, the importance of cell death in hypersensitive response (HR) resistance against viruses in host plants is still unclear. To investigate the role of cell death in virus resistance, we analyzed the response of the Arabidopsis defense, no death (dnd) mutant to Cucumber mosaic virus (CMV) in the presence of RCY1 encoding a CC-NB-LRR class R-protein in the background of ecotype Col-0. The dnd1 phenotype, loss of HR cell death without loss of gene-for-gene resistance to bacterial pathogens, is caused by mutation of the gene encoding the cyclic nucleotide-gated ion channel AtCNGC2. In virus-inoculated leaves of the dnd1 mutant carrying RCY1 (RCY1 dnd1), the induction of HR cell death at the site of virus infection was significantly compromised. However, like wild-type carrying RCY1, local and systemic spread of the virus was restricted and expression of pathogenesis-related (PR) genes was induced in the RCY1 dnd1 mutant. The response to CMV in the RCY1 dnd1 mutant, which was transformed with a salicylic acid-inducible promoter P4::green fluorescence protein-coding region (P4pro::GFP) construct, suggested that the salicylic acid signaling pathway was activated at the site of virus infection without induction of HR cell death. Thus, DND1-mediated HR cell death may not play a critical role in RCY1-conferred resistance to CMV in Arabidopsis. (C) 2012 Elsevier Ltd. All rights reserved.

    Plants harbor microorganisms that are thought to stimulate plant defense systems or promote plant growth. Individual species in these intercellular microbial communities are often not sufficiently abundant to be easily described, although some endophytic microorganisms amenable to culture have been characterized. To better understand the microbial population of plants, we collected intercellular fluid (IF) from leaf blades and sheaths of rice and subsequently isolated DNA from the IF. Denaturing gradient gel electrophoresis (DGGE) analysis of 16S and 18S rDNA fragments amplified from IF DNA by PCR indicated that these band patterns were distinguishable from those of a leaf surface-wash fluid (SF). Analysis of a set of rDNA fragments amplified from IF DNA of rice with different genotypes, paddies or growth stages for the primary survey of overall microbial community in the IF suggested that this approach is suitable for analyzing microbial diversity in the IF from various plant samples. Actually, comparative analysis of amplified rDNA fragments of rice and other five plant species indicated that the microbial diversity in IF is likely to vary substantially among plant species. We can also use sequence analysis of 16S rDNA fragments amplified from rice IF DNA to identify species including unculturable bacteria and proteobacteria and Xanthomonas and 18S rDNA fragments to identify Tilletiaria anomala, Tilletia iowensis, Ustilago maydis and unculturable eukaryotes. Thus, IF DNA analysis seems to be a good tool to further study the microbial ecology of plants.

    Many factors produced by the pathogen Magnaporthe oryzae enhance its ability to infect rice. We found a novel infection-promoting activity in the supernatant of a conidia suspension (SCS) of M. oryzae. The addition of SCS promoted the invasion of excised rice leaf sheaths by infectious hyphae. The activity was heat-stable and was found in SCSs from five virulent and three avirulent isolates of M. oryzae on the rice cv. Nipponbare (Pia). The effect was exclusively detected in compatible interactions. The infection of rice plants by non-rice blast fungi was not enhanced by SCS. These results suggest that SCS includes a heat-stable factor(s) that promotes M. oryzae infection during compatible interactions.

    In clubroot disease, gall formation is induced by infection with the obligate biotroph Plasmodiophora brassicae, and cell hypertrophy is dependent on increased auxin levels. The enzyme nitrilase is suggested to play an important role in auxin biosynthesis in plants. Here, we investigated the expression of nitrilase genes in clubroot disease in Chinese cabbage (Brassica rapa L.). We isolated four isogenes of nitrilase (BrNIT1, BrNIT2, BrNIT3, and BrNIT4) from Chinese cabbage. When a BrNIT2-specific probe was used for Northern blot hybridization, enhanced accumulation of a 1.4 kb mRNA and additional shorter transcripts (1.1 kb) were only detected in clubbed roots 25 days postinoculation (dpi) onward. The expression of BrNIT1 was not strongly affected by infection with P. brassicae. BrNIT3 expression was detected in the roots at 10 and 20 dpi, and the expression was less in clubbed roots than in healthy roots at 20 dpi. Analysis of the transcription initiation point of the BrNIT2 gene suggests that 1.1 kb transcripts were generated by alternative transcription initiation between the second intron and the third exon. The sequence from the second intron to half of the third exon (+415 to +1037, 623 bp) had promoter activity in Arabidopsis during clubroot formation. Therefore, our results suggest that transcriptional regulation of BrNIT2 might be involved in auxin overproduction during clubroot development.

    Prevention of transgene flow from genetically modified crops to food crops and wild relatives is of concern in agricultural biotechnology. We used genes derived from food crops to produce complete male sterility as a strategy for gene confinement as well as to reduce the food purity concerns of consumers. Anther-specific promoters (A3, A6, A9, MS2, and MS5) were isolated from Brassica oleracea and B. rapa and fused to the beta-glucuronidase (GUS) reporter gene and candidate genes for male sterility, including the cysteine proteases BoCysP1 and BoCP3, and negative regulatory components of phytohormonal responses involved in male development. These constructs were then introduced into Arabidopsis thaliana. GUS analyses revealed that A3, A6, and A9 had tapetum-specific promoter activity from the anther meiocyte stage. Male sterility was confirmed in tested constructs with protease or gibberellin insensitive (gai) genes. In particular, constructs with BoCysP1 driven by the A3 or A9 promoter most efficiently produced plants with complete male sterility. The tapetum and middle layer cells of anthers expressing BoCysP1 were swollen and excessively vacuolated when observed in transverse section. This suggests that the ectopic expression of cysteine protease in the meiocyte stage may inhibit programmed cell death. The gai gene also induced male sterility, although at a low frequency. This is the first report to show that plant cysteine proteases and gai from food crops are available as a novel tool for the development of genetically engineered male-sterile plants.

    The phylogenetic relationships of Xanthomonas albilineans strains isolated in Okinawa, Japan, between January and March 2002 were investigated by comparing their gyrB and rpoD gene sequences. Partial gyrB and rpoD sequences from 31 X. albilineans strains were determined, including 21 Okinawa strains and 10 geographically distinct strains. The Okinawa isolates formed a homogenous group and correspond to strains from the Caribbean islands (St Lucia and Barbados), Guyana and Sri Lanka. The level of sequence diversity in the gyrB and rpoD genes depended on the geographic origin of the strains, suggesting that sequence analysis of these genes can be useful for differentiating genetic variants within the X. albilineans species.

    In clubroot disease, gall formation is induced by infection with the obligate biotroph Plasmodiophora brassicae due to increased levels of auxins and cytokinins. Because aldehyde oxidase (AO) may be involved in auxin biosynthesis in plants, we isolated two AO genes (BrAO1 and BrAO2) from Chinese cabbage (Brassica rapa ssp. pekinensis cv. Muso), which are the most similar to AAO1 among Arabidopsis AO genes, and examined their expressions during clubroot development. The expression of BrAO1 was enhanced in inoculated roots from 15 days post-inoculation (dpi) when visible clubroots were still undetectable. Thereafter, BrAO1 expression increased with clubroot development compared with uninoculated roots, although BrAO2 expression was repressed. In situ hybridization revealed that BrAO1 was strongly expressed in tissues that were invaded by immature plasmodia at 35 dpi, suggesting that BrAO1 expression was enhanced by the pathogen in order to establish its pathogenesis. In addition, we detected AO activity, as evidenced by the occurrence of at least six bands (BrAO-a to BrAO-f) in the roots of Chinese cabbage using an active staining method with benzaldehyde and indlole-3-aldehyde as the substrate. Coincidental with BrAO1 expression, the signals of BrAO-a and BrAO-d increased with inoculation by P. brassicae during clubroot development compared with healthy roots, resulting in an increase in total AO activity. By contrast, the band BrAO-b decreased post-inoculation, in parallel with the expression of BrAO2. The other bands of activity were not clearly influenced by the infection. Based on these results, we discuss the involvement of AO in auxin-overproduction during clubroot development in Chinese cabbage.

    Infection of crucifers by the obligate plant pathogen Plasmodiophora brassicae Woron. results in the formation of clubroot disease in these plants. Plasmodiophora brassicae gene expression during disease development was studied by differential display analysis of total RNA extracted from the roots of Chinese cabbage inoculated with the pathogen. In a series of experiments, 30 differentially expressed bands of cDNA were detected, and the expression of clone no. 17 was confirmed in clubbed roots. Southern blot analysis showed that this clone was a single-copy gene in the P. brassicae genome. Putative amino acid sequence analysis of the full-length cDNA of clone no. 17 (4.6 kb, designated PbSTKL1) revealed a serine/threonine kinase-like domain at the C-terminal region and a coiled-coil structure in the middle region of the putative protein. PbSTKL1 expression increased strongly beginning 30 days after inoculation and was coincident with resting spore formation.

    The obligate biotroph Plasmodiophora brassicae induces club formation on roots of various species of the Brassicaceae family. This hypertrophy is dependent on increased amount of auxins and cytokinins. To investigate the involvement of cytokinin biosynthesis by the plant in clubroot development, we isolated cDNA fragments of five putative cytokinin synthase genes from Brassica rapu (BrIPTI, 3, 5 7, and 8) and examined their expression using Northern blot analysis. Observations at 5-day intervals revealed the first visible club formation (only near the base of the root) at 20 days post-inoculation (dpi). Compared with control roots, the expression of BrIPTI, 3, 5, and 7 was increased at 20 dpi in parts of the inoculated root that lacked visible symptoms ( &gt; 2 cm below the base of the root) but at which large clubs formed later in the progress of the disease. However, these genes were less expressed in clubs that formed 0-2 cm below the base of the root than in the control at 20 dpi. These results suggest that P. brassicae infection transiently stimulates the transcription of BrIPTI, 3, 5, and 7 before club formation, after which the expression is repressed in clubs. No signals corresponding to BrIPT8 were detected in either inoculated or uninoculated roots. These results suggest that cytokinin biosynthesis by the plant is related to the primary development of clubroot disease, although it is unlikely to contribute to club enlargement.     The mechanism of telomerase regulation is far better understood in mammalian systems than it is in plant systems. In this study, we investigated the possibility that regulation of telomerase activity occurs at the post-transcriptional level in rice plants (Oryza sativa). Specifically, we used RT-PCR with primers based on the rice telomerase reverse transcriptase (OsTERT) cDNA sequence to detect alternative mRNA splicing products. Five types of DNA product that were shorter than predicted by the cDNA sequence were observed for all the tested cells and tissues. Sequencing of these products indicated that they were alternatively spliced OsTERT fragments lacking some portions of the full-length sequence. These results suggest that regulation of telomerase activity occurs at the post-transcriptional level in rice cells and tissues. The relative amounts of full-length OsTERT mRNA and the shorter, alternatively spliced mRNAs were estimated by quantitative RT-Southern analysis. No correlation was found between levels of telomerase activity and accumulation of full-length OsTERT mRNA in the various cell and tissue types, indicating that splicing of OsTERT mRNAs may be important in telomerase regulation in rice. Further studies are necessary to determine whether the spliced OsTERT mRNAs are translated into proteins and if these putative proteins affect telomerase activity.

    A stretch PCR-based method used to measure human telomerase activity was modified for use with plant telomerase and shown to be useful for quantitative analysis. We isolated a full-length cDNA of a telomerase reverse transcriptase catalytic subunit gene from suspension-cultured cells of rice (Oryza sativa L.). A single copy of this gene is present in the rice genome. Telomerase activity was detected in suspension-cultured O. saliva cells at all growth stages, although the activity was greatest in 2-day-old subcultures and decreased at 6 days. Nevertheless, the accumulation of O. sativa telomerase mRNA was nearly constant in cells at all growth stages. Although no telomerase activity was detected in the lower portions of blades in 3-month-old rice plants, telomerase mRNA accumulated in the blades. Tests of different tissues excised from rice seedlings 3 weeks after sowing showed that root tips had high telomerase activity, while roots without root tips had no telomerase activity. Nevertheless, telomerase mRNA accumulated in both tissues. These results suggest that telomerase activity in tissues of O. saliva is regulated at the post-transcriptional or post-translational levels. (C) 2003 Elsevier Science Ireland Ltd.     Ethylene regulates sex expression in cucumber plants (Cucumis sativus L.). To clarify ethylene-mediated regulation of sex expression, we previously used differential display technology to identify 20 ethylene-responsive genes in the apices of cucumber plants. In this study, we report the analysis of the full-length cDNA from clone #16 and its corresponding gene (designated as ERAF16 ), which encodes a methyltransferase. This type of methyltransferase has recently been identified, and some of them methylate signalling molecules in plants, which include salicylic acid and jasmonic acid. However, the substrate of the ERAF16 protein has not been revealed. We examined the ERAF16 expression pattern using RNA gel blot analysis of total RNA preparations from the apices of a monoecious cucumber (cv. Shimoshirazu-jibai) and a gynoecious cultivar (Rensei). The levels of ERAF16 expression in these cultivars were correlated with the development of female flowers. The induction of ERAF16 transcription by ethephon occurred within 4 h of the start of treatment. The expression of ERAF16 in immature floral buds of the gynoecious cultivar was stronger than in those of the monoecious cultivar. These results suggest that ERAF16 is included in the ethylene-induced formation of female flowers. However, ERAF16 was expressed in opened flowers and vegetative tissues. Therefore, we speculate that the role of ERAF16 protein varies, depending on the tissue and developmental stage.

  47. Isolation of a MADS-box gene (ERAF17) and correlation of its expression with the induction of formation of female flowers by ethylene in cucumber plants (Cucumis sativus L.) 査読有り

    Ethylene regulates sex expression in cucumber (Cucumis sativus L.) plants. When the apices of monoecious cucumber seedlings (cv. Shimoshirazu-jibai) were treated with the ethylene-releasing compound, ethephon, female flowers were induced at the nodes. To clarify the action of ethylene in the regulation of sex expression, we attempted to isolate genes whose expression changed during induction of the formation of female flowers at the apices of these cucumber plants upon treatment with ethephon. Using the differential-display method, we identified 20 clones (#1 to #20) that reflected differences in the accumulation of transcripts in apices treated or not treated with ethephon. Sequence analysis of cDNA fragments revealed that the cDNA #17 had the sequence of a MADS-box gene. We isolated the full-length cDNA and showed that it included both a MAIDS box and a K box, and the corresponding gene was designated ERAF17. We examined the expression of ERAF17 in the apices of cv. Shimoshirazu-jibai and in those of a gynoecious cultivar (Rensei). In these cultivars, the timing and levels of expression of the ERAF17 transcript were correlated with the development of female flowers. Induction of the synthesis of the ERAF17 transcript by ethephon occurred within 4 h of the start of treatment and continued for 4 days at least. Expression of ERAF17 at apices was localized in the floral buds of the gynoecious cultivar, and expression was maintained in female flowers thorough their development. Our results suggest that the induction of the formation of female flowers by ethylene might be regulated by the expression of ERAF17 in floral buds at the apices of cucumber plants and that expression of this gene might also be involved in the development of female flowers.

    The technique of differential display was used to isolate ethylene-responsive cDNAs from the apices of cucumber plants. Differences in RNA populations from apices treated with or without ethephon (an ethylene - releasing compound) were examined using 80 primer combinations. Northern blot analysis confirmed that 20 cDNAs represented mRNAs that were differentially expressed upon ethephon treatment (promoted, 17 suppressed, 3). Sequence analysis of these cDNAs revealed that two clones were identical to the 3′-terminal regions of the CR20 and CUS3 genes of cucumber. Five clones showed significant similarity to the C-terminal regions of short-chain alcohol dehydrogenases, β-1,3-glucanases, S-adenosyl-L-methionine:salicylic acid carboxyl methyltransferase, products of the Aux/IAA gene family, and lipoxygenases. The other 13 clones were found to have no significant homology in the databases. Because ethylene promotes female flower formation in cucumber plants, some of 20 cDNAs might be involved in processes of sex regulation.

    有機農業における無農薬栽培が可能な要因として,有機栽培植物の内生菌叢が病害抵抗性を増強している可能性に着目し,イネを材料として解析を行った。有機栽培イネの地上部に生息する内生菌叢を16S及び18S rDNAを用いたPCR-DGGE法により慣行栽培イネと比較した結果,両者で大きな差異はないが,有機栽培イネにのみ確認できるバンドが存在した。さらに,培養法によって同様に比較したところ,有機栽培イネに特徴的な内生細菌が存在することが示唆された。16S rDNA解析から,それらの中にはPseudomonas属菌やBacillus属菌,Curtobacterium属菌など,過去に病害抵抗性を増強する効果が報告されているものが含まれていた。このうち,AZ2株(Pseudomonas sp.)はいもち病菌の付着器直下の活性酸素生成を促進し,侵入菌糸の伸展を遅延する効果があることが明らかとなった。しかしながら,病斑形成には影響せず,AZ2株単独ではいもち病の発生を抑えることはできないと考えられた。今後,その他の菌の影響を解析すると共に,AZ2株の様な内生菌が有機栽培イネに存在する普遍性を検証する必要がある。

    In this study, we investigated roles of JA in phytoalexin production using the JA-biosynthetic rice mutant cpm2 or the JA-Ile-biosynthetic rice mutant jar1. First we examined phytoalexin production in CuCl_2-treated mutant leaves. The production of the diterpenoid phytoalexins in cpm2 was comparable to that in a wild-type. On the other hand, the production of sakuranetin, a flavonoid phytoalexin, was suppressed in cpm2, and the suppression was restored by the addition of JA. Next, we examined phytoalexin production in Magnaphorthe oryzae infected leaf sheaths. Upon M. oryzae infection production of both diterpenoid phytoalexins and sakuranetin was severely suppressed in cpm2. These data suggest that, under CuCl_2 treatment, JA is essential for the production of sakuranetin, but not for diterpenoid phytoalexins. On the other hand, upon M. oryzae infection, JA is essential for production of both diterpenoid phytoalexins and sakuranetin. We also analyzed phenotypes of rice jar1 mutant defective in JA-Ile production to extend understanding of roles of jasmonates in phytoalexin production. We examined phytoalexin production by JA or JA-Ile treatment in jar1 leaves. Production of both diterpenoid phytoalexins and sakuranetin by JA treatment was suppressed in jar1. On the other hand, production of these phytoalexins by JA-Ile treatment in jarl was comparable to that in wild type. These data suggest that JA-Ile was essential for JA-induced phytoalexin production in rice.

  1. 植物免疫のプライミングを制御するクロマチン構造と転写因子の関係

    本研究は、Defense primingの分子メカニズムの一端を解明するため、RNAサイレンシング関連因子であるArgonaute(AGO)遺伝子のPrimingに着目して解析を行っている。これまでに、シロイヌナズナの10のAGO遺伝子の中でAGO2とAOG3の遺伝子発現が抵抗性誘導剤BTH処理によってPriming(通常よりも刺激に対して強く速く応答可能な状態になること)されることが明らかになっている。 遺伝子発現のプライミングの制御機構にはクロマチン構造の変化が重要な役割を持っていることが示唆されており、AGO2の転写開始点のヒストンメチル化レベル(H3K4me3)はBTH処理によって増加し、活性化状態になっていることが明らかになっている。さらに、本領域へのRNA polymerase IIの結合性はBTH処理によって低下することが示唆された。Priming状態は、刺激に対する応答性は高いが、転写の活性化はまだ起こっていない状態である。従って、RNA polymerase IIの結合性の低下はPriming時のクロマチン構造は活性化状態であるが、実際には転写が活性化されないという状態の形成に関わる可能性がある。 さらに、BTH処理によるPriming誘導時にヒストンのメチル化レベルが変化する領域に結合する転写因子のとしてWRKY25とAtHMGB9が特定されている。WRKY25の遺伝子発現自体もBTH処理によってPriming誘導されたのに対し、AtHMGB9の遺伝子発現はBTH処理によって、Priming誘導されず、やや発現が抑制される傾向が認められた。HMGBはクロマチン構造の制御に関与することが示唆されており、転写因子の相互作用にも影響することが示唆されている。さらにAtHMGB9の機能解析を行うため、HAタグを付加した過剰発現系統の作出を行った。

    ウイルス抵抗性に関与するサイレンシング関連因子AGO2のBTH処理によるプライミング機構におけるmiRNAとヒストン修飾の関与を解析した。BTH処理によるmiRNAの変動を解析した結果、miR403(AGO2制御)やmiR168(AGO1制御)の蓄積量が上昇した他、RNA-seqによりmiRNAの変動が検出された。また、miRNAの機能欠損変異体ではAGO2の恒常的なプライミングが認められた。一方、ヒストン修飾の解析では、AGO2の転写開始点とその上流2kb付近では、H3K4me3レベルが上昇することが確認された。また、RNAPIIの結合性の変化やこの領域に結合する転写因子候補が示された。

    抵抗性対立遺伝子座RCY1/HRT/RPP8は、Cucumber mosaic virus (CMV)、Turnip crinkle virus (TCV)、アブラナ科べと病菌(Hpa)に対するNB-LRR型抵抗性タンパク質をコードしている。同抵抗性座の発現は、プローモーター領域のシトシンメチル化によるエピジェネティックな制御を受けており、抵抗性タンパク質の蓄積量が病害抵抗性と相関していた。さらに、同抵抗性タンパク質は、LRRドメインを介してCMV外被タンパク質(CP)を認識することにより抵抗性を誘導し、その認識には、CPのN末端のαへリックスドメインを介したCP多量体形成が関与していた。

    RCY1とRPP8は、キュウリモザイクウイルス (CMV)とアブラナ科べと病菌の抵抗性を支配する対立遺伝子である。RCY1、RPP8、および両遺伝子の各ドメインを交換したキメラ遺伝子を、N. benthamiana葉で一過的発現し、抵抗性発現を解析したところ、CMVに対する抵抗性応答には、RCY1タンパク質のLRRドメインが関与していた。次に、両遺伝子の発現制御機構の解析では、いずれもイントロンが、RCY1とRPP8タンパク質の高蓄積と病害抵抗性の増強を決定していた。抵抗性タンパク質の機能ドメイン解析とその蓄積量の制御に関わるこれらの新知見は、病害抵抗性の分子基盤の理解に寄与する。

    生物的/非生物的ストレスのクロストークについて新たな知見を得るため、キュウリモザイクウイルス抵抗性タンパク質RCY1の相互作用因子を酵母ツーハイブリッド法により探索したところ、WRKY70が単離された。免疫共沈法によりその相互作用を確認した結果、RCY1全長タンパク質では確認できなかったが、CC-NBドメインとWRKY70の相互作用が確認された。また、wrky70欠損変異体では防御関連遺伝子PR1の発現減少とウイルス感染の促進が認められた。WRKY70は病害応答以外に非生物的ストレスにも重要な機能を持つため、RCY1との相互作用が生物的/非生物的ストレスのクロストークのキーとなる可能性がある。
